Green Tea Catechin Epigallocatechin Gallate Inhibits Vegetative Cell Outgrowth and Expression of Beta-Lactamase Genes in Penicillin-Resistant Bacillus anthracis Strain PCr.

The green tea catechin epigallocatechin gallate (EGCg) has antimicrobial effects on many bacteria. In this study, we investigated the inhibitory effects of EGCg on Bacillus anthracis spores and vegetative cells. The B. anthracis spores were insensitive to EGCg, but the growth of vegetative cells derived from germinated spores was inhibited by EGCg. Moreover, EGCg decreased the minimum inhibitory concentration of penicillin and meropenem for penicillin-resistant B. anthracis. In the penicillin-resistant B. anthracis strain, the transcription levels of the beta-lactamase genes (bla1 and bla2) decreased significantly following the treatment with 50 µg/mL EGCg. These results suggest that the appropriate application of EGCg may effectively control the penicillin-resistant B. anthracis growth and beta-lactamase production.

Effects of Green Tea Extract Epigallocatechin-3-Gallate on Oral Diseases: A Narrative Review.

Oral diseases are among the most prevalent diseases globally. Accumulating new evidence suggests considerable benefits of epigallocatechin-3-gallate (EGCG) for oral health. This review aims to explore the role and application of EGCG in main oral diseases. This narrative review thoroughly examines and summarizes the most recent literature available in scientific databases (PubMed, Web of Science, Scopus, and Google Scholar) reporting advances in the role and application of EGCG within the dental field. The major keywords used included "EGCG", "green tea extract", "oral health", "caries", "pulpitis", "periapical disease", "periodontal disease", "oral mucosa", "salivary gland", and "oral cancer". EGCG prevents and manages various oral diseases through its antibacterial, anti-inflammatory, antioxidant, and antitumor properties. Compared to traditional treatments, EGCG generally exhibits lower tissue irritation and positive synergistic effects when combined with other therapies. Novel delivery systems or chemical modifications can significantly enhance EGCG's bioavailability, prolong its action, and reduce toxicity, which are current hotspots in developing new materials. this review provides an exhaustive overview of the biological activities of EGCG to major oral diseases, alongside an exploration of applications and limitations, which serves as a reference for preventing and managing oral ailments.

Study on the inactivation effect and mechanism of EGCG disinfectant on Bacillus subtilis

The widespread use of chlorine-based disinfectants in drinking water treatment has led to the proliferation of chlorine-resistant bacteria and the risk of disinfection byproducts (DBPs), posing a serious threat to public health. This study aims to explore the effectiveness and potential applications of epigallocatechin gallate (EGCG) against chlorine-resistant Bacillus and its spores in water, providing new insights for the control of chlorine-resistant bacteria and improving the biological stability of distribution systems. The inactivation effects of EGCG on Bacillus subtilis (B. subtilis) and its spores were investigated using transmission electron microscopy, ATP measurement, and transcriptome sequencing analysis to determine changes in surface structure, energy metabolism, and gene expression levels, thereby elucidating the inactivation mechanism. The results demonstrate the potential application of EGCG in continuously inhibiting chlorine-resistant B. subtilis in water, effectively improving the biological stability of the distribution system. However, EGCG is not suitable for treating raw water with high spore content and is more suitable as a supplementary disinfectant for processes with strong spore removal capabilities, such as ozone, ultraviolet, or ultrafiltration. EGCG exhibits a disruptive effect on the morphological structure and energy metabolism of B. subtilis and suppresses the synthesis of substances, energy metabolism, and normal operation of the antioxidant system by inhibiting the expression of multiple genes, thereby achieving the inactivation of B. subtilis.

The antibacterial mechanism of (-)-epigallocatechin-3-gallate (EGCG) against Campylobacter jejuni through transcriptome profiling.

(-)-Epigallocatechin-3-gallate (EGCG) has been shown antibacterial activity against Campylobacter jejuni; however, the relevant antibacterial mechanism is unknown. In this study, phenotypic experiments and RNA sequencing were used to explore the antibacterial mechanism. The minimum inhibitory concentration of EGCG on C. jejuni was 32µg/mL. EGCG-treated was able to increase intracellular reactive oxygen species levels and decline bacterial motility. The morphology and cell membrane of C. jejuni after EGCG treatment were observed collapsed, broken, and agglomerated by field emission scanning electron microscopy and fluorescent microscopy. The RNA-seq analysis presents that there are 36 and 72 differential expressed genes after C. jejuni was treated by EGCG with the concentration of 16 and 32µg/mL, respectively. EGCG-treated increased the thioredoxin expression, which was a critical protein to resist oxidative stress. Moreover, downregulation of the flgH and flgM gene in flagellin biosynthesis of C. jejuni was able to impair the flagella, reducing cell motility and virulence. The primary antibacterial mechanism revealed by RNA-seq is that EGCG with iron-chelating activity competes with C. jejuni for iron, causing iron deficiency in C. jejuni, which potentially impacts the survival and virulence of C. jejuni. The results suggested a new direction for exploring the activity of EGCG against C. jejuni in the food industry. PRACTICAL APPLICATION: A deeper understanding of the antibacterial mechanism of EGCG against C. jejuni was more beneficial in improving the food safety, eliminating concerns about human health caused by C. jejuni in future food, and promoting the natural antibacterial agent EGCG application in the food industry.

The effect of epigallocatechin-3-gallate (EGCG), a main active ingredient in tea residues, on improving fruit quality and prolonging postharvest storage in apple

EGCG, as an active oxygen scavenger, has a significant effect on inhibiting the greasiness of apples. However, the impact of greasiness on fruit storage and how EGCG eliminates this effect, as well as its influence on apple quality formation, have not been clarified. In this study, we found that exogenous application of EGCG effectively improved a series of nutritional growth indicators, including seedling growth status and photosynthetic activity. Additionally, it significantly increased indicators of reproductive growth, such as fruit diameter, soluble solids content, surface wax, and surface smoothness. Through our experiments, we further discovered that greasiness primarily affects fruit respiration, accelerating fruit decay and causing harm during storage, while EGCG mitigates this process. We also found that the application of tea residue had similar effects to exogenous EGCG. Collectively, these studies suggest that EGCG could be used as a novel foliar fertilizer to enhance overall fruit quality. Moreover, the application of tea residue has similar effects to EGCG, providing a cost-effective solution for apple production.

Epigallocatechin-3-gallate improves the quality of maternally aged oocytes.

The decline in female fertility as age advances is intricately linked to the diminished developmental potential of oocytes. Despite this challenge, the strategies available to enhance the quality of aged oocytes remain limited. Epigallocatechin-3-gallate (EGCG), characterised by its anti-inflammatory, antioxidant and tissue protective properties, holds promise as a candidate for improving the quality of maternally aged oocytes. In this study, we explored the precise impact and underlying mechanisms of EGCG on aged oocytes. EGCG exhibited the capacity to enhance the quality of aged oocytes both in vitro and in vivo. Specifically, the application of EGCG in vitro resulted in noteworthy improvements, including an increased rate of first polar body extrusion, enhanced mitochondrial function, refined spindle morphology and a reduction in oxidative stress. These beneficial effects were further validated by the improved fertility observed among aged mice. In addition, our findings propose that EGCG might augment the expression of Arf6. This augmentation, in turn, contributes to the assembly of spindle-associated F-actin, which can contribute to mitigate the aneuploidy induced by the disruption of spindle F-actin within aged oocytes. This work thus contributes not only to understanding the role of EGCG in bolstering oocyte health, but also underscores its potential as a therapeutic intervention to address fertility challenges associated with advanced age.

Dietary Epigallocatechin-3-Gallate (EGCG) Improves Nonspecific Immune Response of Chinese Rice Field Eel (Monopterus albus).

Epigallocatechin-3-gallate (EGCG) has been recognized as a potential additive for aquafeeds due to its beneficial biological functions. In order to evaluate the potential application of EGCG in Chinese rice field eel (Monopterus albus), six isonitrogenous and isolipidic diets containing 0, 25, 50, 100, 200, and 400 mg/kg EGCG were formulated and were fed to Monopterus albus (M. albus) for 9 weeks. The results showed that M. albus fed diets containing 0 and 100 mg/kg EGCG presented higher weight again and specific growth rate than the other groups. Fish fed with 25, 50, and 400 mg/kg EGCG displayed lower whole-body lipid content. Serum aspartate aminotransferase (AST) concentration significantly decreased in EGCG treated groups with the exception of 100 mg/kg group. Hepatic catalase (CAT) activity and glutathione (GSH) concentration decreased as EGCG level increased while malondialdehyde (MDA) concentration showed an opposite trend. EGCG supplementation resulted in a promoted lysozyme (LZM) activity and immunoglobulin M (IgM) level in the liver of M. albus. Furthermore, transcription of three immune related genes including major histocompatibility complex (mhc-2α), hepcidin, and interleukin-8 (il-8) mRNAs was upregulated by EGCG treatment; while transcription of interleukin-6 (il-6) and nuclear factor kappa-B (nf-kb) genes was downregulated. Results also showed a linear relation between EGCG inclusion level and parameters of AST, CAT, GSH, MDA, LZM, IgM, and immune-related genes transcriptions. In summary, it could be suggested that EGCG supplementation enhanced the nonspecific immune response of the Chinese rice field eel. Based on the broken-line regression analysis of IgM, the optimal dietary EGCG supplementation for M. albus was estimated to be 109.81 mg/kg.

Biomodification of eroded and abraded dentin with epigallocatechin-3-gallate (EGCG)

This study aimed to evaluate in vitro the effects of epigallocatechin-3-gallate (EGCG) as a biomodifier of eroded and abraded dentin. Forty dentin specimens were obtained from the buccal surface of bovine teeth. The specimens were randomly distributed in 4 groups according to dentin substrate: sound or eroded/abraded and dentin biomodification: with 0.5% EGCG and no biomodification (control group). Specimens were subdivided according to aging time: 24 h and 3 months for the analysis of microtensile bond strength (n = 10), morphology of the adhesive interface by SEM (n = 3) and dentin micropermeability by fluorescence microscope (n = 8). Statistical analysis was performed using SPSS system version 20.0 with a significance level of 5%. The results revealed that the control group with eroded-abraded dentin exhibited the lowest bond strength values at 24 h and 3 months. However, the application of 0.5% EGCG as a biomodifier significantly increased bond strength on both sound and eroded-abraded substrates. After 3 months, all groups exhibited an adhesive interface with a more intense fluorescence in the adhesive layer, indicating an increase in porosity at the interface. In conclusion, the EGCG application as a biomodifier enhanced bond strength on both sound and eroded-abraded dentin substrates, however, adhesive interfaces are more regular when restorations are performed on sound dentin, regardless of the biomodification with EGCG.

Evaluation of the synergistic effects of epigallocatechin-3-gallate-loaded PEGylated-PLGA nanoparticles with nimodipine against neuronal injury after subarachnoid hemorrhage.

Subarachnoid hemorrhage (SAH) is a devastating subtype of stroke with high mortality and morbidity. Although serious side effects might occur, nimodipine, a second-generation 1,4-dihydropyridine calcium channel blocker, is clinically used to improve neurological outcomes after SAH. Recently, (-)-epigallocatechin-3-gallate (EGCG) has been reported to inhibit Ca2+ overloading-induced mitochondrial dysfunction, oxidative stress, and neuronal cell death after SAH; however, low bioavailability, instability, and cytotoxicity at a high dose limited the clinical application of EGCG. To overcome these limitations, PEGylated-PLGA EGCG nanoparticles (EGCG-NPs) were constructed to enhance the bioavailability by using the double-emulsion method. Antioxidative activity, cytotoxicity, behavioral, and immunohistochemistry studies were carried out to determine the neuroprotective effectiveness after cotreatment with EGCG-NPs (75 mg/kg/d preconditioning for 7 days before SAH) and nimodipine (10 mg/kg/d after 30 min of SAH) by using in vivo SAH models. The optimized EGCG-NPs with a Box-Behnken design showed a small particle size of 167 nm, a zeta potential value of -22.6 mV, an encapsulation efficiency of 86%, and a sustained-release profile up to 8 days in vitro. Furthermore, EGCG-NPs (75 mg/kg/d) had superior antioxidative activity to free EGCG (100 mg/kg/d). EGCG-NPs combined with nimodipine exhibited significant synergistic effects against neuronal cell death by suppressing oxidative stress, Ca2+ overloading, mitochondrial dysfunction, and autophagy after SAH. These results suggest that cotreatment with EGCG-NPs and nimodipine may serve as a promising novel strategy for the treatment of SAH.

Enhancing adhesive–dentin interface stability of primary teeth: From ethanol wet-bonding to plant-derived polyphenol application

ObjectivesTo investigate whether the adhesive–dentin interface stability of primary teeth would be enhanced by epigallocatechin-3-gallate (EGCG) with ethanol wet-bonding. MethodsNon-caries primary molars were sliced to achieve a flat dentin surface and etched then randomly distributed into five groups in accordance with different treatments: group 1, no treatment; group 2, applying absolute ethanol wet-bonding for 60 s; groups 3–5, applying 0.1%, 0.5%, and 1% (w/v) EGCG-incorporating ethanol wet-bonding (0.1%, 0.5%, and 1% EGCG) for 60 s. Singlebond universal adhesive was then applied followed by resin composite construction. Microtensile bond strength, fracture mode, and nanoleakage at adhesive–dentin interface were evaluated after 24 h of water storage or 10,000 times of thermocycling. Zymography of hybrid layer, biofilm formation of Streptococcus mutans by CLSM, FESEM, and MTT test, and cytotoxicity by CCK-8 assay were respectively assessed. ResultsIrrespective of thermocycling, the dentin bond strength was preserved with reduced nanoleakage in the 0.5% and 1% EGCG groups. Furthermore, the activity of endogenous proteases and the growth of Streptococcus mutans biofilm were inhibited after treatment with 0.5% and 1% EGCG/ethanol solutions (groups 4 and 5). CCK-8 results of the 0.1% and 0.5% EGCG groups showed acceptable biocompatibility. ConclusionsTreatment by EGCG/ethanol solutions effectively enhanced the bond stability of primary teeth at the adhesive–dentin interface. Clinical significanceSynergistic application of EGCG and ethanol wet-bonding suggesting a promising strategy to improve dentin bonding durability with bacterial biofilm inhibition, thus increasing resin-based restorations’ service life in primary dentition.

EGCG protects the mouse brain against cerebral ischemia/reperfusion injury by suppressing autophagy via the AKT/AMPK/mTOR phosphorylation pathway.

Stroke remains one of the leading reasons of mortality and physical disability worldwide. The treatment of cerebral ischemic stroke faces challenges, partly due to a lack of effective treatments. In this study, we demonstrated that autophagy was stimulated by transient middle cerebral artery occlusion/reperfusion (MCAO/R) and oxygen-glucose deprivation/reoxygenation (OGD/R). Treatment with (−)-epigallocatechin-3-gallate (EGCG), a bioactive ingredient in green tea, was able to mitigate cerebral ischemia/reperfusion injury (CIRI), given the evidence that EGCG administration could reduce the infarct volume and protect poststroke neuronal loss in MCAO/R mice in vivo and attenuate cell loss in OGD/R-challenged HT22 cells in vitro through suppressing autophagy activity. Mechanistically, EGCG inhibited autophagy via modulating the AKT/AMPK/mTOR phosphorylation pathway both in vivo and in vitro models of stroke, which was further confirmed by the results that the administration of GSK690693, an AKT/AMPK inhibitor, and rapamycin, an inhibitor of mTOR, reversed aforementioned changes in autophagy and AKT/AMPK/mTOR signaling pathway. Overall, the application of EGCG relieved CIRI by suppressing autophagy via the AKT/AMPK/mTOR phosphorylation pathway.

LC-MS Based Metabolomics Study of the Effects of EGCG on A549 Cells.

(−)-Epigallocatechin-3-gallate (EGCG) is the main bioactive catechin in green tea. The antitumor activity of EGCG has been confirmed in various types of cancer, including lung cancer. However, the precise underlying mechanisms are still largely unclear. In the present study, we investigated the metabolite changes in A549 cells induced by EGCG in vitro utilizing liquid chromatography-mass spectrometry (LC-MS)-based metabolomics. The result revealed 33 differentially expressed metabolites between untreated and 80 μM EGCG-treated A549 cells. The altered metabolites were involved in the metabolism of glucose, amino acid, nucleotide, glutathione, and vitamin. Two markedly altered pathways, including glycine, serine and threonine metabolism and alanine, aspartate and glutamate metabolism, were identified by MetaboAnalyst 5.0 metabolic pathway analysis. These results may provide potential clues for the intramolecular mechanisms of EGCG’s effect on A549 cells. Our study may contribute to future molecular mechanistic studies of EGCG and the therapeutic application of EGCG in cancer management.

Competitive binding of synergistic antioxidant chlorogenic acid and (−)-epigallocatechin gallate with lysozyme: Insights from multispectroscopic characterization, molecular docking and activity evaluation

The individual and combined binding of two active polyphenols, (−)-epigallocatechin gallate (EGCG) and chlorogenic acid (CGA), to lysozyme was studied at the molecular level by multispectroscopic techniques and molecular docking. The thermodynamic parameters obtained from fluorescence data indicated that the main driving force for the binding of CGA to lysozyme was electrostatic interaction while the binding of EGCG was mainly driven by hydrogen bonding and van der Waals’ forces. The results of circular dichroism spectroscopy and dynamic light scattering showed that the α-helical content and particle size of lysozyme decreased and increased, respectively, in the presence of EGCG and/or CGA. The competitive binding of EGCG and CGA to lysozyme was confirmed by molecular docking. The binding of EGCG and/or CGA inhibited the activity of lysozyme. The combination of EGCG and CGA had synergistic antioxidant activity and cytotoxicity to HeLa cells. Response surface methodology was used to analyze the maximum synergistic antioxidant activity between EGCG and CGA. The lysozyme + EGCG/CGA interaction weakened and enhanced the antioxidant activity and cytotoxicity of EGCG/CGA, respectively. These results would provide ideas for the combined application of EGCG and CGA in the fields of functional foods and pharmaceuticals.

Evaluation of Epigallocatechin-3-Gallate as a Radioprotective Agent During Radiotherapy of Lung Cancer Patients: A 5-Year Survival Analysis of a Phase 2 Study.

BackgroundPrevious analysis of the study (NCT02577393) had demonstrated the application of epigallocatechin-3-gallate (EGCG) could be safe and effective in the prevention and treatment of acute radiation esophagitis in patients with advanced lung cancer. EGCG seemed to improve the response rate of small cell lung cancer (SCLC) to radiotherapy in a subgroup analysis. This research continued to analyze the impact of EGCG application on cancer-radiation efficacy and patient survival.MethodsAll patients with SCLC in the NCT02577393 study were included. Patients were randomized into EGCG group or conventional therapy group as protocol. The primary endpoints of the study were radiation response rate and progression-free survival (PFS). Overall survival (OS) and the efficacy of EGCG in the treatment of esophagitis were assessed as secondary endpoints.ResultsA total of 83 patients with lung cancer in the NCT02577393 study were screened, and all 38 patients with SCLC were eligible for analysis. No significant differences with regard to baseline demographic and clinical characteristics were observed between the two groups. The objective response rate (ORR) was higher than that of conventionally treated patients (84.6 vs 50%, P = 0.045), while the median PFS and OS were not significantly prolonged. At data cut-off (1 January 2021), 5-year PFS was 33% with EGCG versus 9.3% with conventional treatment, and 5-year OS was 30.3% versus 33.3%, respectively. The mean adjusted esophagitis index and pain index of patients with EGCG application were lower than conventional treatment (5.15 ± 2.75 vs 7.17 ± 1.99, P = 0.030; 8.62 ± 5.04 vs 15.42 ± 5.04, P < 0.001).ConclusionThe study indicates EGCG may alleviate some esophagitis-related indexes in SCLC patients exposed to ionizing radiation without reducing survival. However, this conclusion should be confirmed by further studies with large sample size.

Topical epigallocatechin-3-gallate in the treatment of vitiligo.

To preliminarily assess the efficacy and safety of the topical application of Epigallocatechin-3-gallate (EGCG) in treating vitiligo, a 6-month clinical trial was carried out. Patients were randomly given topical application of EGCG on the assigned lesions, with pimecrolimus being used as the control for twice a day over a 6-month treatment period. Responses to treatment were assessed based on the changes in VASI score for percentage reduction in body surface area and the PGA scores. According to our results, both drugs were discovered to be markedly effective on repigmentation. The VASI of lesion had diminished from 1.19±0.42 to 0.63±0.38, in the EGCG-treated lesions, while from 1.18±0.43 to 0.61±0.36 in the pimecrolimus-treated lesions, and there was no statistically significant difference in VASI score between the EGCG-treated lesions and pimecrolimus-treated lesions (P=0.755). Meanwhile, the mean PGA score on the EGCG applied side was 4.39±2.23, while that was 4.43±2.02 on the pimecrolimus applied side (P=0.886). Furthermore, difference in the improvement degree between pimecrolimus side and EGCG side was not statistically significant (P=0.845). Notably, no serious side effects were observed throughout the study. Findings of the study indicate that topical EGCG can be effective on treating vitiligo.

Effects of epigallocatechin-3-gallate combined with ascorbic acid and glycerol on the stability and uric acid-lowering activity of epigallocatechin-3-gallate

Context Epigallocatechin-3-gallate (EGCG) is unstable and easily oxidized, which limits its applications. Ascorbic acid (Vc) is a natural antioxidant. Objective The effects of EGCG combined with Vc and glycerol on stability and uric acid-lowering activity of EGCG were examined. Materials and methods EGCG (aqueous solution), EGCG + Vc (aqueous solution), EGCG (glycerol solution) and EGCG + Vc (glycerol solution) were prepared and incubated under different conditions in vitro. The recovery rate of EGCG was calculated by HPLC. Kunming mice were randomly divided into normal control group, model group, allopurinol (5 mg/kg), EGCG (10 mg/kg), EGCG + Vc (both 10 mg/kg), EGCG (10 mg/kg) + glycerol (60%), and EGCG (10 mg/kg) + Vc (10 mg/kg) + glycerol (60%) (n = 6). Allopurinol was injected intraperitoneally to mice, others were administered intragastrically to (2 cases) mice. All mice were continuously administrated for 7 days, once a day. Results EGCG recovery rates of EGCG group and EGCG + Vc + glycerol group respectively reached to 32.34 ± 1.86% and 98.90 ± 0.64% when they were incubated for 4 h at 80 °C. EGCG recovery rates reached to 91.82 ± 5.13% (incubated for 6 h at pH 8) and 88.85 ± 2.63% (incubated for 4 h in simulated intestinal fluid) when EGCG incubated with Vc and glycerol. Compared with the model group, UA values of EGCG + Vc + glycerol group reduced by 43.49% while EGCG group reduced by 25.63%. The activities of xanthine oxidase (XOD, 31.41 U/gprot) and adenosine deaminase (ADA, 10.05 U/mgprot), and the mRNA expression levels of glucose transporter 9 (GLUT9, 1.03) and urate transporter 1 (URAT1, 0.44) in EGCG + Vc + glycerol group were notably lower than those of EGCG group (38.12 U/gprot, 13.16 U/mgprot, 1.54, and 0.55). The mRNA expression levels of ATP-binding cassette superfamily G member 2 (ABCG2, 1.39) and organic anion transport 1/2 (OAT1/2, 2.34, 2.53) in EGCG + Vc + glycerol group were notably higher than those of EGCG group (0.57, 1.13, and 1.16). Discussion and conclusions Our findings suggest that when EGCG used in combination with Vc and glycerol could effectively increase its biology activities and can be generalized to the broader pharmacological studies. This sheds light on the development and application of EGCG in the fields of food and medicine.

Efficacy of topical hydrogel Epigallocatechin-3-gallate against neutrophil cells in perforated dental pulp

Background: One cause of pulpitis is mechanical trauma such as pulp perforation. The emergency treatment of pulpitis in a clinic uses eugenol. Eugenol in a high concentration causes cytotoxicity, which causes local necrosis and inhibits the recovery process, while in lower doses it can cause oral mucosal hypersensitivity. Due to these side effects, it is worth considering other biocompatible materials with minimal side effects, such as epigallocatechin-3-gallate (EGCG) which is found in green tea. As a polyphenol, EGCG has a radical scavenging ability, which has an effect on reducing the number of neutrophils. The application of EGCG is expected to reduce neutrophils on the second day after injury so the rehabilitation process is completed more quickly and ongoing inflammation and pulp necrosis is prevented. Purpose: To analyse the efficacy of topical hydrogel EGCG in reducing the number of neutrophils after 48 hours in the perforated dental pulp of Wistar rats. Method: 20 Wistar rats were divided equally into four groups, which were designated control (C) and treatment groups (T1, T2, T3). The upper first molar teeth of each rat were perforated and then T1, T2, and T3 were given 60 ppm, 90 ppm and 120 ppm hydrogel EGCG respectively. On the second day, the rats were sacrificed. HPA preparations were made to calculate the number of neutrophils in each group. Data was analysed using Kolmogorov–Smirnov, Levene’s, one-way ANOVA and Tukey HSD test (p&lt;0.05). Results: There were significant differences between T2 and T3 compared with C and T1 (p&lt;0.05), but no significant differences in the comparison of T1 with C and of T2 with T3 (p&gt;0.05). Conclusion: 90 ppm hydrogel EGCG is effective in reducing the number of neutrophils in the perforated dental pulp of Wistar rats.

Green Tea Catechin (-)-Epigallocatechin-3-Gallate (EGCG) Facilitates Fracture Healing.

Green tea drinking can ameliorate postmenopausal osteoporosis by increasing the bone mineral density. (-)-Epigallocatechin-3-gallate (EGCG), the abundant and active compound of tea catechin, was proven to be able to reduce bone loss and ameliorate microarchitecture in female ovariectomized rats. EGCG can also enhance the osteogenic differentiation of murine bone marrow mesenchymal stem cells and inhibit the osteoclastogenesis in RAW264.7 cells by modulation of the receptor activator of nuclear factor-kB (RANK)/RANK ligand (RANKL)/osteoprotegrin (OPG) (RANK/RANKL/OPG) pathway. Our previous study also found that EGCG can promote bone defect healing in the distal femur partially via bone morphogenetic protein-2 (BMP-2). Considering the osteoinduction property of BMP-2, we hypothesized that EGCG could accelerate the bone healing process with an increased expression of BMP-2. In this manuscript, we studied whether the local use of EGCG can facilitate tibial fracture healing. Fifty-six 4-month-old rats were randomly assigned to two groups after being weight-matched: a control group with vehicle treatment (Ctrl) and a study group with 10 µmol/L, 40 µL, EGCG treatment (EGCG). Two days after the operation, the rats were treated daily with EGCG or vehicle by percutaneous local injection for 2 weeks. The application of EGCG enhanced callus formation by increasing the bone volume and subsequently improved the mechanical properties of the tibial bone, including the maximal load, break load, stiffness, and Young’s modulus. The results of the histology and BMP-2 immunohistochemistry staining showed that EGCG treatment accelerated the bone matrix formation and produced a stronger expression of BMP-2. Taken together, this study for the first time demonstrated that local treatment of EGCG can accelerate the fracture healing process at least partly via BMP-2.

Epigallocatechin-3-gallate enhances tomato resistance to tobacco mosaic virus by modulating RBOH1-dependent H2O2 signaling

Tobacco mosaic virus (TMV) is one of the most damaging plant viruses from an economic and research point of view. Epigallocatechin-3-Gallate (EGCG), a flavonoid type secondary metabolite can selectively improve plant defense against pathogens; however, the effect of EGCG on plant defense against TMV and the underlying mechanism(s) remain elusive. In this study, exogenous EGCG application increased plant resistance to TMV as revealed by significantly decreased transcript levels of TMV-coat protein (CP) in tomato leaves. A time-course of H2O2 concentrations in tomato leaves showed that TMV inoculation rapidly increased the H2O2 accumulation, reaching its peak at 3 days post-inoculation (dpi) which remained the highest until 6 dpi. However, the combined treatment of EGCG and TMV remarkably decreased the concentrations of H2O2 at 3 and 6 dpi. Meanwhile, the transcript levels of RESPIRATORY BURST OXIDASE HOMOLOG 1 (SlRBOH1) were significantly increased by either EGCG or TMV inoculation, but the EGCG treatment along with TMV caused a further upregulation in the SlRBOH1 transcripts compared with that in only TMV-inoculated plants. Chemical scavenging of H2O2 or silencing SlRBOH1 both compromised the EGCG-induced enhanced resistance to TMV. Furthermore, EGCG-induced elevation in the activity of antioxidant enzymes was abolished by SlRBOH1 silencing, suggesting that EGCG enhanced defense against TMV by increasing the antioxidant enzyme activity via RBOH1-dependent H2O2 signaling. Taken together, our results suggest that EGCG functioned to maintain a delicate balance between ROS signaling and ROS scavenging via RBOH1, which enhanced tomato resistance to TMV.

Biofilm inhibition and mode of action of epigallocatechin gallate against Vibrio mimicus

Vibrio mimicus is a relatively rare food-borne pathogen in seafood and water. Rare reports have been published to investigate the biofilm of Vibrio mimicus. (−)-epigallocatechin-3-gallate (EGCG), the major polyphenolic component of tea, can interfere with bacterial biofilm formation. This study showed that Vibrio mimicus was capable of forming high amounts of biofilms in various culture media. Sub-MICs of EGCG significantly reduced the biofilm production of Vibrio mimicus at 15 °C, 28 °C and 37 °C. Confocal laser-scanning microscope (CLSM) observation proved that the architecture of Vibrio mimicus biofilm was affected by EGCG at the concentration of 64 μg/mL (1/4 MIC). EGCG reduced Vibrio mimicus autoaggregation and swimming motility. EGCG also increased membrane permeability and ROS production, caused cell membrane damage and led to potassium leakage. These may contribute to the antibiofilm efficacy of EGCG against Vibrio mimicus. Our work showed the inhibitory effects of sub-MICs of EGCG on Vibrio mimicus biofilm for the first time, supporting the potential application of EGCG as a natural antibiofilm agent in the food industry.
