Testicular morphometry and epididymal sperm qualities of donkeys in Bolgatanga, Ghana

The application of artificial insemination in donkeys must be based on a knowledge of breeding soundness, while other reproductive physiology of the donkey including testicular morphometric parameters and sperm production is scanty in literature. This study sought to assess the testicular morphometry and epididymal sperm qualities of donkeys in Bolgatanga and to determine the effect of season on spermatogenesis and epididymal sperm parameters. Twelve sexually mature donkeys, aging 10 to 15 years old, had their testes and epididymis surgically harvested immediately after slaughter, semen parameters were assessed thereafter and testicular volume calculated. Epididymal sperm was harvested by retrograde flushing technique and sperm parameters were obtained. This study showed no significant difference (p-value >0.05) between the length, width and volume of the left and right testes, and that the rainy season showed greater values in seminiferous tubule diameter, epithelial height, and luminal diameter. The study revealed that donkeys showed a greater level of spermatogenesis in the rainy season compared to the dry season, indicating that donkeys are seasonal breeders.

A Policy Brief on the Application of Artificial Insemination (AI) in Lanao del Sur

This paper is a policy brief regarding the application of the Artificial Insemination (AI) as a mode of reproductive technology in Lanao del Sur. Lanao del Sur is one of the provinces in the Bangsamoro Autonomous Region in Muslim Mindanao (BARMM), the dwelling of Meranao tribes who practice Islam as their religion. The province has favorable climatic conditions for agricultural productions both in crops and animals’ production. Lanao del Sur is a landlocked province and is agriculturally rich having a well-suiting climate and extensively arable land, and it has a growing inventory of livestock such as carabao, cattle, goat, duck and chicken. This policy brief was based on the research findings conducted regarding the perceptions and acceptance of the AI among livestock growers. The goal of this policy brief is to provide baseline data about the farmers perceptions on AI and formulate a research-based policy recommendation to help the LGU, implementing agencies and the farmers for effective and efficient implementation of AI program in the province. The6 following are the salient points: 1) Meranao livestock growers’ perceptions about using Artificial Insemination (AI). 2) Capacitating the farmers towards adoption of artificial insemination, and breeding programs. 3) Ensuring efficiency of livestock production among farmers. 4) Increase the livestock production in Lanao del Sur.

Female Reproductive Organ Monitoring (FROM) as a method of enhancing livestock reproduction

Low conception rate, long calving interval, and poor calf production limits the profitability of livestock production keeping food security a concern. In the ASEAN region, artificial insemination (AI) remains the most common reproductive biotechnology used for livestock breeding to produce up-graded and genetically superior animals that could address the need for food and income to the rural farming communities. Field technicians play a crucial role in carrying out AI to enhance reproduction especially in the remote areas. While ultrasonography emerged as an efficient tool to check the reproductive organ of animals for efficient reproduction, the cost of the machine hinders its availability and access by Field technicians. Skills on palpation per rectum of the ovarian dynamics and tonicity of the uterus are important assets. This paper discusses the importance of Female Reproductive Organ Monitoring or FROM in the economical and efficient application of AI in the field and enhance livestock reproduction. Strategies to develop the skills of AI practitioners are outlined to come out to skilled technicians who could execute an accurately check the reproductive status of the animals and determine the perfect time to carry out an effective and efficient AI. This method is helpful in determining the timing of AI making the resources, time and efforts of the technicians maximized and livestock production more efficient.

43 Establishing Pregnancy Following Artificial Insemination (AI) of Angus Cows: Experience from Fort Valley State University (FVSU)

Abstract Fixed-time artificial insemination (FTAI) provides a practical and feasible option for commercial and stud breeders to implement AI into their herds, as 100% of the herd can be inseminated at a predetermined time. FTAI can suit many herds, whether they are inseminating small numbers or the whole herd. We investigated the effects of sire and inseminator on pregnancy and birth outcomes following FTAI of Angus cows. Three Angus sires; namely GAR Quantum 7AN560, Square B True North 8052, and Tehama Tahoe B767, and two inseminators (1 and 2) were evaluated for differences in pregnancy outcomes and birth rates from 24 inseminations. Angus cows (N = 24) were synchronized with CIDR plus GnRH injection. After 7 days, CIDR was removed and each cow received 25 mg PGF2α. AI was conducted within 72 h from PGF2α injection. Cows were randomly assigned for AI between inseminators (12 cows inseminated by inseminator 1 and 12 cows by inseminator 2) and sires (6, 8, and 10 cows were inseminated by GAR Quantum, Tehama Taho, and True North semen, respectively. Pregnancy was diagnosed at 4-5 months after AI by transrectal ultrasonography. Percentage of re-cycling cows 21 days after AI was 45.8% (11/24). Pregnancy rate was 54.2% (13/24). Pregnancy rate was higher (P<0.05) for cows inseminated by inseminator 1 than by inseminator 2 (69.2% vs. 30.8%). The average age of pregnancy was 5.6 ± 0.2 months. Among sires, 46.2% (6/13) of the pregnant cows were inseminated by True North, 38.5% (5/13) by Tehama Tahoe, and 15.4% (2/13) by GAR Quantum. The pregnant cows gave birth to healthy calves at 280.8 ± 2.1 days after AI. In conclusion, both inseminator and sire affected the pregnancy rates and birth outcomes after AI in beef cattle. This study was conducted to support the establishment of a purebred Black Angus beef herd at FVSU with the application of AI.

Dietary Lasia spinosa Thw. improves reproductive performance of aged roosters.

The application of artificial insemination is particularly, owing to which breeder animals are considered an important resource in breeding farms. However, the reproductive performance of roosters typically declines with age, and the economic loss experienced by breeders is attributable to this shortened reproductive lifespan. Lasia spinosa Thw. (LST) reportedly improved reproductive capacity in male rodents. The objective of this study was to investigate the effects of LST on the reproductive performance of aged roosters. Male Guangxi Partridge chicken (mean weight, 3032.41 ± 34.48 g; age, 500 days; n = 72) randomly received the following three dietary treatments: LST0 group (a basal diet), LST2 group (a basal diet with 2% LST powder), and LST4 group (a basal diet with 4% LST powder). Computer-aided sperm analysis revealed that dietary LST supplementation significantly improved semen volume, sperm motility, and concentration. Furthermore, the most potent effects were observed in the treatment group with the administration of 2% LST, which significantly improved the weight of the testes. Hematoxylin-eosin staining revealed the increase in diameter of the seminiferous tubule and height of the seminiferous tubule epithelium possibly caused as a result of LST treatment. A significant increase in fructose and glucose concentrations were observed in the testis and seminal plasma; in addition, a significant increase was observed in the α-glycosidase levels in the testis and spermatozoa. However, the monoaldehyde levels in the spermatozoa appeared to decline significantly. Additionally, the fertility rate increased significantly following 2% LST supplementation. RNA-seq analysis revealed that 34 and 16 unigenes were upregulated and downregulated, respectively, in testicular tissues from roosters that received dietary supplementation of 2% LST. The assigned functions of the unigenes revealed that LST primarily influenced the mechanisms underlying catalytic activity and cellular processes. Kyoto Encyclopedia of Genes and Genomes enrichment analysis suggested that spermatogenesis-related pathways were significantly enriched, including ABC transporters, ribosome biogenesis in eukaryotes, and VEGF, cAMP, and ErbB signaling pathways.

Application of Artificial Insemination (AI) as a Breeding Method for Dairy Goat Herd Build-Up in Region 3, Philippines

One of the constraints in the development of dairy industry in the country is the limited number of dairy animals of which only 14% are goats providing only 1% of the total volume of local milk production. Meanwhile, increasing dairy goat population is hindered by the lack or the high price of dairy breeder animals that can be used in improving our local stocks. To spur the population of goats with genetic potential for dairying among goat farms in Region 3, Philippines, artificial insemination (AI) was applied in this project. Within the implementation period, a total of 50 participants were trained on AI in goats. They served as AI service providers in their respective and nearby municipalities. Meanwhile, a total of 4 seminars on goat raising was conducted at the different municipalities to promote the use of AI in goat breeding and upgrading which were participated by a total of 166 goat raisers. A total of 346 inseminations were reported by the trained technicians. Based on the total number of inseminations monitored, the observed conception rate was 60.29% ranging from 37.50% in Bulacan to 100% in Zambales. Of the total number of kiddings recorded, the percent of singletons, twins, and triplets observed were 46.38%, 44.93%, and 8.69%, respectively, giving an average kidding size of 1.17. A total of 106 kids were born with 61.32% (65 kids) males and 38.68% (41 kids) females with an overall male to female ratio of 1.59:1. Majority (73.58%) of the total kids born were of upgraded Anglo-Nubian bloodline. Meanwhile, of the five commercial farms visited, the Soliman Goat Farm in Tarlac was selected as the location for the establishment of the farm-based goat semen processing laboratory. Moreover, the owner and the farm staff involved in operating the laboratory had already undergone training on semen collection, processing, and evaluation.

Semen evaluation: methodological advancements in sperm quality-specific fertility assessment - A review.

Assessment of male fertility is based on the evaluation of sperm. Semen evaluation measures various sperm quality parameters as fertility indicators. However, semen evaluation has limitations, and it requires the advancement and application of strict quality control methods to interpret the results. This article reviews the recent advances in evaluating various sperm-specific quality characteristics and methodologies, with the help of different assays to assess sperm-fertility status. Sperm evaluation methods that include conventional microscopic methods, computer-assisted sperm analyzers (CASA), and flow cytometric analysis, provide precise information related to sperm morphology and function. Moreover, profiling fertility-related biomarkers in sperm or seminal plasma can be helpful in predicting fertility. Identification of different sperm proteins and diagnosis of DNA damage has positively contributed to the existing pool of knowledge about sperm physiology and molecular anomalies associated with different infertility issues in males. Advances in methods and sperm-specific evaluation has subsequently resulted in a better understanding of sperm biology that has improved the diagnosis and clinical management of male factor infertility. Accurate sperm evaluation is of paramount importance in the application of artificial insemination and assisted reproductive technology. However, no single test can precisely determine fertility; the selection of an appropriate test or a set of tests and parameters is required to accurately determine the fertility of specific animal species. Therefore, a need to further calibrate the CASA and advance the gene expression tests is recommended for faster and field-level applications.

The Attitude Changes Of Local Farmers Toward The Introduction Of Artificial Insemination In The Extensive Of Cattle Raising In The Seram Bagian Barat District, Maluku Province, Indonesia

Special efforts for cow must be pregnant (Upsus Siwab) is a program for the achievement of meat adequacy in 2022 through the optimization strategy of implementing Artificial Insemination (AI) in 34 provinces including Maluku Province. One of them is Seram Bagian Barat District. The type of cattle that are kept are Bali with extensive management. Maluku Province is The AI introduction area, so there are many challenges to meet the targets set. The purpose of this paper is to know the performance of AI introductions and the effect toward farmer attitudes in the SBB District during the 3 years of assistance. The method of this research was a survey, interviews with farmers, field officers, and district officers. Data with the Ishiknas format for 3 years was collected from the Agriculture services of SBB District. Data was tabulated and analyzed descriptively. The basic problems related to the application of AI are (1) it is difficult to detect the oestrus because all cattle are released, (2) the habits of mating cattle naturally, (3) there is no interest in participating of AI, and (4) the knowledge of oestrus is very minimal. The strategy to solved the problem was introduction the hormone of Oestrus Synchronization (SE). The strategy was quite successful, in 2017, the realization of AI was 85%. Realization in 2018 and 2019 were more than 100%. The S/C score was 1.13 times indicates that the cows have very good reproductive performance. There were 15 variations in gestational age with the range of 8 months 4 days - 9 months 17 days. The most average were 9 months 10 days (16.6%) and 9 months 11 days (16.6%). The pregnancy rates of AI was 10 -311 cows/month with an average of 108.8 cows, while in natural marriages 0 - 32 cows/month with an average of 12.5 cows. The birth weights of AI with Bali bulls straw ranged from 17 to 23 kg (an average of 19.6 kg), while the natural mating was an average of 16 kg. Another results, the birth weight of AI with Ongole bulls straw on male was an average of 32.2 kg and 30 kg on female. In 2017, the implementation of the AI was all done in combination with the SE because no farmers wanted to report the oestrus of cows. In 2018 there were reports from farmers and in 2019, all AI implementations were based on normal oestrus reports. The change in attitudes of farmers regarding adoption of AI was caused by the calves had a higher birth weight and a higher selling price. Implementation of AI in the District of SBB has good prospects for increasing the quantity and quality of cattle and also increasing the income of farmers

Oviductal Extracellular Vesicles Improve Post-Thaw Sperm Function in Red Wolves and Cheetahs.

Artificial insemination (AI) is a valuable tool for ex situ wildlife conservation, allowing the re-infusion and dissemination of genetic material, even after death of the donor. However, the application of AI to species conservation is still limited, due mainly to the poor survival of cryopreserved sperm. Recent work demonstrated that oviductal extracellular vesicles (oEVs) improved cat sperm motility and reduced premature acrosomal exocytosis. Here, we build on these findings by describing the protein content of dog and cat oEVs and investigating whether the incubation of cryopreserved red wolf and cheetah sperm with oEVs during thawing improves sperm function. Both red wolf and cheetah sperm thawed with dog and cat oEVs, respectively, had more intact acrosomes than the non-EV controls. Moreover, red wolf sperm thawed in the presence of dog oEVs better maintained sperm motility over time (>15%) though such an improvement was not observed in cheetah sperm. Our work demonstrates that dog and cat oEVs carry proteins important for sperm function and improve post-thaw motility and/or acrosome integrity of red wolf and cheetah sperm in vitro. The findings show how oEVs can be a valuable tool for improving the success of AI with cryopreserved sperm in threatened species.

Reproductive management in buffalo by artificial insemination

Artificial insemination (AI) is important for genetic improvement and to control the period of breeding in buffalo and has increased significantly over the past 20 years. AI is more difficult in buffalo compared with cattle due to variable estrous cycles, reduced estrous behavior, and reproductive seasonality. The latter is associated with a higher incidence of anestrus and increased embryonic mortality during the nonbreeding season. Protocols to control the stage of the estrous cycle have undergone recent development in buffalo. These protocols are based on the control of both the luteal phase of the cycle, mainly by prostaglandins and progesterone, and follicle development and ovulation, by prostaglandins, progesterone, GnRH, hCG, eCG and estradiol. Protocols that synchronize the time of ovulation enable fixed timed AI, avoiding estrous detection. Factors to consider when selecting an AI protocol include animal category (heifers, primiparous or pluriparous), reproductive status (cyclic or anestrus), and season. This review looks at the current status of estrus synchronization and AI in buffalo and provides some practical suggestions for application of AI in the field.

Adiponectin: Potential Protein Hormone as a Candidate Biomarker for Male Fertility

Reproduction is an important factor that requires attention to increase livestock production. The application of artificial insemination (AI) technology has been developed rapidly in the world, so that availability of fertile superior male is a determinant factor of successful AI. Several recent studies have been aimed to discover male fertility biomarkers, by intense research on fertility-assosiated proteins contained in seminal plasma. This paper describes the role of adiponectin as a candidate of male biomarker fertility. Adiponectin has a positive effect on spermatozoa functionality and steroidogenesis. This has been proven by several studies that found its expression in Sertoli cells and Leydig cells in the reproductive tract. Based on the specific role on spermatozoa functionality and steroidogenesis in supporting male fertility parameter, it is strongly suggested that adiponectin is an excellent candidate biomarker for male fertility.

ABSTRACTCATTLE FARMERS’S RESPONSE ON APPLICATION OF ARTIFICIAL INSEMINATION (AI) TEGNOLOGY IN TONDEGESAN VILLAGE KAWANGKOAN DISTRICT, MINAHASA REGENCY. The government encourages the cattle farming industry through science and technology to increase the productivity of livestock businesses in order to be able to increase the livestock population. The objective of this study was to determine the farmer's response to the adoption of artificial insemination technology (IB) at the village of Tondegesan Dua, Kawangkoan District, Minahasa Regency and to evaluate their knowledge on artificial insemination motivation and technology adoption (IB) in Tondegesan two Village, Kawangkoan District, Minahasa Regency. Data sources were based on primary and secondary data. Data collection were conducted by observation and interview method. The farmer population were involving 42 farmers following artificial insemination and not following artificial insemination activities, but they were aware of artificial insemination technology. The number of samples taken were 30 farmers. Determination of the sample was purposively taken at the village of Tondegesan Dua. The measurement of the research indicator was carried out by applying a Likert scale. Test was done on the relationship between the variables of farmer response to artificial insemination. The results showed that the farmer's responses to artificial insemination were in a fairly good category based on the results of the study. The objective of this study was to compare the reproductive traits of heifers and the development characteristics of their calves following artificial insemination (AI) with sexed and non-sexed semen. The analysed characteristics included conception rate, gestation length, calf birth weight, calf vigour, stillbirth rate, and twinning rate. Data of 530 calves produced with sexed and 1,163 calves produced with non-sexed semen were analysed. The General Linear Model (GLM) was applied to assess the influence of semen type, farm, season of insemination, the calf's sex and the inseminating sire on gestation length and calf birth weight. With the exception of gestation length (P > 0.05), all other traits studied were significantly (P < 0.01) influenced by the type of semen. The conception rate was 55% for conventional and 44% for sexed semen, and the average gestation length was 274.6 and 274.9 days, respectively. The mean calf birth weight was 37.47 kg for non-sexed and 36.75 kg for sexed semen. The stillbirth rate was 6.19% for conventional and 7.54% for sexed semen, while the twinning rate was 3.78% for conventional and 1.13% for sexed semen. The calves produced with non-sexed and sexed semen differed significantly in viability (P < 0.001), the latter having a lower calf vigour score. The use of conventional semen did not affect the ratio of female and male calves (52.7:47.3%; P > 0.05); however, artificial insemination with X-sorted sexed semen significantly altered the sex ratio of calves (85.1:14.9%, P < 0.01). The results obtained in this investigation are in agreement with the majority of studies which compared the fertility traits, sex ratio and calf characteristics depending on the application of artificial insemination with sexed or conventional semen.

A limited number of Y chromosome lineages is present in North American Holsteins

Holsteins are the most numerous dairy cattle breed in North America and the breed has undergone intensive selection for improving milk production and conformation. Theoretically, this intensive selection could lead to a reduction of the effective population size and reduced genetic diversity. The objective of this study was to investigate the effective population size of the Holstein Y chromosome and the effects of limited Y chromosome lineages on male reproduction and the future of the breed. Paternal pedigree information of 62,897 Holstein bulls born between 1950 and 2013 in North America and 220,872 bulls evaluated by multiple-trait across-country genetic evaluations of Interbull (Uppsala, Sweden) were collected and analyzed. The results indicated that the number of Y chromosome lineages in Holsteins has undergone a dramatic decrease during the past 50 years because of artificial selection and the application of artificial insemination (AI) technology. All current Holstein AI bulls in North America are the descendants of only 2 ancestors (Hulleman and Neptune H) born in 1880. These 2 ancestral Y-lineages are continued through 3 dominant pedigrees from the 1960s; namely, Pawnee Farm Arlinda Chief, Round Oak Rag Apple Elevation, and Penstate Ivanhoe Star, with a contribution of 48.78, 51.06, and 0.16% to the Holstein bull population in the 2010s, respectively. The Y-lineage of Penstate Ivanhoe Star is almost eliminated from the breed. The genetic variations in the 2 ancestral Y-lineages were evaluated among 257 bulls by determining the copy number variations (CNV) of 3 Y-linked gene families: PRAMEY, HSFY, and ZNF280BY, which are spread along the majority (95%) of the bovine Y chromosome male-specific region (MSY). No significant difference was found between the 2 ancestral Y-lineages, although large CNV were observed within each lineage. This study suggests minimal genetic diversity on the Y chromosome in Holsteins and provides a starting point for investigating the effect of the extremely limited number of Y-lineages on male reproduction and other traits important for the future of the Holstein breed.

Synchronization of ovulation and fixed-time artificial insemination in beef cattle

The main objective of the implementation of artificial insemination (AI) in cattle is to produce a sustained genetic progress in the herd. Although AI is an old reproductive biotechnology, its widespread implementation is very recent and is mainly because of the use of protocols that allows AI without oestrus detection, commonly called fixed-time artificial insemination (FTAI). The development of FTAI protocols also allowed the application of AI in larger, extensively managed herds and especially in suckled cows instead of restricting the breeding programmes to the heifers. Fixed-time AI treatments are widely used in South America, with about 3 000 000 cows inseminated in the last season in Argentina and about 8 000 000 in Brazil. The objective of this review is to present and describe the various treatments available and some of the factors that may affect pregnancy rates in beef cattle.

Application of estrus synchronization for artificial insemination to improve productivity and quality of beef cattle

Local yellow cattle breed still made up a high percentage of total cattle population in Vietnam. These breed should be inseminated with exotic breed to increase their genetic merit. As the cattle bred in smallholder farmers display estrus sporadic and unconcentrated, application of artificial insemination (AI) tends to limit to areas in close proximity to urban city. Therefore, solutions should be found to enhance the possibilities for using artificial insemination. This paper reports the application of estrus synchronization for artificial insemination of beef cattle. A total of 524 local yellow and Sind crossbreed cattle were assigned randomly into one of two experimental groups. Group 1: cows received 2 ml of PGF2α, followed in 11 d with 2 ml PGF2α and 500 UI PMSG on the same day. Group 2: cows were administered 100 µg GnRH, followed in 7 d with 2 ml of PGF2α. After the last injection in each group, cows were observed for estrus twice daily and those displaying estrus were artificial inseminated. At the end of experiment, rates of detected estrus, conception and parturition in the group 1 and 2 are 84.90 and 82.08%, 82.88 and 80.28%, 93.52 and 92.98% respectively. The results showed that synchronization of estrus has the potential to enhance the possibilities for using AI and help to improve productivity and quality of beef cattle.

Laparoscopic Oviductal Embryo Transfer and Artificial Insemination in Felids – Challenges, Strategies and Successes

Embryo transfer (ET) and artificial insemination (AI) are potentially invaluable techniques for the propagation and management of genetically valuable domestic cat and endangered nondomestic cat populations. Many of the challenges that impair the effective application of ET and AI in felids may be overcome by using laparoscopic oviductal (LO) approaches. LO-ET and LO-AI are minimally-invasive procedures, requiring only two small skin incisions for insertion of a laparoscope and grasping forceps into the abdominal cavity to permit visualization and catheterization of the oviduct for embryo or semen deposition. With concurrent improvements in embryo culture systems and ovarian synchronization protocols, LO-ET has proven effective over the past decade for propagation of laboratory cats, cat models of hereditary disease and nondomestic cats. To date, viable offspring have been produced following LO-ET of non-frozen and frozen-thawed IVF-derived embryos in eight cat hereditary disease models and two nondomestic cat species, the ocelot and sand cat. LO-AI with low sperm numbers (c. 2-8 million motile) has shown similar efficacy to LO-ET, resulting in high pregnancy percentages (50-70%) following insemination of gonadotropin-treated domestic cats. Multiple kittens also have been produced in two hereditary disease models following LO-AI with frozen semen, and both ocelot and Pallas' cat kittens have been born after LO-AI with freshly-collected semen. The application of LO-ET and LO-AI to felids has resulted in substantial improvement in the efficiency of assisted reproduction for genetic management of these invaluable domestic cat and wild cat populations.

Technologies Related with the Artificial Insemination in Buffalo

In buffalo oestrus behaviour has a lower intensity than in cows and is much more difficult to detect, limiting the application of artificial insemination (AI) program. Several methods of heat detection have been developed for use in cattle; these include visual observation, heat mount detectors, tail paint, chin-ball markers, teaser animals and electronic devices. In buffalo, unlike cattle, the female are receptive to mounting activity mainly by the bull and occasionally by other cows. Consequently unless a buffalo bull is to be left running with the herd it can be difficult to know when oestrus is occurring. The presence of a teaser bull is helpful to identify buffaloes in heat; in this case the standing oestrus is the most reliable sign referable to a next ovulation. Other heat detection aids utilized in buffalo include: pedometers; vaginal probes; pressure sensitive telemetry device (Heat Watch®). In order to increase the use of AI easy management schemes, that not require the identification of oestrus, have been studied. These schemes are based on the manipulation of the hormonal events occurring during the oestrous cycle as: manipulate peripheral progesterone concentration (by PGF2a or progesterone releasing device); manipulate follicular growth and timing of ovulation (by GnRH and PGF2a). A brief description of these technologies, with special reference to synchronization protocols to apply fixed time AI in buffalo, are presented in this review. The potential application of predetermining the sex of offspring will be also discussed, with reference to the techniques available for commercial practice in buffalo.
