Application of atmospheric cold plasma for zearalenone detoxification in cereals: Kinetics, mechanisms, and cytotoxicity analysis

IntroductionZearalenone (ZEN) is one of the most widely contaminated mycotoxins in world, posing a severe threat to human and animal health. Atmospheric cold plasma (ACP) holds great penitential in mycotoxin degradation. ObjectivesThis study aimed to investigate the degradation efficiency and mechanisms of ACP on ZEN as well as the cytotoxicity of ZEN degradation products by ACP. Additionally, this study also investigated the degradation efficiency of ACP on ZEN in cereals and its effect on cereal quality. MethodsThe degradation efficiency and products of ZEN by ACP was analyzed by HPLC and LC-MS/MS. The human normal liver cells and mice were employed to assess the cytotoxicity of ZEN degradation products. The ZEN artificially contaminated cereals were used to evaluate the feasibility of ACP detoxification in cereals. ResultsThe results showed that the degradation rate of ZEN was 96.18 % after 30-W ACP treatment for 180 s. The degradation rate was dependent on the discharge power, and treatment time and distance. Four major ZEN degradation products were produced after ACP treatment due to the oxidative destruction of CC double bond, namely C18H22O7 (m/z = 351.19), C18H22O8 (m/z = 367.14), C18H22O6 (m/z = 335.14), and C17H20O6 (m/z = 321.19). L02 cell viability was increased from 52.4 % to 99.76 % with ACP treatment time ranging from 0 to 180 s. Mice results showed significant recovery of body weight and depth of colonic crypts as well as mitigation of glomerular and liver damage. Additionally, ACP removed up to 50.55 % and 58.07 % of ZEN from wheat and corn. ConclusionsThis study demonstrates that ACP could efficiently degrade ZEN in cereals and its cytotoxicity was significantly reduced. Therefore, ACP is a promising effective method for ZEN detoxification in cereals to ensure human and animal health. Future study needs to develop large-scale ACP device with high degradation efficiency.

Effects of the application of low-temperature atmospheric plasma on titanium implants on wound healing in peri-implant connective tissue in rats

PurposeThis study aimed to clarify the effects of surface modification of titanium (Ti) implants by low-temperature atmospheric pressure plasma treatment on wound healing and cell attachment for biological sealing in peri-implant soft tissue.MethodsHydrophilization to a Ti disk using a handheld low-temperature atmospheric pressure plasma device was evaluated by a contact angle test and compared with an untreated group. In in vivo experiments, plasma-treated pure Ti implants using a handheld plasma device (experimental group: PL) and untreated implants (control group: Cont) were placed into the rat upper molar socket, and samples were harvested at 3, 7 and 14 days after surgery. Histological evaluation was performed to assess biological sealing, collagen- and cell adhesion-related gene expression by reverse transcription quantitative polymerase chain reaction, collagen fiber detection by Picrosirius Red staining, and immunohistochemistry for integrins.ResultsIn in vivo experiments, increased width of the peri-implant connective tissue (PICT) and suppression of epithelial down growth was observed in PL compared with Cont. In addition, high gene expression of types I and XII collagen at 7 days and acceleration of collagen maturation was recognized in PL. Strong immunoreaction of integrin α2, α5, and β1 was observed at the implant contact area of PICT in PL.ConclusionsThe handheld low-temperature atmospheric pressure plasma device provided hydrophilicity on the Ti surface and maintained the width of the contact area of PICT to the implant surface as a result of accelerated collagen maturation and fibroblast adhesion, compared to no plasma application.

Evaluation of Low-Pressure Cold Plasma Effect on Phytonutrients, Drying and Microstructural Characteristics, and Changes in Microbial Load of Fresh and Hot-Air-Dried ‘Heidi’ Mango

AbstractAlternative pre-treatment strategies before drying offer the prospect to minimize drying time, replacing the use of chemicals, and preserving quality of dried fresh products. This study explored the application of low-pressure atmospheric cold plasma (CP) for 5- and 10 min (CP5 and CP10) as pre-treatments prior to processing and hot air drying (60 °C) of ‘Heidi’ mango, while non-treated samples served as control. Changes in tissue microstructure and physicochemical properties, bioactive compounds, and microbial load were evaluated, and seven thin layer drying models were applied. Scanned electron microscope images showed that CP pre-treatments altered the tissue microstructure of dried mango slices compared to control. ‘Heidi’ mango slices with the initial moisture content of 80 ± 0.2% on a wet basis was reduced by 81% and 76% in dried CP5 and CP10 samples, respectively. Drying time was reduced by 20% for CP pre-treated samples in comparison to control samples, and the drying behavior of ‘Heidi’ mango slices was best described by the Logarithmic model (R2, 0.9999 and RMSE, 0.0122). Colour attributes were best retained by sodium metabisulphite (SMB) pre-treated samples, followed by CP5 pre-treated, which performed better than CP10 and control (p ≤ 0.05). Highest total flavanols (15.0 ± 0.4 mg CE 100 g−1) and higher total phenolics (1528.2 ± 23.6 mg GA 100 g−1) were found in CP5 pre-treated samples compared to the control (p ≤ 0.05). Lowest antioxidant activities were found in CP10 pre-treated samples compared to the control (p ≤ 0.05). CP-pretreatment and drying resulted in ≥ 2 Log reduction in microbial load on mango slices. These results demonstrate to the role players in mango value addition chain, the potential of low-pressure CP pretreatment in enhancing/maintaining the bioactive compounds, reducing drying time and microbial load.

Inactivation of isolated fungi on Erythrina velutina Willd. seeds through atmospheric plasma.

This study aimed to evaluate the effect of atmospheric plasma application on the inactivation of fungi on the surface of Erythrina velutina seeds and on isolated fungal colonies. Two experiments were conducted using a completely randomized design. First, plasma was applied to the surface of the seeds using helium gas and atmospheric plasma for 3, 6, and 9 min in addition to the control (untreated seeds), constituting seven treatments with five repetitions each. In the second experiment, Petri dishes containing the inoculum of different fungi were treated with atmospheric air plasma for 3, 6, and 9 min (Air-3, Air-6, and Air-9) and were compared with untreated fungi in Petri dishes without treatment (control), totaling four treatments and five repetitions each. We found that the application of atmospheric air plasma to E. velutina seeds for 9 min had an antimicrobial effect on the fungi Aspergillus niger, Aspergillus flavus, Fusarium sp., Brachysporium sp., and Rhizopus sp. The formation of fungal colonies isolated from E. velutina seeds was also inhibited by 3 min of exposure to atmospheric air plasma, except for A. niger, whose inhibition occurred after 6 min of exposure to atmospheric plasma.

Discharge characteristics of an atmospheric pulsed microwave Ar/CF4 plasma jet

The atmospheric fluorocarbon plasma is widely used in surface modification of polymers. Recently, the pulsed microwave Ar/CF4 plasma jet is proved to be a promising atmospheric fluorocarbon plasma source with good performance. In this paper, the discharge characteristics of the pulsed microwave Ar/CF4 plasma jet are studied systematically. The discharge morphologies, ionization processes, optical emission spectra, and electron densities are obtained by a digital camera, an intensified charge coupled device, a fiber spectrometer, and a home-made microwave Rayleigh scattering device, respectively. The influences of the plasma operation parameters on the discharge characteristics are investigated, and the microwave input power and CF4 volume fraction are optimized. The results provide a basis for the generation and surface modification application of high-performance atmospheric fluorocarbon plasma.

The Effect of Atmospheric Dielectric Barrier Discharge Cold Plasma Treatment on the Nutritional and Physicochemical Characteristics of Various Legumes.

High activity of lipoxygenase (LOX) has been identified as a primary cause of oxidative rancidity in legumes. In this study, the application of dielectric barrier discharge atmospheric cold plasma (DBD-ACP) (5 W, 10 min) resulted in an obvious decrease in LOX activity in mung bean (MB), kidney bean (KB), and adzuki bean (AB) flours by 36.96%, 32.49%, and 28.57%, respectively. Moreover, DBD-ACP induced significant increases (p < 0.05) in content of soluble dietary fiber, saturated fatty acids, and methionine. The starch digestibility of legumes was changed, evidenced by increased (p < 0.05) slowly digestible starch and rapidly digestible starch, while resistant starch decreased. Furthermore, DBD-ACP treatment significantly affected (p < 0.05) the hydration and thermal characteristics of legume flours, evidenced by the increased water absorption index (WAI) and gelatinization temperature, and the decreased swelling power (SP) and gelatinization enthalpy (ΔH). Microscopic observations confirmed that DBD-ACP treatment caused particle aggregation.

Inactivation of Alicyclobacillus acidoterrestris vegetative cells and spores induced by atmospheric cold plasma

This study aimed to inactivate Alicyclobacillus acidoterrestris vegetative cells and spores (107–108 CFU/mL) using atmospheric cold plasma (ACP). Alicyclobacillus acidoterrestris vegetative cells decreased by 6.63 and 6.47 log under only ACP treatment of 30 kV for 6 min in saline solution and apple juice, respectively. ACP was less effective against spores, resulting in 2.64 and 2.43 log reductions at the same treatment parameter. However, better inactivation (4.03 log and 3.95 log) was achieved in apple juice and saline solution using ACP at 30 kV combined with mild heat (85 °C) for 6 min. Morphological observation and intracellular substance release revealed severe structural damage to spores induced by ACP. The application of ACP resulted in improved color, reduced pH, and increased levels of total phenolic content and total soluble solids in apple juice. This study offers an effective method for inactivating Alicyclobacillus acidoterrestris in apple juice. Industrial relevanceThe application of atmospheric cold plasma in combination with mild heat to effectively inactivate Alicyclobacillus acidoterrestris in apple juice offers a promising method for ensuring juice product quality. The enhancement of color, reduction in pH, and increase in total phenolic content and total soluble solids further underscore the potential benefits of this technique for enhancing the sensory attributes of apple juice. This study offers a potential approach that may directly contribute to enhancing production practices and product value.

A Review of Cold Atmospheric Plasma Applications in Dermatology and Aesthetics

Nowadays, there is a growing use of plasma technology, especially in medicine and aesthetics. Plasma medicine is an innovative, multidisciplinary science combining plasma physics and technology with medicine and life sciences. Billions of dollars are spent annually in the United States to rejuvenate and reduce signs of aging. For this reason, several methods have been created to perform such operations. Methods with different degrees of invasiveness, from cosmetic surgery, like forehead lifts and blepharoplasty, to noninvasive, such as high-intensity focused ultrasound and nonablative lasers, have been used in the field of aesthetics and skin rejuvenation. Plasma can also be used as a noninvasive, minimally invasive, or even invasive method in cosmetic applications such as removing wrinkles and strengthening the skin, that can somewhat replace cosmetic procedures such as blepharoplasty and rhytidectomy. By using various active species, plasma affects biological activities and increases the proliferation and transfer of skin cells. With local application of heat and an increase in proliferation and activity of fibroblasts, the production of type III collagen and elastin density in the skin increases. Plasma can be used to sublimate the skin and remove excess tissue. This article summarizes the most recent studies and clinical trials in dermatology and skin rejuvenation, in which different cold plasma sources (jet and spark) have been used, and examines their results.

Effect of atmospheric cold plasma (ACP) on chlorine-adapted Salmonella enterica on spring onion.

One of the main drawbacks of chlorine disinfectants is the emergence of chlorine adapted (CA) or resistant microbial cells. This research aimed to investigate the effect of chlorine adaptation on resistance of Salmonella enterica upon atmospheric cold plasma (ACP) application at different voltages (6, 8 and 11 kV) and times (5, 10 and 15 min). Due to higher conversion efficiency and reduced dielectric barrier discharge power consumption, this method was used for cold plasma generation in this study. A higher lethality effect was observed from a higher voltage and longest times (11 kV-15 min) on CA S. enterica than on non-CA (P < 0·05). Still, it induced higher percentages of injured cells in CA (58·77%) than on non-CA (0·61%) (P < 0·05). The highest ACP effect on the inactivation of the indigenous natural flora of onion leaves was observed at the lowest voltage (P < 0·05). More than 3 log CFU/g reduction (P < 0·05) was observed at 6 kV after 5 and 10 min. ACP reduced CA and non-CA S. enterica cells on onion leaf surface to a lower extent than pure-treated cells in broth media. Nevertheless, similar to broth media, a high percentage of injury (61·03%) was induced on CA cells at higher voltage (11 kV-10 min) compared to non-CA (2·15%) (P < 0·05). Biofilm results revealed ACP application (6 kV-5 min) reduced average ODs in CA and non-CA cells (P < 0·05). Chlorine adaptation and ACP treatment influenced the antibiotic resistance pattern according to applied voltage, time and antibiotic type. The finding showed despite the highest lethality of high voltages and long times (11 kV-15 min), given the high percentages of injured cells, lower voltages may offer acceptable inactivation of pathogenic bacteria with lower injury induction. In conclusion, ACP has the potential ability to eliminate CA cells of S. enterica, which are predominant in fresh-cut vegetable outbreaks.

Characteristics of 2.45 GHz Surface-Wave-Sustained Argon Discharge for Bio-Medical Applications

Cold atmospheric plasma (CAP) applications in various fields, such as biology, medicine and agriculture, have significantly grown during recent years. Many new types of plasma sources operating at atmospheric pressure in open air were developed. In order to use such plasmas for the treatment of biological systems, plasma properties should fulfil strong requirements. One of the most important is the prevention from heating damage. That is why in many cases, the post-discharge region is used for treatment, but the short living particles in the active discharge zone and reactions with them are missed in that case. We use the active region of surface-wave-sustained argon plasma for biological systems treatment. The previous investigations showed good bactericidal, virucidal, seeds germination and decontamination effects at a short treatment time, but the discharge conditions for bio-medical applications need specific adjustment. A detailed theoretical and experimental investigation of the plasma characteristics and their possible optimization in order to meet the requirements for bio-medical applications are presented in this paper. The length of the plasma torch, the temperature at the treatment sample position and the microwave radiation there are estimated and optimized by the appropriate choice of discharge tube size, argon flow rate and microwave power.

Recently, the application of atmospheric pressure low temperature plasma to the medical field is expected, and the growth inhibitory action of cancer cells is indicated in the preceding research. In this study, colorectal cancer with many cases and deaths was noticed. The effect of plasma on colorectal cancer cells was evaluated by culturing colorectal cancer cells in a medium irradiated with atmospheric pressure low temperature plasma, expecting to induce proliferation inhibition or cell death. In addition, in this study, in order to examine the components affecting colorectal cancer cells, the concentration measurement of reactive oxygen species generated in the solution irradiated with plasma using the absorbance method was carried out. And, the mechanism of the cell reduction of the colorectal cancer cell induced by plasma was investigated.

Atmospheric plasma overcomes dormancy of Pityrocarpa moniliformis (Benth.) Luckow &amp; R. W. Jobson seeds

Abstract: Atmospheric plasma application technology consists of a fast and practical method, which has the potential to modify the surface of seeds with integumentary hardness. The aim of this research was to evaluate the effect of applying atmospheric cold plasma on Pityrocarpa moniliformis seeds, assessing its implications on the seed coat impermeability, germination and vigor. Cells were subjected to plasma for 1.5, 2.0, 3.0, 4.0 and 5.0 minutes, using seeds without any treatment as a control. After application, the seed coat wettability, imbibition curve and electrical conductivity of the soaking solution were determined. Seeds were also subjected to the germination test, and its results were used to determine viability, median, uniformity and asymmetry of germination. Seeds subjected to plasma for 5.0 and 4.0 minutes showed the lowest apparent contact angles, 64 and 61°, respectively, characterizing greater wettability of the seed coat among the treatments tested. Higher germination was observed in the treatments with plasma application when compared to the control. Atmospheric plasma application technology has the potential to be used as an accelerator of P. moniliformis seed germination.

Protective effects of low-temperature plasma on cisplatin-induced nephrotoxicity

The application of atmospheric pressure low-temperature plasma (LTP) in medical treatment has received extensive attention owing to its redox regulatory and anti-inflammatory properties. Nephrotoxicity due to oxidative stress and inflammation is the main adverse effect of cisplatin. In the present study, rats with cisplatin-induced nephrotoxicity were treated with LTP to investigate its potential protective effect. The results showed that LTP treatment has multiple protective effects on cisplatin-induced nephrotoxicity. It significantly improved clinical indicators such as survival rate, water intake, food intake, body weight, and mobility, as well as physiological indexes such as reduced renal index and levels of serum urea, creatinine, and total bilirubin; pathological indicators such as reduced tubular injury, inflammatory infiltration, tubulointerstitial fibrosis, and apoptosis; cell survival indicators such as decreased protein levels of Caspase-3 and Bax and increased Bcl-2; anti-oxidation status such as reduced malondialdehyde content and increased activities of catalase, superoxide dismutase, and glutathione peroxidase; and reduced inflammatory factors such as TNF-α in kidney tissues. Specially, LTP treatment did not influence the anticancer effect of cisplatin as observed in the solid tumor mouse model established by subcutaneously inoculating H22 cells. Moreover, LTP did not influence the physiological and pathological indicators of normal rats, suggesting its biological safety. In conclusion, LTP can protect against cisplatin-induced nephrotoxicity through its anti-oxidation, anti-inflammation, and anti-apoptosis effects, without influencing the anticancer effect of cisplatin.

Influence of Ti3C2T x (MXene) on the generation of dielectric barrier discharge in air

The formation of homogeneous dielectric barrier discharge (DBD) in air is a key scientific problem and core technical problem to be solved for the application of plasmas. Here, we report the effect of two-dimensional (2D) nanomaterial Ti3C2T x (T x = –F, –O and/or –OH) on regulating the electrical discharge characteristics. The field emission and weak bound state property of Ti3C2T x can effectively increase the seed electrons and contribute to the generation of atmospheric pressure homogeneous air DBD. The electron avalanche development for the uneven electrode structure is calculated, and the discharge mode transition is modeled. The comparative analyses of discharge phenomena validate the regulation of Ti3C2T x on the discharge characteristics of DBD. The light emission capture and the voltage and current waveforms verify that the transition of Townsend discharge to streamer discharge is effectively inhibited. The optical emission spectra are used to characterize the plasma and confirm that it is in a non-equilibrium state and the gas temperature is at room temperature. This is the first exploration of Ti3C2T x on the regulation of electrical discharge characteristics as far as we know. This work proves the feasibility of Ti3C2T x as a source of seed electrons to form homogeneous DBD, establishing a preliminary foundation for promoting the application of atmospheric pressure non-equilibrium plasma.

Antitumor effects of hyperthermia with plasma‐treated solutions on 3D bladder tumor spheroids

AbstractAs a form of cold atmospheric plasma application, plasma‐treated solutions have shown promise in tumor therapy. Especially for the treatment of intracavitary tumors, plasma‐treated solutions combined with traditional perfusion therapy have strong application potential. Here, we assessed the antitumor effects of the plasma‐treated medium combined with hyperthermia on three‐dimensional bladder tumor spheroids in vitro. We found that the combined treatment of plasma‐treated medium and hyperthermia (43°C) presented better antitumor effects than a single treatment. Further research showed that synchronous hyperthermia can promote the penetration of plasma‐generated reactive oxygen species from the plasma‐treated medium into cancer cells. Our results suggest that hyperthermic perfusion therapy using plasma‐treated solutions may serve as a novel and valuable therapeutic strategy for bladder cancer.

Potential application of non-thermal atmospheric plasma in reducing the activity of Pseudomonas-secreted proteases in milk

Application of non-thermal atmospheric plasma was examined against the activity of Pseudomonas-secreted proteases in milk. UHT milk samples were inoculated with crude extract of Pseudomonas-secreted proteases and treated in atmospheric plasma discharge system. Evaluation of the progress in protein hydrolysis of plasma-treated and non-treated milk samples revealed that the degree of hydrolysis was lower in plasma-treated milk samples. The longer the plasma treatment time, the lower the degree of hydrolysis during storage. Furthermore, plasma-induced changes in milk proteins and volatile compounds were also evaluated. Caseins, whey proteins, aldehydes, ketones, free fatty acids and hydrocarbons were all affected by plasma discharge. The amount of increase or decrease in these components was dependent on the duration of plasma treatment. Overall, this method is a promising approach that could be used to inactivate bacterial hydrolytic enzymes in milk.

Dielectric barrier discharge cold atmospheric plasma: Influence of processing parameters on microbial inactivation in meat and meat products.

Decontamination of meat is commonly practiced to get rid of or decrease the microbial presence on the meat surface. Dielectric barrier discharge cold atmospheric plasma (DBD-CAP) as innovative technology is a food microbial inactivation technique considered in high regard by food scientists and engineers in present times. However, cold atmospheric plasma application is at the experimental stage, due to lack of sufficient information on its mode of action in inactivating microbes, food shelf-life extensibility, whereas, the nutritional value of food is preserved. In this review, we have appraised recent work on DBD-CAP concerning the decontamination treatment of meat products, highlighting the processing value results on the efficacy of the DBD-CAP microbial inactivation technique. Also, the paper will review the configurations, proposed mechanisms, and chemistry of DBD-CAP. Satisfactory microbial inactivation was observed. In terms of DBD-CAP application on sensory evaluation, inferences from reviewed literature showed that DBD has no significant effect on meat color and tenderness, whereas in contrast, TBARS values of fresh and processed meat are affected. DBD seems economically efficient and environmentally sustainable.

Application of atmospheric pressure low temperature plasma in cotton fabric desizing

In order to solve the problem of high energy consumption and high pollution in traditional alkali desizing process of cotton fabric, atmospheric pressure low temperature plasma was used to treat pure cotton fabric containing sizing. Through single factor variable experiment, the optimum desizing process was determined. The optimum process conditions were as follows: the optimum treatment power of plasma is 400 W, the optimum treatment time is 15 min, the distance between discharge electrode and grey cloth is 1 mm, the optimum alkali boiling concentration is 8 g/L. Compared to traditional desizing process, the usage of caustic soda was saved by 60%.

Cold atmospheric plasma applications in dermatology: A systematic review.

Cold atmospheric plasma (CAP) applications can potentially lead to effective therapy for numerous skin diseases. Our aim is to systematically review the available data and map the use of CAP in dermatology. PubMed, Embase and Web of science were explored before 2020 for studies regarding the use of CAP in dermatology. A total of 166 studies were finally included. 74.1% of these studies used indirect CAP sources. Most studies used plasma jet (67.5%). Argon was the mostly used working gas (48.2%). Plasma application itself could be direct (89.2%) and indirect (16.3%). The proportion of studies with in vivo results remained 57.2%, of which most concerned direct plasma treatment (97.9%). Analyses performed indicate that CAP has been beneficial in many skin disorders. While, most CAP applications were focused on wound healing and melanoma treatment. This study provides a brief overview of CAP sources and relative medical applications in dermatology.
