The ant’s weapon improves honey bee learning performance

Formic acid is the main component of the ant’s major weapon against enemies. Being mainly used as a chemical defense, the acid is also exploited for recruitment and trail marking. The repelling effect of the organic acid is used by some mammals and birds which rub themselves in the acid to eliminate ectoparasites. Beekeepers across the world rely on this effect to control the parasitic mite Varroa destructor. Varroa mites are considered the most destructive pest of honey bees worldwide and can lead to the loss of entire colonies. Formic acid is highly effective against Varroa mites but can also kill the honeybee queen and worker brood. Whether formic acid can also affect the behavior of honey bees is unknown. We here study the effect of formic acid on sucrose responsiveness and cognition of honey bees treated at different live stages in field-relevant doses. Both behaviors are essential for survival of the honey bee colony. Rather unexpectedly, formic acid clearly improved the learning performance of the bees in appetitive olfactory conditioning, while not affecting sucrose responsiveness. This exciting side effect of formic acid certainly deserves further detailed investigations.

Honey bees can store and retrieve independent memory traces after complex experiences that combine appetitive and aversive associations.

Real-world experiences often mix appetitive and aversive events. Understanding the ability of animals to extract, store and use this information is an important issue in neurobiology. We used honey bees as model organism to study learning and memory after a differential conditioning paradigm that combines appetitive and aversive training trials. First, we used an aversive conditioning paradigm that constitutes a clear opposite of the well-known appetitive olfactory conditioning of the proboscis extension response. A neutral odour is presented paired with the bitter substance quinine. Aversive memory is evidenced later as an odour-specific impairment in appetitive conditioning. Then, we tested the effect of mixing appetitive and aversive conditioning trials distributed along the same training session. Differential conditioning protocols like this were used previously to study the ability to discriminate odours; however, they were not focused on whether appetitive and aversive memories are formed. We found that after differential conditioning, honey bees establish independent appetitive and aversive memories that do not interfere with each other during acquisition or storage. Finally, we moved the question forward to retrieval and memory expression to evaluate what happens when appetitive and the aversive learned odours are mixed during a test. Interestingly, opposite memories compete in such a way that they do not cancel each other out. Honey bees showed the ability to switch from expressing appetitive to aversive memory depending on their satiation level.

Neurotransmitters receptors gene drive the olfactory learning behavior of honeybee

The neurotransmitters receptor gene are multifunctional modulators of behaviors in social insects, with significant effects among species and several dominant roles within the same species. Multiple amines receptors affect diverse behavioral and physiological processes in the honeybee. Apis mellifera ligustica is excellent bee species due to its remarkable learning abilities. Here we analyze whether changes in expression of neurotransmitters receptor genes are related to olfaction learning behavior. We present a behavioral experiment on honeybees’ appetitive olfactory conditioning and examine the relationship between learning success and neurotransmitters receptor gene expression. Bees were trained using proboscis extension response to discriminate learner and failed learner bees and examine the expression of neurotransmitters receptors genes in the brains of learner and failed-learner bees. Real-time quantitative PCR was performed to assess neurotransmitter expression levels. Our finding showed significant results in differentiating the learner and failed-learner bee. The expression level of AmDop1, AmDop2, AmDop3, AmOa1, AmTyr1 and 5-HT showed significantly higher expression in learner and failed-learner bees. Our finding suggests the critical role of neurotransmitters receptors in the olfactory learning behavior of A. mellifera. These significant neurotransmitters receptors could thus act together to define optimal motivational or attentional levels and improve learning success. This study revealed the dynamic modulation of neurotransmitters expression in the brain of learner and failed learner bees and suggested an essential role for neurotransmitters receptors gene in regulating the olfactory learning behavior in honeybees. We suggested that neurotransmitters receptors are expressed in the brain of bees and are likely to be directly influential in regulating olfactory learning success in honeybees.

Cardiac response in aversive and appetitive olfactory conditioning: Evidence for a valence-independent CS-elicited bradycardia.

While the examination of conditioned cardiac responses is well established in human fear conditioning research, comparable studies using less-aversive or rather appetitive unconditioned stimuli (UCS) are sparse and results are mixed. Therefore, the aim of this study was a systematic analysis of cardiac reactions in aversive and appetitive conditioning. Olfactory stimuli were used as unconditioned stimuli as they are suitable reinforcers in both an aversive and an appetitive conditioning offering the opportunity for a comparison between conditioned responses. In total, n=86 participants took part in both an aversive and an appetitive differential conditioning task with a counterbalanced order across participants. Aversive or appetitive odors, respectively, served as UCS and neutral geometrical figures as CS. Subjective ratings, skin conductance response (SCRs), and evoked cardiac reactions were analyzed and compared between tasks. Conditioned responses in subjective ratings could be observed in both aversive conditioning and appetitive conditioning, while SCRs discriminated between CS+ and CS- in aversive conditioning only. Regarding conditioned cardiac responses, the deceleration for the CS+ was longer than for the CS- in both tasks. In addition, a higher deceleration magnitude and a shorter acceleration for the CS+ as compared to the CS- were found in aversive but not in appetitive conditioning. There were medium-size correlations between aversive and appetitive CRs for subjective ratings and none for physiological responses. The results suggest similarities between cardiac response patterns in aversive and appetitive conditioning, which implies that bradycardia in conditioning might not be fear-specific but presents a valence-independent CS-elicited bradycardia.

Appetitive olfactory conditioning in the neonatal male rat facilitates subsequent sexual partner preference

Pairing a neutral odor with a male rat’s initial sexual experiences to ejaculation produces a subsequent conditioned ejaculatory preference (CEP) in which males ejaculate preferentially with receptive females that bear the odor relative to unscented receptive females. In 1986, Fillion and Blass reported that neonatal male rats exposed to a neutral lemon odor (citral) painted on their mother’s ventrum while nursing ejaculated faster as adults with sexually receptive, citral-scented females compared to unscented receptive females. The present study examined whether the same odor paired with tactile reward in neonatal male rats would alter the subsequent expression of a CEP. Newborn Long-Evans male rats were separated from their mothers each day beginning on Postnatal Day 1 and placed into a Plexiglas cage that contained either unscented or citral-scented bedding (N = 8/group). During each trial, rats were stroked from head to toe with a soft, narrow paintbrush, after which they were returned to their mothers. Males were weaned at 21 days of age and housed in same-treatment pairs for an intervening 50 days. Following habituation to a large open field, males were presented with two sexually receptive Long-Evans females, one scented with citral, and the other unscented, for a 30-min test of copulation. Males in the Paired group copulated and ejaculated preferentially with the scented female whereas males in the Unpaired group showed no preference. Pairing a neutral odor with a reward state in infancy generates a preference in male rats to ejaculate with sexually receptive females bearing the same odor in adulthood.

Thymol Affects Congruency Between Olfactory and Gustatory Stimuli in Bees

Honey bees learn to associate sugars with odorants in controlled laboratory conditions and during foraging. The memory of these associations can be impaired after exposure to contaminants such as pesticides. The sub-lethal effects of acaricides such as 5-methyl-2-(propan-2-yl)-phenol (thymol) introduced into colonies to control varroa mites are of particular concern to beekeeping, due to detrimental effects of some acaricides on bees. Here we assess whether various odorant/sugar pairs are identically memorized in a differential appetitive olfactory conditioning experiment and whether this learning is affected by thymol exposure. Responses to odorants in retrieval tests varied according to the sugar they were paired with, a property called congruency. Interestingly, congruency was altered by pre-exposure to some thymol concentrations during retrieval tests, although electroantennography recordings showed it left odorant detection intact. This highlights the importance of taking into account subtle effects such as odor/sugar congruency in the study of the effect of pesticides on non-target insects, in addition to the simpler question of memory impairment.

Petrol exhaust pollution impairs honey bee learning and memory

Volatile organic compounds (VOCs) serve as important infochemicals, mediating several ecological interactions including herbivory and pollination. Atmospheric pollutants including traffic‐related air pollution may impair the detection of VOCs used by insects in insect–plant interactions. We investigated the indirect effect of petrol exhaust pollution on olfactory learning and memory (short and long term) in honey bees. Using appetitive olfactory conditioning, we trained bees to learn one of four floral VOC profiles; linalool, dipentene, myrcene and geranium. VOCs were unpolluted or polluted with exhaust collected from a petrol generator. Exhaust emissions included concentrations of CO (246.07 + 17 ppm), NO (20.50 + 6.90 ppb) and NO2 (20.93 + 0.05 ppb) consistent with those typically encountered in urban areas and near roads. Once bees had learnt the training VOC, we tested whether they could recognise that VOC 1 h, 24 h and 48 h post‐training. Bees took significantly longer to learn polluted VOCs and forgot them faster than unpolluted ones. We also tested the ‘masking’ potential of pollution on floral VOCs. Using gas chromatography mass spectroscopy we noted differences in the chemical profile of polluted versus unpolluted VOCs and tested whether bees could recognise polluted VOCs if trained using unpolluted ones. For several VOCs tested, bees could distinguish between polluted and unpolluted VOCs. Ultimately, our results show that air pollution changes the recognition and retention of floral VOCs by bees which may consequently impact foraging efficiency.

Dopamine interferes with appetitive long-term memory formation in honey bees

Studies in vertebrates and invertebrates have proved the instructive role that different biogenic amines play in the neural representation of rewards and punishments during associative learning. Results from diverse arthropods and using different learning paradigms initially agreed that dopamine (DA) is needed for aversive learning and octopamine (OA) is needed for appetitive learning. However, the notion that both amines constitute separate pathways for appetitive and aversive learning is changing. Here, we asked whether DA, so far only involved in aversive memory formation in honey bees, does also modulate appetitive memory. Using the well characterized appetitive olfactory conditioning of the proboscis extension reflex (PER), we show that DA impairs appetitive memory consolidation. In addition, we found that blocking DA receptors enhances appetitive memory. These results are consistent with the view that aversive and appetitive components interact during learning and memory formation to ensure adaptive behavior.

Appetitive and aversive olfactory learning induce similar generalization rates in the honey bee

Appetitive and aversive learning drive an animal toward or away from stimuli predicting reinforcement, respectively. The specificity of these memories may vary due to differences in cost–benefit relationships associated with appetitive and aversive contexts. As a consequence, generalization performances may differ after appetitive and aversive training. Here, we determined whether honey bees show different rates of olfactory generalization following appetitive olfactory conditioning of the proboscis extension response, or aversive olfactory conditioning of the sting extension response. In both cases, we performed differential conditioning, which improves discrimination learning between a reinforced odor (CS?) and a non-reinforced odor (CS-) and evaluated generalization to two novel odors whose similarity to the CS? and the CS- was different. We show, given the same level of discriminatory performance, that rates of generalization are similar between the two conditioning protocols and discuss the possible causes for this phenomenon.

Focal uncaging of GABA reveals a temporally defined role for GABAergic inhibition during appetitive associative olfactory conditioning in honeybees

Throughout the animal kingdom, the inhibitory neurotransmitter γ-aminobutyric acid (GABA) is a key modulator of physiological processes including learning. With respect to associative learning, the exact time in which GABA interferes with the molecular events of learning has not yet been clearly defined. To address this issue, we used two different approaches to activate GABA receptors during appetitive olfactory conditioning in the honeybee. Injection of GABA-A receptor agonist muscimol 20 min before but not 20 min after associative conditioning affects memory performance. These memory deficits were attenuated by additional training sessions. Muscimol has no effect on sensory perception, odor generalization, and nonassociative learning, indicating a specific role of GABA during associative conditioning. We used photolytic uncaging of GABA to identify the GABA-sensitive time window during the short pairing of the conditioned stimulus (CS) and the unconditioned stimulus (US) that lasts only seconds. Either uncaging of GABA in the antennal lobes or the mushroom bodies during the CS presentation of the CS-US pairing impairs memory formation, while uncaging GABA during the US phase has no effect on memory. Uncaging GABA during the CS presentation in memory retrieval also has no effect. Thus, in honeybee appetitive olfactory learning GABA specifically interferes with the integration of CS and US during associative conditioning and exerts a modulatory role in memory formation depending on the training strength.

Appetitive Learning Requires the Alpha1-Like Octopamine Receptor OAMB in theDrosophilaMushroom Body Neurons

Associative learning is a fundamental form of behavioral plasticity. Octopamine plays central roles in various learning types in invertebrates; however, the target receptors and underlying mechanisms are poorly understood. Drosophila provides a powerful system to uncover the mechanisms for learning and memory. Here, we report that OAMB in the mushroom body neurons mediates the octopamine's signal for appetitive olfactory learning. The octopamine receptor OAMB has two isoforms (OAMB-K3 and OAMB-AS), differing in the third cytoplasmic loop and downstream sequence. The activation of each OAMB isoform increases intracellular Ca(2+) similar to the alpha1 adrenergic receptor, while OAMB-K3 additionally stimulates cAMP production. The oamb-null mutants showed severely impaired learning in appetitive olfactory conditioning that tests flies' capacity to learn and remember the odor associated with sugar reward. This deficit was also seen in the hypomorphic mutant with reduced OAMB expression in the mushroom bodies, the brain structure crucial for olfactory conditioning. Consistently, the oamb mutant's learning phenotype was fully rescued by conditional expression of either OAMB isoform in the mushroom body αβ and γ neurons. These results indicate that the OAMB receptor is a key molecule mediating the octopamine's signal for appetitive olfactory learning and its functional site is the mushroom body αβ and γ neurons. This study represents a critical step forward in understanding the cellular mechanism and neural circuit mediating reward learning and memory.

A Permissive Role of Mushroom Body α/β Core Neurons in Long-Term Memory Consolidation in Drosophila

Memories are not created equally strong or persistent for different experiences. In Drosophila, induction of long-term memory (LTM) for aversive olfactory conditioning requires ten spaced repetitive training trials, whereas a single trial is sufficient for LTM generation in appetitive olfactory conditioning. Although, with the ease of genetic manipulation, many genes and brain structures have been related to LTM formation, it is still an important task to identify new components and reveal the mechanisms underlying LTM regulation. Here we show that single-trial induction of LTM can also be achieved for aversive olfactory conditioning through inhibition of highwire (hiw)-encoded E3 ubiquitin ligase activity or activation of its targeted proteins in a cluster of neurons, localized within the α/β core region of the mushroom body. Moreover, the synaptic output of these neurons is critical within a limited posttraining interval for permitting consolidation of both aversive and appetitive LTM. We propose that these α/β core neurons serve as a "gate" to keep LTM from being formed, whereas any experience capable of "opening" the gate is given permit to be consolidated into LTM.

Remembering Nutrient Quality of Sugar in Drosophila

Taste is an early stage in food and drink selection for most animals [1, 2]. Detecting sweetness indicates the presence of sugar and possible caloric content. However, sweet taste can be an unreliable predictor of nutrient value because some sugars cannot be metabolized. In addition, discrete sugars are detected by the same sensory neurons in the mammalian [3] and insect [4, 5] gustatory systems, making it difficult for animals to readily distinguish the identity of different sugars using taste alone [6-8]. Here we used an appetitive memory assay in Drosophila [9-11] to investigate the contribution of palatability and relative nutritional value of sugars to memory formation. We show that palatability and nutrient value both contribute to reinforcement of appetitive memory. Nonnutritious sugars formed less robust memory that could be augmented by supplementing with a tasteless but nutritious substance. Nutrient information is conveyed to the brain within minutes of training, when it can be used to guide expression of a sugar-preference memory. Therefore, flies can rapidly learn to discriminate between sugars using a postingestive reward evaluation system, and they preferentially remember nutritious sugars.

Searching for learning-dependent changes in the antennal lobe: simultaneous recording of neural activity and aversive olfactory learning in honeybees

Plasticity in the honeybee brain has been studied using the appetitive olfactory conditioning of the proboscis extension reflex, in which a bee learns the association between an odor and a sucrose reward. In this framework, coupling behavioral measurements of proboscis extension and invasive recordings of neural activity has been difficult because proboscis movements usually introduce brain movements that affect physiological preparations. Here we took advantage of a new conditioning protocol, the aversive olfactory conditioning of the sting extension reflex, which does not generate this problem. We achieved the first simultaneous recordings of conditioned sting extension responses and calcium imaging of antennal lobe activity, thus revealing on-line processing of olfactory information during conditioning trials. Based on behavioral output we distinguished learners and non-learners and analyzed possible learning-dependent changes in antennal lobe activity. We did not find differences between glomerular responses to the CS+ and the CS− in learners. Unexpectedly, we found that during conditioning trials non-learners exhibited a progressive decrease in physiological responses to odors, irrespective of their valence. This effect could neither be attributed to a fitness problem nor to abnormal dye bleaching. We discuss the absence of learning-induced changes in the antennal lobe of learners and the decrease in calcium responses found in non-learners. Further studies will have to extend the search for functional plasticity related to aversive learning to other brain areas and to look on a broader range of temporal scales.

A Neural Circuit Mechanism Integrating Motivational State with Memory Expression in Drosophila

Behavioral expression of food-associated memory in fruit flies is constrained by satiety and promoted by hunger, suggesting an influence of motivational state. Here, we identify a neural mechanism that integrates the internal state of hunger and appetitive memory. We show that stimulation of neurons that express neuropeptide F (dNPF), an ortholog of mammalian NPY, mimics food deprivation and promotes memory performance in satiated flies. Robust appetitive memory performance requires the dNPF receptor in six dopaminergic neurons that innervate a distinct region of the mushroom bodies. Blocking these dopaminergic neurons releases memory performance in satiated flies, whereas stimulation suppresses memory performance in hungry flies. Therefore, dNPF and dopamine provide a motivational switch in the mushroom body that controls the output of appetitive memory.

Rapid Consolidation to aradishand Protein Synthesis-Dependent Long-Term Memory after Single-Session Appetitive Olfactory Conditioning inDrosophila

In Drosophila, formation of aversive olfactory long-term memory (LTM) requires multiple training sessions pairing odor and electric shock punishment with rest intervals. In contrast, here we show that a single 2 min training session pairing odor with a more ethologically relevant sugar reinforcement forms long-term appetitive memory that lasts for days. Appetitive LTM has some mechanistic similarity to aversive LTM in that it can be disrupted by cycloheximide, the dCreb2-b transcriptional repressor, and the crammer and tequila LTM-specific mutations. However, appetitive LTM is completely disrupted by the radish mutation that apparently represents a distinct mechanistic phase of consolidated aversive memory. Furthermore, appetitive LTM requires activity in the dorsal paired medial neuron and mushroom body alpha'beta' neuron circuit during the first hour after training and mushroom body alphabeta neuron output during retrieval, suggesting that appetitive middle-term memory and LTM are mechanistically linked. Last, experiments feeding and/or starving flies after training reveals a critical motivational drive that enables appetitive LTM retrieval.

Olfactory memory formation and the influence of reward pathway during appetitive learning by honey bees

Animals possess the ability to assess food quality via taste and via changes in state that occur after ingestion. Here, we investigate the extent to which a honey bee's ability to assess food quality affected the formation of association with an odor stimulus and the retention of olfactory memories associated with reward. We used three different conditioning protocols in which the unconditioned stimulus (food) was delivered as sucrose stimulation to the proboscis (mouthparts), the antennae or to both proboscis and antennae. All means of delivery of the unconditioned stimulus produced robust associative conditioning with an odor. However, the memory of a conditioned odor decayed at a significantly greater rate for subjects experiencing antennal-only stimulation after either multiple- or single-trial conditioning. Finally, to test whether the act of feeding on a reward containing sucrose during conditioning affected olfactory memory formation, we conditioned honey bees to associate an odor with antennal stimulation with sucrose followed by feeding on a water droplet. We observed that a honey bee's ability to recall the conditioned odor was not significantly different from that of subjects conditioned with an antennal-only sucrose stimulus. Our results show that stimulation of the sensory receptors on the proboscis and/or ingestion of the sucrose reward during appetitive olfactory conditioning are necessary for long-term memory formation.

Participation of octopaminergic reward system and dopaminergic punishment system in insect olfactory learning revealed by pharmacological study

Biogenic amines play major roles in the regulation of behavior in vertebrates and invertebrates. Previous studies in honey bees and fruit-flies Drosophila suggested that octopamine (OA, invertebrate counterpart of noradrenaline) and dopamine (DA) participate in appetitive olfactory conditioning with sucrose reward and aversive olfactory conditioning with electric shock punishment, respectively. In order to determine whether the effects of the two chatecholamines on electric shock and sugar learning can be generalized to other kinds of appetitive and aversive reinforcers, we studied the effects of OA and DA receptor antagonists on appetitive olfactory learning with water reward, and aversive olfactory learning with saline punishment in the cricket Gryllus bimaculatus. Crickets injected with epinastine or mianserin, OA receptor antagonists, into the hemolymph exhibited an impairment of appetitive learning with water reward, while aversive learning with saline punishment remained intact. In contrast, fluphenazine, chlorpromazine or spiperone, DA receptor antagonists, impaired aversive learning without affecting appetitive learning. This finding, combined with findings in previous studies, suggests that the octopaminergic reward system and dopaminergic punishment system participate in insect olfactory learning with various appetitive and aversive reinforcements.

Asymmetrical generalisation between pheromonal and floral odours in appetitive olfactory conditioning of the honey bee (Apis mellifera L.).

The capacity to generalise between similar but not identical olfactory stimuli is crucial for honey bees, allowing them to find rewarding food sources with varying volatile emissions. We studied bees' generalisation behaviour with odours having different biological values: typical floral odours or alarm compounds. Bees' behavioural and peripheral electrophysiological responses were investigated using a combined proboscis extension response conditioning-electroantennogram assay. Bees were conditioned to pure linalool (floral) or to pure isoamyl acetate (alarm) and were tested with different concentrations of both compounds. Electrophysiological responses were not influenced by conditioning, suggesting that the learning of individual compounds does not rely on modulations of peripheral sensitivity. Behaviourally, generalisation responses of bees conditioned to the alarm compound were much higher than those of bees conditioned to the floral odour. We further demonstrated such asymmetrical generalisation between alarm and floral odours by using differential conditioning procedures. Conditioning to alarm compounds (isoamyl acetate or 2-heptanone) consistently induced more generalisation than conditioning to floral compounds (linalool or phenylacetaldehyde). Interestingly, generalisation between the two alarm compounds, which are otherwise chemically different, was extremely high. These results are discussed in relation to the neural representation of compounds with different biological significance for bees.
