Neutrophils cultured ex vivo from CD34+ stem cells are immature and genetically tractable

BackgroundNeutrophils are granulocytes with essential antimicrobial effector functions and short lifespans. During infection or sterile inflammation, emergency granulopoiesis leads to release of immature neutrophils from the bone marrow, serving to boost circulating neutrophil counts. Steady state and emergency granulopoiesis are incompletely understood, partly due to a lack of genetically amenable models of neutrophil development.MethodsWe optimised a method for ex vivo production of human neutrophils from CD34+ haematopoietic progenitors. Using flow cytometry, we phenotypically compared cultured neutrophils with native neutrophils from donors experiencing emergency granulopoiesis, and steady state neutrophils from non-challenged donors. We carry out functional and proteomic characterisation of cultured neutrophils and establish genome editing of progenitors.ResultsWe obtain high yields of ex vivo cultured neutrophils, which phenotypically resemble immature neutrophils released into the circulation during emergency granulopoiesis. Cultured neutrophils have similar rates of ROS production and bacterial killing but altered degranulation, cytokine release and antifungal activity compared to mature neutrophils isolated from peripheral blood. These differences are likely due to incomplete synthesis of granule proteins, as demonstrated by proteomic analysis.ConclusionEx vivo cultured neutrophils are genetically tractable via genome editing of precursors and provide a powerful model system for investigating the properties and behaviour of immature neutrophils.

Beta-hydroxybutyrate regulation of metabolic stress responses to Staphylococcus aureusinfection in macrophages

Abstract Macrophages orchestrate the initiation, intensity, and duration of the inflammatory response induced during Staphylococcus aureusinfection. The pleiotropic role of macrophages is dictated by the capacity to adapt to different metabolic cues. During acute inflammation, macrophages are highly glycolytic; during resolution, these cells shift to oxidative metabolism. While glycolysis drives the expression of pro-inflammatory genes, oxidative phosphorylation leads to fatty acid oxidation and synthesis of ketone bodies, such as beta-hydroxybutyrate (BHB). Therefore, identifying the events that drive the shift from glycolytic to oxidative phosphorylation could provide clues for inflammation and homeostasis. If exogenous BHB influences macrophage responses to S. aureusinfection is not well understood. We hypothesize that BHB is a bridge between glycolysis and oxidative phosphorylation during S. aureusinfection. BHB increases mitochondrial respiration in S. aureus-infected immortalized bone-marrow-derived macrophages (iBMDM). During infection, treatment with BHB increased PDHA1(Pyruvate Dehydrogenase E1 Subunit Alpha 1) but decreased HK1 (Hexokinase 1) expression in iBMDMs. Furthermore, BHB decreased macrophage phagocytic capacity in S. aureusinfected peritoneal macrophages, indicating a decrease in glycolytic activity. Cumulatively, these findings suggest that BHB may cause a shift toward aerobic respiration and increase the anti-inflammatory capacity of macrophages. Therefore, exogenous BHB or ketogenic diets could be detrimental during S. aureusinfection by decreasing both antimicrobial effector functions and inflammatory responses. Duvelisib for Critically-Ill Patients with COVID-19 Disease: An Investigator-Initiated, Randomized, Placebo-Controlled Double-Blind Pilot Trial

Background: Severe and critical COVID-19, including acute respiratory distress syndrome (ARDS), cardiomyopathy, and shock, have been associated with a 28-day mortality exceeding 50%. While dexamethasone (dex) improves survival in such patients and highlights a role for anti-inflammatory therapy, mortality remains high especially in those who do not respond initially. Phase II/III studies of the PI3K-d/g inhibitor duvelisib in patients with chronic lymphocytic leukemia or follicular lymphoma have demonstrated that duvelisib significantly downregulates expression of inflammatory cytokines and chemokines (e.g. G-CSF, GM-CSF, macrophage inflammatory protein-1 α and β). These same inflammatory molecules are upregulated in ARDS and COVID-19. Several murine models of acute lung inflammation and sepsis have demonstrated improved markers of inflammation and survival of mice treated with PI3K inhibitors. The original SAR-CoV-1 coronavirus had demonstrated the ability to hijack the PI3K signaling machinery in order to persist within human airway cells. Additionally, in the context of neoplasia, duvelisib polarizes macrophages to the M1 phenotype which has antimicrobial effector functions in contrast to the pro-inflammatory M2 phenotype. In the context of chimeric antigen receptor T (CAR T) therapy, the DiPersio lab has identified duvelisib as a potent mediator of cytokine release syndrome (CRS) without T-cell inhibition, an overlapping concept with COVID-19. We hypothesized that duvelisib could attenuate COVID-19 ARDS and CRS through M1 macrophage polarization, reduction in cytokine signaling/production, and elimination of viral persistence. Methods: We conducted a multicenter, double-blind, placebo-controlled trial in which 28 patients were randomized 1:1 to duvelisib 25mg twice daily or placebo for 10 days. Eligible patients were 18 years or older hospitalized with a primary diagnosis of COVID-19 disease with either respiratory failure (≥ 50% lung involvement by imaging, invasive or non-invasive mechanical ventilation, or extracorporeal membrane oxygenation), new cardiomyopathy, and/or shock requiring vasopressor support. All patients were required to be receiving or have received dex 6mg daily or equivalent for at least 48 hours without improvement. All patients received prophylaxis for Pneumocystis jiroveci. We monitored viral load daily using quantitative RT-PCR to assess for changes in viral replication following initiation of therapy. Primary endpoint was 28-day survival. Secondary endpoints included: 60-day survival; duration on ventilator and vasopressors; hospital and ICU length of stay; viral kinetics; and adverse events. Correlatives included impact on cytokine expression measured by multiplex panel, viral kinetics by RT-PCR, and flow-cytometry to assess immune populations at various time points during therapy. Results: Baseline and disease characteristics were well-balanced with the exception of baseline renal replacement therapy (Table 1). At day 29, 67% of those in the duvelisib arm and 62% of those in the placebo arm were alive. Secondary efficacy endpoints were comparable between the two cohorts. There was a trend toward improved 60-day OS in the duvelisib arm (60% vs. 46%, p = 0.36, Figure 1) and some patients in the treatment arm were noted to have quick radiographic and clinical improvements. Of those on ECMO at initiation 3/5 (60%) in the duvelisib arm and 1/4 (25%) in the placebo arm were successfully weaned. Adverse events (AEs) occurred at similar rates between arms. Among AEs of special interest previously reported with the use of duvelisib, one patient developed colitis ultimately deemed to be stercoral colitis and unrelated to treatment. No CMV reactivation or Pneumocystis jiroveci pneumonia occurred. Eighty percent of overall deaths were due to respiratory failure from disease progression. In the placebo arm, one patient died of Pseudomonas pneumonia and one died of cholecystitis. Conclusion: In our pilot study, duvelisib did not show improvement in 28-day survival in a critically-ill, steroid-refractory population. However, there was a trend toward improved 60-day OS. No added toxicity was observed in those receiving duvelisib. Based on comparable safety, notable improvements in select patients, and a continued unmet need in this population, we believe larger studies are justified. Correlative data will also be presented. Non-apoptotic Cell Death Control of Neutrophil Extracellular Trap Formation.

Neutrophil extracellular traps (NETs) are networks of chromatin and microbicidal proteins released by neutrophils in response to infection and tissue damage. Although classically viewed as a discrete biochemical and cellular process in neutrophils, the effector pathways integrating diverse upstream activating signals to control the formation of NETs (NETosis) are poorly defined. Cell death is one such common unifying endpoint of neutrophils, with several bona fide non-apoptotic cell death agonists now described to initiate NETosis. Integrating these new genetic findings into our existing knowledge of NETosis will likely reveal varied cellular and biochemical processes controlling NET release and specific anti-microbial and inflammatory effector functions of NETs triggered by specific non-apoptotic cell death. To facilitate investigation of regulated cell death pathways in NETosis, we offer a detailed protocol for neutrophil purification from mouse bone marrow and human blood, analysis of NETs by flow cytometry, and validation by immunogold electron microscopy. Future studies may better define cell death-specific forms of NETosis and their influence on inflammation and autoimmunity.

FcγR Genetic Variation and HIV-1 Vaccine Efficacy: Context And Considerations.

Receptors for the crystallisable fragment (Fc) of immunoglobulin (Ig) G, Fcγ receptors (FcγRs), link the humoral and cellular arms of the immune response, providing a diverse armamentarium of antimicrobial effector functions. Findings from HIV-1 vaccine efficacy trials highlight the need for further study of Fc-FcR interactions in understanding what may constitute vaccine-induced protective immunity. These include host genetic correlates identified within the low affinity Fcγ-receptor locus in three HIV-1 efficacy trials – VAX004, RV144, and HVTN 505. This perspective summarizes our present knowledge of FcγR genetics in the context of findings from HIV-1 efficacy trials, and draws on genetic variation described in other contexts, such as mother-to-child HIV-1 transmission and HIV-1 disease progression, to explore the potential contribution of FcγR variability in modulating different HIV-1 vaccine efficacy outcomes. Appreciating the complexity and the importance of the collective contribution of variation within the FCGR gene locus is important for understanding the role of FcγRs in protection against HIV-1 acquisition.

Mechanosensing of Substrate Stiffness Regulates Effector Functions of Human Neutrophils

During a response to injury or infection, neutrophils (PMNs) navigate through a tissue microenvironment that is densely packed with parenchymal cells and extracellular matrix. Within these tissues, PMNs sense the physical property of elasticity as a regulatory element as it pertains to motility and effector functions. Historically, rigid tissue culture surfaces like glass and plastic have been used to study many key anti-microbial effector functions of PMNs; however, these fail to mimic the elasticity of peripheral tissues. We hypothesize that elasticity plays a key regulatory role for PMN anti-microbial effector functions associated with pathogen clearance such as phagocytosis, NETosis, respiratory burst, and degranulation. When comparing phagocytosis by neutrophils on soft substrates and stiff substrates coated with fibronectin, we found that stiffness is selectively regulatory. While Zymosan and Whole Glucan Particles were more readily phagocytosed on soft substrates, IgG-opsonized latex beads were more efficiently taken up on stiff substrates. Further, C3bi-opsonized latex beads and apoptotic bodies were phagocytosed equivalently on all stiffnesses. In addition to a mechanosensitive response to phagocytosed particles, we also saw differential mechanosensitive responses in their mechanism. For example, on stiff substrates, inhibiting Rho-associated protein kinase reduced phagocytosis of Zymosan, while blocking CD45 reduced phagocytosis of IgG-opsonized latex beads. Further, Zymosan uptake was found to increase on soft substrates when actin-related protein Arp2/3 complex was blocked, but have no effect on stiff substrates, further exaggerating the soft/stiff phenotype; conversely, blocking formin-mediated actin had the opposite effect and lowered phagocytosis of Zymosan on soft substrates, attenuating the soft/stiff phenotype. Finally, not all blocking targets showed mechanosensitive responses; while inhibition of Spleen Tyrosine Kinase had no effect on Zymosan uptake on any stiffness, blocking total actin polymerization reduced uptake on all stiffnesses. Taken together, these findings show a highly complex mechanosensitive response to phagocytosis in regards to both particle type and mechanism of uptake. Further, PMNs seeded on Candida albicans hyphae were mechanosensitive in regards to oxidative burst and NETosis; both ROS production and NETosis increased on stiffer substrates. Finally, in addition to phagocytosis and hyphae, we also looked at cytokine release to secondary stimulation by fMLP. We found both Lactoferrin and IL-8 release to increase on stiffer substrates, while elastase release was not mechanosensitive. Taken together, these findings indicate a strong regulatory role of substrate stiffness during PMN effector responses. While extensive work has been done to elucidate PMN biochemical interactions, understanding important mechanosensitive mechanisms underlying phagocytosis, NETosis, respiratory burst, and cytokine release may provide further therapeutic targets to benefit patients in the future by reducing harmful PMN behaviors and/or promoting behaviors crucial to pathogen clearance.

Enhanced Glycolysis Is Required for Antileishmanial Functions of Neutrophils Upon Infection With Leishmania donovani.

Visceral leishmaniasis (VL) is a fatal parasitic disease if untreated. Treatment options of VL diminish due to emerging drug resistance. Although the principal host cells for the multiplication of Leishmania are macrophages, neutrophils are the first cells infected with the parasites rapidly after parasite inoculation. Leishmania can survive in neutrophils despite the potent antimicrobial effector functions of neutrophils that can eliminate the parasites. Recently, the growing field of immunometabolism provided strong evidence for the therapeutic potential in targeting metabolic processes as a means of controlling immune effector functions. Therefore, the understanding of the immunometabolic profile of neutrophils during Leishmania infection could provide new promising targets for host-directed therapies against VL. To our knowledge, this is the first study addressing the bioenergetics profile of L. donovani-infected primary human neutrophils. Transcriptome analysis of L. donovani-infected neutrophils revealed an early significant upregulation of several glycolytic enzymes. Extracellular flux analysis showed that glycolysis and glycolytic capacity were upregulated in L. donovani-infected neutrophils at 6 h post infection. An increased glucose uptake and accumulation of glycolytic end products were further signs for an elevated glycolytic metabolism in L. donovani-infected neutrophils. At the same time point, oxidative phosphorylation provided NADPH for the oxidative burst but did not contribute to ATP production. Inhibition of glycolysis with 2-DG significantly reduced the survival of L. donovani promastigotes in neutrophils and in culture. However, this reduction was due to a direct antileishmanial effect of 2-DG and not a consequence of enhanced antileishmanial activity of neutrophils. To further address the impact of glucose metabolism during the first days of infection in vivo, we treated C57BL/6 mice with 2-DG prior to infection with L. donovani and assessed the parasite load one day and seven days post infection. Our results show, that seven days post-infection the parasite load of 2-DG treated animals was significantly higher than in mock treated animals. This data indicates that glycolysis serves as major energy source for antimicrobial effector functions against L. donovani. Inhibition of glycolysis abrogates important neutrophil effector functions that are necessary the initial control of Leishmania infection.

SOCS-1 inhibition of type I interferon restrains Staphylococcus aureus skin host defense.

The skin innate immune response to methicillin-resistant Staphylococcus aureus (MRSA) culminates in the formation of an abscess to prevent bacterial spread and tissue damage. Pathogen recognition receptors (PRRs) dictate the balance between microbial control and injury. Therefore, intracellular brakes are of fundamental importance to tune the appropriate host defense while inducing resolution. The intracellular inhibitor suppressor of cytokine signaling 1 (SOCS-1), a known JAK/STAT inhibitor, prevents the expression and actions of PRR adaptors and downstream effectors. Whether SOCS-1 is a molecular component of skin host defense remains to be determined. We hypothesized that SOCS-1 decreases type I interferon production and IFNAR-mediated antimicrobial effector functions, limiting the inflammatory response during skin infection. Our data show that MRSA skin infection enhances SOCS-1 expression, and both SOCS-1 inhibitor peptide-treated and myeloid-specific SOCS-1 deficient mice display decreased lesion size, bacterial loads, and increased abscess thickness when compared to wild-type mice treated with the scrambled peptide control. SOCS-1 deletion/inhibition increases phagocytosis and bacterial killing, dependent on nitric oxide release. SOCS-1 inhibition also increases the levels of type I and type II interferon levels in vivo. IFNAR deletion and antibody blockage abolished the beneficial effects of SOCS-1 inhibition in vivo. Notably, we unveiled that hyperglycemia triggers aberrant SOCS-1 expression that correlates with decreased overall IFN signatures in the infected skin. SOCS-1 inhibition restores skin host defense in the highly susceptible hyperglycemic mice. Overall, these data demonstrate a role for SOCS-1-mediated type I interferon actions in host defense and inflammation during MRSA skin infection.

Mucosal-Associated Invariant T (MAIT) Cells Are Highly Activated and Functionally Impaired in COVID-19 Patients.

Coronavirus disease 2019 (COVID-19), caused by infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), comprises mild courses of disease as well as progression to severe disease, characterised by lung and other organ failure. The immune system is considered to play a crucial role for the pathogenesis of COVID-19, although especially the contribution of innate-like T cells remains poorly understood. Here, we analysed the phenotype and function of mucosal-associated invariant T (MAIT) cells, innate-like T cells with potent antimicrobial effector function, in patients with mild and severe COVID-19 by multicolour flow cytometry. Our data indicate that MAIT cells are highly activated in patients with COVID-19, irrespective of the course of disease, and express high levels of proinflammatory cytokines such as IL-17A and TNFα ex vivo. Of note, expression of the activation marker HLA-DR positively correlated with SAPS II score, a measure of disease severity. Upon MAIT cell-specific in vitro stimulation, MAIT cells however failed to upregulate expression of the cytokines IL-17A and TNFα, as well as cytolytic proteins, that is, granzyme B and perforin. Thus, our data point towards an altered cytokine expression profile alongside an impaired antibacterial and antiviral function of MAIT cells in COVID-19 and thereby contribute to the understanding of COVID-19 immunopathogenesis.

Leukotriene B4 licenses inflammasome activation to enhance skin host defense

The initial production of inflammatory mediators dictates host defense as well as tissue injury. Inflammasome activation is a constituent of the inflammatory response by recognizing pathogen and host-derived products and eliciting the production of IL-1β and IL-18 in addition to inducing a type of inflammatory cell death termed "pyroptosis." Leukotriene B4 (LTB4) is a lipid mediator produced quickly (seconds to minutes) by phagocytes and induces chemotaxis, increases cytokine/chemokine production, and enhances antimicrobial effector functions. Whether LTB4 directly activates the inflammasome remains to be determined. Our data show that endogenously produced LTB4 is required for the expression of pro-IL-1β and enhances inflammasome assembly in vivo and in vitro. Furthermore, LTB4-mediated Bruton's tyrosine kinase (BTK) activation is required for inflammasome assembly in vivo as well for IL-1β-enhanced skin host defense. Together, these data unveil a new role for LTB4 in enhancing the expression and assembly of inflammasome components and suggest that while blocking LTB4 actions could be a promising therapeutic strategy to prevent inflammasome-mediated diseases, exogenous LTB4 can be used as an adjuvant to boost inflammasome-dependent host defense.

IL-22 receptor signaling in Paneth cells is critical for their maturation, microbiota colonization, Th17-related immune responses, and anti-Salmonella immunity

Interleukin-22 (IL-22) signaling in the intestines is critical for promoting tissue-protective functions. However, since a diverse array of cell types (absorptive and secretory epithelium as well as stem cells) express IL-22Ra1, a receptor for IL-22, it has been difficult to determine what cell type(s) specifically respond to IL-22 to mediate intestinal mucosal host defense. Here, we report that IL-22 signaling in the small intestine is positively correlated with Paneth cell differentiation programs. Our Il22Ra1fl/fl;Lgr5-EGFP-creERT2-specific knockout mice and, independently, our lineage-tracing findings rule out the involvement of Lgr5+ intestinal stem cell (ISC)-dependent IL-22Ra1 signaling in regulating the lineage commitment of epithelial cells, including Paneth cells. Using novel Paneth cell-specific IL-22Ra1 knockout mice (Il22Ra1fl/fl;Defa6-cre), we show that IL-22 signaling in Paneth cells is required for small intestinal host defense. We show that Paneth cell maturation, antimicrobial effector function, expression of specific WNTs, and organoid morphogenesis are dependent on cell-intrinsic IL-22Ra1 signaling. Furthermore, IL-22 signaling in Paneth cells regulates the intestinal commensal bacteria and microbiota-dependent IL-17A immune responses. Finally, we show ISC and, independently, Paneth cell-specific IL-22Ra1 signaling are critical for providing immunity against Salmonella enterica serovar Typhimurium. Collectively, our findings illustrate a previously unknown role of IL-22 in Paneth cell-mediated small intestinal host defense.

MAIT Cells Display a Specific Response to Type 1 IFN Underlying the Adjuvant Effect of TLR7/8 Ligands.

Mucosal-associated invariant T (MAIT) cells constitute a highly conserved subset of effector T cells with innate-like recognition of a wide array of bacteria and fungi in humans. Harnessing the potential of these cells could represent a major advance as a new immunotherapy approach to fight difficult-to-treat bacterial infections. However, despite recent advances in the design of potent agonistic ligands for MAIT cells, it has become increasingly evident that adjuvants are required to elicit potent antimicrobial effector functions by these cells, such as IFNγ production and cytotoxicity. Indeed, TCR triggering alone elicits mostly barrier repair functions in MAIT cells, whereas an inflammatory milieu is required to drive the antibacterial functions. Cytokines such as IL-7, IL-12 and IL-18, IL-15 or more recently type 1 IFN all display an apparently similar ability to synergize with TCR stimulation to induce IFNγ production and/or cytotoxic functions in vitro, but their mechanisms of action are not well established. Herein, we show that MAIT cells feature a build-in mechanism to respond to IFNα. We confirm that IFNα acts directly and specifically on MAIT cells and synergizes with TCR/CD3 triggering to induce maximum cytokine production and cytotoxic functions. We provide evidences suggesting that the preferential activation of the Stat4 pathway is involved in the high sensitivity of MAIT cells to IFNα stimulation. Finally, gene expression data confirm the specific responsiveness of MAIT cells to IFNα and pinpoints specific pathways that could be the target of this cytokine. Altogether, these data highlight the potential of IFNα-inducing adjuvants to maximize MAIT cells responsiveness to purified ligands in order to induce potent anti-infectious responses.

Different MDSC Activity of G-CSF/Dexamethasone Mobilized Neutrophils: Benefits to the Patient?

Human neutrophils exert a well-known role as efficient effector cells to kill pathogenic micro-organisms. Apart from their role in innate immunity, neutrophils also have the capacity to suppress T cell-mediated immune responses as so-called granulocyte-myeloid-derived suppressor cells (g-MDSCs), impacting the clinical outcome of various disease settings such as cancer. Patients undergoing chemotherapy because of an underlying malignancy can develop prolonged bone marrow suppression and are prone to serious infections because of severe neutropenia. Concentrates of granulocytes for transfusion (GTX) constitute a therapeutic tool and rescue treatment to fight off these serious bacterial and fungal infections when antimicrobial therapy is ineffective. GTX neutrophils are mobilized by overnight G-CSF and/or Dexamethasone stimulation of healthy donors. Although the phenotype of these mobilized neutrophils differs from the circulating neutrophils under normal conditions, their anti-microbial function is still intact. In contrast to the unaltered antimicrobial effector functions, G-CSF/Dexamethasone-mobilized neutrophils were found to lack suppression of the T cell proliferation, whereas G-CSF-mobilized or Dexamethasone-mobilized neutrophils could still suppress the T cell proliferation upon cell activation equally well as control neutrophils. Although the mechanism of how G-CSF/Dex mobilization may silence the g-MDSC activity of neutrophils without downregulating the antimicrobial activity is presently unclear, their combined use in patients in the treatment of underlying malignancies may be beneficial—irrespective of the number of circulating neutrophils. These findings also indicate that MDSC activity does not fully overlap with the antimicrobial activity of human neutrophils and offers the opportunity to elucidate the feature(s) unique to their T-cell suppressive activity.

Leukocyte Immunoglobulin-Like Receptors (LILRs) on Human Neutrophils: Modulators of Infection and Immunity.

Neutrophils have a crucial role in defense against microbes. Immune receptors allow neutrophils to sense their environment, with many receptors functioning to recognize signs of infection and to promote antimicrobial effector functions. However, the neutrophil response must be tightly regulated to prevent excessive inflammation and tissue damage, and regulation is achieved by expression of inhibitory receptors that can raise activation thresholds. The leukocyte immunoglobulin-like receptor (LILR) family contain activating and inhibitory members that can up- or down-regulate immune cell activity. New ligands and functions for LILR continue to emerge. Understanding the role of LILR in neutrophil biology is of general interest as they can activate and suppress antimicrobial responses of neutrophils and because several human pathogens exploit these receptors for immune evasion. This review focuses on the role of LILR in neutrophil biology. We focus on the current knowledge of LILR expression on neutrophils, the known functions of LILR on neutrophils, and how these receptors may contribute to shaping neutrophil responses during infection.

Phagosomal removal of fungal melanin reprograms macrophage metabolism to promote antifungal immunity

In response to infection, macrophages adapt their metabolism rapidly to enhance glycolysis and fuel specialized antimicrobial effector functions. Here we show that fungal melanin is an essential molecule required for the metabolic rewiring of macrophages during infection with the fungal pathogen Aspergillus fumigatus. Using pharmacological and genetic tools, we reveal a molecular link between calcium sequestration by melanin inside the phagosome and induction of glycolysis required for efficient innate immune responses. By remodeling the intracellular calcium machinery and impairing signaling via calmodulin, melanin drives an immunometabolic signaling axis towards glycolysis with activation of hypoxia-inducible factor 1 subunit alpha (HIF-1α) and phagosomal recruitment of mammalian target of rapamycin (mTOR). These data demonstrate a pivotal mechanism in the immunometabolic regulation of macrophages during fungal infection and highlight the metabolic repurposing of immune cells as a potential therapeutic strategy.

Interleukin-22 receptor signaling in Paneth cells is critical for their maturation and antimicrobial function

Abstract Interleukin-22 (IL-22) acts in the intestine to promote critical tissue protective functions. However, since a diverse array of intestinal cell types (absorptive, secretory, and stem cells) express IL-22Ra1, a receptor for IL-22, it has been difficult to determine what cell type(s) specifically respond to IL-22 to mediate mucosal host defense. To address this question, we used entire gut epithelium, intestinal stem cell (ISC)-specific, and Paneth cell-specific IL-22Ra1 knockout mice. Entire epithelium-specific IL-22Ra1 knockout (Il22Ra1fl/fl;Villin-cre) mice displayed defects in Paneth cell function. Using ISC-specific IL-22Ra1 knockout mice (Il22Ra1fl/fl;Lgr5-EGFP-creERT2) and lineage tracing mice, we ruled out the involvement of Lgr5+ ISC-dependent IL-22Ra1 signaling in regulating the lineage commitment of epithelial cells, including Paneth cells. Using novel Paneth cell-specific IL-22Ra1 knockout mice (Il22Ra1fl/fl;Defa6-cre), we show that IL-22Ra1 signaling in Paneth cells is required for small intestinal host defense. We show that Paneth cell maturation, antimicrobial effector functions, gene expression of specific WNTs (Wnt6 and Wnt9b), and enteroid morphogenesis are dependent on cell-intrinsic IL-22Ra1 signaling. Furthermore, IL-22 signaling in Paneth cells regulates the intestinal commensal bacteria and microbiota-dependent IL-17A immune responses. Finally, we show Paneth cell-specific IL-22Ra1 signaling helps provide immunity against Salmonella typhimurium. Collectively, our findings provide a unique and novel role of IL-22 in Paneth cell-mediated small intestinal host defense.

Complement activation by C-reactive protein is essential for protection against pneumococcal infection

Abstract C-reactive protein (CRP), a component of the innate immune system, is an antimicrobial plasma protein. The primary ligand-binding specificity of CRP is for phosphocholine-containing substances and ligand-complexed human CRP activates the complement system in both human and mouse sera. It has been shown that human CRP protects mice against infection with lethal doses of Streptococcus pneumoniae; the protection is due to CRP-dependent decrease in bacteremia and increase in survival time. In this study, we tested a decades old hypothesis that the complement-activating function of ligand-complexed CRP contributes to protection of mice against pneumococcal infection. We employed site-directed mutagenesis of CRP, guided by its three-dimensional structure, and identified a mutant H38R which, unlike wild-type CRP, did not activate C3 in both human and mouse sera. The calcium-dependent binding to phosphocholine on pneumococci, the half-life in mouse circulation, and the overall pentameric structure of H38R mutant CRP were similar to that of wild-type CRP. Employing a murine model of pneumococcal infection, we found that passively administered H38R mutant CRP did not protect mice from infection. Infected mice injected with H38R mutant CRP showed no reduction in bacteremia and did not survive longer, as opposed to infected mice receiving wild-type CRP. Thus, the hypothesis that complement activation by ligand-complexed CRP is an antimicrobial effector function is true. Phagocyte-derived SOCS1 disrupts Staphylococcus aureus skin host defense

Abstract Methicillin-resistant Staphylococcus aureus (MRSA) skin infection is controlled by the actions of skin resident macrophages and the formation of a neutrophilic abscess that prevents bacterial spread and tissue damage. We and others have shown that myeloid differentiation factor 88 (MyD88) is required for the clearance of MRSA skin infection in mice. MyD88 expression is controlled by the balance between STAT1 and the suppressor of cytokine signaling 1 (SOCS1). Here, we hypothesized that SOCS1 inhibits antimicrobial effector functions and the inflammatory response, leading to poor abscess formation and tissue injury during MRSA skin infection. Our data show that MRSA skin infection enhances SOCS1 expression. Infection in myeloid-specific SOCS1 deficient mice displays decreased lesion size, lower bacterial loads, and increased abscess thickness when compared to WT mice. When we treated infected mice with a peptide that specifically inhibits the kinase inhibitory region (KIR) of SOCS1, it also improved infection outcome. Examining the mechanisms by which SOCS1 enhanced skin host defense, we observed increased phagocytosis and bacterial killing in SOCS1 deficient macrophages and KIR peptide-treated cells. Increased antimicrobial effector function correlated with enhanced STAT1 activation and increased production of IFNγ in vivo and in vitro. Next we tested whether IFNγ is crucial to improved host defense in KIR-treated mice. Our data show that the beneficial effect of SOCS1 KIR peptide in skin host defense was abrogated in mice treated with an IFNR blocking antibody and in the IFNR deficient mice. Overall, these data show that preventing SOCS1 actions enhances microbial clearance and host defense during MRSA skin infection.

An Attempt to Polarize Human Neutrophils Toward N1 and N2 Phenotypes in vitro.

Neutrophils act as the first line of defense against invading pathogens. Although traditionally considered in context of their antimicrobial effector functions, the importance of tumor-associated neutrophils (TANs) in the development of cancer has become increasingly clear during the last decade. With regard to their high plasticity, neutrophils were shown to acquire an anti-tumorigenic N1 or a pro-tumorigenic N2 phenotype. Despite the urgent need to get a comprehensive understanding of the interaction of TANs with their tumor microenvironment, most studies still rely on murine tumor models. Here we present for the first time a polarization attempt to generate N1 and N2 neutrophils from primary human neutrophils in vitro. Our results underscore that N1-polarized neutrophils have a pro-inflammatory phenotype characterized among others by a higher level of intercellular adhesion molecule (ICAM)-1 and high secretion of interferon (IFN)γ-induced protein 10 (IP-10)/C-X-C motif chemokine 10 (CXCL10) and tumor necrosis factor (TNF). Further, we demonstrate that neutrophils incubated under a tumor-mimicking in vitro environment show a high cell surface expression of C-X-C motif chemokine receptor 2 (CXCR2) and secrete high levels of interleukin (IL)-8. These findings suggest that it is feasible to polarize blood-derived primary human neutrophils toward N1- and N2-like phenotypes in vitro. Further, we hypothesized that the presence of anti-inflammatory neutrophil phenotype is not a phenomenon limited to cancer but also occurs when neutrophils are infected with intracellular pathogens. Indeed, our findings indicate that N2-polarized neutrophils exert a markedly decreased capacity to kill the protozoan parasite Leishmania donovani and therefore permit parasite persistence.

The Orphan Immune Receptor LILRB3 Modulates Fc Receptor-Mediated Functions of Neutrophils.

Neutrophils are critical to the generation of effective immune responses and for killing invading microbes. Paired immune receptors provide important mechanisms to modulate neutrophil activation thresholds and effector functions. Expression of the leukocyte Ig-like receptor (LILR)A6 (ILT8/CD85b) and LILRB3 (ILT5/CD85a) paired-receptor system on human neutrophils has remained unclear because of the lack of specific molecular tools. Additionally, there is little known of their possible functions in neutrophil biology. The objective of this study was to characterize expression of LILRA6/LILRB3 receptors during human neutrophil differentiation and activation, and to assess their roles in modulating Fc receptor-mediated effector functions. LILRB3, but not LILRA6, was detected in human neutrophil lysates following immunoprecipitation by mass spectrometry. We demonstrate high LILRB3 expression on the surface of resting neutrophils and release from the surface following neutrophil activation. Surface expression was recapitulated in a human PLB-985 cell model of neutrophil-like differentiation. Continuous ligation of LILRB3 inhibited key IgA-mediated effector functions, including production of reactive oxygen species, phagocytic uptake, and microbial killing. This suggests that LILRB3 provides an important checkpoint to control human neutrophil activation and their antimicrobial effector functions during resting and early-activation stages of the neutrophil life cycle.
