Chemical Composition and Antimicrobial Activity of Essential Oils from Three Mediterranean Plants against Eighteen Pathogenic Bacteria and Fungi.

The chemical composition and antimicrobial activity of essential oils (EOs) obtained from three medicinal plants of the Moroccan flora were evaluated. The chemical composition of EOs of Thymus leptobotrys, Laurus nobilis and Syzygium aromaticum was determined using a gas chromatograph coupled with mass spectrometry. Carvacrol (75.05%) was the main constituent of T. leptobotrys EOs, while 1,8-cineole (31.48%) and eugenol (82.16%) were the predominant components of L. nobilis and S. aromaticum EOs, respectively. The antimicrobial activity of the EOs was evaluated qualitatively and quantitatively against 18 microbial strains pathogenic to humans by using the disc diffusion method, and by measuring the minimum inhibitory concentration (MIC) and minimum microbicidal concentration (MMC). The EOs of T. leptobotrys were the most active against the strains tested, with inhibitory zone values ranging from 7.00 to 45.00 mm, and MIC and MMC values ranging from 0.312 to 80.00 mg/mL. In many cases, these EOs exhibited higher antibacterial and antifungal activities than the chemical compounds ciprofloxacin and fluconazole, respectively. This high antimicrobial activity can be ascribed to their richness in carvacrol. The EOs of T. leptobotrys, L. nobilis, and S. aromaticum could be considered a promising alternative to replace chemical antimicrobials, and a readily available natural source of bioactive compounds.

The antimicrobial activity of essential oils from different species of lime, lemon and orange against oral pathogens

This study investigated in vitro the antimicrobial activity against oral pathogens of essential oils (EOs) from different species of lime, lemon, and orange. The EOs were chemically characterized by gas chromatography coupled to mass spectrophotometry (GC-MS) and tested for their antimicrobial activity (MIC and MBC/MFC) against Candida spp., Fusobacteriumnucleatum, Porphyromonasgingivalis, Prevotella intermedia, Streptococcus mutans, Streptococcus mitis and Streptococcus sanguinis. Data were analyzed descriptively and by one-way analysis of Variance (ANOVA) considering a 5% significance level (P < 0.05). Limonene was the major compound tentatively identified in the samples. Among the EOs, C. sinensis showed lower MIC values against C. krusei, C. albicans and C. tropicalis. Nystatin showed MIC of 0.125 mg/mL against all Candida spp. strains and MFC values ≥ 1 mg/mL. C. sinensis EO and chlorhexidine showed MIC/MBC values against bacterial strains ranging from 1 to 8 mg/mL and 2 to 8 mg/mL, respectively. To conclude, C. sinensis EO was the most effective sample against Candida spp. Overall, the EOs showed weak antibacterial activity with bacteriostatic effects, except for C. latifolia, which showed strong antimicrobial activity against P. intermedia.C. sinensis EO showed bactericidal effects against S. mitis.

Chemical composition and antimicrobial activity of essential oils from Mentha pulegium and Rosmarinus officinalis against multidrug-resistant microbes and their acute toxicity study

Abstract This article aimed to study the antimicrobial activity, chemical composition, and acute oral toxicity of essential oils (EOs) of Mentha pulegium and Rosmarinus officinalis, two aromatic and medicinal plants widely used in the traditional Moroccan pharmacopeia. The average content of EOs was 3.2 and 2.5% for M. pulegium and R. officinalis, respectively. The chemical characterization showed a richness in some compounds identified by gas chromatography coupled with mass spectrometry (GC/MS): R(+)-Pulegone (45.48%), Menthone (14.2%), Piperitone (8.15%), and Isomenthone (7.18%) in M. pulegium and 1,8-Cineole (46.32%), Camphene (13.4%), and α-Pinene (9.52%) in R. officinalis. These metabolites showed a significant antimicrobial effect against the tested strains (bacteria and yeasts isolated from the hospital environment) compared to synthetic antibiotics that seem to be ineffective against resistant microorganisms. Based on lethal concentration LD50 >5,000 mg/kg (body weight), the oil was found to be marginally safe according to OECD guidelines and can be further explored (bio-product with low risk).

Antimicrobial Activity of Essential Oils Evaluated In Vitro against Escherichia coli and Staphylococcus aureus.

The spread of extended-spectrum β-lactamase-producing Escherichia coli and methicillin-resistant Staphylococcus aureus has caused a reduction in antibiotic effectiveness and an increase in mortality rates. Essential oils (EOs), known for their therapeutic efficacy, can be configured as novel broad-spectrum biocides. Accordingly, the bacteriostatic–bactericidal activity of Citrus Lemon (LEO), Pinus Sylvestris (PEO), Foeniculum Vulgaris (FEO), Ocimum Basilicum (BEO), Melissa Officinalis (MEO), Thymus Vulgaris (TEO), and Zingiber Officinalis Rosc. (GEO), at concentrations ranging from 1.25 to 40% (v/v), were tested in vitro against different E. coli and S. aureus strains using minimal inhibitory concentrations (MICs) and minimum bactericidal concentrations (MBCs). The chemical compositions of the EOs were analyzed using GC/MS. The major components of all seven tested oils were limonene, α-pinene, anethole, estragole, citral, thymol, and zingiberene, respectively. We found that the bacteriostatic–bactericidal activity of the EOs was related to their chemotypes and concentrations, as well as the strain of the bacteria. A dose–effect correlation was found when testing GEO against S. aureus strains, whilst FEO was found to have no activity regardless of concentration. PEO, MEO, and BEO were found to have bactericidal effect with a MIC and MBC of 1.25% (v/v) against S. aureus strains, and LEO was found to have values of 1.25% (v/v) and 5% (v/v) against ATCC and clinical isolate, respectively. Interestingly, the antimicrobial activity of TEO was not related to oil concentration and the complete inhibition of growth across all E. coli and S. aureus was observed. Although preliminary, our data demonstrate the efficacy of EOs and pave the way for further investigations on their potential synergistic use with traditional drugs in the human and veterinary fields.

In the present study, Salvia fruticosa and Salvia aramiensis essential oils’ chemical composition was compared, and their in vitro antimicrobial activity was tested against Escherichia coli, Klebsiella pneumoniae, Bacillus subtilis, Staphylococcus aureus, Candida albicans and Candida parapsilosis. The major compounds of S. fruticosa oil were eucalyptol (29.04%) and camphor (21.32%). Major components of S. aramiensis essential oil were eucalyptol (59.51%) and β-pinene (20.03%). The antimicrobial activities of essential oils were investigated by using disc diffusion and spectrophotometric broth micro-dilution methods. The highest inhibition zone diameter of S. aramiensis was found at 10.2 mm against C. albicans while the low inhibition zones (IZ) were on E. coli and B. subtilis (0.1 mm). Whereas the highest IZs of S. fruticosa was 8.38 for C. parapsilosis, the lowest IZs were 0.1 mm for E. coli, K. pneumoniae and B. subtilis by disc diffusion method (P˂0.05). The Minimum Inhibitory Concentration (MIC) of S. aramiensis and S. fruticosa EOs ranged from 56.2 to 146 μg/mL for all pathogens (P˂0.05). As a result, the antimicrobial efficacy of S. aramiensis on S. aureus and C. parapsilosis was demonstrated for the first time in this study, furthermore, both essential oils were found to be more effective against studied fungi rather than bacteria.

Determination of Antimicrobial Activities of Essential Oils of Different Thyme Species Grown in Çanakkale Conditions

Turkey is one of the leading countries in the trade of medicinal and aromatic plants due to its geography, and while exporting many medicinal plants, it also imports many plants. Thyme is often used as a spice in meat, fish and many other dishes due to its aromatic properties. It is a scientific fact that the essential oils in thyme have a strong antimicrobial effect. Thymol, which gives the distinctive smell of thyme, is an advanced antimicrobial agent. For this reason, it is recommended by many scientists to use thyme in the preservation of ready-made foods and disinfection of foods. It is an important plant for the country's economy, as it is an important medicinal aromatic plant and meets a large part of the thyme used in the world. Thyme, which is obtained by collecting from nature and cultivating in our country, has been an important research topic for scientists and many positive results have been obtained and continue to be achieved. This research was carried out at Çanakkale Onsekiz Mart University Faculty of Agriculture in 2021 in order to reveal the effect of essential oils obtained from 3 different thyme grown in Çanakkale conditions on 4 different bacteria. As research material, essential oils of Izmir thyme (Origanum onites), Istanbul thyme (Origanum vulgare) and medical thyme (Thymus vulgaris) were obtained by hydro-distillation method, and the anti-bacterial activities of these essential oils were determined against the determined bacteria Pseudomonas aeruginosa, Staphylococcus aureus, Enterobacter cloacaurmicium, Salmonella typhimurium. The antimicrobial activities of essential oils were determined by the disc diffusion method. As a result, according to the data obtained from all thyme essential oils, while all of them showed an antimicrobial effect on bacteria, the most antimicrobial activity was seen in O. onites and O. vulgare essential oils, while the least antimicrobial activity was seen in T. vulgaris essential oil. It showed the best activity in Origanum vulgare, especially on S. aureus (42 mm) and S. typhimurium (39 mm) bacteria. In the study, T. vulgaris had low activity against P. aeruginosa with an inhibition diameter of 19 mm.

Chemical constituents and antimicrobial ability of essential oil from the fruits of Lonicera maackii (Rupr.) Maxim.

Essential oils are the natural products that can pave the way for the creation of effective drugs with various beneficial properties. Plants of Lonicera maackii are well known in the natural growth regions of temperate Asia due to therapeutic effects and a high content of the bioactive compounds. Species diversity of steppe vegetation is enriched by the introduction of plants, but the assessment of this process effectiveness should also take into account the realization of plant genetic potential for the accumulation of secondary metabolites in the new growth conditions. The aim of the work was to establish whether the plants of L. maackii introduced in the steppe zone could reproduce the health promoting properties. The essential oil from the fruits of L. maackii was obtained by hydro distillation method. Chloroform and ethyl acetate fractions isolated from the aqueous distillate of L. maackii fruits were analyzed by gas chromatography combined with mass spectrometry (GC-MS analysis). Chemical constituents identified in ethyl acetate and chloroform fraction represented 98.6% and 94.2 % of the total compounds content, respectively. Among the most abundant components were hexadecane, acetic acid, butyl ester, hexadecanoic acid, methyl ester, linoleic acid, and linoleic acid methyl ester in the and the alkanes heptacosane, hexadecane, and tetradecane in chloroform fraction. Linoleic acid and some minor constituents of L. maackii fruit essential oil, such as ethyl-isoallocholate, arachidic acid, linalool and d-mannitol-1-decylsulfonyl are well known because of their reported bioactivities. The antimicrobial activity of essential oil was assessed against several clinical and collection bacterial and fungal strains. The results of disc-diffusion assays showed low to moderate activity of L. maackii fruit essential oil against Gram-positive and Gram-negative bacterial strains, as well as against fungal itraconazole-resistant clinical strain of Candida albicans. Thus, the essential oil derived from Lonicera maackii fruits may serve as potential resource of bioactive compounds having beneficial properties, in particular, the ability to overcome the resistance of pathogenic microorganisms to drugs. Further research is needed to identify the full range of bioactivities of the essential oil obtained from honeysuckle fruits.

Antimicrobial Activity of Essential Oil from Avocado (Persea americana) Seed and Pulp on Some Pathogenic Organisms

Introduction: Persea americana is regarded as a unique plant in many African countries. Its numerous parts are employed in the treatment of several diseases. The possible function mechanism of P. americana functions is under study.
 Aims: This study was designed to investigate the phytochemical analysis and antibacterial effect of oil extracted from seed and pulp of P. americana using agar diffusion technique.
 Methodology: The seed oil of P. americana was exhaustively extracted with a Soxhlet extractor from 500 g seeds and 200 g seeds of P. americana using di-ethyl ether as solvent. The extraction solvent was removed to obtain the oil which was then subjected to antimicrobial activity test to determine its activity against the following clinical isolates namely Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Streptococcus pneumonia and Candida albicans using conventional antibiotics as control. Phytochemical screening was carried out following standard methods.
 Results: The result of phytochemicals screening was presented with alkaloids, saponin, flavonoids, polyphenol, tannins, steroids as present. The result revealed P. aeruginosa to have the highest diameter of zones of inhibition of 12.00 – 30.00 mm at the concentrations of 0.1 – 0.4 g/mL respectively. S. aureus had diameter of zones of inhibition of 10.00 – 20.00 mm at the concentrations of 0.1 – 0.4 g/mL. E. coli had diameter of zones of inhibition of 10.00 – 15.00 mm at the concentrations of 0.1 – 0.4 g/mL respectively. C. albicans had diameter of zones of inhibition of 8.00 – 13.00 mm at the concentrations of 0.1 – 0.4 g/mL respectively; and S. pneumonia had diameter of zones of inhibition of 8.00 – 12.00 mm at the concentrations of 0.1 – 0.4 g/mL respectively. The results of the antimicrobial test on the seed oil showed appreciable antibacterial activities against the test organisms. The result of the test organisms was susceptible to conventional antibiotics.
 Conclusion: Oil extract of avocado pear (P. americana) will be helpful to many researchers in the field of finding antibacterial activities in plant; and the oil is recommended for treatment of skin infections inherent with these organisms.

Chemical Composition and Antibacterial Activity of Helichrysum italicum (Roth) G. Don Commercial Essential Oil from Serbia

Medicinal plants have been preferred sources against drug-resistant pathogens recently. The antimicrobial activities of essential oils have been known for a long time. In the present research, the chemical composition and antibacterial activity of commercial essential oil of Helichrysum italicum (Roth) G. Don (HIE) were evaluated. Seventy components which constitute 95.7%, were identified in the oil. The major constituents of HIE were neryl acetate (15.6%), α-pinene (11.5%), and γ-curcumene (11.3%). The oilwas screened for antibacterial activity by agar well diffusion and broth microdilution methods. The essential oil displayed remarkable antibacterial activity against test bacteria, except for Escherichia coli, which displayed resistance. The minimum inhibitory concentration (MIC) of essential oil were 5 μg/mL for Staphylococcus aureus and Pseudomonas aeruginosa, 10 μg/mL for Bacillus cereus and B. subtilis. These results suggested that the commercial oil of H. italicum has the potential to be used as a natural antibacterial agent.

Essential Oils of Three Balkan Abies Species: Chemical Profiles, Antimicrobial Activity and Toxicity toward Artemia salina and Drosophila melanogaster.

This study focused on the essential oils (EOs) isolated from needles with twigs of three indigenous Balkan Abies species (A. alba, A. × borisii-regis and A. cephalonica) regarding their chemical composition, antimicrobial activity and toxicity toward crustaceans and insects. Even though distinct phytochemical profiles of dominant volatiles were revealed for each species, β-pinene and α-pinene represented the first two major volatiles in all three EOs. Antimicrobial activity of EOs has shown inhibitory effect against all 17 studied strains (ATCC and respiratory isolates) in the range of 0.62-20.00 mg/mL (MICs). Further, all three EOs exhibited strong toxicity (LC50 <100 μg/mL) in Artemia salina lethality bioassay, but with significant differences that depended on the EO type. Additionally, tested EOs have shown a certain level of toxicity against Drosophila melanogaster, mostly at the highest tested concentration (3 %) which caused significant prolongation of developmental time, larvicidal effect and pupal mortality. In the three biological assays performed, there was no observed inhibitory effect or weakest activity for A. alba EO. Further, A. cephalonica EO has shown the highest levels of antimicrobial activity and toxicity toward A. salina, while in relation to the insecticidal potential, A. cephalonica and A. × borisii-regis EOs exhibited similar level of toxicity against D. melanogaster.

Evaluation of the antimicrobial activity of the crude ethanol extract, essential oil, and fractions from Campomanesia pubescens leaves

Campomanesia pubescens (DC.) O. Berg (Myrtaceae), widely known as “gabiroba”, is a shrub or subshrub used in folk medicine (leaves and barks of the stem) in the form of decoction or infusion in combating urinary tract disorders, diarrhea, and as an astringent. The present work aimed to evaluate the antimicrobial activity of essential oil, crude ethanol extract and hexane fractions, dichloromethane, ethyl acetate, and aqueous from C. pubescens leaves against Gram-positive, Gram-negative bacteria, and fungi. The leaves were collected in Hidrolândia - Goiás and the essential oil was obtained by hydrodistillation in a Clevenger apparatus. The extract and fractions were tested against bacteria and fungi using the microdilution method. The aqueous fractions and ethyl acetate showed strong inhibiting activity (MIC= 8 to 16 μg/mL) against Candida glabrata, Cryptococcus neoformans, and Saccharomyces cerevisiae and moderate activity (MIC= 32 to 64 μg/mL) against Candida krusei, Candida parapsilosis, and Candida tropicalis. There is no report in the literature on the antifungal activity of ethanol extract and fractions of the leaves of C. pubecens. It is concluded through this study that the aqueous fractions and ethyl acetate from C. pubecens leaves presented strong antifungal activity which may justify its popular use in the treatment of urinary tract disorders. Therefore, this species has therapeutic potential that underlies more in-depth studies. This is the first study of the antifungal activity of crude extract and fractions from C. pubecens leaves.

A comparative evaluation of chemical composition and antimicrobial activities of essential oils extracted from different chemotypes of Cinnamomum camphora (L.) Presl

The purpose of this study is to determine the chemical composition of the essential oils of Cinnamomum camphora (L.) Presl leaves (CCPL) from 5 different habitats in China by GC-MS, and to evaluate their antimicrobial activities against 3 foodborne pathogens, using a paper disc diffusion method. A total of 30 compounds were identified with a predominance of oxygenated monoterpenes, including linalool (42.65%-96.47%), eucalyptol (39.07%-55.35%) and camphor (26.08%) as well as monoterpene hydrocarbons such as sabinene (6.18%-12.93%) and α-terpineol (8.19%-13.81%). Through cluster analysis, CCPL from 5 different habitats can be well divided into 2 categories. Combining with principal component analysis, the habitats can be better correlated with the chemical constituents of the essential oils. The antimicrobial activities of 5 extracted essential oils against 2 gram-negative bacteria and one gram-positive bacteria were assessed. It showed that the essential oil extracted from the CCPL harvested in Jinxi had the strongest antibacterial property. The results of this study provided basis for resource identification of CCPL and quality difference identification of essential oils. Research on the antibacterial properties of several pathogenic strains has proved its application value as a natural food preservative.

Chemical Composition and Antimicrobial Activity of Essential Oil of Fruits from Vitex agnus-castus L., Growing in Two Regions in Bulgaria.

The chemical composition of Vitex agnus-castus L. (Verbenaceae family) fruits, collected from two regions in Bulgaria (south-central and north-east Bulgaria), was investigated. The content of proteins (5.3–7.4%), carbohydrates (73.9–78.8%), fiber (47.2–49.9%), ash (2.5–3.0%), essential oils (0.5%), and vegetable oil (3.8–5.0%) were identified in the fruits. The composition of the essential oils (EOs) of Vitex fruits from both regions was determined; the main compounds were 1,8-cineole (16.9–18.8%), α-pinene (7.2–16.6%), sabinene (6.7–14.5%), and bicyclogermacrene (7.3–9.0%), but significant differences in the quantitative and qualitative composition of EOs between the regions were found. The EOs of plants from north-east Bulgaria demonstrated antimicrobial activity against the pathogenic species Salmonella abony, Staphylococcus aureus, and Bacillus subtilis, but the Gram-negative bacteria Esсherichia coli and Pseudomonas aeruginosa exhibited resistance to the oil. Linoleic acid predominated in vegetable oil from both regions, followed by oleic acid. β-sitosterol and γ-tocopherol were the main components in the sterol and tocopherol fraction of the lipids. Phosphatidic acids were the main components in the vegetable oil from north-east Bulgaria, while in the vegetable oil from south-central Bulgaria, all phospholipids were found in almost the same quantity. Overall, significant differences were observed in the chemical composition (proteins, carbohydrates, ash and moisture) of the fruits from the two regions of Bulgaria, as well as in the content of the main components of their essential and vegetable oils.

Comparative Analysis of the Antimicrobial Activity of Essential Oils and Their Formulated Microemulsions against Foodborne Pathogens and Spoilage Bacteria.

The antimicrobial activity of several essential oils (EOs) and their related microemulsions (MEs) was investigated. EOs were obtained from Cannabis sativa L. cv CS (C. sativa), Carum carvi L. (C. carvi), Crithmum maritimum L. (C. maritimum), Cuminum cyminum L. (C. cyminum), x Cupressocyparis leylandii A.B. Jacks & Dallim. (C. leylandii), Cupressus arizonica Greene (C. arizonica), Ferula assa-foetida L. (F. assa-foetida)., Ferula gummosa Boiss. (F. gummosa), Juniperus communis L. (J. communis), Juniperus x pfitzeriana (Spath) P.A. Schmidt (J. pfitzeriana), Pimpinella anisum L (P. anisum). Preliminary screening revealed that Cuminum cyminum, Crithmum maritimum, and Pimpinella anisum (10% v/v) were effective against all tested microorganisms (Escherichia coli ATCC 35218, Listeria monocytogenes ATCC 7644, Staphylococcus aureus ATCC 29213, Pseudomonas fluorescens DSM 4358, and Candida albicans ATCC 10231), with growth inhibition diameter from 10 to 25 mm. These EOs were used to formulate the MEs with an average size < 50 nm and a good stability over 30 days. EOs’ antimicrobial activity was further enhanced in the MEs, with a generalized lowering of minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. C. cyminum-ME reached, in most cases, MIC two times lower (0.312%) than the corresponding EO (0.625%) and even eight times lower against S. aureus (0.156 vs. 1.25%). A more remarkable microbicide effect was noted for C. cyminum-ME, with MBC values eight times lower (from 0.312 to 0.625%) than the corresponding EO (from 2.5 to 5%). Overall, MEs resulted in an efficient system for EOs encapsulation, enhancing solubility and lowering concentration to exert antimicrobial efficacy.

COMPONENT COMPOSITION AND ANTIMICROBIAL ACTIVITY OF THE ESSENTIAL OIL MELILOTUS OFFICI-NALIS (L.) PALL, GROWING IN UZBEKISTAN

The component composition of the essential oil (EO) from the air-dry and fresh aerial parts of the plant Melilotus officinalis (L.) Pall growing on the territory of the Namangan region of the Republic of Uzbekistan during the flowering period has been studied. The GC-MS method identified 49 compounds in the EO from an air-dry plant, while 22 substances were found in the EO from a fresh plant, which is 98.0% and 99.2% of the total amount of EO, respectively.&#x0D; The main components of EO, both air-dry and fresh aboveground parts, is coumarin, the content of which is 83.2% and 87.9%, respectively. EOs also contained alcohols, carboxylic acids, aldehydes and ketones, a small amount of monoterpenes and oxidized monoterpenes, etc. The antimicrobial activity of essential oil against Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Candida albicans, Pseudomonas aeruginosa, Candida albicans was studied.

Antimicrobial Potential of Essential Oils from Aromatic Plant Ocimum sp.; A Comparative Biochemical Profiling and In-Silico Analysis

Medicinal and aromatic plants (MAPs) are a rich source of bioactive compounds that are immensely important due to their potential use in pharmacological and agricultural applications. Here, we have evaluated the antimicrobial activity of essential oils (EOs) from three different species of Ocimum: O. gratissimum (EO1), O. tenuiflorum (EO2), and O. sanctum (EO3). The EOs were screened for antibacterial activity against pathogenic strains of Escherichia coli, Enterobacter cloacae and methicillin-resistant Staphylococcus aureus (MRSA). The essential oils EO1 and EO3 showed significant growth inhibition of the tested bacteria. Likewise, all EOs exhibited antifungal potential against the broad-spectrum plant fungal pathogen Sclerotinia sclerotiorum that causes white-mould disease in plants. Moreover, the antimicrobial potential of the EOs correlates well with their antioxidant activity determined by DPPH free radical scavenging activity. The biochemical analysis of the EOs employing high-performance thin-layer chromatography, gas chromatography-mass spectrometry, and Fourier transform infrared spectroscopy, revealed the presence of distinct phytoconstituents that might be responsible for their differential bioactivity. Furthermore, an in-silico evaluation of the candidate phytoconstituents using molecular docking analysis suggests their potential for antimicrobial applications. Altogether, our results clearly show that EO1 and EO3 possess promising antimicrobial properties, and therefore could be utilized as a potential antimicrobial agent.
