Phytochemical Compositions, Nutritional Contents, Cytotoxicity and Anti-Inflammatory Activity of Different Extracts from Spirogyra neglecta (Hassall) Kützing

Spirogyra neglecta (Hassall) Kützing is an edible freshwater macroalga. It is available and consumed as local food in Thailand, especially in the northern and northeastern parts. To ascertain its good benefit for health and safe for consumption, this study was, therefore carried out to determine the phytochemical compositions, nutritional contents, cytotoxicity, and anti-inflammatory activity of the different extracts from S. neglecta. The extracts were prepared by using different solvents, such as propylene glycol, methanol, and ethanol solutions. GC-MS analysis revealed the presence of 30, 39 and 36 phytochemical compounds in propylene glycol, methanol, and ethanol extracts, respectively. Phytol, which has been reported to have anti-inflammatory activity, was one of the predominant compositions in the extracts. The extracts contained high nutritional contents of protein, lipid, fiber, ash, moisture, and carbohydrate up to 21.68 ± 4.52, 3.53 ± 0.23, 14.77 ± 0.89, 10.73 ± 0.47, 9.79 ± 0.11 and 47.36 ± 3.89 %, respectively. The highest total protein was found in propylene glycol extract. The highest total lipid was in ethanol extract. Meanwhile, the highest fiber, ash and total carbohydrate were in methanol extract. In addition, all the extracts possess relatively low cytotoxicity, with IC50 values greater than 1,000 µg/mL. The extracts 1,000 µg/mL exhibited anti-inflammatory activity via inhibition Nitric oxide (NO) production by 29.87 ± 1.56 to 55.17 ± 1.91 %. The propylene glycol extracts from S. neglecta showed the highest inhibitory activity on NO production compared to others. The overall results indicating the propylene glycol is the solvent that produces the best extraction performance. The extracts from S. neglecta exhibit anti-inflammatory activity by inhibiting NO production. The phytochemical compositions are partly responsible for anti-inflammatory activity of the extracts. The high nutritional contents complement relatively low toxic. Thus, S. neglecta can be used as benefit natural sources for food products, supplements, and medicinal plants. Isolation and Characterization of Flavonoid Naringenin and Evaluation of Cytotoxic and Biological Efficacy of Water Lilly (Nymphaea mexicana Zucc.).

Despite its limited exploration, Nymphaea mexicana Zucc. can be beneficial if pharmacology, isolation, and biological evaluation are given attention. It is an aquatic species that belongs to the family Nymphaeaceae. The thrust area of the work was the extraction, isolation, and biological evaluation of different extracts of the N. mexicana Zucc. plant. The primary goal of this research was to assess the antimicrobial, antioxidant, and anticancer activities of the extracts and to isolate the target naringenin compound. Comparative FT IR analysis of different extracts of this plant revealed the presence of functional groups of plant secondary metabolites, including polyphenols, flavonoids, terpenoids, esters, amines, glycosides, alkanes, alkaloids, fatty acids, and alcohols. Moderate free radical scavenging potential has been achieved for the various extracts via reducing power and DPPH assays. While cytotoxic activity was evaluated by colorimetric and lactate dehydrogenase cell viability tests on potent cancer cell lines. Lung adenocarcinoma epithelial cells (A-549), and breast cells (MC-7) were treated with MeOH extract. The antimicrobial activity against bacterial strains was evaluated using Gram-positive and -negative cultures, where maximum and minimum inhibition zones were recorded for different strains, including 1.6-25.6 μg/mL for Streptococcus aureus, using the agar well diffusion method. In addition, the anti-inflammatory activity of different extracts of N. mexicana Zucc. was evaluated in a nitrite radical scavenging assay with high concentrations of secondary metabolites, which are important against human pathogens and other diseases.

In vitro and In vivo Anti-inflammatory Activity of the Extracts of Whole Plant Argyreia imbricata (Roth) Sant. & Patel

Plants of the genus Argyreia have ethnomedicinal importance, and several pharmacological activities are also reported. In this study, the anti-inflammatory activity of different extracts of Argyreia imbricata was evaluated by in vitro and in vivo methods. In both evaluations, standard, Diclofenac sodium was used for comparative evaluation. In this study, extraction of powdered whole plant material was done with different solvents viz., petroleum ether, chloroform, ethyl acetate and methanol by soxhelation. In vitro anti-inflammatory activity of all the prepared extracts was evaluated by stabilization of human red blood cell (HRBC) membrane in different temperature and tonicity conditions. Among the six different concentrations of four tested extracts, the ethyl acetate and methanol extracts (1000μg/ml) showed significant activity in the in vitro evaluation. They were selected for the in vivo evaluation on the paw oedema induced by carrageenan on Wistar albino rats. Two doses, 200mg.kg-1 and 400mg.kg-1 of the test extracts were subjected to evaluation. Both the tested extracts showed the activity, particularly, the methanol extract in the dose of 400mg.kg-1 showed significant activity. Results of this study strongly supported the anti-inflammatory activity of the tested extracts. Further, studies on toxicity, identification, isolation of the active constituents may give useful results.

The different extracts of Holoptelea integrifolia (H.integrifolia) leaves, stem bark and fruits were studied as a potent natural source of antimicrobial, antioxidant and wound healing potential. This work was carried out to evaluate antimicrobial, antioxidant and anti-inflammatory activity of different extracts of H.integrifolia. The antimicrobial activity of the H.integrifolia ethanolic extract was studied against five fungal and bacterial strains by utilizing the agar well diffusion method and MIC. Among several strain, the ethanolic extract of fruit has shown higher antimicrobial inhibition zone as 9.25-16 mm compare to other two extracts of stem and leaves as 10- 13.25 mm and 6-10.2 mm respectively. The antioxidant activities for different extract were also determined by DPPH free radical assay, Hydroxyl Radical Scavenging and Nitric Oxide Radical Scavenging Activity method. The anti-inflammatory activity also estimated basedon formalin induced paw edema method on Wistar albino rats. The different extracts of leaves, stem bark and fruit parts of Holoptelea integrifolia were estimated for in vivo anti-inflammatory activity against the animal model of female Wistar albino rats. The results of anti-inflammatory activity revealed that the Ethanol extracts showed vital and dose-dependent anti-inflammatory effects. Our findings revealed that aerial parts of H.integrifolia contais potential antimicrobial, antioxidant and anti-inflammatory compounds, which expose the medicinal potential of the selected plant could be a significant drug candidates against microbial, oxidative and inflammation-related pathological processes as a future alternative medicine.

EVALUATION OF IN VITRO ANTIDIABETIC AND ANTI-INFLAMMATORY ACTIVITIES OF LEAVES EXTRACT OF BOEHMERIA RUGULOSA

Objective: The objective of the study is to evaluate in vitro antidiabetic and anti-inflammatory activity of different extracts of leaves of Boehmeria rugulosa by different methods.Methods: In vitro α-glucose and α-amylase were used for antidiabetic activity and lipoxygenase, and protein denaturation method of inhibition assays was used to measure anti-inflammatory activity. Successive extraction of leaves petroleum ether (PE), chloroform (CH), ethyl acetate (EA), acetone (AC), and ethanol (ETH) was performed, and extracts obtained from the extraction were applicable to these activities.Results: The AC extract of leaves shows significantly in vitro antidiabetic activity, and AC has offered significant result 470.07±0.65 μg/mL in the inhibition of α-glucosidase and also for α-amylase assay 698.15±1.71 μg/mL. Acarbose was used as standard. In lipoxidase method, AC had shown better results and in protein denaturation method EA shown the higher inhibition (78.06±0.5 μg/ml) than the other extracts. The standard drug diclofenac sodium also offered significant inhibition against lipoxidase enzyme method with IC50 value 21.76±1.29 μg/mL.Conclusion: These findings suggest that the AC and EA possess potent antidiabetic and anti-inflammatory activities in vitro conditions.

Phytochemical Composition, Antioxidant, Antimicrobial and in Vivo Anti-inflammatory Activity of Traditionally Used Romanian Ajuga laxmannii (Murray) Benth. ("Nobleman's Beard" - Barba Împăratului).

In the Romanian folk medicine, aerial parts of Ajuga laxmannii (“nobleman’s beard,” Romanian – “barba boierului” or “avrămească” or “creştinească”) are traditionally used as galactagogue and anti-inflammatory agents. The present study aimed to evaluate the chemical composition (polyphenols, iridoids, and phytosterols), antioxidant, antimicrobial and in vivo anti-inflammatory activity of different extracts of A. laxmannii aerial parts. The major identified bioactive compounds were rutin, 8-O-acetylharpagide and β-sitosterol. The antioxidant activity of A. laxmannii extracts was evaluated using several methods, and the results showed good antiradical effects. Moreover, the antimicrobial evaluation showed a potent antifungal activity against C. albicans and P. funiculosum. Furthermore, the anti-inflammatory effect was determined by monitoring some parameters involved in the inflammatory process. The results obtained showed differences between the analyzed extracts; and therefore the importance of choosing the best solvent in order to extract the appropriate amount of bioactive compounds. A. laxmannii ethanol extract showed an anti-inflammatory effect by reducing total leukocytes, PMN, phagocytosis, and oxidative stress. Compared to diclofenac, only the 50 mg/mL A. laxmannii extract had better anti-inflammatory and anti-oxidative stress effects, and this could justify the importance of a correlation between the activity and the used concentration. These findings strongly suggest that A. laxmannii could be considered as a valuable source of bioactive compounds, which could be further valued as anti-inflammatory agents in the composition of several herbal drugs.

Evaluation of Anti-inflammatory Activity of Different Extracts of Alpinia galanga

Inflammation is the local response of living mammalian tissues to injury due to any agent. It is the body defense reaction in order to eliminate or limit the spread of injurious agent, followed by removal of the necrosed cells and tissues, but the sign and symptoms which are produced during healing process cannot tolerable. The present study was aimed to investigate the anti-inflammatory activity of different extracts of Alpinia galanga in wistar albino rats. In this study, both acute and sub-acute inflammation models were used to evaluate the anti-inflammatory activity of Alpinia galanga. In acute model, carrageenan was used to induce inflammation in rat hind paw and in subacute inflammation, cotton pellets induced granuloma was performed. The extracts have significantly (p = 0.05) decrease in paw edema volume and wet weight and dry weight of Cotton pellets as compared to the untreated vehicle control group.

Comparative evaluation of different extraction methods for antioxidant and anti-inflammatory properties from Osbeckia parvifolia Arn. – An in vitro approach

The effect of maceration, Soxhlet and fractionation extraction from whole plant of Osbeckia parvifolia was studied for free radical scavenging and anti-inflammatory activities in vitro. The extracts were quantitatively analyzed for total phenolic, tannin and flavonoid contents using spectrophotometric methods. In vitro free radical scavenging activity of extracts were studied for DPPH (1,1-diphenyl-2-picryl-hydrazyl), ABTS+ (2,2′-azinobis (3-ethyl-benzothiozoline)-6-sulfonic acid) scavenging activities, metal ion chelating capability, lipid peroxidation, phosphomolybdenum and FRAP (Ferric reducing/antioxidant power) assays. Protein denaturation and membrane stabilization assays were employed to assess the anti-inflammatory activity of different extracts of O. parvifolia. Quantitative analysis showed that whole plant has high contents of total phenolic, tannin and flavonoid. Antioxidant assessment results registered higher anti-radical property for both macerated and Soxhlet methanol extracts compared to other solvent extracts. Successively extracted methanol extract from Soxhlet apparatus protected protein denaturation and erythrocyte membrane lysis comparable to standard Diclofenac sodium. Whole plant served as a potential source of antioxidant from natural origin and this study also provides a better technique to extract the natural antioxidant and anti-inflammatory substances from O. parvifolia.

Screening of tropical medicinal plants for antiinflammatory activity

AbstractThe antiinflammatory activity of different extracts from 16 tropical plant species belonging to 15 different families has been determined. They were administered topically on TPA‐induced mouse ear oedema and orally on carrageenan‐induced mouse paw oedema. In the latter, extracts from Diospyros leucomelas, Erythrospermum monticolumn, Ficus polita, Heteromorpha trifoliata, Hypericum lalandii, Kirkia acuminata, Pterocarpus angolensis and Zanha africana significantly reduced the oedema 3 h after carrageenan injection. All the extracts were considerably active when assayed topically with only three exceptions.
