Genetic insights of antibiotic resistance, pathogenicity (virulence) and phylogenetic relationship of Escherichia coli strains isolated from livestock, poultry and their handlers - a one health snapshot.

Pathogenic and non-pathogenic strains of Escherichia coli harbouring antibiotic resistance genes (ARGs) from any source (clinical samples, animal settings, or environment) might be transmitted and contribute to the spread and increase of antibiotic resistance in the biosphere. The goal of this study was to investigate the genome to decipher the repertoire of ARGs, virulence genes carried by E. coli strains isolated from livestock, poultry, and their handlers (humans), and then unveil the genetic relatedness between the strains. Whole genome sequencing was done to investigate the genetic makeup of E. coli isolates (n = 20) [swine (n = 2), cattle (n = 2), sheep (n = 4), poultry (n = 7), and animal handlers (n = 5)] from southern India. The detection of resistome, virulome, biofilm forming genes, mobile genetic elements (MGE), followed by multilocus sequence typing (MLST)and phylogenetic analyses, were performed. E. coli strains were found to be multi drug resistant, with a resistome encompassing > 20 ARGs, the virulome-17-22 genes, and > 20 key biofilm genes. MGE analysis showed four E. coli isolates (host: poultry, swine and cattle) harbouring composite transposons with ARGs/virulence genes (blaTEM, dfr, qnr/nleB, tir, eae,and esp) with the potential for horizontal transfer. MLST analyses revealed the presence of ST937 and ST3107 in both livestock/poultry and their handlers. Phylogenomic analyses with global E. coli isolates (human/livestock/poultry hosts) showed close relatedness with strains originating from different parts of the world (the United States, China, etc.). The current study emphasizes the circulation of strains of pathogenic sequence types of clinical importance, carrying a diverse repertoire of genes associated with antibiotic resistance, biofilm formation and virulence properties in animal settings, necessitating immediate mitigation measures to reduce the risk of spread across the biosphere.

In Silico Safety Assessment of Bacillus Isolated from Polish Bee Pollen and Bee Bread as Novel Probiotic Candidates.

Bacillus species isolated from Polish bee pollen (BP) and bee bread (BB) were characterized for in silico probiotic and safety attributes. A probiogenomics approach was used, and in-depth genomic analysis was performed using a wide array of bioinformatics tools to investigate the presence of virulence and antibiotic resistance properties, mobile genetic elements, and secondary metabolites. Functional annotation and Carbohydrate-Active enZYmes (CAZYme) profiling revealed the presence of genes and a repertoire of probiotics properties promoting enzymes. The isolates BB10.1, BP20.15 (isolated from bee bread), and PY2.3 (isolated from bee pollen) genome mining revealed the presence of several genes encoding acid, heat, cold, and other stress tolerance mechanisms, adhesion proteins required to survive and colonize harsh gastrointestinal environments, enzymes involved in the metabolism of dietary molecules, antioxidant activity, and genes associated with the synthesis of vitamins. In addition, genes responsible for the production of biogenic amines (BAs) and D-/L-lactate, hemolytic activity, and other toxic compounds were also analyzed. Pan-genome analyses were performed with 180 Bacillus subtilis and 204 Bacillus velezensis genomes to mine for any novel genes present in the genomes of our isolates. Moreover, all three isolates also consisted of gene clusters encoding secondary metabolites.

Probiotic Properties and Anti-inflammatory Activity of Bacillus spp. Isolated from Ethnically Fermented Soybean

Bacillus spp. probiotics used as feed additives can form spores and tolerate the harsh conditions of the human digestive system and are beneficial for the treatment of inflammatory bowel syndrome. Since reports on probiotics and anti-inflammatory properties of Bacillus spp. isolated from the fermented food of Northeast India have not been explored much. The present study focused on Bacillus spp. BN5, AY5, and AN8, possessing these desired properties. In the probiotics study, the isolates were screened for their tolerance to acid and bile salt, auto-aggregation, hydrophobicity, cholesterol assimilation, antibiotic resistance, and antagonistic properties. It was found that these isolates possessed the desirable probiotic traits. The Bacillus spp. culture and their supernatant were also screened for their ability to reduce LPS-induced inflammation in murine macrophage (RAW 264.7) cells. All the Bacillus spp. culture and their supernatant treatments were found to reduced the Nitric oxide (NO) production by LPS-induced cell lines. The supernatant of LPS-induced cell lines were also analyzed to measure the level of inflammatory cytokine production. It was found that the levels of TNF-α, IL-6, and IL-1β were reduced after co-treatment with LPS and Bacillus spp. culture or LPS and Bacillus spp. supernatant. Results suggested that the Bacillus spp. are potential probiotic candidates with anti-inflammatory properties.

Characterization and efficiency of Ganoderma lucidum biomass as an antimicrobial and anticancer agent

The microconstituents of Ganoderma lucidum biomass (GLB) were evaluated, along with its antimicrobial and anticancer activities. Using gas chromatography-mass spectrometry analysis, 12-octadecadienoic acid (Z,Z)- and n-hexadecanoic acid with area% values of 21.0% and 11.0% were recognized in GLB. Uncooked biomass (UCB) and microwave-cooked (CE) biomass of G. lucidum caused a significant inhibition of human breast cancer (MCF-7) cell line proliferation in a dose-dependent manner. The inhibition of MCF-7 cell proliferation was 27.22 ± 1.64% using 16 µg/mL of CB while it was 52.29 ± 1.09% using 16 µg/mL of UCB. The cytotoxicity test recorded low IC50 (25.63 ± 0.52 µg/mL) of UCE compared to the IC50 value (49.99 ± 0.94 µg/mL) of CB. Highest antimicrobial activities were recorded via using UCE, compared to CE against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Candida albicans, and Aspergillus niger. The 9,12-octadecadienoic acid (Z,Z)- and n-hexadecanoic acid were able to induce both anticancer and antibiotic-resistant properties. A key therapeutic target enzyme for evolving this resistant pathogenic activity on MCF-7 and K. pneumonia, was accessed thoroughly in silico study. The compounds described, therefore, might provide a great potential for the development of new therapeutics such as anticancer and antimicrobial agents.

Drug Resistance, Characterization and Phylogenetic Discrepancy of Salmonella enterica Isolates from Distinct Sources.

Salmonella enterica is one of the foodborne pathogens that can infect humans, spreading from one person to another by contaminated food and water. To identify the pathogenicS. entericafrom the contaminated food product, culture-based and molecular identifications, drug resistance profiling, virulence and genetic traits of the strains have been used. Herein, different animal products was subjected to screen for S. enterica prevalence, pathogenic characterization and compared with clinical Salmonella isolates (human). A total of 173 isolates from animal products and 51 isolates from clinical samples were collected.S. Typhi, S. Agona andS. Ohio were predominant serovars in blood, stool and different animal products. Both, clinical [37% (n = 19/51)] and animal product-associated isolates [21% (n = 37/173)] expressed their highest resistance to nalidixic acid. Thirty-one percentage of (n = 16/51) clinical isolates and 12% (n = 21/173) animal food-associated isolates were resistant to multiple classes of antibiotics. Class 1 integrons encoded byS. Typhi,S. Infantis andS. Emek were screened for sequence analysis, the result revealed that the cassettes encoded-aminoglycoside acetyltransferase and dihydrofolate reductase enzymes. Salmonellapathogenicity island-1 encoded-hilAgene was detected most frequently in all the isolates. PFGE profile revealed the genetic traits of the isolates which were closely linked with antibiotic-resistant properties and virulent characteristics. OnlyS. Enteritidis, collected from different samples had clonal similarities. In summary, drug-resistant pathogenicSalmonellaprevalence was observed in the animal product that could be an important alarm to consumers with the risk of enteric fever and it causes the potential risk to public health.

Antibiotic Resistance/Susceptibility Profiles of Staphylococcus equorum Strains from Cheese, and Genome Analysis for Antibiotic Resistance Genes.

In food, bacteria carrying antibiotic resistance genes could play a prominent role in the spread of resistance. Staphylococcus equorum populations can become large in a number of fermented foods, yet the antibiotic resistance properties of this species have been little studied. In this work, the resistance/susceptibility (R/S) profile of S. equorum strains (n = 30) from cheese to 16 antibiotics was determined by broth microdilution. The minimum inhibitory concentration (MIC) for all antibiotics was low in most strains, although higher MICs compatible with acquired genes were also noted. Genome analysis of 13 strains showed the S. equorum resistome to be composed of intrinsic mechanisms, acquired mutations, and acquired genes. As such, a plasmidic cat gene providing resistance to chloramphenicol was found in one strain; this was able to provide resistance to Staphylococcus aureus after electroporation. An msr(A) polymorphic gene was identified in five strains. The Mrs(A) variants were associated with variable resistance to erythromycin. However, the genetic data did not always correlate with the phenotype. As such, all strains harbored a polymorphic fosB/fosD gene, although only one acquired copy was associated with strong resistance to fosfomycin. Similarly, a plasmid-associated blaR1-blaZI operon encoding a penicillinase system was identified in five ampicillin- and penicillin G-susceptible strains. Identified genes not associated with phenotypic resistance further included mph(C) in two strains and norA in all strains. The antibiotic R/S status and gene content of S. equorum strains intended to be employed in food systems should be carefully determined.

Characterization of Staphylococcus aureus and methicillin‐resistant S. aureus from yak

AbstractStaphylococcus aureus, particularly methicillin‐resistant S. aureus (MRSA), is an important zoonotic pathogen which not only causes significant economic loss in livestock production but also emerges as a potential threat to public health. Compared with bovines, the information on the colonization of S. aureus and MRSA in yaks, the only bovine species providing meat and other necessities for Tibetans living at high altitude, is very limited. Therefore, in the current study, S. aureus and MRSA prevalence, virulence genes, and antibiotic and antiseptic resistance properties in yaks from 18 counties of Ganzi Prefecture, Sichuan province, China were investigated. The results revealed that the overall prevalence of S. aureus during yak breeding was 7.10% among 1281 samples (91/1281), containing 9.44% from nasal (62/657) and 4.65% from anus (29/624). Nearly all of the S. aureus isolates carried at least one staphylococcal enterotoxin (SE) gene and the predominant enterotoxin gene was selx (51.65%). In addition, virulence genes (hlα, hlβ, pvl, and tsst‐1), antiseptic resistance genes (qacA/B, qacC, and qacG), and heavy metal resistance genes (copA, arsB, cadD, and arsA) were also found. Furthermore, 24 MRSA strains were identified. SCCmec, spa, and multilocus sequence typing typing revealed that the predominant types of MRSA were ST59‐IVa‐t437 and ST59‐III‐t437. It was noteworthy that all these methicillin‐resistant strains were resistant to two or more antibiotics and possessed virulence genes. The minimal inhibitory concentration (MIC) values of antiseptic peroxyacetic acid, sodium hypochlorite, and benzalkonium bromide against these MRSA strains were measured and the MIC values of benzalkonium bromide (1.22–9.77 μg/mL) were relatively low. Furthermore, the challenge with 1/4 MIC of oxacillin sodium or ofloxacin or benzalkonium bromide for 216 h increased the resistance of the MRSA strain YB786 to these antibiotics but decreased the MIC value of benzalkonium bromide. The results provide an indication of the health risks associated with these S. aureus strains, especially methicillin‐resistant strains from yaks and new approaches are needed to treat these strains.

Etiology of bacterial community-acquired pneumonia among hospitalized children in Hai Phong Children’s Hospital

The objectives of this study were to determine the etiology of bacterial community-acquired pneumonia (CAP) in children hospitalized at Hai Phong Children’s Hospital (HCH) in 2019 and these bacteria’s antibiotic resistance properties. The subjects were the qualified medical records of all inpatients at the Respiratory Department of Hai Phong Children’s Hospital aging from 1 month to 15 years old diagnosed with CAP according to the diagnostic criteria of the World Health Organization with positive nasopharyngeal swab culture results from January 1st, 2019, to December 31st, 2019. The sample size included all medical records of patients who met the inclusion criteria of this research during the study period. The success rate of nasopharyngeal swab culture test of CAP patients was 638/1605 (39.7%). The most common pathogens of CAP in children at HCH were S. aureus, S. pneumoniae, H. influenzae and M. catarrhalis. Antibiogram revealed that each of them showed high resistance to a various type of antibiotics. Our study also provided valuable data on the antibiotic sensitivity and raised alarm over presence of multidrug-resistant bacteria inducing CAP in children.

Identification of prevalence and antibiotic resistance property as a basis for establishing an efficient treatment of bacteria causing mastitis in beef cows

The purpose of the study was to examine the occurrence of metritis on reproduction of beef cows. A total of 2,962 cows were examined, and samples were collected from those displaying clinical symptoms of bovine metritis for bacterial analysis. Bacterial species identification was performed using culture and PCR techniques. The findings revealed that bovine metritis affected 5.5% of the reproductive cow population with the presence of Streptococcus spp., Staphylococcus aureus, Escherichia coli, and Salmonella spp. at the rates of 87.7%, 61.1%, 59.9% and 17.3%, respectively. The results also demonstrated that Streptococcus spp. was the most prevalent group, while Salmonella spp. had the lowest prevalence across different breeds and litters. However, in the Limousin crossbred cows, the infection rate of E. coli surpassed that of the other bacteria and a significantly higher infection rate for E. coli was observed in litter 4 and litter ≥6 compared to the others. The disk diffusion method was utilized to assess antibiotic resistance patterns of the isolated bacteria. Among the bacteria, marbofloxacin exhibited the lowest prevalence of antibiotic resistance (16.9%), while doxycycline had the highest prevalence (82.2%). All cows recovered from the disease within 3-5 days of treatment, and all of them resumed estrus in the subsequent reproductive cycle. The pregnancy rate ranged from 66.7% to 80.0% for the first insemination in the next reproductive cycle.

Antibacterial Activity of Ulva lactuca against Multidrug Resistant and Betalactamase Producing Isolates from Food Samples- An In-vitro Study

Introduction: Antibiotic resistance is a problem of deep scientific concern, both in hospital and community settings. Due the presence of highly resistant microorganisms in food, it may lead to serious health issues to the human beings and other animals also. This can be controlled by novel control methods using natural products. Algae are one such wonderful source that helps in treating such dreadful microorganisms. Ulva lactuca incorporate discrete active compounds, hence, exhibit antibacterial property to control the multidrug resistant bacteria. The present research aims in exploring the potential antibacterial property of such dreadful bacteria. Aim: To evaluate the potential bioactive compound of Ulva lactuca and its antibacterial activity against Multidrug Resistant (MDR) and betalactamase producing food isolates. Materials and Methods: The present pilot in-vitro study was carried out at Arignar Anna Government Arts College, Namakkal, Tamil Nadu, India, in the period of December 2017 to December 2020. Total 18 bacterial isolates were isolated from five retail chicken meat samples. These were then, tested for its antibiotic resistance property using standard antibiotic discs. The algae Ulva lactuca was isolated and extract was prepared using ethanol and chloroform solvents, followed by which the phytochemical studies were performed. These extracts were then tested, against the selected organisms for its potential activity. Results: Among the 12 antibiotics tested, all isolates were resistance to variety of antibiotic classes, mainly aminoglycosides, cephalosporins, and fluoroquinolones and also 55% of the bacteria were able to produce betalactamase enzyme. The ethanol extract of Ulva lactuca was highly active against all isolates and exhibited a range of 10±1.24 mm to 22±1.24 mm inhibition zone. The chloroform extract, exhibited less potency, which exhibited 10±0.816 mm to 13.16±1.027 mm inhibition zone. Each organic solvent showed positive result for following metabolites- alkaloids, carbohydrates, flavonoids, sterols, tannins and terpenoids. Conclusion: The ethanolic extract of Ulva lactuca was very effective against the selected MDR and betalactamase producing food isolates. Therefore, it could be suggested as an antibacterial agent in the future. Further studies are needed to reveal the behavioural mechanisms of this plant and its pharmacological effects.

A comparative study on antibiotic resistance and virulence properties of Staphylococcus aureus isolated from hospitalized patients and hospital environment

We compared the characteristics of clinical Staphylococcus aureus and S. aureus isolated from environmental surfaces in 3 hospitals. Clinical S. aureus isolates were collected from hospitalized patients. Environmental surfaces were sampled from the rooms of patients infected with S. aureus. After identifying rooms with the target organism, 3-5 high-touch surfaces in patient care areas were sampled using swabs before room cleaning by environmental services. S. aureus isolates were subjected to genotyping, antimicrobial susceptibility testing, and virulence determinant screening. The isolates were analyzed for integron content and sequences of variable region amplification products. There were epidemiologically unrelated 79 clinical and 62 environmental S. aureus isolates. Overall, 11.4% of clinical and 59.7% of environmental isolates were methicillin-resistant. The environmental and clinical S. aureus exhibited very different virulence profiles: 79% of the environmental isolates were negative for virulence genes compared to 2.5% of clinical isolates (P < .001). Environmental isolates were more resistant to antibiotics compared to clinical isolates. Class 1 integrons were only detected in 7 of 62 environmental isolates, of which 3 isolates had integrons with cysteine synthase cassette, 1 had aadA1, and 1 had an unknown cassette. These data indicate the different characteristics between environmental and clinical S. aureus, which may reflect different reservoirs from which the 2 groups acquired the strains.

Antioxidant, Antimicrobial Properties and In Silico Study of a N,N'-(ethane-1,2-diyl)bis(1-(9H-fluoren-2-yl)methanimine)

ABSTRACT This study aimed to determine the antibacterial and antioxidant activities of the newly synthesized Schiff base and to support the laboratory results with molecular modeling studies. Antibacterial activity of schiff bases was demonstrated using Gram (-) Pseudomonas aeruginosa and Acinetobacter baumannii bacterial strains. Minimum Inhibition Concentration (MIC) values were determined to evaluate their antimicrobial activity against Gram-negative P. aeruginosa and A.baumanni bacterial strains. In antioxidant experiments, the responses to DPPH and ABTS radicals were calculated at certain concentration ranges and graphs were drawn. For the molecular modeling study, Autodock Vina and Discovery Studio 2020 package programs were used. The observed bacterial inhibition activity was variable depending on the clinical isolate and the concentration of the tested samples. The highest inhibition activity was achieved at concentration of 75 μl -100 μl. N,N'-(ethane-1,2-diyl)bis(1-(9H-fluoren-2-yl)methanimine) samples. Molecular docking results show that N,N'-(ethane-1,2-diyl)bis(1(9H-fluoren-2-yl) methanimine) binds strongly to the 4ZIY and 4ZHU structures. It has been proven by molecular docking study that the synthesized Schiff base ligand has antibiotic resistance properties. N, N'-(ethane-1,2-diyl)bis(1-(9Hfluoren-2-yl) methanimine) synthesis compound showed moderate activity against A. baumannii and P. aeruginosa strains. It is known that schiff bases have strong biological activities and show antibacterial activity. In this study, it can be said that the synthesis schiff base showed antibacterial and antioxidant activity. In addition, our results were supported by molecular modeling. Our findings can be taken to a higher level with in vivo and in vitro studies.

Assessment of virulence factors and antimicrobial resistance among the Pseudomonas aeruginosa strains isolated from animal meat and carcass samples.

Pseudomonas aeruginosa bacteria are emerging causes of food spoilage and foodborne diseases. Raw meat of animal species may consider a reservoir of P. aeruginosa strains. The present survey was done to assess the prevalence, antibiotic resistance properties and distribution of virulence factors among the P. aeruginosa strains isolated from raw meat and carcass surface swab samples of animal species. Five hundred and fifty raw meat and carcass surface swab samples were collected from cattle and sheep species referred to as slaughterhouses. P. aeruginosa bacteria were identified using culture and biochemical tests. The pattern of antibiotic resistance was determined by disk diffusion. The distribution of virulence and antibiotic resistance genes was determined using polymerase chain reaction. Forty-seven of 550 (8.54%) examined samples were contaminated with P. aeruginosa. The prevalence of P. aeruginosa in raw meat and carcass surface swab samples were 6.57 and 12%, respectively. P. aeruginosa isolates showed the maximum resistance rate toward penicillin (87.23%), ampicillin (85.10%), tetracycline (85.10%), gentamicin (65.95%) and trimethoprim (57.44%). The most commonly detected antibiotic resistance genes were BlaCTX-M (53.19%), blaDHA (42.55%) and blaTEM (27.65%). The most commonly detected virulence factors was ExoS (42.55%), algD (31.91%), lasA (31.91%), plcH (31.91%) and exoU (25.53%). Meat and carcass surface swab samples may be sources of resistant and virulent P. aeruginosa, which pose a hygienic threat in their consumption. However, further investigations are required to identify additional epidemiological features of P. aeruginosa in meat and carcass surface samples.

Characterization of lactic acid bacteria isolated from the poultry intestinal environment with anti-Salmonella activity in vitro.

The purpose of this research was the genotypic identification of lactic acid bacteria (LAB), isolated from the gastrointestinal tract (GIT) of healthy adult birds, and the study of their safety regarding antibiotic resistance, physiological and functional properties involved in the colonization of the GIT of poultry, and Salmonella exclusion, as members of a potential mixed probiotic supplement for poultry. The nucleotidic sequence from Lactobacillus crispatus P1, L. animalis L3, and Enterococcus faecium CRL 1385 (ex-J96) showed 100, 99.8, and 99.3% identity with L. crispatus DSM 20584T, Ligilactobacillus salivarius ATCC 11741T, and E. faecium ATCC 19434T, respectively. These strains showed no resistance to relevant antibiotics usually administered to animals proposed by the European Food Safety Authority. They could endure the detrimental conditions of the gastrointestinal tract (pH 2.6 and oxgall 0.1 and 0.4% w/v). In an ex vivo assay, the LAB showed high adherence to the three sections of the GIT, reaching values higher than 70%. The adhesion to mucus was strain-dependent: L. crispatus CRL 1453 evidenced the highest adhesion (> 19%) while Lig. salivarius subsp. salivarius CRL 1417 and E. faecium CRL 1385 adhered to a lower extent (> 9 and 2%, respectively). Moreover, the LAB elicited remarkable anti-Salmonella activity, taking into account that they could inhibit elevated counts of different Salmonella serovars, especially the host-specific serovars S. Gallinarum and S. Pullorum (up to 8 log CFU/mL decrease in Salmonella counts).

Prevalence, antimicrobial resistance, virulence gene profile and molecular typing of Campylobacter species isolated from poultry meat samples.

Campylobacter jejuni and Campylobacter coli are the most significant Campylobacter species responsible for severe gastrointestinal disorders. Raw poultry meat is considered a source of Campylobacter transmission to the human population. The present study was aimed to assess the prevalence rate, antibiotic resistance properties, virulence characters and molecular typing of C. jejuni and C. coli strains isolated from raw poultry meat samples. Three hundred and eighty raw poultry meat samples were collected and analysed for the presence of Campylobacter spp. using the microbial culture. Species identification was done using the Polymerase Chain Reaction. Disk diffusion was developed to assess the antimicrobial resistance pattern of isolates. The distribution of virulence and antimicrobial resistance genes was determined by PCR. Enterobacterial Repetitive Intergenic Consensus-PCR was used for molecular typing. Campylobacter species were isolated from 6.25% of examined samples. C. jejuni and C. coli contamination rates were found to be 57.44% and 48.14%, respectively. C. jejuni strains harboured the highest resistance rate against serythromycin (42.59%), ampicillin (38.88%), ciprofloxacin (33.33%), chloramphenicol (31.48%) and tetracycline (31.48%). C. coli isolates harboured the highest resistance rate against ampicillin (73.07%), ciprofloxacin (73.07%), erythromycin (65.38%) and chloramphenicol (50%). AadE1 (44.44%), blaOXA-61 (42.59%) and tet(O) (35.18%) were the most commonly detected resistance genes in C. jejuni and cmeB (34.61%) and blaOXA-61 (34.61%) were the most commonly detected among C. coli strains. The most frequent virulence factors among the C. jejuni isolates were flaA (100%), ciaB (100%), racR (83.33%), dnaJ (81.48%), cdtB (81.48%), cdtC (79.62%) and cadF (74.07%). The most frequent virulence factors among the C. coli isolates were flaA (100%), ciaB (100%), pldA (65.38%) and cadF (61.53%). The majority of C. jejuni and C. coli strains had more than 80% similarities in their ERIC-PCR pattern, which may show their common source of transmission. The role of goose and quebec meat samples as reservoirs of virulent and antimicrobial resistant Campylobacter spp. was determined.

Prevalence, Antimicrobial Resistance, and Molecular Description of Pseudomonas aeruginosa Isolated from Meat and Meat Products

Resistant and virulent Pseudomonas aeruginosa (P. aeruginosa) bacteria are measured as the major cause of food spoilage and food-borne diseases. This survey assesses the prevalence, antibiotic resistance properties, and virulence factors distribution in P. aeruginosa bacteria isolated from meat and meat products. A total of 370 raw, frozen, and imported bovine meat samples and diverse types of meat product samples were collected from Alborz province, Iran. P. aeruginosa bacteria were identified by culture. Disk diffusion was used to assess the antibiotic resistance of bacteria. Furthermore, the PCR was used to assess the virulence and antibiotic resistance genes. Twenty nine out of 370 (7.83%) samples were contaminated with P. aeruginosa. Imported frozen bovine meat (20%) harbored the highest distribution, while sausage (2%) harbored the lowest. High resistance rates were observed toward ampicillin (89.65%), penicillin (86.20%), tetracycline (82.75%), cefoxitin (37.93%), gentamicin (34.48%), and clindamycin (31.03%). The most commonly detected antibiotic resistance genes were blaDHA (93.10%), blaCTX-M (83.65%), and blaSHV (48.27%). BlaDHA (93.10%), blaCTX-M (83.65%), and blaSHV (48.27%) were the most frequently detected resistance genes. The most commonly detected virulence genes were exoS (75.86%), lasA (68.96%), exoU (58.62%), lasB (51.72%), plcH (48.27%), and algD (44.82%). Meat and meat product samples may be sources of P. aeruginosa, which show an important threat to their consumption. Nevertheless, additional inquiries are obligatory to find supplementary epidemiological properties of P. aeruginosa in meat and meat product samples.

Isolation of ferric Yersinia bactin A (fyuA) as virulence gene and biofilm forming in Escherichia coli was Collected from patient with UTI.

The focus of the current investigation was applied on determining the presence of ferric yersiniabactin uptake A (fyuA) virulence gene and identifying the antibiotic resistance properties in biofilm forming Escherichia coli­­ isolated from recurrent urinary tract infection patients and healthy individuals. In the Microbiology laboratory at Al Yarmouk hospital in Baghdad, Iraq, 20 (16 females and 4 males) UTI-diagnosed patient urine and stool samples, as well as 20 (10 females and 10 males) healthy individual stool samples, were collected. The samples were exposure to series test including O and H antigen based identification using an antibody (polyvalent and monovalent)-directed agglutination test, antibiotic sensitivity testing using the Kirby Bauer disc diffusion method, Congo-red agar method-based biofilm formation identification, and virulence gene identification using polymerase chain reaction (PCR).Results: 40 and 20 strains were discovered from UTI patients and healthy persons, respectively, in which O serogroups were found in 7(35%) and 3(15%) of those strains, respectively. Furthermore, only two strains of enterohemorrhagic E. coli (O157 H7) and enteroaggregative E. coli (O44 H7) were found in the UTI-based urine samples. Multidrug resistance (MDR) was detected in 60% of the biofilm positive producers (BPPs) isolated from UTI urine samples; however, only 20% of the biofilm negative producers (BNPs) isolated from UTI urine samples displayed MDR features. In the instance of virulence genes, a link was shown between fyuA (a gene that codes for an outer-membrane protein called ferric yersiniabactin uptake) and biofilm in 75% and 60% of UTI urine and stool strains, respectively, and in 55% of healthy individual stool strains. The findings of the antibiotic sensitivity test revealed that the UTI strains were 90 percent, 100 percent, 65 percent, and 40 percent sensitive to imipenem, amikacin, gentamicin, and tobramycin, respectively. Conclusion:The present research offers detailed information on the biofilm, virulence gene, and antibiotic resistance features of Escherichia coli from recurrent urinary tract infection patients and healthy people.

Antibiotic Resistance Properties among Pseudomonas spp. Associated with Salmon Processing Environments.

Continuous monitoring of antimicrobial resistance in bacteria along the food chain is crucial for the assessment of human health risks. Uncritical use of antibiotics in farming over years can be one of the main reasons for increased antibiotic resistance in bacteria. In this study, we aimed to classify 222 presumptive Pseudomonas isolates originating from a salmon processing environment, and to examine the phenotypic and genotypic antibiotic resistance profiles of these isolates. Of all the analyzed isolates 68% belonged to Pseudomonas, and the most abundant species were Pseudomonas fluorescens, Pseudomonas azotoformans, Pseudomonas gessardii, Pseudomonas libanesis, Pseudomonas lundensis, Pseudomonas cedrina and Pseudomonas extremaustralis based on sequencing of the rpoD gene. As many as 27% of Pseudomonas isolates could not be classified to species level. Phenotypic susceptibility analysis by disc diffusion method revealed a high level of resistance towards the antibiotics ampicillin, amoxicillin, cefotaxime, ceftriaxone, imipenem, and the fish farming relevant antibiotics florfenicol and oxolinic acid among the Pseudomonas isolates. Whole genome sequencing and subsequent analysis of AMR determinants by ResFinder and CARD revealed that no isolates harbored any acquired resistance determinants, but all isolates carried variants of genes known from P. aeruginosa to be involved in multidrug efflux pump systems.

Molecular docking analysis of long-chain alkanes with the β-lactamase BEL-1 from P. aeruginosa.

Pseudomonas aeruginosa is a gram-negative opportunistic bacterium that is a concern worldwide due to its innate antibiotic resistance properties. This warrants the development of non-antibiotic compounds that can potentially address the growing concern. Therefore, it is of interest to document the molecular docking analysis data of three long-chain alkanes, namely, eicosane, triacontane, and nonadecane with β-lactamase BEL-1. Data shows that nonadecane have good binding features and drug-likeness when compared to triacontane and nonadecane. It is well known that nonadecane is a compound that is abundantly available from natural resources for further consideration.
