Evaluation of antioxidant and anti-inflammatory activity and identification of bioactive compound from the marine diatom, Odontella aurita extract

Increased production of reactive oxygen species (ROS) leads to oxidative stress, with its damaging effect extending to the mitochondria and plasma membrane. Further, prolonged inflammation can result in chronic disease development. The marine microdiatom Odontella aurita is recognized for its potential in food and pharmaceutical development. Moreover, it contains antioxidant and anti-inflammatory properties. However, studies regarding the efficacy of their varying extract forms and their underlying mechanisms remain scarce. Therefore, this study aims to explore the antioxidant and anti-inflammatory effects of Odontella aurita extracts obtained using various extraction methods (hot water, 70% ethanol, and chloroform:methanol (CM)). Among the three Odontella aurita extracts, the CM extract demonstrated superior efficacy in protecting RAW 264.7 cells from H2O2-induced cytotoxicity. It significantly lowered the levels of ROS and enhanced the expression of superoxide dismutase and glutathione peroxidase. Furthermore, the CM extract outperformed other extracts in inhibiting LPS-induced nitric oxide production, reducing mRNA levels in nitric oxide synthase, cyclooxygenase, and the proinflammatory cytokines interleukin IL-1β, IL-6, TNFα. Additionally, CM extract effectively suppressed the activation of NF-κB/IκBα and JAK2-STAT3 in LPS-induced RAW 264.7 cells. HPLC–UV analysis revealed a remarkable 33-fold higher fucoxanthin content in CM compared to the ethanol extract. GC–MS analysis identified elevated levels of cholest-5-en-3-ol, phytol, eicosapentaenoic acid methyl ester, methyl palmitate, palmitoleic acid methyl ester, and neophytadiene in the CM extract. These findings suggest that Odontella aurita CM extract is a promising antioxidant candidate for preventing or treating inflammatory diseases, consequently emphasizing its potential for further development.Graphical

Evaluation of Anti-Inflammatory Activity in Methanolic Seed Extracts of International Sorghum bicolor L. Resources

Sorghum is an important cereal with high value as a health food ingredient because it contains various phenolic compounds. Anti-inflammatory activity was assessed using 12 sorghum resources collected from various countries to explore their potential as medicinal resources. The findings revealed that, at extract concentrations of 25 µg/mL and 50 µg/mL, cell survival rates were observed to be between 70 and 80% for most varieties, with the exception of K159081. In the analysis of anti-inflammatory activity, measured by the rate of nitric oxide (NO) production, sorghum varieties K159041 and K159081 exhibited NO production rates of 0.46 ± 0.38% and 2.58 ± 0.20%, respectively, indicating significant anti-inflammatory properties. The investigation into anti-inflammatory effects also included examining the expression of the inducible nitric oxide synthase (iNOS) gene, which is related to the inflammatory response triggered by LPS in macrophages. Varieties K159041, K159048, K159077, K159078, K159081, K159089, and K159096 were analyzed for this purpose. Further, an expression test of the cyclooxygenase 2 (COX-2) gene revealed values less than 0.4 in K159077, K159081, and K159089, suggesting these sorghum lines possess higher anti-inflammatory activity compared to others. Additionally, the expression analysis of tumor necrosis factor alpha (TNF-α), a gene identified as an inflammatory cytokine, showed that the mRNA levels in the lines K159048, K159077, K159078, K159088, K159089, K159093, and K159096 were expressed at lower levels relative to other sorghum resources, categorizing them as having high anti-inflammatory activity. Notably, the K159081 line exhibited the lowest expression level of all genes associated with inflammation, marking it as a valuable medicinal resource with potential development as an anti-inflammatory agent.

Preparation of Chitosan Nanoparticle from Crab Shells and Evaluation of ‎Analgesic and Anti-inflammatory Activities of Chitosan-loaded ‎Haematostaphisbarteri Hook. f. Stembark Extract

Haematostaphis barteri Hook. f. is a tree belonging to the Family Anacardiaceae whose morphological parts have been used for decades for treating various diseases in Northern Nigeria. This study aimed to prepare chitosan nanoparticles from crab shells and evaluate the analgesic and anti-inflammatory activities of chitosan loaded with stembark extract from H. barteri. Phytochemical screening of extract, preparation of chitosan, and evaluation of analgesic and anti-inflammatory activities were carried out following standard procedures. The qualitative phytochemical analysis of the extract revealed the presence of alkaloids, saponins, tannins, flavonoids, and cardiac glycosides while steroids, triterpenes and anthracenes were absent. The LD50 of the crude methanol extract in Wistar albino rats was determined to be greater than 5000 mg/kg body weight and after 14 days of single dose oral administration. No signs of toxicity were recorded. Prepared chitosan nanoparticle loaded with extract has a yield of 26.54 %, particle size of 210 ± 1.01 nm, zeta potential of 25.4 ± 1.02 V, drug entrapment efficiency of 68.4 %, cumulative drug release of 88.24 % and swelling index of 58.14 %. The extract produced a dose-dependent analgesic and anti-inflammatory activity. The chitosan-loaded extract produced the highest inhibition of writhing and paw oedema diameter in the rats, which was significant (p < 0.05) when compared with the normal controls and standards. In conclusion, the study showed that chitosan-loaded stembark extract from H. barteri possessed more potential analgesic and anti-inflammatory activities than those delivered without chitosan nanoparticles. The study further affirms the potential usefulness use of the stembark extract for the treatment of pain and inflammation.

Phytochemical analysis and biological investigation of Cheilanthes tenuifolia (Burm.f.) Swartz.

Introduction: Cheilanthes tenuifolia is an evergreen ornamental small fern, belonging to the family Pteridaceae, that grows in warm and rocky regions worldwide. Many species of Cheilanthes genus are evidently endowed with important phytochemicals and bioactivities. This study aimed to perform a preliminary phytochemical analysis of Cheilanthes tenuifolia leaves alongside an evaluation of free radical scavenging, anti-inflammatory, antimicrobial, and clot lysis activities of extract fractions. Materials and methods: A preliminary phytochemical analysis was done after fractionation of ethanolic extract (ECT) with n-hexane (HCT) and chloroform (CCT). Then, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, egg albumin and RBC membrane stabilization tests, disc diffusion, and human blood clot lysis assays were performed. Results: Phytochemical investigations suggested that the plant is rich in alkaloids, glycosides, tannins, and flavonoids. All obtained fractions exhibited concentration-dependent radical scavenging, inhibition of egg protein denaturation and RBC membrane lysis capacities. Except for antifungal tests, ECT exhibited better DPPH radical scavenging, anti-inflammatory, antibacterial, and clot lysis capacities than HCT and CCT fractions. However, all fractions exhibited a mild anti-inflammatory activity. Conclusion: C. tenuifolia might be a good source of antioxidant, anti-microbial, and anti-atherothrombotic agents. Further studies are required to isolate and characterize the active principles liable for each bioactivity, along with possible molecular interactions.

Evaluation of In-vivo Anti-inflammatory activity of methyl 2-(5-butyl-6-thioxo-1, 3, 5-thiadiazinan-3yl), butanoate

Inflammation is the body's way of fending off damage. Nonetheless, continuous and increasing inflammation may lead to the development of some chronic illnesses. It is still needed to evaluate new and safe anti-inflammatory therapies because existing medications have major side effects. Objective: To examine a novel compound's anti-inflammatory properties of methyl 2-(5-butyl-6-thioxo-1, 3, 5-thiadiazinan-3yl), butanoate (MBTTB) in albino mice models. Method: To test the anti-inflammatory effects in vivo, carrageenan-induced paw edema in albino mice of both sexes was utilized, and inflammation was measured by using Digital Plethysmometer. After extraction of carrageenan induced paw edematous, tissue level of inflammatory mediators like TNF-α, IL-1β, NF-ĸβ, and cox2 was ascertained by Western Blot analysis. Result: Based on in-vivo anti-inflammatory effects, compound MBTTB was shown to exhibit a maximum percentage inhibition of 16.7% after 1 hour, which is comparable to the standard drugs 12.8%. Following three hours, compound MBTTB showed the highest percentage inhibition of 23.2%, while the standard was 14.5%. Compound MBTTB showed the highest percentage inhibition after five hours (26.2 %), while the standard was 15.4%. The expression of many mediators' proteins, including TNF-α, IL-1β, NF-ĸβ, and COX-2, was ascertained. The computational analysis was carried out to forecast the level of decline of various mediators, including Tumor Necrosis Factor-α, (TNF) Interleukin-1β, NF-ĸβ, and COX-2. Conclusion: When considered as a whole, our current research indicates that MBTTB is a viable option for preventing inflammation in mice that has been caused by carrageenan in edematous paw.

In vivo Anti-Inflammatory and Antinociceptive Activity Evaluation of Brassica Rapa Ssp. Chinensis Ethanolic Extract with In Vitro Thrombolytic and Anthelmintic Activity Test

Objective: Brassica rapa ssp. chinensis is a well-known vegetable plant (Family: Brassicaceae) often grown as a type of Chinese cabbage. The present study was designed to explore the thrombolytic, anthelmintic, anti-inflammatory, and antinociceptive activities of ethanolic extract of the Brassica rapa ssp. chinensis leaves. Methods: The in vitro thrombolytic activity of the leaf extract was evaluated by clot analysis using Streptokinase as a standard drug. The in vitro anthelmintic activity was assessed by applying four different concentrations of the plant extract (10 mg/ml, 20 mg/ml, 40 mg/ml, and 60mg/ml) and recording the time of paralysis and death of worms. Two different doses (100mg/kg and 250mg/kg b.w.) were administered to evaluate the anti-inflammatory activity of the carrageenan-induced paw edema model. The antinociceptive activity was evaluated by hot plate test in mice by two different doses (200mg/kg and 400 mg/kg b.w.). Results: The ethanolic leave extract had thrombolytic action, which broke up blood clots that had already formed. The ethanolic leaf extract showed both paralysis and faster worm death at those concentrations in the anthelmintic test. The extract showed significant anti-inflammatory action in pedal edema compared to the control group. The results show that the highest level of anti-inflammatory action against carrageenan-induced hind paw edema may be achieved with an ethanolic extract of Brassica rapa ssp. chinensis (250 mg/kg, p.o.). The proportion of maximum possible effect in the hot plate test was 82.49 percent in the antinociceptive evaluation. Conclusion: The pharmacological test of leaf extract endorsed the clinical uses of Brassica rapa ssp. chinensis in treating thrombosis, intestinal worms, and inflammation. Thus, this plant could be subjected to extensive bioactive compounds to discover novel therapeutic agents. Evaluation of analgesic and anti-inflammatory activities of ethanolic fruit extract of Terminalia chebula

Analgesic and Anti-inflammatory activities of ethanolic fruit extract of Terminalia chebula was evaluated using tail-flick method, eddy’s hot plate method, acetic acid induced writhing responses and in-vivo anti-inflammatory property was determined using carrageenan rat paw edema. The ethanolic fruit extract of Terminalia chebula at different concentrations significantly inhibited the nociception in comparision with different standard drugs that showed anti-nociception. The test extract also reduced the edema induced in rats using carrageenan paw edema method as compared with the standard drug Diclofenac sodium 10mg/kg. The results obtained in this research study found to be very significant when compared to standard reference drug.

Anti-Inflammatory and Wound Healing Potential of Comphora Wightii, Herbal Ointment on Wistar Rats

Background:: A herbal approach to Guggulu (Commiphora wightii), as an anti-inflammatory and wound healing agent is anticipated. Phyto-constituents Guggulusterone, Naringenin, and myrrhanol were reported for the anti-inflammatory activity of Guggulu. Studies suggests, that sesame oil (Sesamum indicum L.) and Ratanjot (Arnebia nobilis) also act as potent anti-inflammatory agents.. A combination of all three of these gives a synergistic effect for anti-inflammatory and excisional wound healing activity. Materials and Method:: A simple ointment base, BP and 5% Ratanjot, and Sesame oil were prepared. All the components, i.e., Guggulu, 5% Ratanjot Sesame oil, and simple ointment base, were added and triturated uni-directionally until a smooth, homogenous mixture was obtained. 25% w/w (F1) and 27% w/w (F2) Guggulu ointment were prepared. Four groups, each with three wistar rats, were studied for fourteen days. On the fourteenth day, rats were sacrificed, and tissues were collected for histopathological studies. Result:: F1 was compared against the standard formulation (10% w/w betadine, Win-Medicare) for excisional wound healing and anti-inflammatory activity in rats. On the foutheenth day, the results for percentage wound contraction in groups 1 (Negative control, vaseline), 2 (Controlled, ointment base), 3 (F1), and 4 (positive control, standard) were found to be 12.01, 25.32, 96.14, and 81.23, respectively.. Results of histopathological studies and H&E staining supported the action of F1, as skin sections showed the junction of normal skin and wound area. Sub-epithelial tissues showed low (-) to mild (+) inflammation. Mild oedema (+) was also noted. Conclusion:: The rat group treated with F1 showed maximum wound contraction, healing, and antiinflammatory activity as per percentage wound contraction and histopathological studies

Evaluation of Antioxidant, Anti-inflammatory, and Antimicrobial Activities of Raspberry Fruit Extract: An In Vitro Study.

Introduction Rubus idaeus (R. idaeus)is an edible fruit thatcontains numerous significant bioactive compounds that hold important biological properties and are categorized as nutraceuticals owing to the health benefits it imparts including decreasing the risk of chronic diseases like diabetes mellitus, cancer, heart disease, and many other diseases.The objective of the present research was to explorethe antimicrobial, antioxidant, and anti-inflammatory characteristics of the aqueous raspberry extractthrough in vitro assays. Materials and methods R. idaeus aqueous extract was prepared and examined for its antimicrobial activity against Streptococcus mutans (S. mutans) bacteria and Candida albicans (C. albicans) fungi using the agar-well diffusion method, and the antioxidant activity was evaluated using the DPPH (2, 2-Diphenyl-1-picrylhydrazyl hydrate) radical scavenging assay and the hydrogen peroxide radical scavenging assay. The anti-inflammatory activity of the prepared extract was investigated using bovine serum albumin (BSA) and egg albumin denaturation assays. Results R. idaeus fruit extract displayed strong antimicrobial activity at a higher concentration of 100 µL with a 26 mm zone of inhibition against Streptococcus mutans and 24 mm for Candida albicans. The extract showed 87.42% hydrogen peroxide free radical scavenging activity and inhibited 91.12% DPPH free radicals at the highest concentration of 50 µg/mL. The extract showed effective anti-inflammatory activity by preventing the denaturation of bovine serum albumin (80%) and egg albumin proteins (77%) at the highest concentration of 50 μg/mL.The free radical scavenging activity positively correlates with the increased concentration of the prepared extract against DPPH and hydrogen peroxide free radicals, thus showing the raspberry extract's potent antioxidant activity. Similarly, the anti-inflammatory assay result shows that the prepared raspberry aqueous extract has excellent anti-inflammatory activity by preventing the denaturation of bovine serum albumin and egg albumin protein in a dose-dependent manner. Conclusion The meticulously prepared raspberry extract exhibited noteworthy antimicrobial, antioxidant, and anti-inflammatory characteristics, and owing to its astounding therapeutic benefits it holds a tremendous promise as a natural alternative in the field of oral medicine especially in the management of oral mucosal lesions, oral potentially malignant lesions such as lichen planus and leukoplakia, candidiasis, oral cancer and oral mucositis. Further animal studies and clinical trials are recommended to fully reap the therapeutics potential of raspberry extract.

Multi-level chemical characterization and anti-inflammatory activity evaluation of the polysaccharides from Prunella vulgaris

Prunella vulgaris (PV) is a medicine and food homologous plant, but its quality evaluation seldom relies on the polysaccharides (PVPs). In this work, we established the multi-level fingerprinting and in vitro anti-inflammatory evaluation approaches to characterize and compare the polysaccharides of P. vulgaris collected from the major production regions in China. PVPs prepared from 22 batches of samples gave the content variation of 5.76–24.524 mg/g, but displayed high similarity in the molecular weight distribution. Hydrolyzed oligosaccharides with degrees of polymerization 2–14 were characterized with different numbers of pentose and hexose by HILIC-MS. The tested 22 batches of oligosaccharides exhibited visible differences in peak abundance, which failed to corelate to their production regions. All the PVPs contained Gal, Xyl, and Ara, as the main monosaccharides. Eleven batches among the tested PVPs showed the significant inhibitory effects on NO production on LPS-induced RAW264.7 cells at 10 μg/mL, but the exerted efficacy did not exhibit correlation with the production regions. Conclusively, we, for the first time, investigated the chemical features of PVPs at three levels, and assessed the chemical and anti-inflammatory variations among the different regions of P. vulgaris samples.

Design, synthesis and evaluation of antioxidant and anti-inflammatory activities of novel resveratrol derivatives as potential multifunctional drugs

Oxidative stress and inflammation responses are closely related to the occurrence and development of many diseases. Therefore, anti-oxidation and anti-inflammation have become hot spots in the treatment of diseases. A series of novel resveratrol derivatives which hybrid with benzoylhydrazines were designed, synthesized and assessed for their in vitro antioxidant and anti-inflammatory activity. Initially, the antioxidant abilities of resveratrol derivatives were investigated by DPPH, ABTS radical scavenging and FRAP assays. RAW 264.7 macrophages are routinely used to evaluate the antioxidant and anti-inflammatory activities of drugs, so we used it to construct cell models of oxidative stress and inflammation. Among all the derivatives, compound 5 exhibited superior ROS- and NO-inhibitory activities. The molecular mechanism detected by Western blotting showed that compound 5 could significantly activate the Nrf2 signaling pathway and up-regulate the expression of HO-1 to resist oxidative stress stimulated by H2O2. At the same time, it could down-regulate the expression of apoptosis-related proteins Caspase3 and PARP, alleviating cells damage and apoptosis. In addition, compound 5 dose-dependently inhibited the activation of NF-κB p65/iNOS and MAPKs signaling pathway.

Bioactivity assay of Arundina graminifolia (D.Don) Hochr. extracts from diverse plant parts in Thailand: An assay-based investigation

This research involved surveying and collecting 16 samples of Arundina graminifolia from 8 different subpopulation areas. The main objectives were to analyze the preliminary phytochemical fingerprints and biological activity of the crude extracts obtained from various parts of the plant (roots, rhizomes, stems, and leaves). The findings indicated that rhizome extracts generally yielded a higher percentage of crude extract compared to the dry plant weight in comparison to other plant parts. Moreover, the Thin layer chromatography (TLC) fingerprint data revealed distinct compositions for each part of the plant, with rhizomes exhibiting more prevalent bands of compounds than other parts. Regarding antioxidant activity, the root and rhizome crude extracts from various populations displayed stronger inhibitory activity against DPPH, ABTS, and FRAP methods compared to stems and leaves. High total phenolic content was also found to effectively inhibit free radicals. The lipoxygenase inhibition test, which serves as an in vitro anti-inflammatory activity evaluation, demonstrated that most root extracts exhibited higher lipoxygenase inhibitory activity than the rhizome extracts at a concentration of 0.1 mg/mL. The antibacterial activity demonstrated that rhizome extracts from different regions exerted lower MIC and MBC values against all tested bacteria compared to the root extracts. The findings offer valuable information for selecting specific populations of A. graminifolia as potential sources of bioactive compounds for future nutraceutical applications.

Pharmacological evaluation of 2-(2-hydroxy phenylimino)-N-(2-chlorophenyl)-5-isopropyl-4-methylthiophene-3-carboxamide compound for anti-inflammatory activity in rats

Background: Thiophene, also known as thiofuran, is a heterocyclic molecule represented by the chemical formula C4H4S. Aims and Objectives: The aim of this study was to evaluate the anti-inflammatory activity of 2-(2-hydroxy phenylimino)-N-(2-chlorophenyl)-5-isopropyl-4-methylthiophene-3-carboxamide compound in Wistar rats by acute carrageenan-induced paw edema and chronic cotton pellet-induced granuloma methods. Materials and Methods: This study consists of two stages: An acute inflammation study using carrageenan-induced paw edema in rats, which involves four groups of rats, and a chronic inflammation study using cotton pellet-induced granuloma, which involves three groups of rats. Mean paw edema volume and percentage of inhibition were measured in acute stage. The mean weight of the pellets in each group was determined during the chronic stage. Results: There was increase in the mean paw volume at 3 h and a slow decrease at 6 h and 24 h in control group to thiophene derivative-20 mg/kg/p.o from 0 h to 24 h. The mean difference from control and at 3 h, 6 h, and 24 h showed that mean difference was slowly decreased and it was statistically significant. The mean dry granuloma weight in control group, i.e., 10% tween 80 treated group was 146.6 ± 4.38. The mean dry granuloma weight in standard group, i.e., indomethacin 3 mg/kg treated group was 87.7 ± 3.41. The mean dry granuloma weight in substituted thiophene 10 mg/kg group was 101.91 ± 3.76. Conclusions: The newly synthesized substituted (fused) thiophene compound demonstrated inhibition of inflammation in both acute inflammation induced by carrageenan-induced paw edema and chronic inflammation induced by cotton pellet-induced granuloma in Wistar rats. The compound’s anti-inflammatory effects were comparable to those of the standard drug indomethacin, with a dosage of 10 mg/kg in the acute model and 3 mg/kg in the chronic inflammation model. This concludes that the test compound has anti-inflammatory activity.

Evaluation of Analgesic, Anti-inflammatory and Antipyretic activities of the Methanol Extract of Crinum jagus Bulb in Rats

This study evaluated the analgesic, anti-inflammatory and antipyretic activities of the methanol extract of Crinum jagus bulb. The preliminary phytochemical analysis and acute toxicity studies were carried out following standard procedures. Assessment of analgesic activity in mice was performed using acetic acid-induced writhing and formalin-induced pain tests. Anti-inflammatory and antipyretic activities were performed in rats using egg albumin-induced pedal oedema and Brewer’s yeast-induced pyrexia models respectively. The results of the qualitative phytochemical analysis revealed that the bulb contained alkaloids, flavonoids, steroids, saponins, phenols and terpenoids. The extract was non-toxic; the oral LD was greater than 5000 mgkg-1 at 24 h. The 50 maximal analgesic effect of the extract against acetic acid-induced pain was 47.2% at 400 mgkg-1 compared to 82.2% with aspirin (50 mgkg-1). The extract exerted no significant (p>0.05) analgesic effect different from the control in the first phase (5 min) of formalin-induced pain test. However, there was dose dependent inhibition of pain sensitivity which peaked 85% at 400 mg/kg compared to 94% with piroxicam (500 mgkg-1) in the second phase (15-30 min) of the test indicating a peripherally mediated effect. The lowest test dose (100 mgkg-1) of extract had a comparable oedema inhibition (20%) to aspirin (50 mg/kg). C. jagus bulb extract produced a maximal antipyretic potency (79.5%) at 100 mgkg-1 compared to 83% with paracetamol (50 mgkg-1). Crinum jagus bulb could be a source of potent compounds against pain, inflammation and fever.

Nanofiber (NF) products exhibit outstanding performances in materials science, textiles, and medicine that cannot be realized using conventional technologies. However, the safety of such products is debated because of the potential health risks that nanomaterials pose and the lack of standardized guidelines for the safety evaluation of NF products. The global safety evaluations of nanomaterials have focused on evaluating the cytotoxicity of low-dimensional materials, including nanoparticles and nanotubes, based on OECD (Organization for Economic Co-operation and Development) criteria. NFs are one-dimensional materials with nanometer diameters and considerable lengths. Many fibers are applied in a densely woven web-like form, so assessing cellular penetration and fiber toxicity using the same methods is inappropriate. This study verifies the safety of the polyurethane (PU) and polyvinylidene fluoride (PVDF) polymers currently applied in filters and masks. To this end, polymer NFs were collected from each product, and the NFs were compared with reference samples using FT-IR and Raman spectroscopy. For the safety evaluation, DMSO stocks of varying concentrations of PVDF and PU NFs (at 0.5, 1, 5, and 10 μg/mL) were prepared. The cytotoxicity and inhibitory effects on nitric oxide production and protein expression obtained via Western blot were identified.

Unveiling the therapeutic potential: Evaluation of anti-inflammatory and antineoplastic activity of Magnolia champaca Linn’s stem bark isolate through molecular docking insights

Magnolia champaca Linn. has traditionally been used for medicinal activity in Asia for treating various chronic diseases as well as a source of food, medicines, and other commodities. Due to the long-used history of this plant, the present study was designed to explore the in vitro, in vivo and in silico anti-inflammatory and antineoplastic properties of the methanolic extract and fractions and the pure compound isolated from the most active chloroform fraction (CHF) of the stem bark of the plant. The isolated compound from the most active CHF was characterized and identified as a glycoside, trans-syringin, through chromatographic and spectroscopic (1H-NMR and 13C-NMR) analyses. In the in vitro anti-inflammatory assay, CHF was most effective in inhibiting inflammation and hemolysis of RBCs by 73.91 ± 1.70% and 75.92 ± 0.14%, respectively, induced by heat and hypotonicity compared to standard acetylsalicylic acid. In the egg albumin denaturation assay, CME and CHF showed the highest inhibition by 56.25 ± 0.82% and 65.82 ± 3.52%, respectively, contrasted with acetylsalicylic acid by 80.14 ± 2.44%. In an in vivo anti-inflammatory assay, statistically significant (p < 0.05) decreases in the parameters of inflammation, such as paw edema, leukocyte migration and vascular permeability, were recorded in a dose-dependent manner in the treated groups. In the antineoplastic assay, 45.26 ± 2.24% and 68.31 ± 3.26% inhibition of tumor cell growth for pure compound were observed compared to 73.26 ± 3.41% for standard vincristine. Apoptotic morphologic alterations, such as membrane and nuclear condensation and fragmentation, were also found in EAC cells after treatment with the isolated bioactive pure compound. Such treatment also reversed the increased WBC count and decreased RBC count to normal values compared to the untreated EAC cell-bearing mice and the standard vincristine-treated mice. Subsequently, in silico molecular docking studies substantiated the current findings, and the isolated pure compound and standard vincristine exhibited −6.4 kcal/mol and −7.3 kcal/mol binding affinities with topoisomerase-II. Additionally, isolated pure compound and standard diclofenac showed −8.2 kcal/mol and −7.6 kcal/mol binding affinities with the COX-2 enzyme, respectively. The analysis of this research suggests that the isolated bioactive pure compound possesses moderate to potent anti-inflammatory and antineoplastic activity and justifies the traditional uses of the stem bark of M. champaca. However, further investigations are necessary to analyze its bioactivity, proper mechanism of action and clinical trials for the revelation of new drug formulations.
