Characterization of a bacterial biocontrol strain B106 and its efficacies on controlling banana leaf spot and post-harvest anthracnose diseases

An antagonistic bacterial strain B106 was isolated from the rhizospheric soil of a banana plant in Nanning city, Guangxi, China, and identified as Bacillus subtilis based on its 16S rDNA sequence homology with the related bacteria from GenBank as well as physiological and biochemical characters. The cultural conditions were optimized for enhancing the efficacy of the antagonist against banana leaf spot caused by Pseudocercospora musae (teleomorph: Mycosphaerella musicola) and post-harvest anthracnose by Colletotrichum musae. The optimized cultural condition for strain B106 to express higher antagonistic activity against P. musae was the combination of 31 °C, pH 6.0, EM medium and 5-day-incubation. However, the optimized cultural condition for the bacterium to produce higher biomass was the combination of 31–34 °C, pH 6.5, EM medium and 3-day-incubation. The results based on greenhouse tests showed that 72.3% efficacy of the antagonist on controlling the banana leaf spot disease was obtained 10 days after pathogen inoculation. The efficacies of strain B106 (1 × 10 8 CFU ml −1) on controlling both the banana leaf spot diseases in the field and the anthracnose disease at post-harvest stage were 48.3% and 48.6%, respectively, under the optimized cultural condition for the strain to express higher antagonistic activity. The experimental data indicated that the antagonistic strain was a promising biocontrol agent against the banana diseases.

Characterization of plant growth-promoting rhizobacterium Exiguobacterium NII-0906 for its growth promotion of cowpea (Vigna unguiculata)

A phosphate solubilizing and antagonistic bacterial strain, isolated from a Western Ghat forest soil in Kerala province, India (designated as NII-0906), showed cold tolerance and grew from 10 to 37°C (optimum temperature 30°C). It was a Gram-positive, rod shaped, 0.8–1.6 μm in size, and exhibited tolerance to a wide pH range (5–12; optimum 7.0) and salt concentration up to 7% (w/v). The isolate showed maximum similarity with Exiguobacterium marinum TF-80T based on 16S rRNA analysis. It solubilized tricalcium phosphate under in vitro conditions. The phosphate solubilization was estimated along a temperature range (5–40°C), and maximum activity (84.7 μg mL−1 day−1) was recorded at 30°C after 10 days of incubation. The phosphate solubilizing activity coincided with a concomitant decrease in pH of the medium. The isolate also exhibited antifungal activity against phytopathogenic fungi in Petri dish assays and produced siderophore and hydrogen cyanide. The strain’s plant growth promotion properties were demonstrated through a cowpea-based bioassay under greenhouse conditions. The bacterial inoculation resulted in significant increment in plant root, stem and as well as in plant biomass. Further, scanning electron microscopic study revealed the root colonization in cowpea. These results could offer potential perspective for the strain to be used as plant growth-promoting rhizobacteria, which could be used as an inoculant for regional crops.

Biological control of potato black scurf by rhizosphere associated bacteria

The present work was carried out to study the potential of plant rhizosphere associated bacteria for the biocontrol of potato black scurf disease caused by Rhizoctonia solani Khun AG-3. A total of twenty-eight bacteria isolated from diseased and healthy potato plants grown in the soil of Naran and Faisalabad, Pakistan were evaluated for their antagonistic potential. Nine bacterial strains were found to be antagonistic in vitro, reduced the fungal growth and caused the lysis of sclerotia of R. solani in dual culture assay as well as in extracellular metabolite efficacy test. The selected antagonistic strains were further tested for the production and efficacy of volatile and diffusible antibiotics, lytic enzymes and siderophores against R. solani. Selected antagonistic bacteria were also characterized for growth promoting attributes i.e., phosphate solubilization, nitrogen fixation and indole acetic acid production. Biocontrol efficacy and percent yield increase by these antagonists was estimated in greenhouse experiment. Statistical analysis showed that two Pseudomonas spp. StT2 and StS3 were the most effective with 65.1 and 73.9 percent biocontrol efficacy, as well as 87.3 and 98.3 percent yield increase, respectively. Potential antagonistic bacterial strain StS3 showed maximum homology to Pseudomonas sp. as determined by 16S rRNA gene sequencing. These results suggest that bacterial isolates StS3 and StT2 have excellent potential to be used as effective biocontrol agents promoting plant growth with reduced disease incidence.

Characterization of a Phosphate Solubilizing and Antagonistic Strain of Pseudomonas putida (B0) Isolated from a Sub-Alpine Location in the Indian Central Himalaya

The morphological, biochemical, and physiological characteristics of a phosphate solubilizing and antagonistic bacterial strain, designated as B0, isolated from a sub-alpine Himalayan forest site have been described. The isolate is gram negative, rod shaped, 0.8 x 1.6 microm in size, and psychrotrophic in nature that could grow from 0 to 35 degrees C (optimum temp. 25 degrees C). It exhibited tolerance to a wide pH range (3-12; optimum 8.0) and salt concentration up to 4% (w/v). Although it was sensitive to kanamycin, gentamicin, and streptomycin (<10 microg mL(-1)), it showed resistance to higher concentrations of ampicillin, penicillin, and carbenicillin (>1000 microg mL(-1)). The isolate showed maximum similarity with Pseudomonas putida based on 16S rRNA analysis. It solubilized tricalcium phosphate under in vitro conditions. The phosphate solubilization was estimated along a temperature range (4-28 degrees C), and maximum activity (247 microg mL(-1)) was recorded at 21 degrees C after 15 days of incubation. The isolate also exhibited antifungal activity against phytopathogenic fungi in Petri dish assays and produced chitinase, ss-l,3-glucanase, salicylic acid, siderophore, and hydrogen cyanide. The plant growth promotion and antifungal properties were demonstrated through a maize-based bioassay under greenhouse conditions. Although the bacterial inoculation was found to result in significant increment in plant biomass, it stimulated bacterial and suppressed fungal counts in the rhizosphere. The present study is important with respect to enumerating microbial diversity of the colder regions as well as understanding the potential biotechnological applications of native microbes.

Extracellular Chitinases of Fluorescent Pseudomonads Antifungal to Fusarium oxysporum f. sp. dianthi Causing Carnation Wilt

Vascular wilt of carnation caused by Fusarium oxysporum f. sp. dianthi (Prill. & Delacr.) W. C. Synder & H.N. Hans inflicts substantial yield and quality loss to the crop. Mycolytic enzymes such as chitinases are antifungal and contribute significantly to the antagonistic activity of fluorescent pseudomonads belonging to plant-growth-promoting rhizobacteria. Fluorescent pseudomonads antagonistic to the vascular wilt pathogen were studied for their ability to grow and produce chitinases on different substrates. Bacterial cells grown on chitin-containing media showed enhanced growth and enzyme production with increased anti-fungal activity against the pathogen. Furthermore, the cell-free bacterial culture filtrate from chitin-containing media also significantly inhibited the mycelial growth. Both the strains and their cell-free culture filtrate from chitin-amended media showed the formation of lytic zones on chitin agar, indicating chitinolytic ability. Extracellular proteins of highly antagonistic bacterial strain were isolated from cell-free extracts of media amended with chitin and fungal cell wall. These cell-free conditioned media contained one to seven polypeptides. Western blot analysis revealed two isoforms of chitinase with molecular masses of 43 and 18.5 kDa. Further plate assay for mycelial growth inhibition showed the 43-kDa protein to be antifungal. The foregoing studies clearly established the significance of chitinases in the antagonism of fluorescent pseudomonads, showing avenues for possible exploitation in carnation wilt management.

Biocontrol of Korean Ginseng Root Rot Caused by Phytophthora cactorum Using Antagonistic Bacterial Strains ISE13 and KJ1R5

In this biocontrol research, we evaluated disease suppressive effects of antagonistic bacterial strains ISE13 and KJ1R5 against Korean ginseng root rot caused by P. eaetorum. We also examined the effects of nutrient solution in the hydroponic culture system for Korean ginseng on biological activity of the bacterial strains. As results of dual culture tests of the bacterial strains on <TEX>$V_{8}$</TEX> juice agar, the strain ISE13 showed antifungal activity against P. eaetorum and other plant pathogenic fungi, but the strain KJ1R5 did not. When their inhibitory effects against infection of P. eaetorum on the roots grown in either nutrient solution or water were tested, the strains ISE13 and KJ1R5 inhibited the disease severity of Korean ginseng roots only grown with water, compared to buffer-treated, inoculated controls. However, the nutrient solution used for hydroponic cultures of ginseng in pots caused higher levels of disease severity by the strains ISE13 and KJ1R5 from <TEX>418.8\%$</TEX> to <TEX>40.0\%$</TEX> and from <TEX>24.3\%$</TEX> to <TEX>45.0\%$</TEX>, respectively. In this study, the bacterial strains ISE13 and KJ1R5 could be potentially biocontrol agents to suppress Korean ginseng root rot caused by P. eaetorum. However, more attention using nutrient solution in hydroponic cultures for Korean ginseng production should be applied in biocontrol of plant diseases using the antagonistic microorganisms.

Integration of Biological Control Agents and Systemic Acquired Resistance Inducers Against Bacterial Spot on Tomato.

Two strains of plant growth-promoting rhizobacteria, two systemic acquired resistance inducers (harpin and acibenzolar-S-methyl), host-specific unformulated bacteriophages, and two antagonistic bacteria were evaluated for control of tomato bacterial spot incited by Xanthomonas campestris pv. vesicatoria in greenhouse experiments. Untreated plants and plants treated with copper hydroxide were used as controls. The plant growth-promoting rhizobacteria or a tap water control were applied as a drench to the potting mix containing the seedlings, while the other treatments were applied to the foliage using a handheld sprayer. The plant growth-promoting rhizobacteria strains, when applied alone or in combination with other treatments, had no significant effect on bacterial spot intensity. Messenger and the antagonistic bacterial strains, when applied alone, had negligible effects on disease intensity. Unformulated phage or copper bactericide applications were inconsistent in performance under greenhouse conditions against bacterial spot. Although acibenzolar-S-methyl completely prevented occurrence of typical symptoms of the disease, necrotic spots typical of a hypersensitive reaction (HR) were observed on plants treated with acibenzolar-S-methyl alone. Electrolyte leakage and population dynamics experiments confirmed that acibenzolar-S-methyl-treated plants responded to inoculation by eliciting an HR. Application of bacteriophages in combination with acibenzolar-S-methyl suppressed a visible HR and provided excellent disease control. Although we were unable to quantify populations of the bacterium on the leaf surface, indirectly we determined that bacteriophages specific to the target bacterium reduced populations of a tomato race 3 strain of the pathogen on the leaf surface of acibenzolar-S-methyl-treated plants to levels that did not induce a visible HR. Integrated use of acibenzolar-S-methyl and phages may complement each other as an alternative management strategy against bacterial spot on tomato.

Biological treatment of clogged emitters in a drip irrigation system

This study was conducted to investigate microbial organisms that can be used for preventing clogging in drip irrigation systems caused by biological factors. A total of 25 fungi isolate and 121 bacterial strains were isolated from water samples collected from drip irrigation systems in tomato greenhouses in the eastern Anatolia region of Turkey in the spring season of 2001. Biological clogging of emitters in a model drip irrigation system was experimentally caused by application of the microorganisms (fungi and bacteria) isolated in the study. Three antagonistic bacterial strains in the genus Bacillus spp (ERZ, OSU-142) and Burkholdria spp (OSU-7) were used for treatment of biological clogging of the emitters. The results showed that the antagonistic bacterial strains tested have the potential to be used as anti-clogging agents for treatment of emitters in drip irrigation system. This is the first study that demonstrated that antagonistic microorganisms can be utilized for treatment of clogging in drip irrigation systems.

Strategy to select and assess antagonistic bacteria for biological control of Rhizoctonia solani Kühn.

A screening strategy was developed to assess the potential of plant-associated bacteria to control diseases caused by Rhizoctonia solani Kühn. About 434 already characterized antagonistic bacterial strains isolated from diverse plant species and microenvironments were evaluated for biocontrol and plant growth promotion by a hierarchical combination of assays. Analyzing in vitro antagonism towards different Rhizoctonia isolates resulted in a selection of 20 potential biocontrol agents. The strains were characterized by their antagonistic mechanisms in vitro as well as their production of the plant growth hormone indole-3-acetic acid. The plant growth promoting effect by antagonistic bacteria was determined using a microtiter plate assay on the basis of lettuce seedlings. Lettuce and sugar beet as host plant were included in the biocontrol experiments in which the antagonistic effect of 17 bacterial isolates could be confirmed in vivo. Sequencing of the 16S rDNA gene and (or) fatty acid methyl ester gas chromatography was used to identify the antagonistic isolates. Molecular fingerprints of isolates obtained by BOX-polymerase chain reaction were compared to avoid further investigation with genetically very similar strains and to obtain unique molecular fingerprints for quality control and patent licensing. According to our strategy, an assessment scheme was developed and four interesting biological control agents, Pseudomonas reactans B3, Pseudomonas fluorescens B1, Serratia plymuthica B4, and Serratia odorifera B6, were found. While S. plymuthica B4 was the best candidate to biologically control Rhizoctonia in lettuce, P. reactans B3 was the best candidate to suppress the pathogen in sugar beet.

Influence of inoculum density of the antagonistic bacteria Pseudomonas fluorescens and Pseudomonas corrugata on sugar beet seedling colonisation and suppression of Pythium damping off

The effect of initial inoculum density of the antagonistic bacterial strains Pseudomonas fluorescens B5 and Pseudomonas corrugata 2140 (103 to 108 CFU per seed pellet) on sugar beet seedling colonisation, in situ bioluminescence and antagonistic activity towards Pythium ultimum was investigated. Populations of the bacteria colonising sugar beet root systems approached an apparent carrying capacity of 105 to 106 CFU per plant after 12 d growth, irrespective of inoculum density. This meant an up to 320-fold population increase at low inoculum densities and a decrease at high densities. Population densities of both bacteria and their corresponding in situ bioluminescence (resulting from luciferase enzyme expression from the inserted luxAB genes) reached highest levels in the hypocotyl region and in the upper root region 0–20 mm below seed level (104–106 CFU/cm section, 101–103 RLU/cm section) and decreased with root depth. In situ bioluminescence, which indicates physiological activity, was measurable at lowest antagonist initial inoculum density (103 CFU per seed pellet) and did not increase significantly with increasing inoculum density. Bioluminescence was also significantly correlated with population density. For Pseudomonas fluorescens B5, the total population size per plant and downward colonisation of the root (below 40 mm depth) increased significantly with antagonist inoculum density applied to the seeds. For Pseudomonas corrugata 2140, no significant influence of initial inoculum density on root colonisation was observable. Survival and dry weight of sugar beet seedlings in Pythium infested soil increased significantly with increasing inoculum density of Pseudomonas fluorescens B5, whereas for Pseudomonas corrugata 2140, initial densities of 104 to 106 CFU per seed resulted in maximal survival of plants.
