Integrated set-up of sampling, sample treatment and assay of an intracellular protein during fermentation

A complete system for on-line sampling, sample handling and analysis of fermentation processes has been set up. The system is designed such that a sample is taken from the fermentor, the metabolism of microbial cells is instantaneously arrested, the cells are lysed in order to release intracellular compounds and in a subsequent binding assay the content of target molecules is monitored. The system is constructed based on the flow-injection analysis concept. Good agreement between on-line analysis and off-line analysis is reported.

Analysis of a fermentation recycle loop for on-line measurements

Recycle lines have been widely used in on-line analysis of fermentation processes. However, like most tools, many practical considerations can remain overlooked. Upon reflecting on this technique, some of these overlooked considerations have been studied. Because of the importance of a debubbler to a recycle line, an improved design was made and tested which can produce a bubble free high flow rate stream to reduce residence times. The effect of the recycle line on the culture growth, and DO was also investigated and found to be negligible.

Some practical aspects of computer applications in a fermentor hall

The use of computers in a fermentation pilot plant is described from a practical point of view. The aim is not the application of a computer to a single special process but a general application of computers to prepare and present data for the following analysis of fermentation processes. The hardware is normally bought from computer manufacturers, but some additional installations are useful. Application software has to be developed by the biotechnologist, therefore the software structure is the most important part of the computer application. Data storage is divided into three parts: short-time memory, long-time storage, and a medium memory mainly to be used in process analyses and process control. Four types of programs are used: main schedule tasks, low-priority on-line tasks, sense-switch routine, and different off-line programs. A table of all programs is presented, the main schedule task is described in detail to demonstrate the software structure.

Analysis of fermentation processes using flow microfluorometry: Amylase and protease activities in Bacillis subtilis batch cultures

AbstractFlow microfluorometry, which provides detailed information on the state of a microbial population, has been employed to characterize the Bacillus subtilis population during time intervals in which significant changes in the culture amylase activity occur. Four different batch experiments have been conducted, and the influences of inoculum age, fermentation temperature, and aeration rate on microbial population dynamics and amylase activity have been examined. Relatively high rates of amylase activity increase are observed twice during the batch, first as double cells initiate sporulation and later during germination. Rapid decreases in amylase activity are observed in highly (25–50%) sporulated populations, and in at least one experiment, during a transition from large, rounded protoplast forms to normal rod morphology. Amylase and protease activities do not follow parallel nor proportional trajectories in these 72 hr batch fermentations.
