Producing natural functional and low-carbon milk by regulating the diet of the cattle-The fatty acid associated rumen fermentation, biohydrogenation, and microorganism response.

Conjugated linoleic acid (CLA) has drawn significant attention in the last two decades for its various potent beneficial effects on human health, such as anticarcinogenic and antidiabetic properties. CLA could be generally found in ruminant products, such as milk. The amount of CLA in ruminant products mainly depends on the diet of the animals. In general, the fat content in the ruminant diet is low, and dietary fat supplementation can be provided to improve rumen activity and the fatty acid (FA) profile of meat and milk. Especially, dietary 18-carbon polyunsaturated FA (C18 PUFA), the dominant fat source for ruminants, can modify the milk FA profile and other components by regulating the ruminal microbial ecosystem. In particular, it can improve the CLA in milk, intensify the competition for metabolic hydrogen for propionate producing pathways and decrease methane formation in the rumen. Therefore, lipid supplementation appears to be a promising strategy to naturally increase the additional nutritional value of milk and contribute to lower methane emissions. Meanwhile, it is equally important to reveal the effects of dietary fat supplementation on rumen fermentation, biohydrogenation (BH) process, feed digestion, and microorganisms. Moreover, several bacterial species and strains have been considered to be affected by C18 PUFA or being involved in the process of lipolysis, BH, CLA, or methane emissions. However, no review so far has thoroughly summarized the effects of C18 PUFA supplementation on milk CLA concentration and methane emission from dairy cows and meanwhile taken into consideration the processes such as the microorganisms, digestibility, rumen fermentation, and BH of dairy cattle. Therefore, this review aims to provide an overview of existing knowledge of how dietary fat affects rumen microbiota and several metabolic processes, such as fermentation and BH, and therefore contributes to functional and low-carbon milk production.

Relationship between the Composition of Lipids in Forages and the Concentration of Conjugated Linoleic Acid in Cow's Milk: A Review.

The content of these precursors will depend on factors, such as age and nitrogen fertilization, since the high availability of nitrogen stimulates the synthesis of metabolic components, such as leaf protein. The amounts of precursors in the diets of animals are related to the amounts of CLA in milk. In the literature review, it was found that the milk of cows fed fresh forage has a higher amount of CLA because they have a higher amount of linoleic acid and α-linolenic acid compared to other foods used in the diets of cows. The amount of CLA precursors in pastures can be increased through agronomic practices, such as nitrogen fertilization, and regrowth age. It is also a technique used to increase the amount of CLA in milk to obtain a greater benefit regarding its nutritional value.

Probiotic Properties of Lactic Acid Bacteria with High Conjugated Linoleic Acid Converting Activity Isolated from Jeot-Gal, High-Salt Fermented Seafood.

Conjugated linoleic acid (CLA) isomers are potent health-promoting fatty acids. This study evaluated the probiotic properties of 10 strains of high CLA-producing lactic acid bacteria (LAB) isolated from Jeot-gal, a high-salt, fermented seafood. Two isolates, Lactiplantibacillus plantarum JBCC105683 and Lactiplantibacillus pentosus JBCC105676, produced the largest amounts of CLA (748.8 and 726.9 μg/mL, respectively). Five isolates, L. plantarum JBCC105675, L. pentosus JBCC105676, L. pentosus JBCC105674, L. plantarum JBCC105683, and Lactiplantibacillus paraplantarum JBCC105655 synthesized more cis-9, trans-11-CLA than trans-10, cis-12-CLA (approximately 80:20 ratio). All the strains survived severe artificial acidic environments and showed antimicrobial activity and strong adhesion capability to Caco-2 cells as compared to the commercial strain Lactocaseibacillus rhamnosus GG. Among them, Pediococcus acidilactici JBCC105117, L. paraplantarum JBCC105655, and L. plantarum JBCC105683 strongly stimulated the immunological regulatory gene PMK-1 and the host defense antimicrobial peptide gene clec-60 in Caenorhabditis elegans. Moreover, three strains showed a significant induction of tumor necrosis factor-α, interleukin (IL)-1β, IL-6, IL-12, and IL-10 production in RAW 264.7 macrophages, indicating that they were promising candidates for probiotics with high CLA-converting activity. Our results indicate that the newly isolated CLA-producing LAB might be useful as a functional probiotic with beneficial health effects that modulate the immune system.

Impact of winterization on fatty acids' composition, isomers, and oxidative stability of conjugated linoleic acids produced from selected vegetable oils

Degummed, refined, winterized (at −60°C), and non-winterized soybean, sunflower, cottonseed, and safflower oil samples were used for the production of conjugated linoleic acid (CLA) by urea treatment. Fatty acids' composition and CLA isomers of winterized and non-winterized versions of oil were analyzed on GC-MS. Winterization improved the concentration of linoleic acids (LAs) in all vegetable oils. Safflower oil exhibited the highest concentration of LAs at 84.19% followed by sunflower oil (68.54%), cottonseed oil (63.74%), and soybean oil (59.88%), respectively. In safflower oil derived CLAs, concentrations of ∆9c,11t-18:2, ∆10t,12c-18:2, ∆9c,11c-18:2, ∆10c,12t-18:2, ∆8,9,11,10,12c-c18:2, and ∆8,9,11,10,12t-t18:2 were calculated 47.68%, 39.95%, 5.42%, 1.89%, 3.06%, 0.55%, and 1.45%, while in winterized safflower oil derived CLAs, these concentrations were 55.67%, 41.92%, 0.19%, 0.78%, 0.65%, 0.47%, and 0.32%, respectively, with significant difference in induction period and peroxide value. Yield of CLA from winterize soybean, sunflower cottonseed, and safflower oil was 22.58%, 24.95%, 23.16%, and 36.68%. Practical applications Conjugated linoleic acids (CLAs) are naturally present in milk, dairy products, and beef. However, the amount of CLAs in dairy products and beef accounts limited moreover, the yield of CLA supplement produced from safflower oil is about 20%. To make the industrial processes economical, and sustainable fortification strategies, yield of CLA supplement should be increased. In the current study, winterized version of sunflower, soybean, cottonseed, and safflower oils were used to produce CLA supplement. Using winterized version, the yield of CLA supplement from safflower oil was increased to 36.68%. This work is highly suitable for the food industries to produce functional foods containing CLA.

Effect of Rearing System on the Straight and Branched Fatty Acids of Goat Milk and Meat of Suckling Kids

Goat meat is considered healthy because it has fewer calories and fat than meat from other traditional meat species. It is also rich in branched chain fatty acids that have health advantages when consumed. We studied the effects of maternal milk and milk replacers fed to suckling kids of four breeds on the straight and branched fatty acid compositions of their muscle. In addition, the proximal and fatty acid compositions of colostrum and milk were studied. Goat colostrum had more protein and fat and less lactose than milk. Goat milk is an important source of healthy fatty acids such as C18:1 c9 and C18:2 n–6. Suckling kid meat was also an important source of C18:1c9. Dairy goat breeds had higher percentages of trans monounsaturated fatty acids (MUFAs) and most of the C18:1 isomers but lower amounts of total MUFAs than meat breeds. However, these dairy kids had meat with a lower percentage of conjugated linoleic acid (CLA) than meat kids. The meat of kids fed natural milk had higher amounts of CLA and branched chain fatty acids (BCFAs) and lower amounts of n–6 fatty acids than kids fed milk replacers. Both milk and meat are a source of linoleic, α-linolenic, docosahexaenoic, eicosapentaenoic and arachidonic fatty acids, which are essential fatty acids and healthy long-chain fatty acids.

Effect of Conjugated Linoleic Acid on Memory and Reflex Maturation in Rats Treated During Early Life.

In the critical period of neurodevelopment (gestation and lactation), maternal consumption of essential fatty acids (FAs) can alter the offspring cognitive function permanently causing damage. Lipids can regulate neurotrophin and compose brain tissue. However, the effects of maternal consumption of a mixture of conjugated linoleic acid (CLA) on an offspring nervous system are not completely clear. We aimed to investigate the impacts of different CLA concentrations mixed into the maternal diet during early life on neonatal reflex maturation and cognitive functions of the offspring. Three groups were formed: control (CG): receiving a standard diet; CLA1: receiving a diet containing 1% of CLA, and CLA3: receiving a diet containing 3% of CLA, offered during gestation and lactation. After birth, the reflex responses of the offspring were observed from the 1st to the 21st day. After weaning, the animals’ anxiety and memory were assessed using open field (OF) and novel object recognition tests. Fatty acids in the breast milk and the offspring’s brain were also quantified. The data were analyzed using one-way ANOVA and the Kruskal–Wallis test. CLA1 presented accelerated palmar grasp disappearance versus CLA3 and negative-geotaxis versus CG; and the CLA3 presented increases for most reflexes (cliff-avoidance, vibrissa-placing, negative-geotaxis, and auditory-startle response), and decrease in reflexes palmar grasp and free-fall righting versus CG (p < 0.05). CLA3 group explored less of the OF in the second exposure. CLA1 and CLA3 presented an increased exploration ratio for new objects, which indicates memory improvement. The milk tested from CLA3 demonstrated an increase in polyunsaturated fatty acids (PUFAs), and a decrease in monounsaturated fatty acids. The amount of CLA in milk was greater in CLA1 and CLA3 and in the brain offspring both presented moderated amounts of CLA. Maternal treatment with the CLA mixture induced anticipated reflex maturation and improved memory in the offspring. Even though CLA was detected in the brains in only trace amounts, offspring’s brain PUFA and SFA levels were increased. Further studies aimed to delineate the effect of maternal CLA supplementation on offspring’s brain lipid metabolism and long-term neurologic outcome are needed to confirm these findings.

Milk fatty acid profile from cows fed with mixed rations and different access time to pastureland during early lactation.

Milk fatty acid (FA) profiles were determined in Holstein cows (n=27) fed total mixed rations (TMR) ad libitum (G0) or diet composed by TMR (50% dry matter [DM] offered) plus grazing of pasture with 6hr of access time to paddock in one session (G1) or 9hr in two sessions (G2) at 45days in milk (DIM). Moreover, milk FA was determined at 65 DIM when G0 cows turned out to G1 diet without adaptation period (Post-G0), G1 remained as controls. Milk FA was quantified using gas chromatography and mass spectrometry. Preformed FA at 45 DIM was greater (+27%) for G2 than G0 cows (p<.05). Stearic acid (C18:0) was 30% greater for G2 cows (p<.05). De novo FA was lowest for G2 cows (p<.05). Conjugated linoleic acid (CLA) did not differ (p<.12), while vaccenic acid (C18:1trans) was twofold greater for grazing treatments (p<.01). Linolenic acid [C18:3(n-3)] was greatest for G2 and lowest for G0 cows (p<.01). Omega 6 FA was greater for G0 than grazing cows, mainly due to linoleic acid [18:2cis(n-6); p<.05]. These results determined that n-6/n-3 ratio was almost threefold greater for G0 than grazing cows (p<.001). When diet of G0 cows changed to include pasture (Post-G0), preformed FA increased (p<.05), explained mainly by the increase (p<.05) of stearic (C18:0) and C18:1trans, while de novo FA tended to decrease (p<.1). Moreover, the amount of CLA and C18:3(n-3) tended to increase (p<.1) in Post-G0 cows. Offering 50% of dietary DM from pasture modified milk FA profile in early lactation potentially beneficial for human health. When TMR-fed cows were turned out to 50% pasture, milk FA profile reflected dietary change without need of an adaptation period.

Effect of pre- and postpartum supplementation with lipid-encapsulated conjugated linoleic acid on reproductive performance and the growth hormone–insulin-like growth factor-I axis in multiparous high-producing dairy cows

The objective of the study was to evaluate the effect of prepartum and postpartum (PP) supplementation with 2 isomers of conjugated linoleic acid (CLA) on reproductive parameters and some related metabolic factors in dairy cows. High-producing, multiparous Holstein Friesian cows (n = 60) were allotted to 3 treatment groups: the CLA1 group (n = 20) was supplemented with 70 g of lipid-encapsulated CLA providing 7 g each of cis-9,trans-11 and trans-10,cis-12 CLA from d 21 (d 21) before expected calving until d 7 after artificial insemination (AI), that is, until 77 to 91 d PP; the CLA2 group (n = 20) was supplemented with the same amount of CLA beginning at calving until d 7 after AI; and the control group (n = 20) received an isocaloric, isonitrogenous, and isolipidic diet. Blood samples were taken weekly to measure glucose, insulin, insulin-like growth factor-I (IGF-I), and leptin. Liver biopsy was performed in 10 cows per group for growth hormone receptor 1A and IGF-I mRNA analyses. At d 49 to 63 PP, ovulation was synchronized with the Pre-Synch protocol followed by fixed-time AI. Milk progesterone was monitored from calving until d 35 post-AI. Cows returning to estrus following AI were inseminated. Supplementation with CLA before calving improved the recovery of plasma leptin levels in the early PP period (from the day of calving until wk 3 PP; treatment effect). Later PP (wk 5), plasma IGF-I, and leptin remained significantly higher in both CLA1 and CLA2 groups compared with control, although hepatocellular IGF-I mRNA was not different among groups. Plasma IGF-I levels remained higher in both CLA-treated groups on the day of AI. Growth hormone receptor 1A mRNA levels in hepatic tissue decreased in all groups, reaching a nadir in the first week PP. Days to first PP ovulation did not differ between groups; however, both supplemented groups conceived earlier compared with control (d 97 ± 19, d 97 ± 23, and d 113 ± 30 for CLA1, CLA2, and control, respectively). Plasma progesterone concentration was higher in both supplemented groups on d 2 to 5 following the synchronized ovulation than in controls. We concluded that CLA supplementation around calving alters PP metabolic signals as reflected by higher plasma leptin and IGF-I levels. Conjugated linoleic acid stimulated early luteal function and reduced the PP interval to conception.

Innovative superparamagnetic iron-oxide nanoparticles coated with silica and conjugated with linoleic acid: Effect on tumor cell growth and viability

One of the goals for the development of more effective cancer therapies with reduced toxic side effects is the optimization of innovative treatments to selectively kill tumor cells. The use of nanovectors loaded with targeted therapeutic payloads is one of the most investigated strategies. In this paper superparamagnetic iron oxide nanoparticles (SPIONs) coated by a silica shell or uncoated, were functionalized with single-layer and bi-layer conjugated linoleic acid (CLA). Silica was used to protect the magnetic core from oxidation, improve the stability of SPIONs and tailor their surface reactivity. CLA was used as novel grafting biomolecule for its anti-tumor activity and to improve particle dispersibility. Mouse breast cancer 4T1 cells were treated with these different SPIONs.SPIONs functionalized with the highest quantity of CLA and coated with silica shell were the most dispersed. Cell viability was reduced by SPIONs functionalized with CLA in comparison with cells which were untreated or treated with SPIONs without CLA. As regards the types of SPIONs functionalized with CLA, the lowest viability was observed in cells treated with uncoated SPIONs with the highest quantity of CLA.In conclusion, the silica shell free SPIONs functionalized with the highest amount of CLA can be suggested as therapeutic carriers because they have the best dispersion and ability to decrease 4T1 cell viability.

Isolation, molecular characterization and screening of indigenous lactobacilli for their abilities to produce bioactive conjugated linoleic acid (CLA).

Ingestion of conjugated linoleic acid poised many health benefits; however, amount of CLA one can get through generalized diet in is inadequate in exerting the desired benefits. Therefore, presence of CLA producing lactobacilli in dairy fermented foods has a tremendous potential to increase the CLA content. Therefore, present study was focused to isolate and characterize CLA producing lactobacilli from different dairy products and human faeces. Arguably, 283 lactobacilli were isolated from various sources and tested for CLA production. Fifty-seven CLA producing (≥20µg/ml) lactobacilli were selected from screening in de Man, Rogosa and Sharpe (MRS) broth and reconstituted with skim milk (SM), supplemented with 0.5mg/ml of linoleic acid. Positive strains were classified into-L. plantarum (44%), L. gasseri (30%), L. fermentum (21%) and L. salivarius (5%) species. Nineteen most efficient strains (CLA ≥25µg/ml) were further assessed in SM for CLA production. Total 08 strains produced significantly higher CLA in SM than MRS and also produced cis 9, trans 11, trans 10, cis 12 and trans 9, trans 11 isomers. Overall, L. plantarum HIF15 was reported as the best producer of CLA and other 08 lactobacilli may be utilized for the formulation of CLA-enriched functional foods to support these bacteria to synthesize CLA in the human gut.

Scientific Advances of Milk Enrichment with Conjugated Linoleic Acid to Produce Anti-Cancer Milk

Background: Conjugated linoleic acid (CLA) is an unsaturated fatty acid that after the discovery of its anti-cancerous properties numerous researches conducted to increase its concentration in the food products. The current study aimed to review researches in the field of milk enrichment with CLA to identify the main actors and highlight the achievements in the field; therefore, the effectiveness of these researches for producing anti-cancer milk was surveyed. Methods: The journal of dairy science was selected for data extraction and related papers were identified via review of all published papers during years 2003 - 2013 and then were subjected into content analysis. Statistical analysis was done with the MEANS procedure of SAS software (Version 9.1). Results: Among all collected papers, only 72 were eligible. Data analysis showed that the USA, Canada and France were the most productive countries in the field of milk enrichment with CLA. Surprisingly, Iran did not have any paper in this field in the journal of dairy science. The supplements that were added to the animal diets were enriched by linoleic or linolenic acids. Enhancing the milk cis-9, and trans-11 CLA content were the main objectives of researches. More than 70% of treatments led to the supply of the minimum amount of CLA to prevent cancer with serving milk twice a day. Conclusions: Considerably, milk enrichment with CLA mostly has being attracted by developed countries. Enriching milk with CLA is a suitable scientific strategy to fight against cancer. It is important to increase researches in increasing trans- 10, and cis- 12 CLA.

Conjugated linoleic acids influence fatty acid metabolism in ovine ruminal epithelial cells

Conjugated linoleic acids (CLA), particularly cis-9,trans-11 (c9t11) and trans-10,cis-12 (t10c12), are used as feed additives to adapt to constantly increasing demands on the performance of lactating cows. Under these feeding conditions, the rumen wall, and the rumen epithelial cells (REC) in particular, are directly exposed to high amounts of CLA. This study determined the effect of CLA on the fatty acid (FA) metabolism of REC and expression of genes known to be modulated by FA. Cultured REC were incubated with c9t11, t10c12, and the structurally similar FA linoleic acid (LA), oleic acid (OA), and trans-vaccenic acid (TVA) for 48 h at a concentration of 100µM. Cellular FA levels were determined by gas chromatography. Messenger RNA expression levels of stearoyl-CoA desaturase (SCD) and monocarboxylate transporter (MCT) 1 and 4 were quantified by reverse transcription-quantitative PCR. Fatty acid evaluation revealed significant effects of CLA, LA, OA, and TVA on the amount of FA metabolites of β-oxidation and elongation and of metabolites related to desaturation by SCD. The observed changes in FA content point (among others) to the ability of REC to synthesize c9t11 from TVA endogenously. The mRNA expression levels of SCD identified a decrease after CLA, LA, OA, or TVA treatment. In line with the changes in mRNA expression, we found reduced amounts of C16:1n-7 cis-9 and C18:1n-9 cis-9, the main products of SCD. The expression of MCT1 mRNA increased after c9t11 and t10c12 treatment, and CLA c9t11 induced an upregulation of MCT4. Application of peroxisome proliferator-activated receptor (PPAR) α antagonist suggested that activation of PPARα is involved in the changes of MCT1, MCT4, and SCD mRNA expression induced by c9t11. Participation of PPARγ in the changes of MCT1 and SCD mRNA expression was shown by the application of the respective antagonist. The study demonstrates that exposure to CLA affects both FA metabolism and regulatory pathways within REC.

In-vitro microbial production of conjugated linoleic acid by probiotic L. plantarum strains: Utilization as a functional starter culture in sucuk fermentation

Twenty-three probiotic Lactobacillus plantarum strains were screened in-vitro to determine their ability to produce conjugated linoleic acid (CLA). L. plantarum AA1-2 and L. plantarum AB20-961 were identified as potential strains for CLA production. Optimum conditions for these strains to produce high levels of CLA were determined by evaluating the amount of added hydrolyzed sunflower oil (HSO) and initial pH levels in a nutrient medium. The highest CLA production was obtained in medium with pH6.0 and 2% HSO (P<0.05). Those strains were then used as starter culture in sucuk fermentation. Five sucuk treatments included a control (no starter culture), two sucuk groups with L. plantarum AA1-2 at the initial pH of 5.8 or 6.0 and two sucuk groups with L. plantarum AB20-961 at the initial pH of 5.8 or 6.0. Results indicate that L. plantarum AB20-961 produced higher amount of CLA in sucuk at initial pH of 5.8 and 6.0 levels during first 24h of fermentation compared with other groups. CLA isomer concentration decreased in all sucuk groups during the rest of the fermentation period (P<0.05) and remained quite stable during the storage. This study demonstrated that probiotic L. plantarum AB20-961 can be used in sucuk manufacturing without posing any quality problems.

Concentrations of retinol and tocopherols in the milk of cows supplemented with conjugated linoleic acid.

This study was performed to investigate the hypothesis that supplementation of conjugated linoleic acid (CLA) changes the concentrations of retinol and tocopherols in the milk of cows. To investigate this hypothesis, Holstein cows received daily from 3 weeks ante-partum to 14 weeks post-partum either 172 g of a CLA-free rumen-protected control fat (control group, n = 20) or the same amount of a rumen-protected CLA fat, supplying 4.3 g of cis-9, trans-11 CLA and 3.8 g of trans-10, cis-12 CLA per d (CLA group, n = 20). Milk samples (collected at weeks 1, 3, 5, 8 and 11 of lactation) were analysed for retinol, α- and γ-tocopherol concentrations. Milk of cows supplemented with CLA had higher concentrations of retinol (+34%), α-tocopherol (+44%) and γ-tocopherol (+21%) than milk of control cows (p < 0.05). The daily output of these vitamins via milk was also greater in cows of the CLA group than in cows of the control group (+36, 50 and 24% for retinol, α-tocopherol and γ-tocopherol, respectively, p < 0.05). In agreement with higher concentrations of tocopherols, concentrations of thiobarbituric acid-reactive substances, determined in milk of week 5, were lower in cows of the CLA group than in control cows, indicative of a lower susceptibility of milk lipids to peroxidation. Plasma concentrations of retinol and α-tocopherol, determined at 1 and 5 weeks post-partum, were not different between the two groups of cows. In conclusion, this study shows that supplementing dairy cows with a moderate amount of CLA causes an increase of the concentrations of vitamins A and E in the milk and results in an increased output of those vitamins via milk. These effects might be beneficial with respect to the nutritional value of dairy products and the susceptibility of milk fat to oxidative deterioration.

Effect of dietary conjugated linoleic acid supplementation on the technological quality of backfat of pigs

Pigs were fed diets containing 0, 0.25, 0.5 and 1% conjugated linoleic acid (CLA). Compared to controls, backfat from CLA fed pigs was firmer and extracted lipid contained increasing amounts of CLA, but a ±11% overall decrease in unsaturated fatty acids and a ±5% overall increase in each of C16:0 and C18:0 saturated fatty acids were noted. This resulted in a change in the melting properties of fat. The onset setting temperature increased from ±14°C to ±18°C for lipid of backfat of pigs from the 0.25 and 0.5% CLA supplementation groups, and to ±26°C for lipid from the 1% CLA supplementation group. The final melting temperatures increased from ±37°C to ±43°C and ±45°C, respectively. The presence of β′-crystals of C18:0–C16:0–C18:1c9 triacylglycerides in fat from CLA fed pigs and β-crystals in fat from 1% CLA fed pigs was observed. Fatty acid and melting point results explained the improvement in the technological quality of backfat as a result of dietary CLA supplementation.

Conjugated linoleic acids and conjugated vegetable oils: From nutraceutical to bio‐polymer

Recent interest from academia, nutritionists, the chemical as well as the feed and food industry in conjugated linoleic acids (CLAs) and conjugated vegetable oils has grown spectacularly. CLA isomers, either in their natural or synthetic forms, have not only been associated with diverse health and physiological effects, but they are also interesting renewable compounds in the production of industrial products such as paints, glues, and polymers, due to their very reactive conjugated double bond system. Due to the depletion of the world crude oil reserves and the increasing trend to use renewable feedstock in the chemical industry, it is to be expected that the use of conjugated fatty acids and oils will continue to grow in the near future. As high amounts of CLAs and conjugated vegetable oils will be needed and natural resources are limited, efficient production processes are urgently needed. An efficient process for the production of CLA from methyl linoleate, using the Wilkinson catalyst, is described by Behr et al. in this issue of European Journal of Lipid Science and Technology.

Effect of long term feeding with conjugated linoleic acid (CLA) in growth performance and lipid metabolism of European sea bass (Dicentrarchus labrax)

In the present study, the effects of long term feeding dietary conjugated linoleic acid (CLA) on growth, liver and perivisceral fat morphology, lipid metabolism enzymes and prostaglandin production potential were assessed. Two hundred and forty-three European sea bass (152.4±1.96g) were randomly allocated in 9 indoor 1m3 fiberglass tanks (4.1kg/m3) and assigned three diets containing graded amounts of CLA (0, 0.5 and 1%) for 20weeks. Dietary CLA did not affect growth performance or tissue proximate composition although increased the saturated fatty acid fraction in fish fed with 1% CLA in muscle. Muscle and liver fatty acid profiles were slightly altered by CLA inclusion, mainly by the increase of the CLA isomers and by the increase of 18:0 and the decrease of 16:1n-7. Liver lipogenic and lipolytic enzymes were not affected by dietary CLA. Hepatocyte area was reduced in fish fed with 1% of CLA, as well as adipocyte area in fish fed with 0.5% of CLA where adipocyte number by unit of area also increased. Head kidney prostaglandins' production potential was reduced in fish fed with 1% of CLA, having also a negative correlation with dietary CLA level of inclusion. The present results suggest that CLA can be incorporated up to 1% in 150g European sea bass for 20weeks without affecting fish performance or enzyme activities and at the time improves liver and adipose tissue morphology.

Appraisal of Conjugated Linoleic Acid Production by Probiotic Potential of Pediococcus spp. GS4

Probiotics with ability to produce conjugated linoleic acid (CLA) is considered as an additive health benefit property for its known role in colon cancer mitigation. The conversion involves the biohydrogenation of the unsaturated fatty acid into conjugated form. Probiotic strain Pediococcus spp. GS4 was efficiently able to biohydrogenate linoleic acid (LA) into its conjugated form within 48h of incubation. Quantum of CLA produced with a concentration of 121μg/ml and sustained cell viability of 8.94log cfu/ml maximally. Moreover, antibacterial effect of LA on the strain ability for biohydrogenation was examined at different concentrations and concluded to have a direct relationship between LA and amount of CLA produced. The efficiency of the strain for CLA production at different pH was also estimated and found maximum at pH 6.0 with 149μg/ml while this ability was reduced at pH 9.0 to 63μg/ml. Sesame oil, which is rich in the triacylglycerol form of LA, was also found to act as a substrate for CLA production by Pediococcus spp. GS4 with the aid of lipase-catalyzed triacylglycerol hydrolysis and amount of CLA produced was 31μg/ml at 0.2% while 150μg/ml at 1.0% of lipolysed oil in skim milk medium. Conjugated form was analyzed using UV scanning, RP-HPLC, and GC-MS. This study also focused on the alternative use of lipolysed sesame oil instead of costly LA for biohydrogenation and could be a potential source for the industrial production of CLA.

Cross-sectional study of conjugated linoleic acid in adipose tissue and risk of diabetes

Some experimental studies on conjugated linoleic acid (CLA) and insulin regulation suggested that CLA could be associated with risk of diabetes, but epidemiologic studies are lacking. The aim of the study was to test whether the amount of CLA in adipose tissue is associated with risk of diabetes. A cross-sectional design was used to test the study hypothesis in 232 adults with diabetes and 1512 adults without diabetes who lived in Costa Rica. The cis-9, trans-11 and trans-10, cis-12 CLA isomers in adipose tissue and 48 other fatty acids were assessed by using gas chromatography. Prevalence ratios (PRs) and 95% CIs were estimated by using Poisson regression adjusted for potential confounders. The mean (±SD) percentage of total fatty acids of CLA for the cis-9, trans-11 isomer in adipose tissue was 0.57 ± 0.18% in adults without diabetes and 0.53 ± 0.17% in adults with diabetes (P = 0.0078). The trans-10, cis-12 CLA isomer was not detected in adipose tissue. The cis-9, trans-11 CLA isomer was associated with a lower risk of diabetes. In comparison with the first quintile, the PR (95% CI) for the fifth quintile was 0.48 (0.31, 0.76) (P-trend = 0.0005) in the basic and 0.46 (0.29, 0.72) (P-trend = 0.0002) in the multivariable model. Additional adjustment for other fatty acids in adipose tissue including trans-9 16:1, which is a fatty acid that was previously associated with diabetes, did not modify the results. The observed inverse association between the cis-9, trans-11 CLA in adipose tissue and diabetes risk is consistent with the hypothesis that CLA may be involved in insulin regulation.

Backfat Fatty Acid Profile of Crossbred Pigs Fed a Diet Supplemented with Conjugated Linoleic acid or Sunflower Oil

Backfat Fatty Acid Profile of Crossbred Pigs Fed a Diet Supplemented with Conjugated Linoleic acid or Sunflower OilThe aim of the study was to investigate the impact of feeding pigs with different levels of conjugated linoleic acid (CLA) or sunflower oil (SFO) on the backfat fatty acid profile. The subjects of research were 60 crossbred gilts divided into 6 groups, which were fed different levels of conjugated linoleic acid or sunflower oil (0.5, 1.0 and 2.0%, respectively). All fatteners were kept and fed under standardized conditions. Animals were slaughtered at 95 kg of body weight. Fatty acid profile was determined in samples of backfat from each animal using gas chromatography. The significance of differences between groups was verified by Duncan's test. In the present study, a beneficial effect of adding CLA was that it reduced saturated fatty acids and increased unsaturated fatty acids in the adipose tissue (backfat) of pigs as compared to fatteners receiving SFO. The amount of monounsaturated fatty acids in the backfat decreased with increasing amounts of CLA or SFO. Modifying the fatty acid profile of pig backfat through addition of CLA or SFO would be beneficial to the health of consumers because relatively large amounts of fat pork, including backfat, are added to the sausages.
