Amino-acid sequence of fragment A, an enzymically active fragment from diphtheria toxin.

The amino-acid sequence of Fragment A from diphtheria toxin is reported. Fragment A (molecular weight, Mr, 21,145) is the major enzymically active fragment produced upon activation of the intact toxin (Mr about 60,000) by limited tryptic digestion and reduction. It, or a similar fragment, is believed responsible for the inhibition of protein synthesis in animal cells exposed to the toxin. Fragment A, which corresponds to the amino terminus of the toxin, is shown here to consist of three major forms (190, 192, and 193 residues) resulting from cleavage by trypsin adjacent to any of three closely spaced arginine residues. All three forms are enzymically active.

The NH 2-terminal amino acid sequence of a major allergenic fragment (TM2) from cod was elucidated and shown to have the following structure: ▪ Tryptic hydrolysis of the molecule yielded 6–8 oligopeptides. No allergenicity was found for either of these peptides or for fragment TM 2 after the removal of the 17 NH 2-terminal residues.

Macroglobulin Structure: Homology of Mu and Gamma Heavy Chains of Human Immunoglobulins

The amino acid sequence of fragments obtained by cyanogen bromide cleavage of the mu-chain of a human gammaM-globulin is homologous to the NH(2)-terminal sequences of the gamma-chain of human and rabbit gammaG-globulins and is related to that of human light chains. This supports the hypothesis that light and heavy chains evolved from a common ancestral gene.
