Transgenerational Impacts of Oxidative Damage Induced by Prenatal Ethanol Exposure on Spatial Learning/Memory and BDNF in the Hippocampus of Male Rats

Alcohol consumption during pregnancy harms fetal development, leading to various physical and behavioral issues. This study investigates how prenatal ethanol exposure triggers oxidative stress (OS) and affects neurotrophic factors (NTFs), particularly brain-derived growth factor (BDNF) gene expression in the hippocampus, influencing learning and memory decline across two generations of male offspring from ethanol-exposed female rats. A rat model of fetal alcohol spectrum disorder (FASD) was initially generated to reflect on the deficits in the first generation, and then those transmitted via the male germline to the unexposed male ones. The pregnant rats were thus divided into four groups, namely, the control group (CTRL) receiving only distilled water (DW), and three groups being exposed to ethanol (20 %, 4.5 g/kg) by oral gavage, during the first 10-day gestation (FG), the second 10-day gestation (SG), and the entire gestation (EG) periods. Subsequent Morris water maze (MWM) tests on male offspring revealed spatial learning deficits during the second and entire gestational periods in both generations. Analysis of antioxidant enzyme activity including glutathione peroxidase (GPx), superoxide dismutase (SOD), and malondialdehyde (MDA), and BDNF gene expression in the hippocampus further highlighted the impacts of prenatal ethanol exposure. The study results demonstrated that prenatal ethanol exposure caused spatial learning/memory deficits during the SG and EG, altered antioxidant enzyme activity, and reduced BDNF gene expression in both generations. The findings underscore the role of OS in developmental and behavioral issues in FASD rat models and suggest that lasting transgenerational effects in the second generation may stem from alcohol-induced changes.

Effects of diuron and two of its metabolites in biochemical markers and behavior of zebrafish larvae.

Diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is an herbicide used in many crops, including sugar cane cultivation. It is commonly found in aquatic ecosystem and is of high concern due to its ability to persist in the environment. Diuron metabolites include DCA (3,4-dichloroaniline) and DCPMU (3-(3,4-dichlorophenyl-1-methylurea). The objective of this study was to evaluate the effects of diuron and two of its metabolites in zebrafish (Danio rerio) developing embryos, from biochemical to individual level. Activities of the enzymes acetylcholinesterase (AChE), catalase (CAT), glutathione-S-transferase (GST), andlactate dehydrogenase (LDH) and the levels of lipid peroxidation (LPO), swimming activity, and body length were investigated after an exposure of 120h, and the heart rate was determined after 48h of exposure. The range of concentrations tested was 0.003-3.000mg/L diuron, 0.020-1.500mg/L DCA, and 0.020-2.100mg/L DCPMU. Results showed that AChE activity was inhibited by diuron (3.000mg/L) and DCPMU (0.326, 0.828mg/L). However, the swimming activity of fish larvae exposed to diuron or its metabolites was not affected. The CAT was induced by DCPMU, and GST was induced by diuron. This suggests that CAT is acting to cope with oxidative stress induced by DCPMU and GST might have a role in the detoxification of diuron. In addition, larvae exposed to DCA (0.633 and 1.500mg/L) had a reduction in their length, and larvae exposed to diuron (0.754 and 3.000mg/L) and DCA (0.267, 0.633, and 1.500mg/L) presented bradycardia, suggesting cardiotoxicity. Overall, results indicate that diuron, DCA, or DCPMU induces adverse effects during the early phases of zebrafish development, such as the impairment of neurotransmission and cardiovascular function and alterations in antioxidant enzymes and growth. Diuron appeared as more toxic than its metabolites since the lowest LOEC (0.012mg/L) and higher integrated biomarker response (IBR) values were obtained with exposure to this herbicide. Furthermore, as it is fast degraded into DCA and DCPMU, which also affected the zebrafish developing embryos at environmentally relevant concentrations, its use might be of concern in ecosystems that receive agriculture runoff due to their potential adverse effects to aquatic biota.

The importance of oxidative stress in the pathogenesis and treatment of bipolar affective disorder: a review of the literature

Introduction: Bipolar affective disorder (BD), also known as manic-depressive illness, is a chronic and recurrent psychiatric disorder characterised by significant mood disturbances. It is one of the leading causes of disability worldwide and is associated with a high risk of suicide. Recent studies highlight the role of oxidative stress (OS) in the pathogenesis of BD. The body's pro/antioxidant imbalance adversely affects cellular and molecular processes. Aim: The aim of this review is to synthesise the current state of knowledge on the role of OS in the aetiology and course of BD, including key biomarkers and potential therapeutic interventions. Methods: A review of the scientific literature was conducted, including articles published between 2000 and 2024. Searches were conducted in PubMed, Scopus and Web of Science databases, using the following keywords: 'bipolar disorder', 'oxidative stress', 'antioxidants', 'biomarkers', 'mitochondrial dysfunction', 'redox homeostasis', 'treatment'. Results: Results indicate that patients with BD have elevated levels of OS markers, including increased lipid peroxidation, altered antioxidant enzyme activity and impaired redox homeostasis. Treatment with lithium and other mood stabilisers may modulate levels of OS markers, which is one potential mechanism of drug action. However, inconclusive data suggest the need for further research to clarify the relationship between OS and BD. Conclusions: OS plays an important role in the pathophysiology of BD, offering potential directions for therapeutic interventions. Understanding the complex interactions between OS and BD may lead to the development of more targeted therapies aimed at reducing oxidative damage and improving patient health. Pseudochrobactrum asaccharolyticum mitigates arsenic induced oxidative stress of maize plant by enhancing water status and antioxidant defense system

BackgroundOxidative stress mediated by reactive oxygen species (ROS) is a common denominator in arsenic toxicity. Arsenic stress in soil affects the water absorption, decrease stomatal conductance, reduction in osmotic, and leaf water potential, which restrict water uptake and osmotic stress in plants. Arsenic-induced osmotic stress triggers the overproduction of ROS, which causes a number of germination, physiological, biochemical, and antioxidant alterations. Antioxidants with potential to reduce ROS levels ameliorate the arsenic-induced lesions. Plant growth promoting rhizobacteria (PGPR) increase the total soluble sugars and proline, which scavenging OH radicals thereby prevent the oxidative damages cause by ROS. The main objective of this study was to evaluate the potential role of Arsenic resistant PGPR in growth of maize by mitigating arsenic stress.MethodologyArsenic tolerant PGPR strain MD3 (Pseudochrobactrum asaccharolyticum) was used to dismiss the ‘As’ induced oxidative stress in maize grown at concentrations of 50 and 100 mg/kg. Previously isolated arsenic tolerant bacterial strain MD3 “Pseudochrobactrum asaccharolyticum was used for this experiment. Further, growth promoting potential of MD3 was done by germination and physio-biochemical analysis of maize seeds. Experimental units were arranged in Completely Randomized Design (CRD). A total of 6 sets of treatments viz., control, arsenic treated (50 & 100 mg/kg), bacterial inoculated (MD3), and arsenic stress plus bacterial inoculated with three replicates were used for Petri plates and pot experiments. After treating with this MD3 strain, seeds of corn were grown in pots filled with or without 50 mg/kg and 100 mg/kg sodium arsenate.ResultsThe plants under arsenic stress (100 mg/kg) decreased the osmotic potential (0.8 MPa) as compared to control indicated the osmotic stress, which caused the reduction in growth, physiological parameters, proline accumulation, alteration in antioxidant enzymes (Superoxide dismutase-SOD, catalase-CAT, peroxidase-POD), increased MDA content, and H2O2 in maize plants. As-tolerant Pseudochrobactrum asaccharolyticum improved the plant growth by reducing the oxidation stress and antioxidant enzymes by proline accumulation. PCA analysis revealed that all six treatments scattered differently across the PC1 and PC2, having 85.51% and 9.72% data variance, respectively. This indicating the efficiency of As-tolerant strains. The heatmap supported the As-tolerant strains were positively correlated with growth parameters and physiological activities of the maize plants.ConclusionThis study concluded that Pseudochrobactrum asaccharolyticum reduced the ‘As’ toxicity in maize plant through the augmentation of the antioxidant defense system. Thus, MD3 (Pseudochrobactrum asaccharolyticum) strain can be considered as bio-fertilizer.

Understanding the interplay between strigolactone and nitric oxide in alleviating cadmium-induced toxicity in Artemisia annua

Metal stress severely limits the development and productivity of medicinal plants like A. annua. Strigolactone (SL), a relatively new plant hormone, and nitric oxide (NO) have demonstrated potential in mitigating HM-related symptoms in plants. Both SLs and NO are regulatory signals that play different roles under stressful conditions. At the moment, it is unclear how SLs and NO interact in plants under cadmium (Cd) stress. In this study, Artemisia annua was utilized to study the role and interaction of SL and NO for Cd stress tolerance. To investigate this, we undertook a detailed physiological, anatomical, and histochemical examination to determine the mechanisms by which SLs and NO reduce the effects of Cd stress on A. annua plants. Cadmium exposure hindered the growth of A. annua plants, influenced several chlorophyll fluorescence attributes, altered antioxidant enzyme activity, lowered cell viability, and caused lipid peroxidation, oxidative damage, and chloroplast ultrastructure abnormalities. A. annua plants grew better when treated with 4 μM synthetic SL analogue GR24 and 200 μM NO donor sodium nitroprusside. Meanwhile, under Cd stress, GR24 and/or SNP spray effectively boosted chlorophyll and carotenoid content, increased antioxidant enzyme activity, and elevated proline and glutathione levels. Treatment with SNP and/or GR24 improved chloroplast ultrastructure and chlorophyll fluorescence characteristics in plants under Cd-stress, resulting in better photosynthetic performance. It also reduced Cd accumulation, increased membrane permeability, and regulated stomatal behavior, resulting in enhanced stomatal conductance in Cd-stressed plants. The application of these growth regulators to plants under Cd-stress mitigated the decrease in glandular trichome dimensions and artemisinin content as observed from the HPLC results. Our results indicate that GR24 and NO, either individually or in combination, can be very effective in reducing Cd-induced damage in A. annua.

Cultivar-specific regulation of antioxidant enzyme activity and stomatal closure confer tolerance of wheat to elevated ozone: A two-year open-field study with five cultivars

Elevated concentrations of tropospheric ozone (O3) pose a significant threat to food production in many regions of the world. We conducted a two-year experiment with five winter wheat (Triticum aestivum L.) cultivars, Yangmai16 (Y16), Yangmai23 (Y23), Zhenmai12 (Z12), Yangmai29 (Y29), and Yangmai30 (Y30), to assess photosynthetic traits, lipid peroxidation, and antioxidant systems under ambient (A-O3) or elevated (E-O3; 1.5 times A-O3) O3 in a Free-Air O3Concentration Enrichment system (FACE). E-O3 accelerated leaf senescence, manifested by marked reductions in photosynthetic rate, stomatal conductance, total antioxidant capacity, and photosynthetic pigment content, accompanied by altered antioxidant enzyme activity and a significant increase in malondialdehyde content (an indicator of lipid peroxidation). Most parameters showed significant inter-cultivar differences, as well as interactions between O3 and cultivar. The response of the antioxidant system in wheat flag leaves was influenced by O3 exposure levels. The response of antioxidant enzymes at the grain filling stage and the timely closure of stomata conferred tolerance to O3. This tolerance was associated with higher antioxidant enzyme activity at the anthesis stage but was not related to the size of stomata. This study provides a crucial theoretical foundation for further breeding O3-tolerant cultivars.

Antidiarrhoeal screening of Himalayan edible plant Begonia rubrovenia and its marker followed by its validation using computational analysis

BackgroundDiarrhoea has become one of the major areas of concern due to its high mortality rate contributing it to be the second largest cause of death in world. To explore the effectiveness of medicinal plant, the present investigation was undertaken to scientifically justify the traditional claim of the ethanolic root extract of the plant Begonia rubrovenia (EBV) against diarrhoea.ResultsEBV was standardized using HPLC with quercetin as marker and was further subjected to normal fecal excretion study at 100, 200 and 300 mg/kg, p.o. along with quercetin and loperamide. The study confirmed the effectiveness of EBV at 200 and 300 mg/kg followed by quercetin. In castor oil induced diarrhoea rat model, EBV at 200 and 300 mg/kg significantly delayed onset of diarrhoea, reduced the diarrhoeal faecal output which contributed in higher % protection. The effectiveness of EBV at 200 mg/kg was also confirmed through gastrointestinal motility, fluid accumulation and PGE2 induced enteropooling tests. EBV and its marker quercetin also reduced the elevated level of NO and cytokines and restored the alterations in antioxidant enzymes, ions and enhanced Na+/K+–ATPase activity. Molecular docking, dynamics and network pharmacology study confirmed the role of quercetin in modulating the inflammatory mediators IL-1β, TNF-α and EP3 prostanoid receptor, where quercetin formed more stable complex with EP3 prostanoid receptor.ConclusionThe study has scientifically justified the traditional use of the plants B. rubrovenia in treating diarrhoea, where quercetin played a critical role in the observed antidiarrhoeal potential of B. rubrovenia contributing in maintaining electrolyte balance, antioxidant status and inhibiting inflammatory mediators.

Mitigation of lens opacification by a functional food in a diabetic rodent model

The current study was designed to test a functional food (FF) mixture containing aldose reductase inhibitors and antiglycation bioactive compounds for suppressing the onset and progression of cataracts in a diabetic rat model. Two-month-old Sprague Dawley rats were grouped as control (C), diabetes untreated (D), and diabetic rats treated with FF at two doses (FF1 = 1.35 g and FF2 = 6.25 g/100g of diet). Diabetes was induced by a single injection of streptozotocin. The FF is a mixture of amla, turmeric, black pepper, cinnamon, ginger, and fenugreek added to the rodent diet. The status of cataracts was monitored weekly by a slit lamp examination for 20 weeks, after which animals were sacrificed to collect eye lenses. Feeding FF1 and FF2 to diabetic rats yielded a significant anti-hyperglycaemic effect and marginally prevented body weight loss. FF delayed cataract progression, and FF2 showed better efficacy than FF1. FF prevented the loss of lens crystallins and their insolubilization in diabetic rats. The antioxidant potential of FF was evident with the lowered protein carbonyls, lipid peroxidation, and prevention of altered antioxidant enzyme activities induced by diabetes. These studies demonstrate the efficacy of plant-derived dietary supplements against the onset and progression of cataracts in a well-established rat model of diabetic eye disease.

Lipid changes and molecular mechanism inducing cuproptosis in Cryptocaryon irritans after copper–zinc alloy exposure

Cryptocaryons irritans is a ciliate parasite responsible for cryptocaryoniasis, leading to considerable economic losses in aquaculture. It is typically managed using a copper–zinc alloy (CZA), effectively diminishing C. irritans infection rates while ensuring the safety of aquatic organisms. Nevertheless, the precise mechanism underlying cuproptosis induced C. irritans mortality following exposure to CZA remains enigmatic. Therefore, this study delves into assessing the efficacy of CZA, investigate cuproptosis as a potential mechanism of CZA action against C. irritans, and determine the alterations in antioxidant enzymes, peroxidation, and lipid metabolism. The mRNA expression of dihydrolipoamide S-acetyltransferase was upregulated after 40 and 70 min, while aconitase 1 was implicated in cuproptosis following 70 min of CZA exposure. Furthermore, the relative mRNA levels of glutathione reductase experienced a significant increase after 40 and 70 min of CZA exposure. In contrast, the relative mRNA levels of glutathione S-transferase and phospholipid-hydroperoxide glutathione peroxidase were significantly decreased after 70 min, suggesting a disruption in antioxidant defense and an imbalance in copper ions. Lipidomics results also unveiled an elevation in glycerophospholipids metabolism and the involvement of the lipoic acid pathway, predominantly contributing to cuproptosis. In summary, exposure to CZA induces cuproptosis in C. irritans, impacts glutathione-related enzymes, and alters glycerophospholipids, consequently triggering lipid oxidation.

Physiological and Transcriptome Analysis of the Effects of Exogenous Strigolactones on Drought Responses of Pepper Seedlings.

Drought stress significantly restricts the growth, yield, and quality of peppers. Strigolactone (SL), a relatively new plant hormone, has shown promise in alleviating drought-related symptoms in pepper plants. However, there is limited knowledge on how SL affects the gene expression in peppers when exposed to drought stress (DS) after the foliar application of SL. To explore this, we conducted a thorough physiological and transcriptome analysis investigation to uncover the mechanisms through which SL mitigates the effects of DS on pepper seedlings. DS inhibited the growth of pepper seedlings, altered antioxidant enzyme activity, reduced relative water content (RWC), and caused oxidative damage. On the contrary, the application of SL significantly enhanced RWC, promoted root morphology, and increased leaf pigment content. SL also protected pepper seedlings from drought-induced oxidative damage by reducing MDA and H2O2 levels and maintaining POD, CAT, and SOD activity. Moreover, transcriptomic analysis revealed that differentially expressed genes were enriched in ribosomes, ABC transporters, phenylpropanoid biosynthesis, and Auxin/MAPK signaling pathways in DS and DS + SL treatment. Furthermore, the results of qRT-PCR showed the up-regulation of AGR7, ABI5, BRI1, and PDR4 and down-regulation of SAPK6, NTF4, PYL6, and GPX4 in SL treatment compared with drought-only treatment. In particular, the key gene for SL signal transduction, SMXL6, was down-regulated under drought. These results elucidate the molecular aspects underlying SL-mediated plant DS tolerance, and provide pivotal strategies for effectively achieving pepper drought resilience.

Nano silicon dioxide reduces cadmium uptake, regulates nutritional homeostasis and antioxidative enzyme system in barley seedlings (Hordeum vulgare L.) under cadmium stress

Cadmium (Cd) toxicity is one of the most severe environmental threats inhibiting crop growth and productivity. Strategies to mitigate the adverse effects of Cd stress on plants are under scrutiny. Nano silicon dioxide (nSiO2) is an emerging material and could protect plants against abiotic stress. Can nSiO2 alleviate Cd toxicity in barley, and the possible mechanisms are poorly understood. A hydroponic experiment was conducted to study the mitigation effects of nSiO2 on Cd toxicity in barley seedlings. The results showed that the application of nSiO2 (5, 10, 20, and 40mg/L) increased barley plant growth and chlorophyll and protein content, improving photosynthesis, compared with Cd-treated alone. Specifically, 5-40mg/L nSiO2 addition increased net photosynthetic rate (Pn) by 17.1, 38.0, 30.3, and - 9.7%, respectively, relative to the Cd treatment alone. Furthermore, exogenous nSiO2 reduced Cd concentration and balanced mineral nutrient uptake. The application of 5-40mg/L nSiO2 decreased Cd concentration in barley leaves by 17.5, 25.4, 16.7, and 5.8%, respectively, relative to the Cd treatment alone. Moreover, exogenous nSiO2 lowered malondialdehyde (MDA) content by 13.6-35.0% in roots, and by 13.5-27.2% in leaves, respectively, compared with Cd-treated alone. Besides, nSiO2 altered antioxidant enzyme activities and alleviated detrimental effects on Cd-treated plants, attaining maximal values at 10mg/L nSiO2. These findings revealed that exogenous nSiO2 application may be a viable option for addressing Cd toxicity of barley plants.

Anti-epileptic Activity of Rosmarinus offi cinalis Extracts with Male Rats Experiment

Rosmarinus offi cialis L., Lamiaceae, has been used to treat a range of illnesses in Iraq for decades and is regarded as a desirable species due to its medicinal properties and antioxidant properties. Aim of study: The main aim of this study was to study more about how the R. offi cialis L plant may help to reduce the negative eff ects of epilepsy. Materials and methods: To estimate the alteration of antioxidant enzymes and lipid peroxidation. Fifty male rats were split into fi ve groups: control (no additions), FeCl3 group, and the rest of the groups were fed R. offi cialis L. components (fl avonoid, glycoside, and alkaloids). Results: β-asecretase, Na and K levels raised in FeCl3 and other R. offi cialis compounds groups while. In contrast to the FeCl3 group and FeCl3 groups containing R. offi cialis L. plant components reduced fucose, vit-C, and Zn levels. In compared to the control group, the activity of oxidant enzymes and lipid peroxidation resulted in a signifi cant amount of glutathione (GSH), malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT) in the FeCl3 group; however, R. offi cialis components increased the levels of MDA and SOD, whilst reducing GSH, and CAT. Conclusion: R. offi cinalis extraction components have an eff ects on -asecretase, fucose, and vit-C in male rats, in addition to antioxidant activity and may increase or decrease lipid peroxidation while also improving antioxidative enzymes significantly. The active components of the R. offi cinalis L. plant might be used as one of the next generation of anti-epileptic medications.

Impact of Bariatric Surgery on the Stability of the Genetic Material, Oxidation, and Repair of DNA and Telomere Lengths

Obesity causes genetic instability, which plays a key-role in the etiology of cancer and aging. We investigated the impact of bariatric surgery (BS) on DNA repair, oxidative DNA damage, telomere lengths, alterations of antioxidant enzymes and, selected proteins which reflect inflammation. The study was realized with BS patients (n = 35). DNA damage, base oxidation, BER, and NER were measured before and 1 month and 6 months after surgery with the single-cell gel electrophoresis technique. SOD and GPx were quantified spectrophotometrically, malondealdehyde (MDA) was quantified by HPLC. Telomere lengths were determined with qPCR, and plasma proteome profiling was performed with high-resolution mass spectrophotometry. Six months after the operations, reduction of body weight by 27.5% was observed. DNA damage decreased after this period, this effect was paralleled by reduced formation of oxidized DNA bases, a decline in the MDA levels and of BER and NER, and an increase in the telomere lengths. The activities of antioxidant enzymes were not altered. Clear downregulation of certain proteins (CRP, SAA1) which reflect inflammation and cancer risks was observed. Our findings show that BS causes reduced oxidative damage of DNA bases, possibly as a consequence of reduction of inflammation and lipid peroxidation, and indicate that the surgery has beneficial long-term health effects.

Neuroprotective Effect of Vitamin D on Behavioral and Oxidative Parameters of Male and Female Adult Wistar Rats Exposed to Mancozeb (manganese/zinc ethylene bis-dithiocarbamate)

The constant exposure of rural workers to pesticides is a serious public health problem. Mancozeb (MZ)is a pesticide linked to hormonal, behavioral, genetic, and neurodegenerative effects, mainly related to oxidative stress. Vitamin D is a promising molecule that acts as a protector against brain aging. This study aimed to evaluate the neuroprotective role of vitamin D in adult male and female Wistar rats exposed to MZ. Animals received 40mg/kg of MZ i.p. and 12.5μg/kg or 25μg/kg vitamin D by gavage, twice a week, for 6weeks. The concentration of manganese had a significant increase in the hippocampus of both sexes and in the striatum of females, unlike zinc, which did not show a significant increase. MZ poisoning led to mitochondrial changes in brain tissues and promoted anxiogenic effects, especially in females. Alterations in antioxidant enzymes, mainly in the catalase activity were observed in intoxicated rats. Taken together, our results showed that exposure to MZ leads to the accumulation of manganese in brain tissues, and the behavior and metabolic/oxidative impairment were different between the sexes. Furthermore, the administration of Vitamin D was effective in preventing the damage caused by the pesticide.

Impact of exogenous hydrogen peroxide on osteogenic differentiation of broiler chicken compact bones derived mesenchymal stem cells.

The effects of hydrogen peroxide (H2O2) on the osteogenic differentiation of primary chicken mesenchymal stem cells (MSCs) were investigated. MSCs were subjected to an osteogenic program and exposed to various concentrations of H2O2 for 14 days. Results showed that high concentrations of H2O2 (200 and 400nM) significantly increased pro-apoptotic marker CASP8 expression and impaired osteogenic differentiation, as indicated by decreased mRNA expression levels of osteogenesis-related genes and reduced in vitro mineralization. In contrast, long-term H2O2 exposure promoted basal expression of adipogenic markers at the expense of osteogenesis in MSCs during osteogenic differentiation, and increased intracellular reactive oxygen species (ROS) production, as well as altered antioxidant enzyme gene expression. These findings suggest that long-term H2O2-induced ROS production impairs osteogenic differentiation in chicken MSCs under an osteogenic program.

EFFECTS OF NAUCLEA HATIFOLIA ON OXIDATIVE STRESS IN WISTER ALBINO RATS IN CRUDE OIL TOXICITY

Crude oil toxicity created significant inhibition (p<0.05) of antioxidant enzymes(superoxide dismutase and peroxidase) but activated catalase (CAT) and malondialdehyde(MDA) levels in hepatic cells when compared with control. Crude oil toxicity also inducedsignificant increase in aspartate aminotransferase (AST), alanine aminotransferase (ALT) andalanine phosphatase (ALP) in the serum. The alteration of antioxidant enzymes and other liverparameters may be attributed to oxidative stress and adaptive response. The rats that weresimultaneously treated with crude oil and N. hatifolia stem bark extract however, maintained or recorded relatively no significant (p>0.05) alteration in biochemical parameters in the serum and hepatic cells when compared with control. These non-significant (p>0.05) alteration may be attributed to the antioxidant and anti-inflammatory properties of N. hatifolia which may explain the probable therapeutic and protective role of N. hatifolia in crude oil toxicity.

Effects of diluted seawater on the growth, antioxidation capacity and accumulation of bioactive compounds of the brown alga, Sargassum muticum

Environmental condition is a significant factor that directly affects the growth, metabolism and productivity of marine algae. This study examined the influences of diluted seawater on the growth, antioxidation capacity and accumulation of bioactive compounds of the marine macroalga, Sargassum muticum. Compared to the natural seawater (30 ppt, parts per thousand) control, the specific growth rate was clearly elevated at diluted seawater with 15 and 20 ppt during the initial 3 days, and was not statistically different at diluted seawater from 15 to 30 ppt at 15 days, while it decreased significantly at a low seawater concentration of 10 ppt. Chlorophyll a, carotenoids and total protein content fluctuated slightly under the different seawater solutions, suggesting that the algal cells underwent metabolic regulation during the period of adaptation to culture media changes. Changes in seawater concentration altered antioxidant enzyme activity, for instance, total superoxide dismutase, catalase and peroxidase, especially at low seawater solutions with 10 and 15 ppt. The malondialdehyde content also increased substantially, indicating that oxidative damage occurred in diluted seawater below 15 ppt. Moreover, the alginate content increased with decreasing seawater concentration, the alginate productivity was significantly increased by 22.32%–54.79% in all dilutions at 3 days and this promotion continued only at 15 and 20 ppt salinities after 7 days. The fucoxanthin content reduced significantly in the 10 and 15 ppt dilutions at 15 days, however, its productivity increased slightly at 10 and 15 ppt salinities in the first 3 days. The phlorotannin content increased slightly in the seawater dilutions with 20 ppt at 3 and 7 days, meanwhile, its productivity was significantly elevated during the initial 3 days at all seawater dilutions and the highest value was found at 20 ppt treatment. These results provide valuable data for the development and management of the aquaculture of S. muticum.

Oxidative and osmoregulatory effects of imidacloprid, cadmium, and their combinations on Daphnia magna

Oxidative stress and osmoregulatory system damage-inducing potential of binary mixtures of neonicotinoid insecticide imidacloprid (IMI) and Cd2+ in Daphnia magna were evaluated. Animals were subjected to subchronic (7 days) and acute (48 h) of IMI and Cd2+ effects with single and binary mixtures. ATPase and antioxidant enzyme activities with lipid peroxidation were measured. Morphometric characteristics were also evaluated. Response patterns showed variability due to the duration, concentration, and toxicant type. While the enzyme activities mostly showed a decreasing trend upon the subchronic IMI effect, there was an increasing trend after the Cd2+. Declined enzyme activities were more pronounced with the acute higher IMI+Cd2+ exposure. Ca2+-ATPase and CAT were the most sensitive biomarkers in the toxicity response. IMI+Cd2+ exposures are appeared to increase their toxic effects due to their oxidative potential. ATPase inhibition and antioxidant enzyme alterations with a decrease in morphometric characteristics in Daphnia even at their low concentrations of IMI and Cd2+ show evidence of their toxicities on aquatic life. It was emphasized that investigating the combined effects of toxicants at their environmental level based on the multi-biomarker approach is essential in toxicity evaluation.

Application of gamma irradiation on morphological, biochemical, and molecular aspects of wheat (Triticum aestivum L.) under different seed moisture contents

Nuclear technology is currently used as a tool in mutation breeding to improve crops by increasing genetic variation. The ionization of gamma rays produces large amounts of free radicals, simulating stressors in the natural environment. To avoid gamma-ray-induced oxidative stress, plants use antioxidant defense systems. Exposure of plants to irradiation can affect the germination, growth, and production of metabolites. Plants' sensitivity to irradiation depends on genetic and environmental factors such as moisture content. For this purpose, the effects of different gamma irradiation doses [0, 100, 200, 300, and 400 Gray (Gy)] and different seed moisture contents (7, 13, and 19%) on traits such as seed germination, seedling growth, molecular and biochemical alterations in antioxidant enzymes were examined in the current study. Based on the results, the highest seed germination percentage was observed in the interaction effect of seed moisture at 13% with an irradiation dose of 400 Gy (98.89%). Seedling survival percent and seedling length decreased with increasing doses of gamma irradiation at different seed moisture contents. Increasing gamma irradiation doses were reduced root and stem fresh and dry weight, and root and stem length. The highest level of catalase enzyme activity and expression was observed at 200 and 300 Gy irradiation doses at different moisture contents. The peroxidase and polyphenol oxidase gene expression were reduced at all contents of gamma irradiation doses and seed moisture compared to the control. It can be concluded that the dose of 200–300 Gy of gamma irradiation reduced plant growth by 30% in terms of fresh and dry weight and length of plants, as well as enhanced the expression of antioxidant enzymes. The results of this study could help plant breeders select an appropriate dose rate in wheat for further research.
