Biogenic synthesis, characterization, and in vitro biological investigation of silver oxide nanoparticles (AgONPs) using Rhynchosia capitata

The current research aimed to study the green synthesis of silver oxide nanoparticles (AgONPs) using Rhynchosia capitata (RC) aqueous extract as a potent reducing and stabilizing agent. The obtained RC-AgONPs were characterized using UV, FT-IR, XRD, DLS, SEM, and EDX to investigate the morphology, size, and elemental composition. The size of the RC-AgONPs was found to be ~ 21.66 nm and an almost uniform distribution was executed by XRD analysis. In vitro studies were performed to reveal biological potential. The AgONPs exhibited efficient DPPH free radical scavenging potential (71.3%), reducing power (63.8 ± 1.77%), and total antioxidant capacity (88.5 ± 4.8%) to estimate their antioxidative power. Antibacterial and antifungal potentials were evaluated using the disc diffusion method against various bacterial and fungal strains, and the zones of inhibition (ZOI) were determined. A brine shrimp cytotoxicity assay was conducted to measure the cytotoxicity potential (LC50: 2.26 μg/mL). In addition, biocompatibility tests were performed to evaluate the biocompatible nature of RC-AgONPs using red blood cells, HEK, and VERO cell lines (< 200 μg/mL). An alpha-amylase inhibition assay was carried out with 67.6% inhibition. Moreover, In vitro, anticancer activity was performed against Hep-2 liver cancer cell lines, and an LC50 value of 45.94 μg/mL was achieved. Overall, the present study has demonstrated that the utilization of R. capitata extract for the biosynthesis of AgONPs offers a cost-effective, eco-friendly, and forthright alternative to traditional approaches for silver nanoparticle synthesis. The RC-AgONPs obtained exhibited significant bioactive properties, positioning them as promising candidates for diverse applications in the spheres of medicine and beyond.

Quantification of the Anti-diabetic Effect of Allium cepa.

Allium cepa, or onion, boosts numerous health benefits, including anti-diabetic effects. Its rich array of antioxidants and sulfur compounds not only aids heart health by lowering cholesterol and blood pressure but also exhibits anti-inflammatory properties. Onion's antibacterial and antiviral properties help combat infections, while its compounds like quercetin show promise in cancer prevention. Additionally, Allium cepa supports respiratory health by relieving coughs and colds and aids digestion with its prebiotic properties. Incorporating onions into a balanced diet can enhance overall well-being, including managing blood sugar levels in individuals with diabetes. This study aims to determine if the ethanolic extract from the dried peel of Allium cepa holds potential as an anti-diabetic agent, with a focus on its ability to manage diabetes and reduce blood sugar levels. To prepare the ethanolic extract from dried onion peel, the peel was finely ground and soaked in ethanol. The mixture was then agitated and filtered to separate the liquid extract. Finally, the filtrate was concentrated using methods such as rotary evaporation or vacuum distillation to obtain a concentrated extract for further analysis like alpha-amylase inhibition assay and alpha-glucosidase inhibition assay. The ethanolic extracts derived from dried onion peel demonstrate inhibition of alpha-glucosidase, leading to reduced blood glucose levels. Additionally, this inhibition prompts an increase in insulin production. The study underscores that the efficacy of the ethanolic extract of dried onion peel increases with concentration. It highlights the presence of beneficial compounds like total phenolics, flavonoids, quercetin, and its derivatives in onion peel, known for their therapeutic roles in cardiovascular health, weight management, diabetes control, cancer prevention, and antimicrobial activity. These findings affirm the hypoglycemic and anti-diabetic properties of Allium cepa's ethanolic leaf extract.

Alpha-amylase and Alpha-glucosidase Inhibitory Activities of Philippine Indigenous Medicinal Plants

In the Philippines, medicinal plants still provide the first line of therapeutic remedies and their reported efficacy in traditional treatments provides an opportunity for drug discovery and development. The study aimed to evaluate in vitro commonly used Philippine medicinal plants for their alpha-amylase and alpha-glucosidase inhibitory activities as potential sources of antidiabetic agents. The study emphasised the inhibitory activities of 54 medicinal plants against α-amylase and α-glucosidase enzymes. The findings revealed 7 medicinal plants with the highest alpha-glucosidase inhibitory activity of ≥ 50% and 3 with alpha-amylase inhibitory activity of ≥ 20%. Ethyl acetate extract of Cycas sp. exhibited the highest α-glucosidase inhibitory activity with 83.87% ± 1.52 followed by the aqueous extract of cf. Calyptranthera sp. With 82.07% ± 0.14 with Acarbose as the reference standard (99.71% ± 0.63) at 10µg/mL (w/v) concentration. For the alpha-amylase inhibitory assay, the highest bioactivity was observed in ethyl acetate extract of Curcuma longa L. (39.44% ± 1.56) with Acarbose as the reference standard (51.59% ± 0.98) at 10µg/mL (w/v) concentration. These findings suggest the potential of the above-mentioned plants as sources of alpha-amylase and alpha-glucosidase inhibitors that may be used as antidiabetic agents.

Biogenic Synthesis and Characterisation of Flueggea leucopyrus Willd Leaf Mediated Copper Nanoparticles for Antibacterial, Antioxidant, and Antidiabetic Activities

IntroductionThe chemical synthesis of metal Nanoparticles could be changed into green synthesis if solvents, stabilising agents, and reducing agents are environmentally friendly. Our aims were to synthesise and characterise bioconjugated copper nanoparticles (CuNPs) and evaluate their antibacterial, antioxidant, and antidiabetic activities. MethodsThis study was made to synthesise CuNPs using Flueggea leucopyrus Willd leaf extract. The produced CuNPs were characterised using UV-visible, Fourier Transform Infrared Spectroscopy (FTIR), Field Emission Scanning Electron Microscopy (FESEM), TEM, EDX, and Zeta-potential analytical methods. The antibacterial, antioxidant, and antidiabetic properties of CuNPs were screened through disc diffusion methods, 2,2-diphenyl-1-picrylhydrazyl radical quenching, and alpha-amylase inhibition assay, respectively. ResultsThe CuNPs are poly-dispersed, spherical, crystalline, and stable with an average particle size of 20 nm. These nanoparticles show antibacterial activity and exhibit the same inhibition zone against S aureus and P aruginosa (13 mm), a strong zone for E faecalis, and no inhibition zone for E coli bacteria. The CuNPs demonstrated dose-dependent sequencing and inhibition activities with an IC50 value of 100.06 ppm and 435.68 ppm, respectively. ConclusionsThis study shows that these poly-disperse CuNPs could act as a bacterial resistance agent, antioxidant, and antidiabetic drug. It concluded that CuNPs made from medicinal plant leaf extract have good inhibitory potential against alpha-amylase, one of the main pharmacological targets for treating type 2 diabetes.

&lt;i&gt;In vitro&lt;/i&gt; studies on the antidiabetic and antibacterial activity of biosynthesized silver nanoparticles with aqueous extract of &lt;i&gt;Moringa oleifera&lt;/i&gt; leaf

Silver nanoparticles having applications in the field of medicine and biology are shown to have tremendous health benefits. Diabetes is still on rampage and antibacterial resistance is currently a global health challenge. This study focuses on the in-vitro antidiabetic and antibacterial activity of biosynthesized silver nanoparticles with aqueous extract of Moringa oleifera leaf. An aliquot (5 mL) of extract sample was added to 50 mL of 1 mM aqueous AgNO3. Reaction mixtures were heated and maintained at 700C for 10 minutes. Colour change to dark brown solution and a UV-vis spectrum peak at 400 nm confirmed silver nanoparticle synthesis. The antidiabetic activity of the nanoparticle was studied using an in vitro alpha-amylase inhibition assay. Acarbose was used as antidiabetic control drug. Disc diffusion method was used for antibacterial susceptibility testing on Mueller-Hinton agar medium with ampiclox as control antibiotic. Results of the analysis showed significant inhibition of alpha amylase that resembled the activity of acarbose. The highest percentage inhibition of alpha amylase by AgNPs was observed at 65.625% while that of acarbose was 90.357%. Antibacterial inhibition assay revealed that 100% biosynthesized silver nanoparticles had significant (p&lt;0.05) inhibitory effects on Eschericia coli and Streptococcus pyogenes.

Evaluation of Antioxidant and Anti-diabetic Activity of Stem Bark of &lt;i&gt;Ziziphus jujuba&lt;/i&gt; (L.) Gaertn

Diabetes Mellitus (DM), a significant metabolic disorder that increases mortality and morbidity, affects people all over the world. Recently, many individuals are exploring CAM treatment that may have fewer side effects due to the prominent side effects of allopathic medications. Ziziphus jujuba (L.) Gaertn. (Rhamnaceae) stem bark has been a vital part of the conventional system of medicine for the management of diabetes, inflammation, wounds, fever, asthma and liver disorders. In spite of its pharmacological significance, Ziziphus jujuba stem bark lacks scientific evidence of its antidiabetic and antioxidant potential. Hence, an attempt has been made to evaluate the anti-diabetic effect of the stem bark of Ziziphus jujuba using alpha-amylase inhibition assay. In this research work, we further investigated the antioxidant activity of the Ziziphus jujuba using DPPH and Nitric oxide radical scavenging assay. The Chloroform (CEZJ), ethyl acetate (EAZJ) and Ethanol (EEZJ) extracts of Ziziphus jujuba stem bark were used in the study. In the DPPH radical scavenging assay, EEZJ had an IC50 value of 117.1 µg/ml, however, the value for the nitric oxide scavenging assay was 64.65 µg/ml. The Ziziphus jujuba was additionally examined for its in-vitro antidiabetic potential using an alpha-amylase inhibition assay, and it was discovered to possess a considerable percentage of alpha-amylase inhibition. The IC50 of EEZJ was found to be 34.68 µg/ml. These results imply that the Ziziphus jujuba possesses considerable antioxidant and anti-diabetic properties. To thoroughly establish the anti-diabetic potential of Ziziphus jujuba, additional in vitro experiments utilizing cell lines and other enzymes may be conducted.

ISOLATION, CHARACTERIZATION OF Β-SITOSTEROL FROM THE HERBAL EXTRACTS AND EVALUATION OF IN VITRO ANTI-OXIDANT AND ANTI-DIABETIC ACTIVITIES

Plants naturally produce phytochemicals, physiologically active chemical compounds that serve as a natural defence mechanism and have considerable potential for treating a wide range of ailments. Cyperus rotundus and Tinospora cordifolia are the potent traditional herbs used in Ayurveda medicine, especially for diabetes. The current study aimed to isolate and characterize the bioactive compound β-sitosterol from the combined ethanolic extracts of the rhizome of Cyperus rotundus (C. rotundus) and the stem of Tinospora cordifolia (T. cordifolia). Furthermore, the in vitro antioxidant and anti-diabetic activities of isolated compounds are tested by DPPH assay, alpha amylase inhibition assay, alpha glucosidase inhibition assay and glucose uptake activity in 3T3L1 cells. The combined ethanolic extract of both medicinal plants was initially subjected to column and preparative thin layer chromatography for the isolation of bioactive compounds, followed by their characterization by spectral methods like UV, FT-IR, 1H and 13C NMR. The structure of the isolated compound is identified as β-sitosterol and further confirmed by structural elucidation. Similarly, the isolated compound β-sitosterol exhibited higher antioxidant scavenging activity of 64.54±1.38 µg/ml against DPPH at a concentration 100µg/ml. The study also confirmed that β-sitosterol showed good inhibitory activities towards alpha-amylase (62.21±1.25 µg/ml) and alpha-glucosidase (65.48±1.47 µg/ml). β-sitosterol showed significantly increased glucose uptake (28% at 100μg/ml). Based on the conclusion that the organic compound β-sitosterol exhibited good antioxidant and anti-diabetic efficacy, it could have been used in the treatment of free radical disease and can be utilized in the management of diabetes mellitus.

Pharmacognostical and Pharmacological Evaluation of leaves of Citrullus lanatus L.

Citrullus lanatus (Watermelon) is commonly available throughout India and traditionally used in treatment of various ailments. This plant is well-explored for its fruit but the potential of its leaves is still unexplored. Thus, the present study focuses on pharmacognostical and pharmacological evaluation of leaves of Citrullus lanatus. In pharmacognostical evaluation, all macroscopic, microscopic, qualitative, quantitative and microbiological analysis were performed. In pharmacological evaluation, the study mainly focuses on the potential of leaves for antioxidant and antidiabetic activities. The antioxidant activity has been performed by various methods such as DPPH and phospho-molybdenum method. The antidiabetic activity has been performed by non-enzymatic glycosylation of hemoglobin assay method, glucose uptake in yeast cells method, alpha amylase inhibition assay method and alpha glucosidase inhibition assay method. The results of antioxidant activity shows that the plant extract possess good free radical scavenging properties due to the presence of vitamin C, polyphenol and flavonoid content. The results of antidiabetic activity by non-enzymatic glycosylation method shows that the plant extract of C. lanatus exhibit significant inhibition of glycosylation as compared with standard drug alpha tocopherol. In glucose uptake by yeast cells method, the extract showed greater efficiency in increasing the glucose uptake by yeast cells as compared to standard drug acarbose. In alpha amylase inhibition method, Alpha amylase inhibitors bind to alpha bond of polysaccharide and prevent breakdown of polysaccharide in mono and disaccharide which shows significant activity of plant extract as compared to an acarbose standard drug. Thus from the above findings, it can be concluded that C. lanatus leaves possess a great potential for antioxidant and antidiabetic activity and this plant should be more researched and formulation should be marketed in the recent future.&#x0D; Keywords: Citrullus lanatus (Cucurbitaceous), Watermelon, Anti-oxidant Activity, Anti-diabetic Activity

Synthesis, characterization, DFT studies and biological assessment of palladium(II) complexes with imidazole derivatives

Six palladium(II) complexes (1–6) with 2-phenylimidazole (L1) and 1,2-dimethylimidazole (L2), were synthesized and characterized by elemental analyses, IR and NMR spectroscopy. The crystal structure of one of them ([Pd(2-phenylimidazole)2Br2], 1) was determined by X-ray crystallography. The palladium(II) ion in complex 1 is coordinated by two nitrogen atoms of 2-phenylimidazole and two bromide ions at trans positions, adopting a nearly regular square planar geometry. The structures of PdBr2 complexes {[Pd(2-phenylimidazole)2Br2] (1) and [Pd(1,2-dimethylimidazole)2Br2] (4)} as well as their chloride analogues, {[Pd(2-phenylimidazole)2Cl2] (1a) and [Pd(1,2-dimethylimidazole)2Cl2] (4a)} were predicted by DFT calculations. Complexes 1 and 1a appeared to be stabilized by a weak agostic interaction between palladium and hydrogen atoms. The energy of Pd⋯H bond was estimated to be 1.6 kcal/mol for both complexes that was comparable to the strength of Cl⋯H hydrogen bond in 1a (1.7 kcal/mol). The complexes were examined for various biological activities including; antimicrobial, antioxidant and anti-diabetic assays. The complexes exhibited moderate antimicrobial activities but they were not good as antioxidants. However, the alpha amylase inhibition assay showed the excellent anti-diabetic effect of the complexes.

Analysis of phytochemical constituent and biological significance of rhododendrons collected from capital of Laliguras, Laliguras municipality, Tehrathum district, Nepal

The largest genus of the Ericaceae family, Rhododendron includes an inspiring number of species. Some of which have been used in the treatment of various ailments such as diseases associated with heart, dysentery, diarrhea, detoxification, fever, constipation, bronchitis and asthma around the world including Nepal. However, systematic study on phytochemistry and pharmacology of Rhododendrons grown in the Capital of Laliguras, Laliguras Municipality, Tehrathum of Nepal is lacking. So this study was conducted to evaluate the antioxidant activity, α-amylase inhibition activity, total phenolic and flavonoid contents of Rhododendron arboretum and Rhododendron campanulatum from Laliguras Municipality, Tehrathum of Nepal. Plant extracts were prepared in methanol by cold percolation method. Phytochemical constituents were analyzed by following standard methods. Antioxidant activity was measured through 2, 2-Diphenyl-1-Picrylhydrazyl assay. Total phenolic and flavonoid content were estimated using Folin–Ciocalteau and aluminum chloride method respectively. Alpha amylase inhibition assay was carried out by starch iodine method. Phytochemical screening of methanolic extract of flower and leaf of R. arboretum and leaves of R. campanulatum displayed the presence of different chemical constituents such as flavonoids, polyphenols, terpenoids, saponins and quinins. Samples showed dose-dependent radical scavenging and α-amylase inhibition activity. Radical scavenging activity and α-amylase inhibition activity of the methanolic extracts of different parts of R. arboretum and R. campanulatum ranged from 19.46 to 95.88% and 13.23 to 68.44% respectively. Flower extracts of R. arboreum showing the strongest radical scavenging activity and α-amylase inhibition with IC50 values 33.61 and 443.44 μg/mL respectively. Among the tested samples, flower extract of R. arboretum showed the strongest antioxidant activity, alpha amylase inhibition activity and contained the highest amount of total phenolic and flavonoid content.&#x0D;

Purification of ursolic acid and β-sitosterol from endophytic Alternaria alternata for their alpha-amylase inhibitory activity

Fungal endophytes are a known warehouse of bioactive compounds with multifarious applications. In the present investigation two compounds, β-Sitosterol (1) and ursolic acid (2), were isolated from Alternaria alternata, an endophytic fungus associated with Morus alba Linn for the first time. The structure of the compounds was elucidated on the basis of comprehensive spectral analysis (UV, IR, 1 H-, 13 C- and 2D-NMR, as well as HRESI–MS). In the in vitro alpha amylase inhibitory assay both compounds (1) and (2) show potent antidiabetic activity. In support, Docking studies indicate significant binding affinity of the isolated compounds. Hence from the present study, it can be concluded that endophytic fungi in Morus alba Linn can find use in antidiabetic drug development in the medicinal industry. Communicated by Ramaswamy H. Sarma

Fabrication of protein crocin nanocomplex and its addition in proso millet flour for development of functional snacks with anti‐diabetic, anti‐hypertensive and antioxidant properties

SummaryThe current study was undertaken to develop novel gluten‐free functional snacks using proso millet flour with the addition of nano‐encapsulated crocin in sorghum (SPCS), pearl millet (PPCS) and foxtail millet (FPCS) protein nanoparticles. The crocin content was well retained (58.08%–77.67%) in the developed snacks. Characteristic properties like nutraceutical potential, rheological, sensory and physical attributes were evaluated. The extruded snacks showed good antioxidant (DNA damage inhibition assay), anti‐diabetic (alpha amylase inhibition assay) and anti‐hypertensive (angiotensin converting enzyme assay) properties. The developed snacks (P &gt; 0.05) showed higher elastic behaviour (G′) compared to viscous behaviour (G″). Hardness value for functional snacks were found to be in the range of 407–115 N. The control snacks showed significantly higher expansion ratio (348.76%) compared to functional snacks. The developed novel functional snacks appeared darker than control snacks as evaluated by colorimetric assay. The overall acceptability of the snacks was found to be equal on a 9‐point hedonic scale.

Phytochemical Screening and Evaluation of Antioxidant, Antidiabetic, and Antimicrobial Potential of Bergenia ligulata

Bergenia ligulata, locally known as “Pashanbheda,” belongs to the family Saxifragaceae and is a perennial herb with a short and thick stem. This plant has numerous medicinal properties including antioxidant, antidiabetic, antibacterial, and anti-inflammatory properties. The qualitative and quantitative analysis shows that the rhizome of this plant contains various phytochemical constituents. Qualitative analysis shows that it contains phenols, flavonoids, tannins, terpenoids, alkaloids, steroids, carbohydrates, and glycosides. Thirty- two bioactive compounds were identifiedduring GC-MS analysis. Dihydro-3-methylene-5-methyl-2-furanon, Stigmast-5-en-3-ol, 9-Octadecenamide(Z), 6,9-Octadecadienoic acid, methyl ester, 9,12-Octadecadienoic acid (Z, Z)-, methyl compounds have antioxidant, antibacterial and anti-inflammatory activities. The total phenol and flavonoid content from the quantitative analysis revealed a significant amount of phenol i.e., 67.1±0.02*6mg/g, and flavonoid content i.e., 89.11±0.01* mg/g. In vitro, antioxidant activities were estimated by diphenyl-1-picrylhydrazyl assay showing the highest % inhibition of 47.24±0.06* for plant extract and 41.63±0.057* for standard respectively. The in vitro, antidiabetic activitieswere estimated by alpha-amylase inhibition assay showing the highest % inhibition of 67.14±0.05* at 250 μg/ ml for methanolic plant extract and 61.42±0.01* at 250 μg/ ml for standard and in vitro antibacterial potential were detected by using Agar well diffusion process. The anti-bacterial action was found maximum in aqueous plant extract against staphylococcus aureus bacteria.

Biogenic Synthesis of Multifunctional Silver Oxide Nanoparticles (Ag2ONPs) Using Parieteria alsinaefolia Delile Aqueous Extract and Assessment of Their Diverse Biological Applications.

Green nanotechnology has made the synthesis of nanoparticles a possible approach. Nanotechnology has a significant impact on several scientific domains and has diverse applications in different commercial areas. The current study aimed to develop a novel and green approach for the biosynthesis of silver oxide nanoparticles (Ag2ONPs) utilizing Parieteria alsinaefolia leaves extract as a reducing, stabilizing and capping agent. The change in color of the reaction mixture from light brown to reddish black determines the synthesis of Ag2ONPs. Further, different techniques were used to confirm the synthesis of Ag2ONPs, including UV-Visible spectroscopy, Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), scanning electron microscope (SEM), Energy-dispersive X-ray spectroscopy (EDX), zeta potential and dynamic light scattering (DLS) analyses. The Scherrer equation determined a mean crystallite size of ~22.23 nm for Ag2ONPs. Additionally, different in vitro biological activities have been investigated and determined significant therapeutic potentials. Radical scavenging DPPH assay (79.4%), reducing power assay (62.68 ± 1.77%) and total antioxidant capacity (87.5 ± 4.8%) were evaluated to assess the antioxidative potential of Ag2ONPs. The disc diffusion method was adopted to evaluate the antibacterial and antifungal potentials of Ag2ONPs using different concentrations (125-1000 μg/mL). Moreover, the brine shrimp cytotoxicity assay was investigated and the LC50 value was calculated as 2.21 μg/mL. The biocompatibility assay using red blood cells (<200 μg/mL) confirmed the biosafe and biocompatible nature of Ag2ONPs. Alpha-amylase inhibition assay was performed and reported 66% inhibition. In conclusion, currently synthesized Ag2ONPs have exhibited strong biological potential and proved as an attractive eco-friendly candidate. In the future, this preliminary research work will be a helpful source and will open new avenues in diverse fields, including the pharmaceutical, biomedical and pharmacological sectors.

Microencapsulation of &lt;i&gt;Ruellia tuberosa&lt;/i&gt; L. Extracts Using Alginate: Preparation, Biological Activities, and Release

The bioactive compounds naturally present in plants have great importance due to their biological characteristics. These substances could lose their active characteristics since they are highly unstable. Microencapsulation is one of the techniques to improve stability and protect these compounds. In this work, Ruellia tuberosa L. ethanolic extracts microcapsules were prepared using a freeze-drying method by varying pH, alginate concentration, and stirring time. The encapsulation efficiency (EE), characteristics, alpha-amylase inhibition activity, and release behavior of the microcapsules were investigated. The results highlighted that the highest encapsulation efficiency for the microcapsules was obtained at pH 6, alginate concentration of 1% (w/v), and 30 min of stirring time (51.63% EE). The microcapsules mostly had spherical shapes with a mean diameter of 197.53 μm. The alpha-amylase inhibition assay from microcapsules resulted in the IC50 value of 46.66 ± 0.13 μg/mL, demonstrating high biological activity. The bioactive substances from microcapsules were released during intervals of 30–120 min at pH values of 1.2 and 7.4. Only 3.51% of the bioactive substances were released at pH 1.2 after 120 min, compared to 55.78% at pH 7.4. Overall, this work confirms the possibility of developing plant extracts with preserved biological activity using the produced microcapsules.

Bioactive compound profile and their biological activities of endogenous black rice from Java and East Nusa Tenggara

ABSTRACT Indonesia has an abundance of pigmented rice cultivars with high nutritional value. The purpose of this study was to identify and characterize the phytochemical profiles, anthocyanin properties, and biological activities of endogenous Indonesian black rice (Oryza sativa L.) cultivars from Java and East Nusa Tenggara (NTT) Islands. Thirteen black rice cultivars from eastern Nusa Tenggara and West, Central, and East Java were examined. The secondary metabolites of the black rice samples included flavonoids, tannins, phenolics, quinones, glycosides, anthraquinones, leucoanthocyanidins, alkaloids, and proteins. According to the FRAP (ferric reducing antioxidant power) assay, various levels of antioxidant activity were present. The results of an alpha amylase inhibition assay indicated that the black rice samples might have antidiabetic properties. We conclude that the Java and NTT endogenous black rice cultivars have rich phytochemical compounds with high nutritional value, and that their antioxidant and antidiabetic biological activities can promote a healthy life.

Chemical profiling and in-vitro α-amylase inhibitory activity of Sesbania sesban and Sesbania grandiflora seeds

ABSTRACT S. sesban is mainly used as economical purpose and S. grandiflora is known for medicinal as well as economical. Chemical characterization and antidiabetic activity of seed extracts were not explored yet. Seeds of S. sesban and S. grandiflora were separated in seed coat, germ, and endosperm by domestic grinder mixer. Successive soxhlet extraction of separated parts of Sesbania seed were carried out. Chemical profiling of methanolic extract was carried out by GC-MS, HPLC, and in vitro alpha amylase inhibitory assay. D-pinitol was found to be maximum (48.39%) in seed coat and minimum in seed (8.35%) of methanolic extracts of Sesbania seed. Alpha-amylase inhibitory activity of methanolic extracts of different parts were also studied in which DSSh, DSSp, & DSSe showed similar IC50 to acarbose, antidiabetic drugs. The result showed that S. sesban and S. grandiflora seed and endosperm is rich in health promoting mineral like Na (241 mg/100 gm and 238 mg/100 gm), K (103.1 mg/100 gm) and (60.2 mg/100 gm), Ca (525 mg/100 gm) and (500 mg/100 gm) and Fe (67.5 and 91.9 mg/100 gm). Hence, it is similar to some edible legumes which possess bioactive compound with therapeutic value and can be efficiently explored for the development of new functional or nutraceutically valued food products.

Bioactivity evaluation of least explored traditionally acclaimed medicinally potent herb Nanorrhinum ramosissimum(Wall.) Betsche

Present study analysed the therapeutic potential of traditionally acclaimed medicinal herb Nanorrhinum ramosissimum, using plant parts extracted with different solvents (10 mg/mL). Shoot extracts exhibited comparatively better antimicrobial properties, in comparison to root extracts. Total phenolic content was estimated, to ascertain its dependency on antioxidant properties of plant extracts. Antioxidant assay revealed promising results in comparison to IC50 value of standard ascorbic acid (52.2±0.07 µg/mL), for methanolic extracts of shoot (61.07±0.53 µg/mL and 64.33±0.33 µg/mL) and root (76.705±0.12 µg/mL and 89.73±0.28 µg/ mL) for in vivo and in vitro regenerants respectively. Correlation coefficient R2 values ranged between 0.90-0.95, indicating a positive correlation between phenolic contents and antioxidant activity. Plant extracts were also able to inhibit DNA oxidative damage again indicating their antioxidative potential. Antidiabetic potential was confirmed by alpha amylase inhibition assay where shoot methanolic extracts (invivo, in vitro) exhibited the best IC50 values (54.42±0.16 µg/mL, 66.09±0.12 µg/mL) in comparison to standard metformin (41.92±0.08 µg/mL). Ethanolic extracts of roots (in vitro, invivo) exhibited the relative IC50 values (88.97±0.32µg/mL,96.63±0.44 µg/mL) indicating that shoot parts had a better alpha amylase inhibition property; thus proving the herb’s bioactive potential and its prospective therapeutic source for curing various ailments.

Mangosteen Metabolites as Promising Alpha-Amylase Inhibitor Candidates: In Silico and In Vitro Evaluations.

Diabetes is a chronic metabolic disorder characterized by raised glucose levels in the blood, resulting in grave damage over time to various body organs, including the nerves, heart, kidneys, eyes, and blood vessels. One of its therapeutic treatment approaches involves the inhibition of enzymes accountable for carbohydrate digestion and absorption. The present work is aimed at evaluating the potential of some reported metabolites from Garcinia mangostana (mangosteen, Guttiferae) as alpha-amylase inhibitors. Forty compounds were assessed for their capacity to inhibit alpha-amylase using in silico studies as well as in vitro assays. Molecular docking was carried out to analyze their binding capacities in the 3D structure of alpha-amylase (PDB ID: 4GQR). Among the tested compounds, 6-O-β-D-glucopyranosyl-2,4,6,3',4',6'-hexahydroxybenzophenone (8), aromadendrin-8-C-glucoside (5), epicatechin (6), rhodanthenone (4), and garcixanthone D (40) had a high XP G.score and a Glide G.score of -12.425, -11.855, -11.135, and -11.048 Kcal/mol, respectively. Compound 8 possessed the XP and Glide docking score of -12.425 Kcal/mol compared to the reference compounds myricetin and acarbose which had an XP and Glide docking score of -12.319 and 11.201 Kcal/mol, respectively. It interacted through hydrogen bond formations between its hydroxyl groups and the residues His 101, Asp 197, Glu 233, Asp 300, and His 305, in addition to water bridges and hydrophobic interactions. Molecular mechanics-generalized born surface area (MM-GBSA) was used to calculate the binding free energy and molecular dynamic studies that indicated the stability of the alpha-amylase-compound 8 complex during the 100 ns simulation in comparison with myricetin- and acarbose-alpha-amylase complexes. Additionally, the in vitro alpha-amylase inhibition assay findings validated the in silico study's findings. This could further validate the potential of G. mangostana as a candidate for diabetes management.

In Vitro Hypoglycemic and Radical Scavenging Activities of Certain Medicinal Plants

Abstract The purpose of this study is to investigate in vitro hypoglycemic and free radical scavenging activities of some medicinal plants including Ficus glomerata (FG), Pandanus amaryllifolia (PaA), Artocarpus altilis (AA), Gomphrena celosioides (GC) and Gynostemma pentaphyllum (GP). Alpha-amylase inhibitory assay was examined by dinitrosalicylic acid reaction. Glucose uptake assay was investigated by LO-2 cell model. DPPH and ABTS+ scavenging assays were performed by spectrophotometry. Cell viability was determined by MTT method. It was found that the extracts including FG, PaA, AA, GC and GP were able to inhibit alphaamylase activity up to 38.4 ± 4.2%, 47.8 ± 4.3%, 49.3 ± 3.5%, 40.1 ± 4.4% and 38.5 ± 3.8%, respectively. Moreover, glucose adsorption and glucose uptake capacity of these extracts were evidenced. In addition, free radical scavenging activity of these extracts was indicated in a range of 30.6-54.5% for DPPH radical and 31.8-51.1% for ABTS+ radical. Especially, these extracts exhibited no cytotoxicity effect on human hepatic LO-2 cells and human gastric BGC-823 cells at the concentration of 100 μg/ml. The results indicated that A. altilis leaves were effective in inhibiting alpha-amylase activity, increasing glucose adsorption and glucose uptake and scavenging free radicals. Therefore, it could be suggested to be a promising hypoglycemic agent for managing type 2 diabetes.
