Amidephrine mesylate, a new potent sympathomimetic amine, was compared with phenylephrine using several cardiovascular preparations. The vasopressor molar potency (amidephrine: phenylephrine) was as follows: anesthetized dogs (i.v.) 1:·9; anesthitized cats (i.v.) 1:2·1; unanesthetized rats (i.p.) 5·7:1; unanesthetized rats (p.o.) 3·7:1. The duration of action in each case was longer than phenylephrine. No tolerance to the pressor action was observed in monkeys which had received the drug daily for 6 months prior to blood pressure reactivity studies.Amidephrine induced reflex bradycardia in dogs and rats. This was abolished by atropine sulfate or bilateral vagotomy. Amidephrine elicited only minor alterations in heart rate, myocardial contractile force and coronary flow in the isolated rabbit heart preparation.Intranasal pressure was reduced in anesthetized dogs by the intravenous administration of doses of amidephrine which did not alter systemic arterial blood pressure. No evidence of after-congestion was observed. Phenylephrine was 1·4 times more active than amidephrine in this preparation.Amidephrine administered directly into the artery increased femoral arteriolar and renal arteriolar resistance. Chez tous les animauxe´tudie´s, la dure´e de l'action ae´te´plus longue que celle de la phe´nyle´phrine.Pas de manifestation d'intole´rancea`l'effet vasopresseur du me´dicamenta`l'administration chronique chez le singe, pendant une dure´e de six mois qui a pre´ce´de lese´tudes sur la tension arte´rielle.Chez le chien et le chat, l'amidephrine a de´termine´une bradycardie re´actionnelle, pouvanteˆtre abolie par le sulfate d'atropine ou la vagotomie bilate´rale.Etudie´sur le coeur isole´de lapin, l'amidephrine n'a de´termine´que des changements mineurs en ce qui concerne le rythme cardiaque, la contractilite´du myocarde et le flux coronaire.Chez le chien anesthe´sie´, l'administration intraveineuse de faibles doses d'amidephrine ne posse`dant aucun effet sur la tension arte´rielle, font diminuer la tension intranasale, sans de´terminer de congestion local re´actionnelle.Il esta`noter qu'au cours de ces expe´riences, la phe´nyle´phrine s'est re´ve´le´e 1·4 fois plus active que l'amidephrine.Injecte´e par voie intra-arte´rielle, l'amidephrine augmente la re´sistance capillaire au niveau des arte`res fe´morales et des reins.Teste´e chez le chien ete´tudie´e au niveau de l'arte`re fe´morale, la phe´nyle´phrine posse`de un effet vasoconstricteur 1·4 fois plus marque´que l'amidephrine; en revanche, au niveau des reins, c'est l'amidephrine qui s'est re´ve´le´e 2·5 fois plus active que son analogue hydroxylique.Amidephrin mesylat, ein neues sympathicomimetisch stark wirkendes Amin wurde mit Phenylephrin auf seine Herz- und Kreislaufwirkung hin verglichen. Die Wirkungsdauer war in allen Versuchen im Vergleich zu Phenylephrin verla¨ngert. Bei Affen, die die Substanz ta¨gl.u¨ber einen Zeitraum von 6 Monaten vor den Versuchen erhalten hatten entwickelte sich keine Toleranz gegenu¨ber den Ausgangswerten der pressorischen Wirkung gemessen an den Blutdruckreaktionen.Amidephrin lo¨ste bei Hunden und Ratten eine reflektorische Bradykardie aus. Diese Wirkung konnte durch Atropin-Sulfat oder bilaterale Vagotomie aufgehoben werden. Amidephrin verursachte nur geringe Vera¨nderungen der Herzfrequenz, der Kontraktilita¨t des Herzens und der Coronardurchblutung isolierter Kaninchenherzen.Intraveno¨se Applikationen von Amidephrin in Dosen, welche den arteriellen Blutdruck unbeeinflußt lassen, verringerten den intranasalen Druck an narkotisierten Hunden. Eine reaktive Hypera¨mie konnte nicht beobachtet werden. In dieser Versuchsanordnung war Phenylephrin 1·4 mal sta¨rker als Amidephrin.Nach arteriellen Injektionen des Amidephrins wurde der periphere Gefa¨sswiderstand der Arteriola femoralis und renalis erho¨ht. Die Vasokonstriktion nach Phenylephrin war im arteriellen Bereich der Femoralis 1·4 mal sta¨rker als nach Amidephrin, wa¨hrend andererseits das Amidephrin an den Nieren 2·5 mal sta¨rker wirkte als sein Hydroxyl-Analogum.
