AcEXPA1, an α-expansin gene, participates in the aluminum tolerance of carpetgrass (Axonopus compressus) through root growth regulation.

AcEXPA1, an aluminum (Al)-inducible expansin gene, is demonstrated to be involved in carpetgrass (Axonopus compressus) root elongation under Al toxicity through analyzing composite carpetgrass plants overexpressing AcEXPA1. Aluminum (Al) toxicity is a major mineral toxicity that limits plant productivity in acidic soils by inhibiting root growth. Carpetgrass (Axonopus compressus), a dominant warm-season turfgrass widely grown in acidic tropical soils, exhibits superior adaptability to Al toxicity. However, the mechanisms underlying its Al tolerance are largely unclear, and knowledge of the functional genes involved in Al detoxification in this turfgrass is limited. In this study, phenotypic variation in Al tolerance, as indicated by relative root elongation, was observed among seventeen carpetgrass genotypes. Al-responsive genes related to cell wall modification were identified in the roots of the Al-tolerant genotype 'A58' via transcriptome analysis. Among them, a gene encoding α-expansin was cloned and designated AcEXPA1 for functional characterization. Observed Al dose effects and temporal responses revealed that Al induced AcEXPA1 expression in carpetgrass roots. Subsequently, an efficient and convenient Agrobacterium rhizogenes-mediated transformation method was established to generate composite carpetgrass plants with transgenic hairy roots for investigating AcEXPA1 involvement in carpetgrass root growth under Al toxicity. AcEXPA1 was successfully overexpressed in the transgenic hairy roots, and AcEXPA1 overexpression enhanced Al tolerance in composite carpetgrass plants through a decrease in Al-induced root growth inhibition. Taken together, these findings suggest that AcEXPA1 contributes to Al tolerance in carpetgrass via root growth regulation.

From stress to responses: aluminium-induced signalling in the root apex.

Aluminium (Al) toxicity is one of the major constraints for crop growth and productivity in most of the acid soils worldwide. The primary lesion of Al toxicity is the rapid inhibition of root elongation. The root apex, especially the transition zone (TZ), has been identified as the major site of Al accumulation and injury. The signalling, in particular through phytohormones in the root apex TZ in response to Al stress, has been reported to play crucial roles in the regulation of Al-induced root growth inhibition. The binding of Al in the root apoplast is the initial event leading to inhibition of root elongation. Much progress has been made during recent years in understanding the molecular functions of cell wall modification and Al resistance-related genes in Al resistance or toxicity, and several signals including phytohormones, Ca2+, etc. have been reported to be involved in these processes. Here we summarize the recent advances in the understanding of Al-induced signalling and regulatory networks in the root apex involved in the regulation of Al-induced inhibition of root growth and Al toxicity/resistance. This knowledge provides novel insights into how Al-induced signals are recognized by root apical cells, transmitted from the apoplast to symplast, and finally initiate the defence system against Al. We conclude that the apoplast plays a decisive role in sensing and transmitting the Al-induced signals into the symplast, further stimulating a series of cellular responses (e.g. exudation of organic acid anions from roots) to adapt to the stress. We expect to stimulate new research by focusing on the signalling events in the root apex in response to Al stress, particularly taking into consideration the signal transduction between the meristem zone, TZ, and elongation zone and the apoplast and symplast.

Using brefeldin A to disrupt cell wall polysaccharide components in rice and nitric oxide to modify cell wall structure to change aluminum tolerance.

The components and structure of cell wall are closely correlated with aluminum (Al) toxicity and tolerance for plants. However, the cell wall assembly and function construction in response to Al is not known. Brefeldin A (BFA), a macrolide, is used to disrupt cell wall polysaccharide components, and nitric oxide (NO), a signal molecule, is used to modify the cell wall structure. Pretreatment with BFA accelerated Al accumulation in root tips and Al-induced inhibition of root growth of two rice genotypes of Nipponbare and Zhefu 802, and significantly decreased the cell wall polysaccharide content including pectin, hemicellulose 1, and hemicellulose 2, indicating that BFA inhibits the biosynthesis of components in the cell wall and makes the root cell wall lose the ability to resist Al. The addition of NO donor (SNP) significantly alleviated the toxic effects of Al on root growth, Al accumulation, and oxidative damage, and decreased the content of pectin polysaccharide and functional groups of hydroxyl, carboxyl, and amino in the cell wall via FTIR analysis, while had no significant effect on hemicellulose 1 and hemicellulose 2 content compared with Al treatment. Furthermore, NO didn't change the inhibition effect of BFA-induced cell wall polysaccharide biosynthesis and root growth. Taken together, BFA disrupts the integrity of cell wall and NO modifies partial cell wall composition and their functional groups, which change the Al tolerance in rice.

Silicon confers aluminium tolerance in rice via cell wall modification in the root transition zone.

The root-apex transition zone (TZ), the major perception site for aluminium (Al) toxicity, is crucial for the Al-induced root-growth inhibition, while the mechanism underlying silicon-mediated alleviation of Al toxicity in the TZ is largely unknown. In this study, the role of silicon (Si) in alleviating Al-induced damage in the TZ and root-growth inhibition of rice was investigated. We found that Si had direct alleviative effect on Al toxicity as revealed by less root growth-inhibition, Al accumulation, and callose formation. Si reversed Al-induced decreases of the cell wall elongation and extensibility, and reduced Al-induced increments of cell wall polysaccharides in the TZ. The similar distribution patterns of Al and Si in the cell wall indicated that Si might detoxify Al by forming hydroxyaluminumsilicates in the apoplast of the root-apex TZ. Moreover, the wall-bound form of Si reduced Al binding sites, thereby reducing the capability of Al bound to the cell wall. These results suggest that Si-mediated cell wall modification in the TZ alleviates Al-induced root-growth inhibition in rice involving the promotion of cell wall extensibility and the decrease of Al accumulation in the cell wall.

Local regulation of auxin transport in root‐apex transition zone mediates aluminium‐induced Arabidopsis root‐growth inhibition

Aluminium (Al) stress is a major limiting factor for worldwide crop production in acid soils. In Arabidopsis thaliana, the TAA1-dependent local auxin biosynthesis in the root-apex transition zone (TZ), the major perception site for Al toxicity, is crucial for the Al-induced root-growth inhibition, while the mechanismunderlying Al-regulated auxin accumulation in the TZ isnot fully understood. In the present study, the role of auxin transport in Al-induced local auxin accumulation in the TZ and root-growth inhibition was investigated. Our results showed that PIN-FORMED (PIN) proteins such as PIN1, PIN3, PIN4 and PIN7 and AUX1/LAX proteins such as AUX1, LAX1 and LAX2 were all ectopically up-regulated in the root-apex TZ in response to Al stress and coordinately regulated local auxin accumulation in the TZ and root-growth inhibition. The ectopic up-regulation of PIN1 in the TZ under Al stress was regulated by both ethylene and auxin, with auxin signalling acting downstream of ethylene. Al-induced PIN1 up-regulation and auxin accumulation in the root-apex TZ was also regulated by the calossin-like protein BIG. Together, our results provide insight into how Al stress induces local auxin accumulation in the TZ and root-growth inhibition through the local regulation of auxin transport.

Phytohormone signalling and cross-talk to alleviate aluminium toxicity in plants.

Aluminium (Al) is one of the most abundant metals in earth crust, which becomes toxic to the plants growing in acidic soil. Phytohormones like ethylene, auxin, cytokinin, abscisic acid, jasmonic acid and gibberellic acid are known to play important role in regulating Al toxicity tolerance in plants. Exogenous applications of auxin, cytokinin and abscisic acid have shown significant effect on Al-induced root growth inhibition. Moreover, ethylene and cytokinin act synergistically with auxin in responding against Al toxicity. A number of studies showed that phytohormones play vital roles in controlling root responses to Al toxicity by modulating reactive oxygen species (ROS) signalling, cell wall modifications, organic acid exudation from roots and expression of Al responsive genes and transcription factors. This review provides a summary of recent studies related to involvement of phytohormone signalling and cross-talk with other pathways in regulating response against Al toxicity in plants.

Heat shock induces cross adaptation to aluminum stress through enhancing ascorbate-glutathione cycle in wheat seedlings

Aluminum (Al), a neurotoxin agent, is universal in the earth crust, but its bioavailability and toxicity are manifested under acidic conditions. Up to 60% of the acid soils are distributed in tropical and subtropical regions, where crops simultaneously experience heat-shock stress. Here, we investigated the effects of heat shock-priming on Al tolerance in two different wheat genotypes. Conditioning of wheat seedlings with short period high temperature significantly alleviated Al-induced root growth inhibition, but did not significantly affect Al accumulation. However, we observed that heat shock-primed roots maintained lower levels of lipid peroxidation and higher cell viability. These priming-triggered effects were associated with reactive oxygen species (ROS) homeostasis. Furthermore, conditioning of plants with high temperature increased the contents of reduced ascorbate and glutathione, and ratios of reduced to oxidized forms of these molecules in wheat roots. However, ascorbate or glutathione biosynthesis inhibitors markedly prevented heat shock priming-induced ROS reduction accompanied by aggravated root elongation. Moreover, heat shock-priming enhanced the metabolic intensity of ascorbate-glutathione cycle, as activities of the cycle-allied enzymes were significantly increased. These results suggest that heat-shock induces cross adaptation to Al toxicity through sustaining efficient ascorbate-glutathione cycle operation in wheat plants.

Jasmonate and aluminum crosstalk in tomato: Identification and expression analysis of WRKYs and ALMTs during JA/Al-regulated root growth

The WRKY transcription factors (TFs) are involved in aluminum (Al) stress and jasmonic acid (JA)-regulated resistance responses. WRKYs act as regulators of Al-activated malate transporter (ALMT) proteins (anion channels) by directly binding to their promoters and altering malate efflux, thereby regulating Al ion toxicity in plant roots. JA enhances Al-induced root growth inhibition in Arabidopsis. However, the relationship between WRKY and ALMT genes and their involvement in JA-mediated root growth inhibition during Al stress in tomato remain unknown. Here, we demonstrate a similar phenomenon that JA enhances Al-induced root growth inhibition in tomato (Solanum lycopersicum). By analyzing RNA-seq data and tissue-specific expression data from public databases, we selected 17 WRKY and 6 ALMT family genes to identify the genes participated in this process. The promoters of many of the selected genes contained MeJA responsive element, G-box (target site of MYC2, a core TF of JA signaling), and W-box (target site for WRKY). Quantitative real-time PCR was performed to evaluate the expression levels of selected WRKY and ALMT genes under AlCl3 and Methyl jasmonate (MeJA) treatment. SlMYC2-VIGS seedlings and jasmonic acid-insensitive1 (jai1) mutant were also employed to analyze the expression patterns of selected genes. We find that SlALMT3 is responsible for the crosstalk regulatory mechanism between Al and JA in root growth inhibition, and 6 SlWRKYs may act as the upstream regulators of SlALMT3 in this crosstalk response. This study is initial and informative in exploring the crosstalk regulatory mechanism between JA and Al in tomato.

The auxin influx carrier, OsAUX3, regulates rice root development and responses to aluminium stress

In rice, there are five members of the auxin carrier AUXIN1/LIKE AUX1 family; however, the biological functions of the other four members besides OsAUX1 remain unknown. Here, by using CRISPR/Cas9, we constructed two independent OsAUX3 knock-down lines, osaux3-1 and osaux3-2, in wild-type rice, Hwayoung (WT/HY) and Dongjin (WT/DJ). osaux3-1 and osaux3-2 have shorter primary roots (PRs), decreased lateral root (LR) density, and longer root hairs (RHs) compared with their WT. OsAUX3 expression in PRs, LRs, and RHs further supports that OsAUX3 plays a critical role in the regulation of root development. OsAUX3 locates at the plasma membrane and functions as an auxin influx carrier affecting acropetal auxin transport. OsAUX3 is up-regulated in the root apex under aluminium (Al) stress, and osaux3-2 is insensitive to Al treatments. Furthermore, 1-naphthylacetic acid accented the sensitivity of WT/DJ and osaux3-2 to respond to Al stress. Auxin concentrations, Al contents, and Al-induced reactive oxygen species-mediated damage in osaux3-2 under Al stress are lower than in WT, indicating that OsAUX3 is involved in Al-induced inhibition of root growth. This study uncovers a novel pathway alleviating Al-induced oxidative damage by inhibition of acropetal auxin transport and provides a new option for engineering Al-tolerant rice species.

GmGRP-like gene confers Al tolerance in Arabidopsis

Aluminium (Al) toxicity restrains water and nutrient uptake and is toxic to plant roots, ultimately inhibiting crop production. Here, we isolated and characterized a soybean glycine-rich protein-like gene (GmGRPL) that is mainly expressed in the root and that is regulated by Al treatment. Overexpression of GmGRPL can alleviate Al-induced root growth inhibition in Arabidopsis. The levels of IAA and ethylene in GmGRPL-overexpressing hairy roots were lower than those in control and RNA interference-exposed GmGRPL hairy roots with or without Al stress, which were mainly regulated by TAA1 and ACO, respectively. In transgenic soybean hairy roots, the MDA, H2O2 and O2–·content in GmGRPL-overexpressing hairy roots were less than that in control and RNA interference-exposed GmGRPL hairy roots under Al stress. In addition, IAA and ACC can enhance the expression level of the GmGRPL promoter with or without Al stress. These results indicated that GmGRPL can alleviate Al-induced root growth inhibition by regulating the level of IAA and ethylene and improving antioxidant activity.

Melatonin alleviates aluminum-induced root growth inhibition by interfering with nitric oxide production in Arabidopsis

Root growth inhibition is the primary symptom of aluminum (Al) toxicity. In this study, the roles of nitric oxide (NO) and melatonin, two ubiquitous signal molecules involved in diverse plant physiological processes and stress responses, were investigated in Arabidopsis responses to Al toxicity. The results showed that nitric reductase- and nitric oxide synthase-dependent NO production contributed to Al-induced root growth inhibition through interfering with the root cell cycle progression and the quiescent center cell homeostasis. The Al-induced NO production was decreased by exogenous melatonin, which was associated with abolishment of Al-induced root elongation and cell cycle arresting, and with the quiescent center activity returning to the Al-free control levels. The expression of AtSNAT (Arabidopsis thalianaserotonin N-acetyltransferase) was down-regulated by Al, which coincided with decreased melatonin accumulation in Arabidopsis. Pharmacological and genetic analyses further confirmed that a decrease in melatonin synthesis enhanced NO production and Al sensitivity. Taken together, the results presented here suggest that melatonin alleviates Al-induced root growth inhibition by interfering with NO-mediated reduction of cell division cycle progression and the quiescent center cellular activity in Arabidopsis roots.

Auxin Efflux Carrier ZmPGP1 Mediates Root Growth Inhibition under Aluminum Stress.

Auxin has been shown to enhance root growth inhibition under aluminum (Al) stress in Arabidopsis (Arabidopsis thaliana). However, in maize (Zea mays), auxin may play a negative role in the Al-induced inhibition of root growth. In this study, we identified mutants deficient in the maize auxin efflux carrier P-glycoprotein (ZmPGP1) after ethyl methanesulfonate mutagenesis and used them to elucidate the contribution of ZmPGP1 to Al-induced root growth inhibition. Root growth in the zmpgp1 mutant, which forms shortened roots and is hyposensitive to auxin, was less inhibited by Al stress than that in the inbred line B73. In the zmpgp1 mutants, the root tips displayed higher auxin accumulation and enhanced auxin signaling under Al stress, which was also consistent with the increased expression of auxin-responsive genes. Based on the behavior of the auxin-responsive marker transgene, DR5rev:RFP, we concluded that Al stress reduced the level of auxin in the root tip, which contrasts with the tendency of Al stress-induced Arabidopsis plants to accumulate more auxin in their root tips. In addition, Al stress induced the expression of ZmPGP1 Therefore, in maize, Al stress is associated with reduced auxin accumulation in root tips, a process that is regulated by ZmPGP1 and thus causes inhibition of root growth. This study provides further evidence about the role of auxin and auxin polar transport in Al-induced root growth regulation in maize.

The Cell Cycle Checkpoint Regulator ATR Is Required for Internal Aluminum Toxicity-Mediated Root Growth Inhibition in Arabidopsis.

Aluminum (Al) can target multiple sites of root cells for toxicity, including the cell wall, the plasma membrane and symplastic components. Previous work revealed that the cell cycle checkpoint regulator (ATR) Ataxia Telangiectasia-mutated and Rad3-related is required for Al toxicity-induced root growth inhibition in als3 and that the symplastic component DNA is an important target site of Al for the toxicity. However, whether monitoring DNA integrity through ATR-regulated pathway is required for Al-induced root growth inhibition in other Al-sensitive mutants remains unknown. In this study, we demonstrated that the atr mutation could also rescue the Al hypersensitivity and Al-induced cell cycle arrest in star1, which supports the hypothesis that ALS3 and STAR1 function together to be involved in the detoxification of Al in Arabidopsis. However, mutation of ATR could not rescue the Al-sensitive phenotype of almt1 or stop1, both of which are defective in external detoxification mechanisms of Al. We further showed that the Al hypersensitivity and Al-induced quiescent center (QC) differentiation in als1 could also be rescued by the atr mutation. Therefore, our results suggest that ATR-regulated pathway is involved in the modulation of internal Al toxicity-mediated root growth inhibition in Arabidopsis.

Boron supply alleviates Al-induced inhibition of root elongation and physiological characteristics in rapeseed (Brassica napus L.)

ABSTRACTAluminum (Al) toxicity is one of the major problems affecting crop production. Boron (B) is an essential micronutrient for higher plants. In the present study, we investigated the alleviation of Al-induced inhibition of root growth and physiological characteristics by B in rapeseed. The rapeseeds were grown in different Al concentrations (0 and 300 μM), and for every concentration, two B treatments (2.5 and 25 µM as H3BO3) were applied. The results showed that Al toxicity under low B drastically inhibited root growth. The supply of B improved root length, photosynthesis, root activity, total chlorophyll by 60.15%, 104.7%, 102%, and 106.3%, respectively under Al toxicity. This further resulted in improvement of peroxidase, catalase, and ascorbate peroxidase activities while decreasing malondialdehyde, H2O2, and Al contents in roots and leaves. It might be supposed that B alleviates Al toxicity by less mobilization of Al in plant parts and through improving antioxidant enzyme activities.

A proteomic approach to the mechanisms underlying activation of aluminium resistance in roots of Urochloa decumbens

The mechanisms of extreme Al-resistance in Urochloa decumbens are not established. Full resistance expression requires a lag time of 72–96h and is preceded by a sensitive phase (24–48h) with Al-induced root growth inhibition. The aim here was to identify key processes of the activation phase of Al-resistance analysing both root exudates and comparative root proteome. Samples were taken after 0, 24 and 96h exposure to 0 or 200μM Al. Al-induced stimulation of citrate and oxalate efflux was limited to the sensitive phase. Only 11 proteins revealed Al-induced abundance differences; six were identified. After 24h, phenylalanine ammonium lyase (PAL), methionine synthase (MS), and deoxymugineic acid synthase (DMAS) decreased, while acid phosphatase (APase) abundance increased. Coincident with growth recovering, PAL and MS, but not DMAS, returned to initial levels. After 96h, γ‑carbonic anhydrase (γ‑CA) and adenylate kinase (AK) along with two unidentified proteins were more abundant. In conclusion, few protein changes characterize the initial response to Al in signalgrass. During the alarm phase, changes are related to P-mobilization, downregulation of Fe-acquisition, reduction of phenolic biosynthesis, and small stimulation of organic acid exudation. After recovering (resistant phase), biosynthesis of phenolics and methionine, but not Fe-mobilization are re-established. Full expression of Al-resistance is characterized by enhanced γ‑CA mediating mitochondrial complex I assembly and increased AK abundance indicating higher root respiration and better provision of ADP and Mg2+ to ATP synthase, respectively. The unidentified proteins and the specific role of γ‑CA in Al resistance of U. decumbens will centre future research.

Apple MdMYC2 reduces aluminum stress tolerance by directly regulating MdERF3 gene

Jasmonate (JA) and ethylene are involved in the regulation of the aluminum (Al)-induced growth inhibition. Although it has been reported that JA enhances Al-induced root-growth inhibition, its role in the regulation of growth interplaying with ethylene is still not well understood. In this study, we investigated the mechanism underlying the effect of apple MdMYC2 transcription factor on Al stress. Overexpression lines were used for functional analysis. Real-time quantitative RT-PCR was used to examine the expression level of ethylene responsive genes. ChIP-PCR, EMSA, and Y1H assays were used to test whether MdMYC2-GST fusion protein could directly bind to MdERF3 promoter. Transient transactivation assays in tobacco leave were conducted to confirm whether MdMYC2 positively regulated the expression of MdERF3. MdMYC2 negatively regulated Al tolerance with up-regulating the expression of ethylene responsive genes. Moreover, MdMYC2 was observed to bind to the promoter of MdERF3, a positive regulator of ethylene biosynthesis, and directly activated its transcription. And applying the antagonist of ethylene biosynthesis, AVG, alleviated MdMYC2-modulated growth inhibition in Al stress. We consider that MdMYC2 protein directly interacts and promotes the transcript of MdERF3 to affect ethylene biosynthesis, thereby regulating the Al-mediated stress response. Our findings provide a deeper understanding of the crosstalk between JA and ethylene as well as JA-mediated growth inhibition in apple.

LEUNIG_HOMOLOG transcriptional co-repressor mediates aluminium sensitivity through PECTIN METHYLESTERASE46-modulated root cell wall pectin methylesterification in Arabidopsis.

A major factor determining aluminium (Al) sensitivity in higher plants is the binding of Al to root cell walls. The Al binding capacity of cell walls is closely linked to the extent of pectin methylesterification, as the presence of methyl groups attached to the pectin backbone reduces the net negative charge of this polymer and hence limits Al binding. Despite recent progress in understanding the molecular basis of Al resistance in a wide range of plants, it is not well understood how the methylation status of pectin is mediated in response to Al stress. Here we show in Arabidopsis that mutants lacking the gene LEUNIG_HOMOLOG (LUH), a member of the Groucho-like family of transcriptional co-repressor, are less sensitive to Al-mediated repression of root growth. This phenotype is correlated with increased levels of methylated pectin in the cell walls of luh roots as well as altered expression of cell wall-related genes. Among the LUH-repressed genes, PECTIN METHYLESTERASE46 (PME46) was identified as reducing Al binding to cell walls and hence alleviating Al-induced root growth inhibition by decreasing PME enzyme activity. seuss-like2 (slk2) mutants responded to Al in a similar way as luh mutants suggesting that a LUH-SLK2 complex represses the expression of PME46. The data are integrated into a model in which it is proposed that PME46 is a major inhibitor of pectin methylesterase activity within root cell walls.

Nitric oxide-mediated cytosolic glucose-6-phosphate dehydrogenase is involved in aluminum toxicity of soybean under high aluminum concentration

Glucose-6-phosphate dehydrogenase (G6PDH) has been reported to be involved in resistance to various environmental stresses. However, the role of G6PDH in aluminum (Al) toxicity remains unclear. Physiological and biochemical methods together with histochemical analysis were used to investigate the participation of G6PDH in Al-induced inhibition of root growth. Exposure to high Al concentration caused a significant increase in the activities of total and cytosolic G6PDH in roots of soybean. Al-induced inhibition of root growth and oxidative stress were alleviated by a G6PDH inhibitor. Reactive oxygen species (ROS) accumulation in Al-treated root apexes could be abolished by a NADPH oxidase inhibitor. Furthermore, treatment with a G6PDH inhibitor reduced NADPH content and NADPH oxidase activity in Al-treated root apexes. Further investigation demonstrates that nitric oxide (NO) mediates Al-induced increase in cytosolic G6PDH activity by modulating the expression of genes encoding cytosolic G6PDH. In addition, nitrate reductase pathway is mainly responsible for Al-induced NO production in root apexes. These results indicate that NADPH produced by NO-modulated cytosolic G6PDH in root apexes is responsible for ROS accumulation mediated by NADPH oxidase under Al stress, subsequently suffering from oxidative stress and thus causing the inhibition of root elongation.

MiR393-Mediated Auxin Signaling Regulation is Involved in Root Elongation Inhibition in Response to Toxic Aluminum Stress in Barley.

High-throughput small RNA sequencing has identified several potential aluminum (Al)-responsive microRNAs (miRNAs); however, their regulatory role remains unknown. Here, we identified two miR393 family members in barley, and confirmed two target genes-HvTIR1 and HvAFB-through a modified form of 5'-RACE (rapid amplification of cDNA ends) as well as degradome data analysis. Furthermore, we investigated the biological function of the miR393/target module in root development and its Al stress response. The investigation showed that miR393 affected root growth and adventitious root number by altering auxin sensitivity. Al3+ exposure suppressed miR393 expression in root apex, while overexpression of miR393 counteracted Al-induced inhibition of root elongation and alleviated reactive oxygen species (ROS)-induced cell death. Target mimic (MIM393)-mediated inhibition of miR393's activity enhanced root sensitivity to Al toxicity. We also confirmed that auxin enhanced Al-induced root growth inhibition in barley via application of exogenous 1-naphthaleneacetic acid (NAA), and the expression of auxin-responsive genes in the root apex was induced upon Al treatment. Overexpression of miR393 attenuated the effect of exogenous NAA on Al-induced root growth inhibition, and down-regulated the expression of auxin-responsive genes under Al stress, implying that miR393 regulates root sensitivity to Al through the alteration of auxin signaling output in barley. Therefore, these data indicate that miR393 acts as an integrator of environmental cues in auxin signaling, and suggest a new strategy to improve plant resistance to Al toxicity.

Jasmonic Acid Enhances Al-Induced Root Growth Inhibition.

Phytohormones such as ethylene and auxin are involved in the regulation of the aluminum (Al)-induced root growth inhibition. Although jasmonate (JA) has been reported to play a crucial role in the regulation of root growth and development in response to environmental stresses through interplay with ethylene and auxin, its role in the regulation of root growth response to Al stress is not yet known. In an attempt to elucidate the role of JA, we found that exogenous application of JA enhanced the Al-induced root growth inhibition. Furthermore, phenotype analysis with mutants defective in either JA biosynthesis or signaling suggests that JA is involved in the regulation of Al-induced root growth inhibition. The expression of the JA receptor CORONATINE INSENSITIVE1 (COI1) and the key JA signaling regulator MYC2 was up-regulated in response to Al stress in the root tips. This process together with COI1-mediated Al-induced root growth inhibition under Al stress was controlled by ethylene but not auxin. Transcriptomic analysis revealed that many responsive genes under Al stress were regulated by JA signaling. The differential responsive of microtubule organization-related genes between the wild-type and coi1-2 mutant is consistent with the changed depolymerization of cortical microtubules in coi1 under Al stress. In addition, ALMT-mediated malate exudation and thus Al exclusion from roots in response to Al stress was also regulated by COI1-mediated JA signaling. Together, this study suggests that root growth inhibition is regulated by COI1-mediated JA signaling independent from auxin signaling and provides novel insights into the phytohormone-mediated root growth inhibition in response to Al stress.
