Hexamitiasis and an emaciation syndrome in pheasant poults: clinical aspects and differential diagnosis.

Clinical hexamitiasis was recorded in pheasant poults between six and 12 weeks old, after placing the birds into release pens, and was characterised by reduced appetite, lethargy and emaciation. Post mortem the carcases were dehydrated. The presence in the lumen of the gut of characteristic motile organisms which could often be found several hours after death, provided a good clinical diagnosis. An emaciation syndrome, clinically similar but not associated with hexamitiasis or other pathogens also occurs in poults. It is characterised by extreme emaciation, largely confined to the pectoral muscles, and dehydration although the birds continue to eat and drink; the cause is unknown. Both hexamitiasis and the emaciation syndrome can cause high morbidity and mortality.

Temporal changes after death in primate diaphragm muscle oxidative enzyme activity.

We have previously reported that muscle fibers in fresh samples of the diaphragm muscle of prenatal baboons have moderate to high oxidative capacity and are resistant to fatigue in vitro. These conclusions are in conflict with those of others on diaphragmatic muscle fibers studied in autopsy specimens from human infants. Reasons for these divergent interpretations could include species difference and the use of autopsy specimens rather than fresh tissue samples. We have, therefore, tested whether characteristics of human infant diaphragm muscle fibers differ from those of premature baboons, and whether the use of autopsy specimens alters interpretations of histochemical results. Samples obtained from premature, newborn, or adult baboons were quick-frozen immediately after death or after storage for as long as 24 h. Samples were obtained at autopsy from human infants at 4 to 24 h after death. Histochemical assay for NADH-TR activity was performed on cross sections. Samples from baboons at any age showed deterioration with storage, but the muscles from premature and newborn animals were considerably more susceptible to damage than those of adults. Fibers in human infant diaphragm obtained within 10 h of death looked remarkably similar to those of the infant baboons. However, samples obtained at later times after death showed deterioration and loss of oxidative enzyme activity. We conclude that diaphragmatic muscle fibers of humans and nonhuman primates are similar in enzymatic profile, but that elapsed time after death can reduce the intensity of mitochondrial enzyme assays. The decrement in tissue preservation with elapsed time after death is less pronounced in more mature muscles.

Postmortem fixation of human lungs for electron microscopy.

A simple method of lung tissue preservation immediately post mortem by transthoracic injection of stained glutaraldehyde is described. By repetitive injections at varying time intervals after death it can be shown qualitatively and quantitatively by morphometry that normal lung tissue shows good preservation for at least 8 hours, whereas autolysis sets in shortly after death in diseased lungs.

RAPID NEEDLE SAMPLING OF THE BRAIN AFTER DEATH FOR PATHOLOGY AND VIROLOGY

Vital reaction as enzymatic response to injury--with regard to distinction between antemortem and postmortem skin wounds by histochemical methods.

In order to discover early vital reactions the authors investigated the changes in the activity of alkaline phosphatase, acid phosphatase and esterase in ante ­ mortem and postmortem skin wounds. Dermal connective tissues adjacent to the wound edge showed a decrease in the activity of the three enzymes from 2 hours after vital wounding. Surround­ ing the decreased zone, along the wound edge, the activit y of the three enzymes increased simultaneously in local fibroblasts from 2 hours and in immigrating leucocytes from 4 hours after vital wounding. The authors consider that these phenomena belong to the intravital reactions, since there were no such changes in postmortem wounds. Epidermis and hair follicles in the vicinity of the wound edge revealed an increased esterase activity as early as 30 minutes after vital wounding. Such a phenomenon was also seen even in postmortem skin wounds which were inflicted within 2 hours after death and preserv ed for a certain time inter val. On a lkaline phosphatase and acid phosphatase, increased reactions in the epidermis and hair follicles could not be observed. It seems that this increase in esterase activity corresponds to the intermediate reactions. Accordingly, whether a wound is of vital origin or not, might not be decided solely by the earlier increase in esterase a ctivity. Further, because of the simultaneous activation of the three enzymes, the estimation of the age of wounds should b e based rather on the intensity and localization of the enzyme activit ies.
