Inhibition of the extraneuronal metabolism of noradrenaline in the isolated heart by adrenergic blocking agents.

ADRENERGIC blocking agents inhibit the response of effector cells to noradrenaline and related amines1; several of these drugs also act as inhibitors of the uptake of noradrenaline into sympathetic nerves2,3. This communication describes a novel action of adrenergic blocking agents, the ability to inhibit the extraneuronal metabolism of noradrenaline in a peripheral tissue.

Pharmacological studies of spinal cord adrenergic and cholinergic mechanisms and their relation to physical dependence on morphine.

Several investigators (M~YzeE~ and KLI~G)~, 1962; Gu~-~TE~ 1963; SLoA~ et al., 1963) have demonstrated that chronic intoxication with morphine causes an increase in both brain and adrenal catecholamine levels. In addition, abrupt withdrawal of morphine in chronically intoxicated animals results in a fall in brain levels and increased excretion of" cateeholamines. These findings have suggested that there may be adaptive changes in cateeholamine metabolism during addiction to morphine, as well as increased utilization of catecholamines both in the brain and in the adrenal medulla during withdrawal. Because the spinal cord reflexes seem to be more amenable to physiological and pharmacological analysis than more complex supraspinal functional systems, and because both the effects of morphine and morphine dependence on spinal cord reflexes have been characterized (WmLEa and FRANK, 1948), it was felt that the importance and the necessity of adrenergic synapses and cateeholamines in the functioning of the spinal cord and in the genesis of physical dependence could be definitively assessed in this preparation. Because of possible interactions between eholinergie and adrenergic neurones in the central nervous system, pharmacological experiments were a]so conducted to assess the role of atropine sensitive mechanisms and presumably eholinergie synapses in the genesis of physical dependence. To these ends, three different series of experiments were conducted: (1) A study of the effects of peripherally active adrenergie and adrenergic-blocking agents on spinal cord reflexes in chronic spinal dogs, (2) a study of the effects of peripherally and centrally active cholinergie and cholinergic-blocking drugs in chronic spinal dogs, and

Effect of catecholamines and adrenergic blocking agents on oxidative phosphorylation in rat heart mitochondria.

Polarographic methods were used to examine oxidative phosphorylation in mitochondria isolated from hearts of rats treated with 5 mg/kg epinephrine, norepinephrine, or isoproterenol in oil given subcutaneously. Uncoupling of oxidative phosphorylation occurred (P/O ratio 2.1 ± .04 compared to P/O ratio of controls, 2.9 ± .03). Other indices of mitochondrial function, including oxygen consumption, respiratory control, adenosinetriphosphatase activity, and swelling were normal. The time course of depression of P/O ratio was parallel to that of myocardial catecholamine uptake, yet normal mitochondria were not affected by catecholamines in vitro (10 -3 M). Adrenergic blocking agents in vitro did not improve the function of mitochondria from catecholamine-treated animals. No free fatty acids were detectable in extracts of mitochondria from either group by gas liquid chromatography, and the lipid profiles of extracts from each were similar by thin layer chromatography. The data indicate a means by which large quantities of exogenous or endogenous catecholamines could lead to increased oxygen consumption in vivo, or contribute to impairment of myocardial function.

Acute and chronic effects of an adrenergic beta-receptor blocking agent (propranolol) in treatment of cardiac arrhythmias.

The effects of intravenous and oral administration of propranolol, an adrenergic beta-receptor blocking agent, have been studied in 29 patients with various cardiac arrhythmias. The ventricular responses in chronic or paroxysmal ectopic supraventricular arrhythmias were decreased at rest or during exercise. Sinus tachycardias were regularly slowed. Two supraventricular tachycardias and one ventricular tachycardia, all consistently precipitated by exogenous stimuli, were prevented. Six instances of digitalis-induced arrhythmias were responsive to treatment. Propranolol, especially when given orally, is of definite value in selected disturbances of cardiac rhythm. Reasonable caution should be exercised, however, because of the risk of precipitation of congestive heart failure or hypotension in patients with limited cardiac reserves.

Inhibitory Action of Adrenergic Blocking Agents on Catecholamine Release and Uptake in Isolated Nerve Granules

AbstractA series of adrenergic α‐and β‐blockers have been found to decrease the release rate of nor‐adrenaline from a suspension of bovine splenic nerve granules incubated in phosphate buffer at pH 7.5. The same blockers also inhibited the uptake of noradrenaline in partially depleted nerve granules in the presence of ATP—Mg++. It is tentatively suggested that the interactions of the adrenergic blocking agents with uptake mechanisms for noradrenaline in the nerve granules and at the receptor level in effector cells involve certain common features.

Antagonism of anticonvulsants by adrenergic blocking agents.

SummaryAlpha and beta adrenergic receptor blocking agents were tested for their ability to block the anticonvulsant effect of diphenylhydantoin, acetazolamide and chlor-diazepoxide. The anticonvulsant effect of acetazolamide was significantly antagonized by all the blocking agents but DCI, while the anticonvulsant activity of diphenylhydantoin and chlordiazepoxide was only antagonized by dibenzyline. The blockade of the anticonvulsant effect of acetazolamide by adrenergic blocking agents supports the hypothesis that catechola mines are involved in the anticonvulsant effect of this compound.

Endotoxin-induced susceptibility to staphylococcal infection and its reversal by adrenergic blocking agents.

J. Bacteriol. 90:1001-1006. 1965.-The transient phase of increased susceptibility to bacterial infection in mice provoked by prior administration of small doses of endotoxin was investigated for possible mediation by vasoactive substances. Animals were given endotoxin intravenously shortly before intraperitoneal injection of Staphylococcus aureus Smith, thereby lowering the lethal inoculum 10-fold. To determine whether this susceptibility state could be obviated, mice were pretreated with phenoxybenzamine or dibenzylchlorethylamine. Mortality decreased from an average of 81% in the endotoxin control groups to about 23% in the treated mice, closely approximating the mortality in control mice injected with saline and staphylococci. Neither antiadrenergic agent independently altered the resistance of mice to a higher lethal staphylococcal challenge, nor did these materials induce extravascular leukocyte mobilization into the peritoneal cavity. The results suggest a possible role of vasoactive staphylococcal alpha-toxin, as well as epinephrine or epinephrine-like factors, in this altered state of resistance to staphylococcal infection.

Effect of Certain Drugs on Perfused Human Placenta V: Use of α and β Adrenergic Blocking Agents to Determine the Specific Effector Site of Vasoactive Substances

In this investigation pretreatment of placental vessels with both an <i>α</i> (phentolamine) and a <i>β</i> (dichloroisoproterenol, DCI) adrenergic blocking agent was performed to determine the specific nature of the effector site upon which epinephrine and isoproterenol act. The results of this combination of blocking agents in producing an almost complete inhibition of the diphasic action of epinephrine provided further support to the hypothesis that both <i>α</i> and <i>β</i> adrenergic receptors exist in placental vessels. In addition, DCI alone had relatively little effect on the action of large doses of isoproterenol in these vessels, but did reverse the vasodilation produced by small doses of this agent. The effects of DCI on the activity of vasodilators and phentolamine on vasoconstrictors were also investigated. DCI appeared to have only a slight, if any, blocking action on the vascular responses to nitroglycerin, papaverine, isosorbide dinitrate, sodium nitrite, sodium cobaltinitrite, and cyclandelate. Likewise, phentolamine had relatively little blocking action on the vaso-constricting effect of vasopressin and oxytocin.
