Response of C-6 glioma to isoproterenol and papaverine in vivo depends on beta-adrenergic receptor density.

Rat C-6 glioma serves as an experimental model for human glioma. C-6 glioma cells carried to high culture passages in medium containing 10% fetal bovine serum when injected in vivo are unresponsive to treatment with the beta-adrenergic agonist isoproterenol and the phosphodiesterase inhibitor papaverine. When C-6 glioma cells are kept in culture in serum-containing medium, beta-adrenergic receptor density falls and, concomitantly, ability to accumulate cyclic adenosine monophosphate in response to stimulation with catecholamines declines. Responsiveness to treatment in vivo with a beta-adrenergic agonist was restored when C-6 glioma cells were cultured in serum-free defined medium prior to systemic injection into rats. Culturing of C-6 glioma cells in serum-free medium significantly increases the number of beta-adrenergic receptors when compared with C-6 glioma cells grown in serum-containing medium.

Inhibition by lithium of PGE1-sensitive adenylate cyclase in neuroblastoma X glioma hybrid cells: approach to the attenuation of the opiate withdrawal syndrome.

Exposure of neuroblastoma X glioma hybrid cells to LiCl (Li+) for 48 h significantly reduces prostaglandin E1 (PGE1)-dependent cyclic adenosine monophosphate (AMP) accumulation. When applied simultaneously with morphine for the same period, Li+ inhibits the excessive accumulation of cyclic AMP in response to PGE1 characteristically exhibited by these cells upon opiate withdrawal. Likewise, Li+ inhibits, in vitro, PGE1-dependent cyclic AMP accumulation in tissue slices from rat corpus striatum. Prophylactic administration of Li+ in the diet attenuates the withdrawal symptoms in morphine-addicted rats. These findings support clinical reports that suggest that prophylactic treatment with Li+ attenuates the severity of the opiate withdrawal syndrome.

Metabolic Studies in a Case of Hypocalcemia and Osteoblastic Metastases

Metabolic studies were performed in a case of severe hypocalcemia and extensive osteoblastic metastases from prostatic adenocarcinoma in a 75-year-old man. The predominant abnormality appeared to be an increase in bone avidity for calcium, phosphorus, and magnesium, as evidenced by increased retention rates of infused calcium and magnesium and low urinary and stool values for calcium, phosphorus, and magnesium. Other aberrations included the presence of a relative hypoparathyroidism, peripheral resistance to parathyroid injection (United States Pharmacopeia), and subnormal plasma levels of magnesium and vitamin D metabolites. Prior to magnesium therapy there was no phosphaturic or cyclic adenosine monophosphate (cAMP) response to exogenous PTE, whereas after therapy there was normal cAMP but absent phosphaturic response. Hypomagnesemia together with relative parathyroid insufficiency could be responsible for the low 25-hydroxyvitamin D (25-[OH]-D) and 1,25-dihydroxyvitamin D (1,25-[OH]2D) levels. The sporadic elevation in thyrocalcitonin levels was consistent with the possibility of ectopic production of this polypeptide. These study results strongly suggest that the hypocalcemia associated with osteoblastic metastases results from a number of interrelated abnormalities, chief of which is the great avidity of bone for calcium. Cyclic 3′,5′-adenosine monophosphate response in the rabbit lung—Adult properties and development

When various doses of catecholamines (norepinephrine, epinephrine and isoproterenol) and histamine were added to incubated rabbit lung slices, there was a dose-related increase in levels of cyclic 3′,5′-adenosine monophosphate (cAMP). Isoproterenol was the most potent and histamine the least potent agent capable of eliciting this response. The time course of this hormonally induced cAMP response was maximal from 6–36 min. Propranolol but not phentolamine significantly antagonized the catecholamine-induced cAMP response, while tripelennamine blocked the effects of histamine. Aminophylline enhanced the catecholamine and histamine stimulation of cAMP. During lung development, a cAMP response to epinephrine was detected at fetal day 21. By fetal day 25 a prominent cAMP response to epinephrine, norepinephrine and histamine occurred. The response to catecholamines dropped slightly at birth, and the highest cAMP levels were seen at 4 days postpartum. The histamine-induced response was maximal at fetal day 28, was diminished at birth and another peak response was observed at 20 days postpartum.
