Assessing the performance of polyphosphate accumulating organisms in a full-scale side-stream enhanced biological phosphorous removal.

Phosphorous (P) removal in wastewater treatment is essential to prevent eutrophication in water bodies. Side-stream enhanced biological phosphorous removal (S2EBPR) is utilized to improve biological P removal by recirculating internal streams within a side-stream reactor to generate biodegradable carbon (C) for polyphosphate accumulating organisms (PAOs). In this study, a full-scale S2EBPR system in a water resource recovery facility (WRRF) was evaluated for 5 months. Batch experiments revealed a strong positive correlation (r = 0.91) between temperature and C consumption rate (3.56-8.18 mg-COD/g-VSS/h) in the system, with temperature ranging from 14°C to 18°C. The anaerobic P-release to COD-uptake ratio decreased from 0.93 to 0.25 mg-P/mg-COD as the temperature increased, suggesting competition between PAOs and other C-consumers, such as heterotrophic microorganisms, to uptake bioavailable C. Microbial community analysis did not show a strong relationship between abundance and activity of PAO in the tested WRRF. An assessment of the economic feasibility was performed to compare the costs and benefits of a full scale WRRF with and without implementation of the S2EBPR technology. The results showed the higher capital costs required for S2EBPR were estimated to be compensated after 5 and 11 years of operation, respectively, compared to chemical precipitation and conventional EBPR. The results from this study can assist in the decision-making process for upgrading a conventional EBPR or chemical P removal process to S2EBPR. PRACTITIONER POINTS: Implementation of S2EBPR presents adaptable configurations, exhibiting advantages over conventional setups in addressing prevalent challenges associated with phosphorous removal. A full-scale S2EBPR WRRF was monitored over 5 months, and activity tests were used to measure the kinetic parameters. The seasonal changes impact the kinetic parameters of PAOs in the S2EBPR process, with elevated temperatures raising the carbon demand. PAOs abundance showed no strong correlation with their activity in the full-scale S2EBPR process in the tested WRRF. Feasibility assessment shows that the benefits from S2EBPR operation can offset upgrading costs from conventional BPR or chemical precipitation.

Coupled In-Situ Fermentation for Enhanced Biological Phosphorus Removal from Digested Swine Wastewater

This study demonstrated the feasibility of enhanced biological phosphorus removal coupled with in-situ fermentation (EBPR-F) to improve phosphorus removal from real digested swine wastewater. We used fermentable substrates (casein hydrolysate and glucose) as the external carbon sources to promote in-situ fermentation and enhance biological phosphorus removal. Compared with conventional EBPR dominated by Candidatus Accumulibacter, EBPR-F had significantly better phosphorus removal with enriched polyphosphate-accumulating organisms (PAOs). Under supplementation with 100 mg/L glucose, total phosphorus (TP) removal was over 95% in EBPR-F, with an average TP concentration in the effluent below 1.0 mg/L, satisfying the discharge standard (8 mg P/L) in China. The PAO activity and relative abundance of Candidatus Accumulibacter (44.7% ± 3.1%) and Tetrasphaera (18.1% ± 6.6%) in EBPR-F were much higher than those in EBPR. The improvement in phosphorus removal of EBPR-F was due to the enrichment of Tetrasphaera through the enhanced in-situ fermentation, as Tetrasphaera can efficiently ferment complex organic matter and provide bioavailable organics for phosphorus removal.

Assessing the impact of increasing phenol loads on the performance and stability of an aerobic granular sludge system

This work evaluated the impact of phenol on the performance and stability of an aerobic granular sludge (AGS) sequencing batch reactor (SBR) operated under alternating anaerobic-aerobic conditions for simultaneous organic matter, nitrogen, and phosphorus removal. The reactor was fed with synthetic wastewater whose phenol concentration was gradually increased from 2 to 200 mg L−1, while the other carbon source (acetate) was reduced proportionally to keep the organic loading rate invariable. Up to 20 mg L−1 of phenol, the average COD removal was above 90%, while practically full ammonium, phosphorus, and phenol removal was achieved. However, at higher phenol concentrations, the activity of polyphosphate-accumulating organisms and nitrifiers was affected. Although the overall COD removal was only slightly affected by increasing phenol loads, replacing the main carbon source from acetate to phenol changed the organic matter removal profile over the cycle, with the latter being converted mainly within the aerated stage of the SBR. As a result, there was filamentous growth that deteriorated the settling properties of the granules. The establishment of an anaerobic feast-aerobic famine regime over the non-aerated and aerated phases of the AGS SBR and the presence of a readily biodegradable substrate (such as acetate) proved to be crucial for maintaining granules stability. Having phenol as the sole organic carbon source led to complete process failure regarding nutrient removal capability.

Sulfide inhibition on polyphosphate accumulating organisms and glycogen accumulating organisms: Cumulative inhibitory effect and recoverability

Sulfate in wastewater can be reduced to sulfide and its impact on the stability of enhanced biological phosphorus removal (EBPR) is still unclear. In this study, the metabolic changes and subsequent recovery of polyphosphate accumulating organisms (PAOs) and glycogen accumulating organisms (GAOs) were investigated at different sulfide concentrations. The results showed that the metabolic activity of PAOs and GAOs was mainly related to H2S concentration. Under anaerobic conditions, the catabolism of PAOs and GAOs was promoted at H2S concentrations below 79 mg/L S and 271 mg/L S, respectively, and inhibited above these concentrations; whereas anabolism was consistently inhibited in the presence of H2S. The phosphorus (P) release was also pH-dependent due to the intracellular free Mg2+ efflux from PAOs. H2S was more destructive to the esterase activity and membrane permeability of PAOs than those of GAOs and prompted intracellular free Mg2+ efflux of PAOs, resulting in worse aerobic metabolism and subsequent recovery of PAOs than GAOs. Additionally, sulfides facilitated the production of extracellular polymeric substances (EPS), especially tightly bound EPS. The amount of EPS in GAOs was significantly higher than that in PAOs. The above results indicated that sulfide had a stronger inhibition to PAOs than GAOs, and when sulfide was present, GAOs had a competitive advantage over PAOs in EBPR.

Enhancing phosphorus removal of photogranules by incorporating polyphosphate accumulating organisms

Photogranules are a novel wastewater treatment technology that can utilize the sun's energy to treat water with lower energy input and have great potential for nutrient recovery applications. They have been proven to efficiently remove nitrogen and carbon but show lower conversion rates for phosphorus compared to established treatment systems, such as aerobic granular sludge. In this study, we successfully introduced polyphosphate accumulating organisms (PAOs) to an established photogranular culture. We operated photobioreactors in sequencing batch mode with six cycles per day and alternating anaerobic (dark) and aerobic (light) phases. We were able to increase phosphorus removal/recovery by 6 times from 5.4 to 30 mg/L/d while maintaining similar nitrogen and carbon removal compared to photogranules without PAOs. To maintain PAOs activity, alternating anaerobic feast and aerobic famine conditions were required. In future applications, where aerobic conditions are dependent on in-situ oxygenation via photosynthesis, the process will rely on sunlight availability. Therefore, we investigated the feasibility of the process under diurnal cycles with a 12-h anaerobic phase during nighttime and six short cycles during the 12 h daytime. The 12-h anaerobic phase had no adverse effect on the PAOs and phototrophs. Due to the extension of one anaerobic phase to 12 h the six aerobic phases were shortened by 47% and consequently decreased the light hours per day. This resulted in a decrease of phototrophs, which reduced nitrogen removal and biomass productivity up to 30%. Finally, we discuss and suggest strategies to apply PAO-enriched photogranules at large-scale.

Partitioning of nutrient removal contribution between granules and flocs in a hybrid granular activated sludge system

Sludge granulation in continuous-flow systems is an emerging technology to intensify existing activated sludge infrastructure for nutrient removal. In these systems, the nutrient removal contributions and partitioning of microbial functions between granules and flocs can offer insights into process implementations. To this end, a reactor system that simulates the continuous-flow environment using an equal amount of initial granule and floc biomass was investigated. The two operational strategies for maintaining granule growth in the continuous-flow system were (a) the higher solids retention time (SRT) for the granules versus flocs, as well as (b) selective feeding of carbon to the granules. The SRT of the large granule fractions (>425 µm, LG) and floc/small granule fractions (<425 µm, FSG) were controlled at 20 and 2.7–6.0 days, respectively. Long term operation of the hybrid granule/floc system achieved high PO43- and NH4+ removal efficiencies. Higher polyphosphate-accumulating organisms (PAO) activity was observed in the FSG than LG, while ammonia-oxidizing bacteria (AOB) activities were similar in the two biomass fractions. Nitrite shunt was observed in the FSG, possibly due to out-competition by the high NOB activity in LG. More importantly, washing out the FSG caused a reduction in LG's AOB and PAO activity, indicating a possible dependency of LG on FSG for maintaining its nutrient removal capacity. Our findings highlighted the partitioning and potential competition/cooperation of key microbial functional groups between LG and FSG, facilitating nutrient removal in a hybrid granular activated sludge system, as well as implications for practical application of the treatment platform.

Nitrite and nitrate inhibition thresholds for a glutamate-fed bio-P sludge

Enhanced biological phosphorus removal (EBPR) is an efficient and sustainable technology to remove phosphorus from wastewater. A widely known cause of EBPR deterioration in wastewater treatment plants (WWTPs) is the presence of nitrate/nitrite or oxygen in the anaerobic reactor. Moreover, most existing studies on the effect of either permanent aerobic conditions or inhibition of EBPR by nitrate or free nitrous acid (FNA) have been conducted with a “Candidatus Accumulibacter” or Tetrasphaera-enriched sludge, which are the two major reported groups of polyphosphate accumulating organisms (PAO) with key roles in full-scale EBPR WWTPs. This work reports the denitrification capabilities of a bio-P microbial community developed using glutamate as the sole source of carbon and nitrogen. This bio-P sludge exhibited a high denitrifying PAO (DPAO) activity, in fact, 56% of the phosphorus was uptaken under anoxic conditions. Furthermore, this mixed culture was able to use nitrite and nitrate as electron acceptor for P-uptake, being 1.8 μg HNO2–N·L−1 the maximum FNA concentration at which P-uptake can occur. Net P-removal was observed under permanent aerobic conditions. However, this microbial culture was more sensitive to FNA and permanent aerobic conditions compared to “Ca. Accumulibacter”-enriched sludge.

Side-stream enhanced biological phosphorus removal (S2EBPR) process improves system performance - A full-scale comparative study

To address the common challenges in enhanced biological phosphorus removal (EBPR) related to stability and unfavorable influent carbon to phosphorus ratio, a side-stream EBPR (S2EBPR) process that involves a side-stream anaerobic biological sludge hydrolysis and fermentation reactor was proposed as an emerging alternative. In this study, a full-scale pilot testing was performed with side-by-side operation of a conventional anaerobic-anoxic-aerobic (A2O) process versus a S2EBPR process. A comparison of the performance, activity and microbial community between the two configurations was performed. The results demonstrated that, with the same influent wastewater characteristics, S2EBPR configuration showed improved P removal performance and stability than the conventional A2O configuration, especially when the mixers in the side-stream anaerobic reactor were operated intermittently. Mass balance analysis illustrated that both denitrification and EBPR were enhanced in S2EBPR configuration, where return activated sludge was diverted into the anaerobic zone to promote fermentation and enrichment of polyphosphate accumulating organisms (PAOs), and the influent was bypassed to the anoxic zone for enhancing denitrification. A relatively higher PAO activity and total PAO abundance were observed in S2EBPR than in A2O configuration, accompanied by a higher degree of dependence on glycolysis pathway than tricarboxylic acid cycle. No significant difference in the relative abundances of putative PAOs, including Ca. Accumulibacter and Tetrasphaera, were observed between the two configurations. However, higher microbial community diversity indices were observed in S2EBPR configuration than in conventional one. In addition, consistently lower relative abundance of known glycogen accumulating organisms (GAOs) was observed in S2EBPR system. Extended anaerobic retention time and conditions that generate continuous and more complex volatile fatty acids in the side-stream anaerobic reactor of S2EBPR process likely give more competitive advantage for PAOs over GAOs. PAOs exhibited sustained EBPR activity and delayed decay under extended anaerobic condition, likely due to their versatile metabolic pathways depending on the availability and utilization of multiple intracellular polymers. This study provided new insights into the effects of implementing side-stream EBPR configuration on microbial populations, EBPR activity profiles and resulted system performance.

Removal of nutrients and emission of nitrous oxide during simultaneous nitrification, denitrification and phosphorus removal process with metal ions addition

The impacts of metal ions (Cu2+ and Fe3+) on nutrients removal and nitrous oxide (N2O) emission during the simultaneous nitrification, denitrification and phosphorus removal (SNDPR) process were evaluated and the mechanism was further investigated. Results showed that both Cu2+ and Fe3+ addition at moderate concentrations enhanced nitrogen removal by reinforcing the denitrification process. The long-term addition of Fe3+ weakened the removal of phosphorus by 2.3%–11.7%, due to the suppression of polyphosphate-accumulating organisms (PAOs) growth and activity. N2O production and emission factor decreased greatly by the addition of Cu2+. This decrease was caused by the enhancement of nitrite reductase and N2O reductase (N2OR) activities. On the contrary, Fe3+ addition at moderate concentrations stimulated the N2O production. Fe3+ enhanced the nitric oxide reductase (NOR) and increased the NOR/N2OR ratio, leading to the accumulation of N2O. Moreover, addition of Fe3+ caused a reduction in polyhydroxyalkanoates synthesis at the anaerobic stage and renforced glycogen-accumulating organisms (GAOs) activity instead of PAOs activity, resulting in increased N2O accumulation. Addition of high concentrations of metal ions (5 mg/L Cu2+ and 60 mg/L Fe3+ in this study) suppressed the nutrients removal and stimulated the N2O production, partly due to the inhibition of the bacterial activity.

Effects of carbon supplementary modes on phosphorus bioaccumulation/phosphorus harvesting process in a tidal flow constructed wetland.

We explored the effects of carbon supplementary modes on operational performance in a tidal flow constructed wetland (TFCW) utilizing phosphorus bioaccumulation/phosphorus harvesting (PB-PH) process. Three different carbon supplementary modes were adopted during the periodical phosphorus harvesting process. The results showed that the carbon supplementary mode significantly affected the performances of phosphorus bioaccumulation and phosphorus harvesting throughout the experiment. The optimization of carbon supplementary mode would facilitate the maintenance of abundance and activities of polyphosphate-accumulating organisms (PAOs). Subsequently, the amounts of phosphorus release and PHB synthesis in PAOs, as well as the utilization rate of supplementary carbon, could be effectively enhanced during the phosphorus harvesting process. The efficient and stable phosphorus removal effects of the TFCW could be ensured during the phosphorus bioaccumulation process. When the PB-PH cycle length was 30 d and using the continuous circulation carbon supplementary mode, the amount of phosphorus retained in the bed and the mean phosphorus harvesting efficiency of the TFCW were 26994.88 mg and 70.8%, respectively. Regarding a typical PB-PH cycle, the mean phosphorus removal efficiency of the TFCW could achieve (91.4±2.1)% during the phosphorus bioaccumulation process. The amounts of phosphorus release and PHB synthesis in PAOs, and the utilization rate of supplementary carbon in the system could reach up to (1563.72±127.84) mg, (4.52±0.39) mmol C·g-1 VSS, and (97.3±1.6)%, respectively.

Sulfide effects on the anaerobic metabolism of polyphosphate-accumulating organisms

Sulfate rich wastewaters can be generated from industry, use of seawater in urban environments, or by saline water infiltration into the sewerage. Under anaerobic conditions sulfate can be converted to sulfide, which may affect micro-organisms performing biological nutrient removal. The objective of this study was to evaluate the effect of sulfide on the activity of polyphosphate-accumulating organisms (PAO) in the anaerobic stage of the enhanced biological phosphorus removal process (EBPR). In this regard, a highly enriched culture of PAO was exposed in short-term activity tests to a range of sulfide concentrations at different operational pH values. The PAO activity was mainly affected by un-dissociated H2S. The specific acetate uptake rate was inhibited by 50% at around 60mg H2S.L−1. With increasing H2S concentrations, higher phosphate release rate to acetate uptake rate ratios were observed, possibly due to increased energy requirements for cell detoxification. Mathematical expressions were developed, which satisfactorily described the sulfide effects on the acetate uptake rate and phosphate release rate. The results show that, dependent on the pH, EBPR might be negatively affected by total sulfide concentrations exceeding 275mg SO4.L−1 at pH 6.5 or 1200mg SO4.L−1 at pH 7.8mg SO4.L−1 or when freshwater is partially replaced by seawater more than 45% (pH 7.8) or 10% (pH 6.5) used as secondary quality water. The findings of this study imply that sulfide, which is commonly found in different type of wastewaters, affects the anaerobic metabolism of PAO and may play an important role in the process performance of treatment plants treating wastewaters with high sulfide content.

Modeling the Effect of External Carbon Source Addition under Different Electron Acceptor Conditions in Biological Nutrient Removal Activated Sludge Systems.

The aim of this study was to expand the International Water Association Activated Sludge Model No. 2d (ASM2d) to predict the aerobic/anoxic behavior of polyphosphate accumulating organisms (PAOs) and "ordinary" heterotrophs in the presence of different external carbon sources and electron acceptors. The following new aspects were considered: (1) a new type of the readily biodegradable substrate, not available for the anaerobic activity of PAOs, (2) nitrite as an electron acceptor, and (3) acclimation of "ordinary" heterotrophs to the new external substrate via enzyme synthesis. The expanded model incorporated 30 new or modified process rate equations. The model was evaluated against data from several, especially designed laboratory experiments which focused on the combined effects of different types of external carbon sources (acetate, ethanol and fusel oil) and electron acceptors (dissolved oxygen, nitrate and nitrite) on the behavior of PAOs and "ordinary" heterotrophs. With the proposed expansions, it was possible to improve some deficiencies of the ASM2d in predicting the behavior of biological nutrient removal (BNR) systems with the addition of external carbon sources, including the effect of acclimation to the new carbon source.

Phosphate release involving PAOs activity during anaerobic fermentation of EBPR sludge and the extension of ADM1

Anaerobic fermentation of the enhanced biological phosphorus removal (EBPR) sludge was investigated in terms of phosphate release and volatile fatty acids (VFAs) production regarding polyphosphate accumulating organisms (PAOs) activity. PAOs decay rate during fermentation was determined as 0.35±0.03d−1. Sludge lysis was enhanced with an increase in polyhydroxyalkanoate (PHA) content. Moreover, the phosphate release profiles and the VFAs production as well as the individual VFA fractions varied with different acetate concentrations added initially. Based on these observations, anaerobic digestion model No. 1 was extended and modified by introducing: (1) processes that PAOs store 4 VFA species as PHA, which can be degraded into varied fractions of individual VFA in dependence on PHA composition, (2) the effect of PHA content on disintegration rate, and (3) phosphorus precipitation. The proposed model adequately fitted a multi-experiment, multi-variable data set, indicating that it plays an important role in predicting phosphate and VFAs variations during EBPR sludge fermentation.

Assessment of crude glycerol for Enhanced Biological Phosphorus Removal: Stability and role of long chain fatty acids

Enhanced Biological Phosphorus Removal (EBPR) of urban wastewaters is usually limited by the available carbon source required by Polyphosphate Accumulating Organisms (PAO). External carbon sources as volatile fatty acids (VFA) or other pure organic compounds have been tested at lab scale demonstrating its ability to enhance PAO activity, but the application of this strategy at full-scale WWTPs is not cost-effective. The utilization of industrial by-products with some of these organic compounds provides lower cost, but it has the possible drawback of having inhibitory or toxic compounds to PAO. This study is focused on the utilization of crude glycerol, the industrial by-product generated in the biodiesel production, as a possible carbon source to enhance EBPR in carbon-limited urban wastewaters. Crude glycerol has non-negligible content of other organic compounds as methanol, salts, VFA and long chain fatty acids (LCFA). VFA and methanol have been demonstrated to enhance PAO activity, but there is no previous study about the effect of LCFA on PAO. This work presents the operation of an EBPR SBR system using crude glycerol as sole carbon source, studying also its long-term stability. The effect of LCFA is evaluated at short and long-term operation, demonstrating for the first time EBPR activity with LCFA as sole carbon source and its long-term failure due to the increased hydrophobicity of the sludge.

Phosphorus removal in a membrane-assisted BNR process with focus on evolutions of PAOs and DPAOs

A bench-scale UCT (University of Cape Town)-type membrane bioreactor (UCT-MBR) fed with low-strength synthetic wastewater was operated to investigate phosphorus removal with reference to poly-phosphate accumulating organisms (PAOs) and denitrifying poly-phosphate accumulating organisms (DPAOs). A series of kinetic assays of PAOs and DPAOs were also conducted to analyze the metabolic activities of PAOs and DPAOs. Results showed that 93% of chemical oxygen demand (COD) and 77% of total nitrogen could be removed at 0.08 kgCOD kg(-1) MLSS d(-1) and 0.015 kgN kg(-1) MLSS d(-1) loading (MLSS: mixed liquor suspended solids). Removal efficiencies of total phosphorus increased during the experimental phase, with an ultimate removal efficiency of 96.1%. Kano and Kaer increased from 1.95 and 6.29 mgPO4(3-)-P g(-1) MLSS h(-1) to 5.47 and 11.13 mgPO4(3-)-P g(-1) MLSS h(-1) for DPAOs and PAOs respectively, with the increased ratio of DPAOs to PAOs from 31 to 49% implying DPAO metabolic activity increased faster than that of PAOs during the DPAO accumulation phase. Pano-uptake increased by 6.6 mg L(-1) and the ratio of PTano-uptake to PTupt increased from 58.97 to 91.62%. The ratio of DPAOs to PAOs tended to stabilize at around 50% over time.

Experimental evaluation of decrease in the activities of polyphosphate/glycogen‐accumulating organisms due to cell death and activity decay in activated sludge

Decrease in bacterial activity (biomass decay) in activated sludge can result from cell death (reduction in the amount of active bacteria) and activity decay (reduction in the specific activity of active bacteria). The goal of this study was to experimentally differentiate between cell death and activity decay as the cause of decrease in bacterial activity. By means of measuring maximal anaerobic phosphate release rates, verifying membrane integrity by live/dead staining and verifying presence of 16S rRNA with fluorescence in situ hybridization (FISH), the decay rates and death rates of polyphosphate-accumulating organisms (PAOs) in a biological nutrient removal (BNR) system and a laboratory phosphate removing sequencing batch reactor (SBR) system were determined, respectively, under famine conditions. In addition, the decay rate and death rate of glycogen-accumulating organisms (GAOs) in a SBR system with an enrichment culture of GAOs were also measured under famine conditions. Hereto the maximal anaerobic volatile fatty acid uptake rates, live/dead staining, and FISH were used. The experiments revealed that in the BNR and enriched PAO-SBR systems, activity decay contributed 58% and 80% to the decreased activities of PAOs, and that cell death was responsible for 42% and 20% of decreases in their respective activities. In the enriched GAOs system, activity decay constituted a proportion of 74% of the decreased activity of GAOs, and cell death only accounted for 26% of the decrease of their activity.

Optimization of biological nutrient removal in a pilot plant UCT-MBR treating municipal wastewater during start-up

This study shows that an MBR pilot plant with UCT configuration is able to obtain high nutrient removal efficiency already during start-up. The biological nutrient removal (BNR) efficiencies significantly increased towards the end of the experimental run, achieving a COD removal efficiency exceeding 94% and N removal efficiency in the range of 89 to 93%. P removal efficiencies in the range of 80 to 92% have been obtained. During the experimental period (4 months) the evolution of the activity of polyphosphate-accumulating organisms, obtained from P release and P uptake rates, showed a small increase in the activity of polyphosphate-accumulating organisms (PAOs) and denitrifying polyphosphate-accumulating organisms (DPAOs). The specific phosphate accumulation at the end of the experimental run amounted to 8.0 mg P g − 1 VSS h − 1 and 3.29 mg P g − 1 VSS h − 1 , for the PAOs and DPAOs respectively. Moreover, the DPAOs activity increased faster than PAOs activity, i.e. from 0.36 to 0.41 of phosphate uptake rate (PUR) ratio.

Development of the modified activated sludge model describing nitrite inhibition of aerobic phosphate uptake

Since metabolic pathway and enzymatic clarification of poly-phosphate accumulating organisms (PAOs) are still unclear, biological phosphorus removal (BPR) sometimes become unstable. We have focused on nitrite as one of unknown factors deteriorating BPR performance. And we obtained some findings from previous studies, namely 1) nitrite inhibits phosphate uptake and growth of PAOs, 2) nitrite inhibition persists even after nitrite disappearance, 3) PAOs with the higher relative anoxic activity are less sensitive to nitrite exposure. This study provides the modified Activated Sludge Model No.2d (ASM2d) to express nitrite inhibition properly by incorporating the assumed intracellular reaction product (reaction complex) of nitrite. The model also considers the tolerance mechanism of nitrite inhibition by the denitrifying activity of PAOs. From the results of previous experiments and relevant references, the modified model called "Nitrite-Complex model" incorporates new components, namely nitrite (S(NO(2) )) and reaction complex (X(complex)), to ASM2d. The model incorporation only nitrite inhibition was able to fit with measured phosphate concentration while nitrite exists. Whereas, this model couldn't describe the persistence of nitrite inhibition after nitrite disappeared. However, result of nitrite-complex model incorporating nitrite and reaction complex inhibition, it fits well with measured phosphate concentration not only before nitrite disappeared but also after nitrite disappeared.

Modeling the Aerobic Metabolism of Polyphosphate‐Accumulating Organisms Enriched with Propionate as a Carbon Source

In enhanced biological phosphorus removal (EBPR) systems, polyphosphate-accumulating organisms (PAOs) are primarily responsible for removing phosphate from wastewater. Propionate is an abundant carbon substrate in many EBPR plants and has been suggested to provide PAOs an advantage over their carbon competitors--the glycogen-accumulating organisms (GAOs). The aerobic metabolism of PAOs enriched with a propionate carbon source is studied in this paper. A metabolic model is proposed and experimentally validated to characterize the aerobic biochemical transformations by PAOs. The model predicts very well the experimental data obtained from the enriched PAO culture through solid-, liquid-, and gas-phase analyses. This model may be combined with previously formulated metabolic models to better describe the biochemical activity of PAOs with acetate and propionate as the primary carbon sources. Furthermore, it can also facilitate the study of the effect of different carbon sources on PAO-GAO competition.
