Antidiabetic, antioxidant, and phytochemical profile of Pennisetum glaucum cultivated in central-southern Morocco and imported from India

Alternative methods of producing food sustainably are being considered due to rapid expanding population, increasing food needs, declining agricultural soils fertility, and ongoing drought. To achieve that, we examined the pharmacological, functional, and nutritional characteristics of two cultivars of pearl millet (Pennisetum glaucum), which are distinguished by their ability to tolerate a variety of climates compared to other cereal grains. In this study, we evaluated the antioxidant and antidiabetic effects of two seeds cultivars: one cultivated in Morocco and the other grown in India. In addition to having a higher energy value (403.91 Kcal), higher protein (9.13 %) and lipid content (7.11 %), and a lower capacity for self-oxidation than the Indian variety due to its fatty acid composition, the content of phenolic compounds of Moroccan millet is well concentrated in its seeds, making it more powerful as an antioxidant and hypoglycemic plant. The ethanolic extract of Moroccan pearl millet showed the greatest total phenolic content (36.278 mg GAE/g extract), DPPH and ABTS radical scavenging activity (IC50 = 152.286 and 252.480 μg/mL respectively), α-amylase and α-glucosidase inhibitory activity (IC50 = 84.439 μg/mL and 224.103 μg/mL respectively). The evidence gathered in this study indicates that P.glaucum possesses characteristics that qualify it as a food, medicinal, and productive plant.

Free radical scavenging, α-amylase, α-glucosidase, and lipase inhibitory activities of metabolites from strawberry kombucha: Molecular docking and in vitro studies

Obesity, a global issue, is linked to cardiometabolic syndrome. Dietary modification is one of the recommended modes for managing cardiometabolic syndrome. Strawberries, a functional food, and kombucha, a fermented tea beverage, have gained attention for their health benefits. This study investigated the bioactive components of strawberry kombucha drink (SKD) and their effects on antioxidant activities and improving metabolic disorder markers. An in vitro experiment was performed to determine the effect of SKD on enzymatic parameters: lipase, α-glucosidase, and α-amylase activities. In addition, antioxidant activity using the DPPH method and quantification of the radical scavenging activity were also measured. Furthermore, untargeted metabolomic profiling of SKD and molecular docking simulation were conducted. The findings suggest that SKD, rich in secondary metabolites, can inhibit lipase, α-glucosidase, and α-amylase activities. It demonstrated in vitro anti-obesity, anti-diabetic, and antioxidant properties, potentially reducing metabolic and inflammatory issues. Thus, SKD could be a therapeutic beverage to alter metabolic issues associated with obesity. Nevertheless, further preclinical study is warranted to determine SKD’s potential in vivo.

Investigation of novel benzimidazole-based indole/thiazole hybrids derivatives as effective anti-diabetics and anti-alzheimer's agents: Structure-activity relationship insight, in vitro and in silico approaches

Alzheimer's disease (AD) and type 2 diabetes mellitus (T2DM) are common in developed countries and their incidence is increasing. The research is mostly concentrated on the exploration of novel therapies to remedy or avert these ailments. AD and T2DM have several molecular pathways in common that contributes to their degenerative development.A novel series of ten hybrid analogues (1-10) based on benzimidazole bearing indole analogues were synthesized and these scaffolds were characterized by spectroscopic techniques like HR-EIMS, 13CNMR and 1HNMR. Furthermore,the synthesized moieties were screened for α-amylase and α-glucosidase inhibitory activities. All these synthesized analogues showed good to moderate α-amylase and α-glucosidase inhibition potency ranging (4.90 ± 0.10 to 15.30 ± 0.60 µM) and (5.30 ± 0.10 to 16.10 ± 0.60 µM) while acarbose was used as a reference standard (IC50 = 10 .30 ±0.20µM), (IC50 = 9.80 ± 0.20 µM). Another series of ten hybrid analogues (11-20) based on benzimidazole bearing thiazole moieties were synthesized and were evaluated for AChE and BuChE inhibition activities. All these newly afforded analogues showed good to moderate AChE and BuChE inhibitory potential ranging of (IC50 = 0.20 ± 0.01 µM), (IC50 = 0.15 ± 0.01 µM) to (IC50 = 3.90 ± 0.20 µM), (IC50 = 3.80 ± 0.10 µM) while Donepezil was taken as a reference standard with (IC50 = 0.016 ± 0.01 µM), (IC50 = 0.30 ± 0.010 µM).The scaffolds were also characterized using spectroscopic techniques such as HR-EIMS, 13CNMR and 1HNMR for a better understanding of their structure. SAR was done for all synthesized analogues for substituents.In addition, docking research was conducted to determine the binding mode of interaction between the active site of analogues and targeted enzymes. This work aims to estimate the potential of the synthesized scaffolds as competitors to existing drugs. Incorporating molecular docking analysis with in vitro activities improves the accuracy of predictions and validates the possibility of these compounds as options for treating type-2 diabetes and Alzheimer's disease.

Nutritional composition, bioactive compounds, and pharmacological activities of tossa jute sprout (Corchorus olitorius L.): A potential functional food

This study investigated the food properties, phytochemical profiles, and pharmacological activities of tossa jute sprout, underscoring the exploration of its potential as a functional food. Tossa jute sprout is rich in protein, carbohydrate, fiber, total soluble solids, color content, total energy, vitamins, and minerals, with a lower fat and sugar content. Bioactive compound profiling found that it contains a significant quantity of total phenolics and flavonoids. The HPLC-DAD polyphenol profile detected ten compounds, with 3,4 dihydroxybenzoic acid being the most abundant compound, followed by (-) epicatechin, rosmarinic acid, kaempferol, catechin hydrate, caffeic acid, myricetin, catechol, trans-ferulic acid, and rutin hydrate. It has strong radical scavenging, reducing power, metal chelating power, and total antioxidant power, suggesting this sprout's ability to repair cellular oxidative damage or restore impaired cellular oxidative homeostasis. It also possesses strong α-glucosidase and pancreatic lipase inhibition activities, but mild α-amylase, acetylcholinesterase (AChE), and butyrylcholinesterase (BChE) inhibition activities support the sprout’s ability to lower blood sugar, suppress postprandial hyperglycemia, downregulate fat metabolism, and maintain or restore acetylcholine. Overall findings established tossa jute sprout’s high potential as a functional food in eliminating malnutrition, maintaining quality health, and eradicating or moderating ailments, including diabetes, obesity, and Alzheimer’s.

Compositional, structural, and functional characterization of fucoidan extracted from Sargassum polycystum collected from Saint Martin's Island, Bangladesh

Fucoidan is a sulphated polysaccharide with diversified biological activities commonly found in brown seaweeds. In our present study, fucoidan was extracted from Sargassum polycystum collected from Saint Martin's Island in three different extraction methods. The composition of the fucoidans was determined by measuring the total carbohydrate, sulfate, uronic acid, fucose, and protein contents. Fourier-transform infrared spectroscopy (FT-IR) and Proton nuclear magnetic resonance (1H NMR) were studied to determine the structural groups of extracted fucoidans. Functional activities were analyzed by antioxidant activity and anti-diabetic activity assay. Fucoidan yield was obtained highest in the acid extraction method (0.061 ± 0.007 g/g), followed by the water extraction method (0.058 ± 0.005 g/g) and salt extraction method (0.041 ± 0.005 g/g). Water extracted fucoidan showed the highest carbohydrate content (62.97 %), uronic acid content (6.46 %), and protein content (2.62 %). Sulphate content (28.56 %) was highest in salt extracted fucoidan, whereas fucose content (24.0 %) was highest in water extracted fucoidan. FT-IR and 1H NMR analysis confirm several characteristic picks for the extracted fucoidans. Extracted fucoidans presented comparable antioxidant activity, but salt-extracted fucoidan presented the highest DPPH radical scavenging assay (86.85 ± 0.74 %, IC50 = 0.03 mg/mL), ABTS radical scavenging assay (65.13 ± 0.92 %, IC50 = 1.94 mg/mL), Phosphomolybdenum assay (4.356 ± 0.11) and Nitric Oxide (NO) radical scavenging assay (57.08 ± 0.52 %, IC50 = 3.73 mg/mL) in dose-dependency manner. Further anti-diabetic activity assay resulted in the highest α-amylase inhibition activity (62.82 ± 1.36 %, IC50 = 2.30 mg/mL) and α-glucosidase inhibition activity (55.73 ± 0.84 %, IC50 = 3.71 mg/mL) by salt-extracted fucoidan. These functional potentials of seaweed may add significant value to the development of functional food and pharmaceutical materials.

Phytochemical profiling, antidiabetic, antiobesity, and antibacterial activities of Cometes abyssinica

This study aims to complete my chemical and biological studies on Cometes abyssinica (CA). The chemical profile was reported by LC-ESI-TOF-MS and GC-MS analyses. Antidiabetic (α-Glucosidase and a-amylase inhibitory activity assay), antiobesity (Lipase inhibitory activity assay), and antibacterial activity (agar well diffusion assay) were determined. The GC-MS analysis led to the identification of 27 hydrocarbons representing 93.4% of the total hydrocarbon content in hexane extract. 11-Octadecenoic acid, methyl ester (28.07%), citronellol (9.31%), hexadecanoic acid, methyl ester (7.64%), phytol (5.93%), and 10-epi-γ-eudesmol (4.91%) were the major components. The LC-ESI-TOF-MS analyses of aqueous-methanolic extract led to the identification of 99 secondary metabolites including phenolics, flavonoids, alkaloids, and vitamins. The percentages of inhibition of the n-hexane, chloroform, ethyl acetate, and methanol extracts of CA were evaluated against α-glucosidase, α-amylase, and pancreatic lipase enzymes at different concentrations. Hexane extract showed the most powerful α-glucosidase and α-amylase inhibitory activity with IC50 value of 0.22 and 0.27 mg/ml comparing with acarbose (IC50 0.43 and 0.38), respectively. Among all CA extracts, only ethyl acetate extract revealed potent antiobesity as a lipase inhibitory activity with IC50 value of 0.079 mg/ml comparing with orlistat (IC50 0.11 mg/ml). CA extracts showed mild-weak antibacterial effect against Pseudomonas aeruginosa and Bacillus cereus. In Vitro α-Glucosidase, α-Amylase Inhibitory and Antioxidant Activities of Root Crude Extract and Solvent Fractions of Arbutus unedo L. (Ericaceae)

Roots of Arbutus unedo (Ericaceae) have been used traditionally to treat diabetes in some countries. Conducting more experiments is needed to support this plant's potential antidiabetic properties, though. Ethanolic extract yielded by cold maceration was subjected into fractionation to get hexane, chloroform, ethyl acetate, butanol, and aqueous fractions. Total phenolic contents and antioxidant capacity have been estimated utilizing 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and ferric ion reducing antioxidant potential (FRAP) assay models, respectively. Antidiabetic activity of A. unedo extracts were evaluated using in vitro α-glucosidase, α-amylase inhibition activity. The ethanol extract had the highest antioxidant activity among used extracts. While the results of both α-glucosidase, α-amylase enzyme inhibition activity was detected to be dose-dependent, the strongest inhibition activity for α-glucosidase and α-amylase was shown by ethanol extract (526.65 and 522.66 µg mL-1 respectively) compared to the standard acarbose. Results demonstrated the beneficial effects of A. unedo extracts by showing antioxidant and antidiabetic activities.

Peptidomics-based identification of antihypertensive and antidiabetic peptides from sheep milk fermented using Limosilactobacillus fermentum KGL4 MTCC 25515 with anti-inflammatory activity: in silico, in vitro, and molecular docking studies.

This study investigated the synthesis of bioactive peptides from sheep milk through fermentation with Limosilactobacillus fermentum KGL4 MTCC 25515 strain and assessed lipase inhibition, ACE inhibition, α-glucosidase inhibition, and α-amylase inhibition activities during the fermentation process. The study observed the highest activities, reaching 74.82%, 70.02%, 72.19%, and 67.08% (lipase inhibition, ACE inhibition, α-glucosidase inhibition, and α-amylase inhibition) after 48h at 37°C, respectively. Growth optimization experiments revealed that a 2.5% inoculation rate after 48h of fermentation time resulted in the highest proteolytic activity at 9.88mg/mL. Additionally, fractions with less than 3kDa of molecular weight exhibited superior ACE-inhibition and anti-diabetic activities compared to other fractions. Fermentation of sheep milk with KGL4 led to a significant reduction in the excessive production of NO, TNF-α, IL-6, and IL-1β produced in RAW 267.4 cells upon treatment with LPS. Peptides were purified utilizing SDS-PAGE and electrophoresis on 2D gels, identifying a maximum number of proteins bands ranging 10-70kDa. Peptide sequences were cross-referenced with AHTPDB and BIOPEP databases, confirming potential antihypertensive and antidiabetic properties. Notably, the peptide (GPFPILV) exhibited the highest HPEPDOCK score against both α-amylase and ACE.

Chemical composition and biological activities of essential oils and extract of Eucalyptus citriodora Hook

In this study, the leaves of Eucalyptus citriodora Hook (Lemon-Scented Eucalyptus) were harvested and collected from Üzümlü neighborhood of Fethiye district of Muğla in 2021. Chemical content analysis of steam distillation and hydrodistillation of essential oils were determined by GC-MS, while phenolic content of methanol extract was determined by HPLC-DAD. Antioxidant activities of essential oils and methanol extracts were determined by DPPH radical removal, ABTS cation removal, β-carotene linoleic acid, and CUPRAC activity methods; Anticholinesterase activity against AChE and BChE enzymes was determined by Ellman method; and tyrosinase inhibition associated with melanin hyperpigmentation, α-amylase inhibition, and α-glucosidase inhibition activities associated with diabetes were determined as an in vitro. The bioactivities and chemical contents of E. citriodora species, a great value of, Türkiye, were determined, bringing new natural products to organic chemistry. As a result of the study, new bioactive extracts would be obtained and thus, they can effectively reveal the potential of new business opportunities. Since methanol extract is effective against incurable diseases such as Alzheimer's and diabetes, it will also be possible to develop therapeutics of such diseases by investigating the advanced chemistry and in vivo activities of the extracts with new projects.

Enzyme-mediated ultrasound-assisted extraction of rutin, quercetin, and other antioxidant and antidiabetic compounds from Holarrhena antidysenterica seeds and optimization as per RSM

Rutin and quercetin are valuable bioactive natural products. The current study successfully discovered an efficient extraction protocol for these and other bioactive compounds from Holarrhena antidysenterica (HA) seeds based on an enzyme-mediated ultrasound-assisted extraction (EM-UAE). Extraction process optimization was done based on response surface methodology with seven response factors including total extraction yield (%), amount of rutin (AOR, mg/g), amount of quercetin (AOQ, mg/g), total phenolic content (TPC, mg gallic acid equivalents/g dry weight (mg GAE/g DW), total flavonoids content (TFC, mg rutin equivalents/g DW (mg RE/g DW) anti-radical activity (ARA, %), and α-amylase inhibitory activity (AIA, %). High-performance liquid chromatography (HPLC) was used for quantification of rutin and quercetin. The optimal extraction yield, AOR, AOQ, TPC, TFC, ARA, and AIA were 32.39 %, 71.43 mg/g, 23.68 mg/g, 178.63 mg GAE/g DW, 100.22 mg RE/g DW, 76.41 %, and 77.48 %, respectively. The optimal conditions were 91 min time, 6.1 mL enzyme cocktail, pH 4.8, and 48 ℃ temperature. For ARA, EC50 (µg/mL) of HA was 9.75 ± 0.27 compared to 6.10 ± 0.38 of ascorbic acid standard. For AIA, IC50 (µg/mL) of HA and acarbose standard were 51.97 ± 0.21 and 47.89 ± 0.61 μg/mL, respectively. The validation study strongly supported the predicted model with a very low relative standard deviation (0.45–3.37 %). The model was statistically significant and efficient for obtaining rutin, quercetin, and other antioxidant and antidiabetic compounds from Holarrhena antidysenterica seeds on the industrial level.

Anti-hyperglycemic effect of herbal formula of Moringa oleifera, Vernonia amygdalina and Centella asiatica extracts in streptozotocin-induced hyperglycemic mice

IntroductionMoringa oleifera, Vernonia amygdalina, and Centella asiatica are herbs used in traditional Chinese medicine and are commonly used in Southeast Asian countries to treat a variety of diseases. The experimental evidence showed that M. oleifera leaves, V. amygdalina leaves, and C. asiatica had the effect of supporting their treatment of diabetes. This study investigated the anti-hyperglycemic effect of a novel herbal formula from M. oleifera leaf, V. amygdalina leaf, and C. asiatica aerial part extracts. MethodsThe extract was examined for anti-hyperglycemic capacity in oral glucose tolerance test in normal mice. To confirm their role in a higher model, the extract was studied in the streptozotocin (STZ)-induced hyperglycemic model. Plasma glucose concentration, body weight, organ weight, malondialdehyde, glutathione in liver and kidneys, and pancreatic histology were also evaluated. Besides, in vitro properties of the extract were tested including α-amylase and α-glucosidase inhibitory, and antioxidant activities. ResultsAfter 7-day of treatment, the extract showed strong anti-hyperglycemic activity in the STZ-induced hyperglycemic mice, plasma glucose levels were reduced by 48.75 % and 54.13 % in the group administered with 200 mg/kg and 400 mg/kg of the extract, respectively, compared with the hyperglycemic control. The extract had the effect to protect the liver and kidneys against oxidation stress by enhancing glutathione levels in the liver and decreasing malondialdehyde levels in the liver and kidneys. The pancreatic β-cell protection was also observed by improving the size and number of pancreatic islets by the extract compared to the hyperglycemic control. The extract showed a significant α-amylase and α-glucosidase inhibitory activities, and antioxidant activity through DPPH and ABTS quenching and reducing power abilities. DiscussionThese results demonstrated remarkable potential of the combined extract of M. oleifera leaves, V. amygdalina leaves, and aerial part of C. asiatica in the management of diabetes.

Aloe juvenna Brandham & S.Carter as α-Amylase Inhibitor and Hypoglycaemic Agent with Anti-inflammatory Properties for Diabetes Management.

Despite Aloe's traditional use, Aloe juvenna Brandham & S.Carter is poorly characterized. Other Aloes are known for their antidiabetic activity. This study describes the antidiabetic potentials and phytoconstituents of the A. juvenna leaves methanolic extract (AJME). Twenty-six phytoconstituents of AJME were described using HPLC/MS-MS. Lupeol and vitexin were isolated using column chromatography. The antidiabetic activity of AJME was investigated using an in vivo high-fat diet/streptozotocin-induced diabetic rat model and in vitro α-glucosidase and α-amylase inhibitory activity assays. AJME demonstrated its α-amylase inhibitory activity (IC50=313±39.9 ppm) with no effect on α-glucosidase. In vivo, AJME dose-dependently improved hyperglycaemia in a high-fat diet/streptozotocin-induced diabetic rat model. Notably, the higher dose (1600 mg/kg) of AJME significantly downregulated serum interleukin-6, tumor necrosis factor-α, and matrix metalloproteinase-1 genes, suggesting its anti-inflammatory effect. These findings indicate AJME's potential as a significant antidiabetic agent through its α-amylase inhibition, hypoglycaemic, and anti-inflammatory properties.

Pyrano[2,3-c]pyrazole fused spirooxindole-linked 1,2,3-triazoles as antioxidant agents: Exploring their utility in the development of antidiabetic drugs via inhibition of α-amylase and DPP4 activity

Environment-benign, multicomponent synthetic methodologies are vital in modern pharmaceutical research and facilitates multi-targeted drug development via synergistic approach. Herein, we reported green and efficient synthesis of pyrano[2,3-c]pyrazole fused spirooxindole linked 1,2,3-triazoles using a tea waste supported copper catalyst (TWCu). The synthetic approach involves a one-pot, five-component reaction using N-propargylated isatin, hydrazine hydrate, ethyl acetoacetate, malononitrile/ethyl cyanoacetate and aryl azides as model substrates. Mechanistically, the reaction was found to proceed via in situ pyrazolone formation followed by Knoevenagel condensation, azide alkyne cycloaddition and Michael's addition reactions. The molecules were developed using structure-based drug design. The primary goal is to identifying anti-oxidant molecules with potential ability to modulate α-amylase and DPP4 (dipeptidyl-peptidase 4) activity. The anti-oxidant analysis, as determined via DPPH, suggested that the synthesized compounds, A6 and A10 possessed excellent anti-oxidant potential compared to butylated hydroxytoluene (BHT). In contrast, compounds A3, A5, A8, A9, A13, A15, and A18 were found to possess comparable anti-oxidant potential. Among these, A3 and A13 possessed potential α-amylase inhibitory activity compared to the acarbose, and A3 further emerged as dual inhibitors of both DPP4 and α-amylase with anti-oxidant potential. The relationship of functionalities on their anti-oxidant and enzymatic inhibition was explored in context to their SAR that was further corroborated using in silico techniques and enzyme kinetics.

Four Undescribed Stilbenoid Derivatives from the Aerial Parts of Dendrobium officinale with their α-Glucosidase and α-Amylase Inhibitory Activity.

In this study, four undescribed bibenzyl derivatives (1-4), together with seven known compounds (5-11) were isolated from the aerial parts of Dendrobium officinale. Their chemical structures were determined to be (7'S,8'S) -9''-acetyldendrocandin U (1), (7'S,8'S) -4'-methoxydendrocandin T (2), (7'R,8'S) -dendrocandin B (3), (1S,2R) -5'''-methoxydendrofindlaphenol C (4) by analyzing of the spectroscopic data including HR-ESI-MS, 1D-, and 2D-NMR spectra. The absolute configurations of compounds 1-4 were determined by the electronic circular dichroism (ECD) spectra. Compounds 1-3, 5, 10 and 11 inhibited α-glucosidase with the IC50 values ranging from 56.3 to 165.3 μM, compounds 1-3, 5, 7-10 inhibited α-amylase with the IC50 values ranging from 65.2 to 177.6 μM.

Investigation of the antidiabetic and probiotic properties of lactic acid bacteria isolated from some ethnic fermented foods of Darjeeling District

BackgroundIndigenous communities residing in the Darjeeling Himalayan region and its adjacent hilly areas have a deeply rooted cultural tradition of consuming a diverse range of vegetable and milk-based fermented products, believed to confer various health advantages. With this traditional knowledge, lactic acid bacteria (LAB) were isolated from popular fermented foods such as Chhurpi (derived from Bos grunniens milk), Gundruk (made from Brassica juncea leaves), Sinki (derived from Raphanus sativus taproots), and Kinema (produced from Glycine max beans). This study aimed to investigate the probiotic properties of the prevalent LABs, including aggregation properties, bile salt hydrolase activities, survival under gastro-inhibitory conditions, safety evaluations, and their potential health-promoting attributes, with a specific focus on inhibiting α-amylase and α-glucosidase enzymes.ResultsFive of the LAB isolates demonstrated notable viability rates exceeding 85% when exposed to gastro-inhibitory challenges. Based on 16S rRNA gene sequencing, these isolates were identified as Pediococcus pentosaceus (isolate GAD), Lactobacillus plantarum (isolates KAD and CAD), Lactobacillus brevis (isolate SAD), and Lactiplantibacillus plantarum (isolate CMD). These LAB isolates exhibited versatile carbon source utilization, significant auto- and co-aggregation, and bile salt hydrolase (BSH) properties. Auto-aggregation capacity notably increased over time, ranging from 30 to 150 min, with percentage increments from 4.83 ± 1.92% to 67.60 ± 5.93%. L. brevis SAD displayed the highest co-aggregation increment (%) against Staphylococcus aureus, while L. plantarum KAD demonstrated potent antimicrobial activity. In vitro analyses postulated potential health benefits related to antidiabetic properties, particularly inhibiting α-amylase and α-glucosidase enzymes. L. brevis SAD exhibited the highest α-glucosidase inhibitory activity, while L. plantarum KAD displayed the most potent α-amylase inhibitory activity. Comprehensive safety assessments, including antibiotic susceptibility profiling, hemolytic activity evaluation, and in vivo acute toxicity studies, confirmed the suitability of these LAB isolates for human consumption.ConclusionsThe isolates show promising probiotic characteristics and significant potential in addressing metabolic health. These results carry substantial scientific implications, suggesting the pharmaceutical-based applications of these traditional fermented foods. Further in vivo investigation is recommended to fully elucidate and exploit the health benefits of these LAB isolates, opening avenues for potential therapeutic interventions and the development of functional foods.

Enrichment and purification of Torreya grandis peptides by macroporous resin and its hypoglycemic mechanism revealed by transcriptome analysis

The objective of this study was to develop an efficient strategy to prepare high-hypoglycemic efficacy Torreya grandis meal peptides with macroporous resin and investigate the underlying mechanisms through transcriptome analysis. The evaluation of adsorption and desorption abilities of Torreya grandis meal peptides (TGPs) on six different microporous resins were compared. The hypoglycemic activity of prepared TGPs was further measured in a Zebrafish diabetic model, and the underlying mechanisms were investigated through transcriptome analysis. Results showed that DA201-C exhibited the highest adsorption and desorption capacities. After DA201-C resin enrichment, the α-glucosidase inhibition and α-amylase inhibition activities of TGPs in 75% ethanol eluate (TGPs-75) both significantly increased (the IC50 values are 0.41 mg/mL and 1.08 mg/mL, respectively). The in vivo data showed that TGPs-75 remarkably reduced blood glucose concentrations to 7.8, 15.6, and 31.2 μg/mL in an high-sugar-high-fat-treated (HSHF-treated) Zebrafish diabetic model and exerted obvious hypoglycemic activity. According to transcriptome analysis, 382 genes were significantly differentially expressed (229 up-regulated and 153 down-regulated) after TGPs-75 pretreatment. The main functions of these genes are related to gluconeogenesis and insulin resistance. The findings from the present study suggested that TGPs-75 produced positive hypoglycemic efficacy and showed potential to provide a scientific basis for its further utilization as a dietary supplement or an ingredient in functional foods.

Green Synthesis and Anti-Diabetic Potential of Berberis vulgaris Silver Nanoparticles: A Promising Approach for Natural Therapeutics

This study reports the green synthesis of Berberis vulgaris root water-mediated silver nanoparticles (BvAgNPs) showcasing high purity, size-controlled morphology, and potential anti-diabetic activity. Characterization techniques confirmed the successful synthesis of predominantly spherical BvAgNPs within the 15–70 nm range, coated with organic moieties from the extract. Remarkably, BvAgNPs exhibited significantly enhanced α-amylase and α-glucosidase inhibitory activity compared to the extract alone, with inhibition higher for α-amylase. This superior anti-diabetic activity, even comparable to the standard drug Acarbose for α-amylase inhibition, highlights the potential of BvAgNPs as natural and potentially less-toxic alternatives in diabetes management. Further research on their mechanisms and in vivo efficiency is warranted to pave the way for their future application as promising anti-diabetic therapeutics.

Effect of three modification methods on physical and functional properties of flaxseed cake dietary fiber

Flaxseed cake dietary fiber (FCDF) is an important by-product of the oil manufacturing industry, but the low bioactivity limits its application. To improve the physical and functional properties of FCDF, it was separately treated using acidic (AC), cellulase (CE), and acrylate-grafting (AG) methods. Compared with the untreated FCDF, FCDF modified by AC (FCDF-AC), CE (FCDF-CE), and AG (FCDF-AG) had looser surface structures and greater flow characteristics. The hydration capacities were higher for FCDF-AC and FCDF-AG than for FCDF-CE. After modifications, FCDF showed high adsorption capacities to lipids, cholesterol, and nitrite ions. Moreover, the hypoglycemic properties of FCDF-CE and FCDF-AG, including glucose absorption capacity (GAC), glucose dialysis retardation index (GDRI), and α-amylase inhibition activity (AIA), were higher than those of untreated FCDF. In contrast, the GAC, GDRI, and AIA were not significantly different between FCDF-AC and FCDF (p > 0.05). Therefore, these results suggest that modified FCDF can be used as an additive in hypolipidemic and hypoglycemic foods.

Phytochemical Screening and Antidiabetic Activity of Aqueous Extract of Evolvulus Alsinoides Leaves: An In Vitro and In Silico Study

ABSTRACT Background: Nowadays, diabetes mellitus has become common worldwide due to changes in lifestyle, sedentary life, alterations in food habits, and other genetic and environmental factors. It is necessary to create awareness about the growing pandemic. Aim: To evaluate the antidiabetic potential of Evolvulus alsinoides. Methods: In vitro α-amylase inhibition and α-glucosidase inhibition activity. Molecular docking analysis. Results: The plant is rich in naturally occurring phytocompound. The results of the study showed that E. alsinoides has the potential to inhibit the activity of alpha-amylase and alpha-glucosidase. Moreover, this result was validated using in silico molecular docking studies that showed a good binding affinity of one of the major phytocompound, caffeic acid, with molecules in the insulin signaling cascade. Conclusion: E. alsinoides has a potent antidiabetic activity and can be further employed in animal and human studies.

Evaluation of the Anticancer, Antidiabetic, and In vitro Wound Healing Properties of the Aqueous and Ethanolic Extract of Hibiscus rosa-sinensis L

ABSTRACT Cancer, diabetes, and wounds are critical health challenges affecting millions of people worldwide. Cancer arises from the transformation of normal cells into tumor cells, leading to uncontrolled growth and potential spread to other parts of the body. Diabetes is a chronic metabolic disorder characterized by elevated blood sugar levels, and wounds can result from various injuries and diseases. In this study, we investigated the therapeutic potential of Hibiscus rosa-sinensis for its anticancer, antidiabetic, and in vitro wound healing activities. The anticancer activity of the ethanolic extract of H. rosa-sinensis was evaluated using the MTT (3- [4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay, which showed a concentration-dependent decrease in cell proliferation and growth. The extract exhibited promising anticancer potential, which could help control cancer progression. For the antidiabetic activity, we assessed α-amylase inhibition using a colorimetric method. The extract demonstrated substantial α-amylase inhibitory activity, comparable with the standard metformin. This indicates its potential in reducing postprandial blood sugar spikes in diabetic patients. In the in vitro wound healing activity, a scratch assay was performed to measure cell migration and growth. The results showed that H. rosa-sinensis extract promoted wound closure similar to the control. The extract’s wound healing properties suggest its potential use in managing various types of wounds. Phytochemical analysis confirmed the presence of key bioactive compounds in both ethanolic and aqueous extracts of H. rosa-sinensis, supporting the observed therapeutic effects. In conclusion, H. rosa-sinensis holds promise as a potential source of novel therapeutic agent for cancer, diabetes, and wound management. The results from this study provide valuable insights into the plant’s pharmacological activities and may pave the way for the development of new herbal-based treatments for these conditions.
