Evaluation of Immunomodulatory activity of Salvia hispanica. L Seeds

Background: Immune system plays a vital role in protective mechanism against the foreign particles and its modulation either suppression or stimulation was implicated in several human diseases like inflammatory, infectious and autoimmune diseases etc. for which several allopathic immunomodulators has been used for treatment, exhibits potential side effects. Biological active compounds from natural sources used in the treatment of immune related disorder. Objective: Being the importance of natural compound, the present study was aimed to evaluate the immunomodulatory activity of Salvia hispanica.L seeds by in-vitro methods. Method: The immunomodulatory effect of hexane extract on phagocytosis and stimulation and activation of neutrophils were studied by using bactericidal activity, NBT Test, phagocytosis of killed Candida albicans and congo red stained yeast cells methods. Results: The extract exhibited the immunomodulatory effect by causing significant increase in phagocytosis and stimulation and their intercellular killing property of neutrophils support the immunomodulation due to presence of active compounds in extract. Conclusion: Results summarises the immunomodulatory activity of hexane extract of Salvia hispanica.L seeds due to presence of active constituents like fatty acids, essential oils etc.

Inhibition of Corn Oil Peroxidation by Extracts from Defatted Seeds ofCamellia oleiferaAbel

The defatted seed ofCamellia oleiferaAbel is an underexploited byproduct. In the present study, 3 solvents (isopropanol, hexane, and diethyl ether) were used to extract active compounds from defatted seeds ofCamellia oleiferaAbel. The qualitative and quantitative characterization of the main phenolic compounds were performed using the HPLC-DAD. In addition, the antioxidant efficacy of defatted seed extracts in the stabilization of the corn oil was estimated by monitoring its primary and secondary oxidation products. The results showed that isopropanolic extract (IE) exhibited the highest yield of total phenolic compounds (9.23±0.33 mg/g) compared with other solvents. The predominant phenolic compounds identified by HPLC-DAD in defatted seed extracts were epicatechin, naringenin, and catechin. In addition, IE exhibited the best inhibition effect on both primary and secondary oxidation products. The results reveal that IE of defatted seed ofCamellia oleiferaAbel is a useful antioxidant for the stabilization of corn oil.

Using wheat bran and soybean meal as solid state fermentation substances for the production of Xylaria nigripes with bioactivities

Abstract Different combinations of wheat bran (WB) and soy meal (SM) were used as solid state fermentation substrates for the growth of functional Xylaria nigripes ( XN, Wu Ling Shen , in Chinese). Hot water as well as ethanol (70%) were used to extract active compounds with antioxidant, anti-inflammatory and neuroprotective properties. In general, ethanol was a more effective solvent than hot water to extract active compounds. When WB was used as the sole fermentation substrate, the ethanol extract of XN -fermented substances obtained the highest antioxidant activities, especially DPPH radical scavenging and reducing activities; anti-inflammation, as measured by the inhibition of COX-2 activity, was used as an active index. Fermentation substrates containing equal amounts of WB and SM increased the protective effects against H 2 O 2 -induced damage in neuronal cells (PC12 cells) for 70% ethanol-extracted XN . Both XN and residues of fermentation substrates may have contributed to the biological activities of XN -fermented substances.

Trimer procyanidin oligomers contribute to the protective effects of cinnamon extracts on pancreatic β-cells in vitro.

Cinnamon extracts rich in procyanidin oligomers have shown to improve pancreatic β-cell function in diabetic db/db mice. The aim of this study was to identify the active compounds in extracts from two species of cinnamon responsible for the pancreatic β-cell protection in vitro. Cinnamon extracts were prepared from Cinnamomum tamala (CT-E) and Cinnamomum cassia (CC-E). Six compounds procyanidin B2 (cpd1), (-)-epicatechin (cpd2), cinnamtannin B1 (cpd3), procyanidin C1 (cpd4), parameritannin A1 (cpd5) and cinnamtannin D1 (cpd6) were isolated from the extracts. INS-1 pancreatic β-cells were exposed to palmitic acid (PA) or H2O2 to induce lipotoxicity and oxidative stress. Cell viability and apoptosis as well as ROS levels were assessed. Glucose-stimulated insulin secretion was examined in PA-treated β-cells and murine islets. CT-E, CC-E as well as the compounds, except cpd5, did not cause cytotoxicity in the β-cells up to the maximum dosage using in this experiment. CT-E and CC-E (12.5-50 μg/mL) dose-dependently increased cell viability in both PA- and H2O2-treated β-cells, and decreased ROS accumulation in H2O2-treated β-cells. CT-E caused more prominent β-cell protection than CC-E. Furthermore, CT-E (25 and 50 μg/mL) dose-dependently increased glucose-stimulated insulin secretion in PA-treated β-cells and murine islets, but CC-E had little effect. Among the 6 compounds, trimer procyanidins cpd3, cpd4 and cpd6 (12.5-50 μmol/L) dose-dependently increased the cell viability and decreased ROS accumulation in H2O2-treated β-cells. The trimer procyanidins also increased glucose-stimulated insulin secretion in PA-treated β-cells. Trimer procyanidins in the cinnamon extracts contribute to the pancreatic β-cell protection, thus to the anti-diabetic activity.

Methyl Anthranilate as a Repellent for Western Corn Rootworm Larvae (Coleoptera: Chrysomelidae).

Methyl anthranilate was identified as the active compound in extracts of maize (Zea mays L.) roots that were shown to be repellent to neonate western corn rootworm (Diabrotica virgifera virgifera LeConte) larvae. A bioassay-driven approach was used to isolate the active material from diethyl ether extracts of roots from germinating maize seeds. Separation of the extract on a Florisil column yielded an active fraction of 90:10 hexane:diethyl ether. Analysis with gas chromatography-mass spectrometry identified two compounds in the active fraction: indole (2,3-benzopyrrole) and methyl anthranilate (methyl 2-aminobenzoate). When tested in behavioral bioassays, methyl anthranilate elicited a significant (P < 0.05) repellent response at doses of 1, 10, and 100 µg. In subsequent single-choice bioassays, 1, 10, and 100 µg of methyl anthranilate prevented larvae from approaching 10 mmol/mol concentrations of carbon dioxide, which is normally highly attractive to the larvae. Indole, the other compound identified from the active fraction, did not elicit a behavioral response by the larvae. Methyl anthranilate has potential for development as a management tool for western corn rootworm larvae and may be best suited for use in a push-pull control strategy.

Extraction, Toxicity Assay and Identification of Active Compounds of Red Algae (Eucheuma cottonii) from Sumenep Madura

&lt;p&gt;Extraction, Toxicity Assay and Identification ofActive Compounds of Red Algae Extract &lt;em&gt;(Eucheuma&lt;/em&gt;&lt;em&gt; &lt;/em&gt;&lt;em&gt;cottonii&lt;/em&gt;&lt;em&gt;)&lt;/em&gt; from Tanjung Sumenep Madura was analysed. Extraction of active compound was conducted by maceration method using methanol as a solvent. The methanol extract was hydrolyzed with HCl 2 N. The result was partitioned using five solvents (1-butanol, ethyl acetate, chloroform, petroleum ether and n-hexane). Toxicity level was determined by Brine Shrimp Lethality Test (BSLT). The identification of the active compound was analyzed by phytochemical test, qualitatively. The result showed that the 1-butanol is the best solvent to extract active compounds of red algae &lt;em&gt;E.&lt;/em&gt;&lt;em&gt; &lt;/em&gt;&lt;em&gt;Cottonii&lt;/em&gt; in partition process. 1-Butanol fraction gives the highest activity of toxicity with LC&lt;sub&gt;50&lt;/sub&gt;70,32 ppm. LC&lt;sub&gt;50&lt;/sub&gt; value for ethyl acetate, chloroform, petroleum ether and n-hexane was 143,43; 194,40; 195,28; 303,66; and 634,97 ppm, respectively. Subcritical Water Technology for Enhanced Extraction of Biochemical Compounds fromChlorella vulgaris

Subcritical water extraction (SWE) technology has been used for the extraction of active compounds from different biomass materials with low process cost, mild operating conditions, short process times, and environmental sustainability. With the limited application of the technology to microalgal biomass, this work investigates parametrically the potential of subcritical water for high-yield extraction of biochemicals such as carbohydrates and proteins from microalgal biomass. The SWE process was optimized using central composite design (CCD) under varying process conditions of temperature (180–374°C), extraction time (1–20 min), biomass particulate size (38–250 μm), and microalgal biomass loading (5–40 wt.%). Chlorella vulgaris used in this study shows high volatile matter (83.5 wt.%) and carbon content (47.11 wt.%), giving advantage as a feedstock for biofuel production. The results showed maximum total carbohydrate content and protein yields of 14.2 g/100 g and 31.2 g/100 g, respectively, achieved under the process conditions of 277°C, 5% of microalgal biomass loading, and 5 min extraction time. Statistical analysis revealed that, of all the parameters investigated, temperature is the most critical during SWE of microalgal biomass for protein and carbohydrate production.

New Alternative in the Methodology of Extraction and Dyeing with Active Molecules Derived from Vegetal Sources

The general objective of this study refers to the identification of a sustainable and physical methodology of extraction of active compounds, envisaging the preservation of the high purity active natural dye molecule from nut shell (juglone), even under the conditions of an extraction performed in a mixt solvent medium (water-ethanol). The second major objective of the study consists of the application of these above mentioned dyes onto natural and synthetic substrates, thus making a correlation between their colour attributes and the fibrous composition of the substrates they are applied on. The motivation of this research was given by the identification of an improved extraction methodology that can increase the percentage of active parts of the plant cell wall. The efficacy of the extraction protocol in a water-ethanol solvent medium from nut shells is evidenced by the complex system of investigations: HPLC chromatography, FT-Raman and IR spectroscopy. HPLC chromatography reveals an efficient ecologic extraction procedure. Moreover, the refluxing duration in the experimental protocol of juglone extraction in a water-ethanol solvent medium from nut shells is almost the same as that belonging to synthetic juglone.

Analysis of Microbial Biochemical Inducting Nutritional Contents in Barley

Current study focuses the biochemical analysis of the metabolites of Acetobacter aceti to explore bioactive compounds inducing nutritional elements (including vitamins) in barley seeds. Antibacterial activity from Rhanterium adpressum flowers extracts, depending on seasonal variations

The main purpose of this study was to evaluate the effect of solvent extraction (acetone or methanol) on total phenolic content and antibacterial potential of Rhanterium adpressum flowers extracts. Flowers of the plant were harvested during three different months (April, May and June) for two years 2011 and 2012. The Folin–Ciocalteu method was used to determine the total phenolic content (TPC) in organic extracts; however, the antibacterial activity was tested using disc diffusion method. The highest phenolic content was detected in R. adpressum methanolic extracts collected in April. Same was evident in the antibacterial assay and it was also remarkable that the extracts activity was related to the concentration of phenolic compounds in active extracts, antibacterial activity was more efficient against Staphylococcus aureus and Salmonella typhi Gram+Bacteria. This is the first report on seasonal changes of phenolic compounds and antibacterial activity on R. adpressum and might be interesting to prospection of new molecules with antibacterial properties.

Anticancer Activity against Breast Cancer Cells T47D and Identification of Its Compound from Extracts and Fractions of Leaves Bamboo Grass (Lophaterum gracile B.)

Bamboo grass (Lophatherum gracile B.) is one of weed plants which frequently used as a medicine because it provides active compounds, for instance steroids and tannins. This research was conducted to determine the anticancer activity of extracts and fractions of leaves bamboo grass in inhibiting the growth of breast cancer cells T47D, and to determine the class of the active compounds contained in extracts and fractions which have the lowest IC&lt;sub&gt;50&lt;/sub&gt; value from phytochemical test and identification with UV-Vis instruments and FTIR. The extraction of active compound leaves of bamboo grass is done by maceration method using ethanol 80%, hydrolysis and partition with n-hexane and chloroform. Four of the extracts is phytochemical tested and anticancer activity tested towards breast cancer cells T47D with MTT method. The best results of the test activities are performed by the separation of classes of active compounds KLTP and are identified using UV-Vis spectrophotometer and FTIR. The value of IC&lt;sub&gt;50&lt;/sub&gt; from 80% ethanol extraction is 321,389, from hydrolysis extraction is 481.984, from the fraction of chloroform is 177.852, and from n-hexane fraction is 300.681 µg / mL. The lowest IC50 which have the best cytotoxic is the fraction of chloroform and n-hexane. The results of phytochemical test fractions of chloroform and n-hexane fraction indicates the tannin and steroids. The identification result by using UV-Vis and FTIR shows that the fraction of chloroform and n-hexane fraction is containing tannin, which isproven by maximum wavelength at 331.6 nm and 331.6 nm allegations of -OH, CH, C=C. Moreover, the existence of steroid compounds in fraction chloroform and n-hexane is shown by the absorption at 279.1 nm and 278.0 nm of wavelength with allegations of -OH, CH, C=C, C-O.

Identification of Microbial Metabolites Elevating Vitamin Contents in Barley Seeds.

The current investigation analyzes metabolites of Acetobacter aceti to explore chemical compounds responsible for the induction of vitamins in barley seeds. A bioactivity guided assay of bacterial extracts and chromatographic analyses of barley produce revealed 13 chemical compounds, which were subjected to principal component analysis (PCA). PCA determined four chemical compounds (i.e., quinolinic acid, pyridoxic acid, p-aminobenzoate, and α-oxobutanoic acid) highly associated with increased quantities of vitamins. Further experimentations confirmed that quinolinic acid and p-aminobenzoate were the most efficient vitamin inducers. The results indicated chloroform/ethanol (4:1) as the best solvent system for the extraction of active compounds from crude metabolites of A. aceti. Significant quantities of mevalonic acid were detected in the extracted fraction, indicating the possible induction of the isoprenoid pathway. Altogether, the current investigation broadens the frontiers in plant-microbe interaction.

Optimization of ultrasound assisted extraction of anthocyanins from red cabbage using Taguchi design method

There is a growing demand for developing suitable and more efficient extraction of active compounds from the plants and ultrasound is one of these novel methodologies. Moreover, the experimental set up to reach an appropriate condition for an optimum yield is demanding and time consuming. In the present study, Taguchi L9 orthogonal design was applied to optimize the process parameters (output power, time, temperature and pulse mode) for ultrasound assisted extraction of anthocyanins from red cabbage and the concluding yield of anthocyanin was measured by pH differential method. The statistical analysis revealed that the most important factors contributing to the extraction efficiency were time, temperature and power, respectively and the optimum condition was at 30min, 15°C and 100W which could result the maximum anthocyanin yield of about 20.9mg/L. The theoretical result was confirmed experimentally by carrying out the trials at the optimum condition and evaluating the actual yield.

Chemical composition and antimicrobial activity of Aloysia gratissima (Verbenaceae) leaf essential oil

Gas chromatography–mass spectrometry (GC–MS) analysis of Aloysia gratissima (Gill. et Hook) Tronc. essential oil obtained by hydrodistillation lead to the identification of twenty-two compounds. The major compounds found in this study were 1,8-cineole (13.7%), germacrene D (13.4%), β-caryophyllene (12.7%) and β-pinene (11.7%). The oil demonstrated antibacterial and antifungal activities against mainly Bacillus cereus and Trichophyton mentagrophytes. Aloysia gratissima oil was also screened for its antioxidant activity by 2,2-diphenylpicrylhydrazyl (DPPH) free radical; however, it was less effective than butylated hydroxytoluene (BHT). These findings suggest feasible application of A. gratissima essential oil as an antimicrobial agent and its chemical characteristic may lead to the extraction of active compounds in isolated or combined forms with useful applications.

Mechanistic Study of Silver Nanoparticle's Synthesis by Dragon's Blood Resin Ethanol Extract and Antiradiation Activity.

Biological synthesis of nanoparticles is best way to avoid exposure of hazardous materials as compared to chemical manufacturing process which is a severe threat not only to biodiversity but also to environment. In present study, we reported a novel method of finding antiradiation compounds by bioreducing mechanism of silver nanoparticles formation using 50% ethanol extract of Dragons blood, a famous Chinese herbal plant. Color change during silver nanoparticles synthesis was observed and it was confirmed by ultra violet (UV) visible spectroscopy at wave length at 430 nm after 30 min of reaction at 60 °C. Well dispersed round shaped silver nanoparticles with approximate size (4 nm to 50 nm) were measured by TEM and particle size analyser. Capping of biomolecules on Ag nanoparticles was characterized by FTIR spectra. HPLC analysis was carried out to find active compounds in the extract. Furthermore, antiradiation activity of this extract was tested by MTT assay in vitro after incubating the SH-SY5Y cells for 24 h at 37 °C. The results indicate that presence of active compounds in plant extract not only involves in bioreduction process but also shows response against radiation. The dual role of plant extract as green synthesis of nanoparticles and exhibit activity against radiation which gives a new way of fishing out active compounds from complex herbal plants.

Homogenate-assisted negative pressure cavitation extraction of active compounds from Pyrola incarnata Fisch. and the extraction kinetics study

Abstract A new extraction method, homogenate-assisted negative pressure cavitation extraction, was designed to extract the main active compounds from Pyrola incarnata Fisch. The influential parameters were optimized by single factor experiments and extraction kinetics. Optimal conditions were set as follows: homogenate time of 120 s, negative pressure of − 0.05 MPa, liquid/solid ratio of 30:1 mL/g, ethanol concentration at 60%, extraction temperature of 50 °C and extraction time of 25 min. Maximum extraction yields of hyperin, 2″-O-galloylhyperin and chimaphilin reached 1.205 ± 0.054, 4.961 ± 0.108 and 0.291 ± 0.016 mg/g, respectively. Furthermore, 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay was used to assess the antioxidant activity of the extracts. HNPCE extract had the best antioxidant activity with IC50 value of 0.144 ± 0.017 mg/mL. The present results showed that HNPCE could be a promising extraction technique used in the extraction of active compounds from plants. Industrial relevance In this work we showed a new extraction method, homogenate-assisted negative pressure cavitation extraction. This novel extraction technique also processes the advantages of potential eco-friendly technology, lower energy consumption, easy operation, shorter time and higher efficiency. It is promising for extensive industry applications in the nature product extraction fields. The results may provide valuable data for industrial scale-up applications.

In vitroantipsoriatic study of successive solvent extracts ofBarleria gibsoniDalz. using HaCa T keratinocyte cells

The present study was to investigate the in vitro antipsoriatic activity of the successive solvent extracts of Barleria gibsoni Dalz. leaves against HaCa T keratinocyte cells. Asiaticoside was given as reference drug. The plant material was dried under shade, powdered and extracted with different solvent extract like Petroleum ether, chloroform, ethyl acetate and ethanol. Preliminary phytochemical screening was done. 50% cell growth inhibition (IC50) of successive solvent extract of Barleria gibsoni and Asiaticoside was 576.82 μg/ml and 460.13μg/ml against the HaCa T keratinocyte cells. Petroleum ether and ethanol extract of Barleria gibsoni and Asiaticoside was effective cytotoxic drugs against HaCa T keratinocyte cells. It is due to the extraction of active compounds in the Petroleum ether and ethanolic extract. Therefore, it is suggested that evaluation of active constituents and clinical evaluation of petroleum ether extract of Barleria gibsoni would give a encouraging lead in the successful management of psoriasis.

Alkaline phosphatase activity-guided isolation of active compounds and new dammarane-type triterpenes from Cissus quadrangularis hexane extract

Ethnopharmacological relevanceThe stem of Cissus quadrangularis L. (CQ) is used in traditional medicine to treat bone fractures and swelling. Anti-osteoporotic activity of CQ hexane extract has been reported, but the active compounds in this extract remain unknown. Thus, we aimed to identify the active compounds in CQ hexane extract using bioassay-guided isolation. Materials and methodsThe CQ hexane extract was fractionated sequentially with benzene, dichloromethane, ethyl acetate, and methanol. The examination of CQ extract and its fractions was guided by bioassays for alkaline phosphatase (ALP) activity during the differentiation of MC3T3-E1 osteoblastic cells. The cells were treated with or without the CQ extract and its fractions for a period of time, and then the stimulatory effect of the alkaline phosphatase enzyme, a bone differentiation marker, was investigated. The compounds obtained were structurally elucidated using spectroscopic techniques and re-evaluated for activity during bone differentiation. ResultsA total of 29 compounds were isolated, viz., triterpenes, fatty acid methyl esters, glycerolipids, steroids, phytols, and cerebrosides. Four new dammarane-type triterpenes were isolated for the first time from nature, and this report is the first to identify this group of compounds from the Vitaceae family. Seven compounds, viz., glycerolipids and squalene, stimulated ALP activity at a dose of 10μg/mL. Moreover, the synergistic effect of these compounds on bone formation was demonstrated. ConclusionThis report describes, for the first time, the isolation of active compounds from CQ hexane extract; these active compounds will be useful for the quality control of extracts from this plant used to treat osteoporosis.

Abstract 3216: Anti-proliferative properties of Euphorbia mauritanica medicinal plant against breast cancer

Abstract Over the decades, the increased interest in the use of natural resources in pharmaceutical and phytotherapeutic research has led to manufacture of drugs (including anti-cancer drugs) derived from natural sources With our increased understanding of the molecular mechanisms underlying cancer progression and cell death, it is therefore necessary to test if there are any natural products that can be used to act as antitumor agents. As breast cancer continues to be the leading cause of cancer in females, we aimed to exploit the use of South African traditional medicine, Euphorbia mauritanica as a tool for anti breast cancer therapy. Euphorbia mauritanica was tested for anti-proliferative properties against MCF-7 and Cama-1 breast cancer cell lines. The following solvents were used to extract active compounds from E. mauritanica: n-hexane, dichloromethane, chloroform, ethyl acetate, acetone, methanol and butanol. A real time cell analyzer (Xcelligence) was used to monitor cell growth over a period of 48 hours following treatment of cells with the extracts. Butanolic extracts showed the highest level of cytotoxicity on both MCF-7 and Cama-1 cell lines with IC50 of 15 and 30 µg/ml respectively. These were followed by hexanolic extracts with IC50 of 30 and 50 µg/ml and methanolic extracts with IC50 of 50 and 100 µg/ml. Flow cytometry using annexin V was further employed to determine if cell death was as a result of apoptosis and the data confirmed that this was most likely the case. On investigating whether the observed apoptosis was via p53 mediated pathway, western blot analysis of p53, bcl-2, caspase 9, caspase3, RBBP6 and mdm2 showed that treatment with the 3 extracts did not have a significant effect on any of these genes, suggesting that their mode of apoptosis induction maybe through an alternative apoptotic pathway. Practice and Synthetize Molecularly Imprinted Polymers for Seperating Target Neuro-Protective Compounds from TCM with Less Pollution

Molecularly imprinted polymers (MIPs) are synthetic materials that can be the environmental protection extraction method in TCM research and industry. They can overcome the defects of traditional extract methods and environmental pollution. In our research, MIPs were prepared by precipitation polymerization with neuro-protective picroside I and ginsenoside Rb1 as the template molecule. Moreover, the morphology of MIPs was characterized by electron microscope scanning and the static adsorption capacity was measured by the Scatchard equation. Finally, MIPs were made into MIP-SPE columns to enrich the template molecule and its analogues comparing with C18-SPE column and the results show that MIPs have good affinity and selectivity towards the Rb1 and Picroside I in SPE columns. This research may offer a more environmentally friendly method to extract active compounds in the traditional herbal.
