Virus membrane fusion

Membrane fusion of enveloped viruses with cellular membranes is mediated by viral glycoproteins (GP). Interaction of GP with cellular receptors alone or coupled to exposure to the acidic environment of endosomes induces extensive conformational changes in the fusion protein which pull two membranes into close enough proximity to trigger bilayer fusion. The refolding process provides the energy for fusion and repositions both membrane anchors, the transmembrane and the fusion peptide regions, at the same end of an elongated hairpin structure in all fusion protein structures known to date. The fusion process follows several lipidic intermediate states, which are generated by the refolding process. Although the major principles of viral fusion are understood, the structures of fusion protein intermediates and their mode of lipid bilayer interaction, the structures and functions of the membrane anchors and the number of fusion proteins required for fusion, necessitate further investigations.

Inhibition of Various Steps in the Replication Cycle of Vesicular Stomatitis Virus Contributes to Its Photoinactivation by AIPcS4 or Pc4 and Red Light

Abstract— Vesicular stomatitis virus (VSV) was used as a model virus to study the processes involved in photoinactivation by aluminum phthalocyanine tetrasulfonate (AlPcS4) or silicon phthalocyanine HOSiPcOSi(CH3)2(CH2)3N(CH3)2 (Pc4) and red light. Previously a very rapid decrease in the intracellular viral RNA synthesis after photodynamic treatment was observed. This decrease was correlated to different steps in the replication cycle. Binding of VSV to host cells and internalization were only slightly impaired and could be visualized by electron microscopy. The capability of the virus to fuse with membranes in an acidic endosomal environment was studied using both pyrene‐labeled liposomes and a hemolysis assay as a model. These tests indicate a rapid decrease of fusion capacity after AlPcS4 treatment, which correlated with the decrease in RNA synthesis. For Pc4 treatment no such correlation was found. The fusion process is the first step in the replication cycle, affected by AlPcS4 treatment, but also in vitro RNA polymerase activity was previously shown to be inhibited. Inactivation of VSV by Pc4 treatment is apparently caused by damage to a variety of viral components. Photodynamic treatment of virus suspensions with both sensitizers causes formation of 8‐oxo‐7,8‐dihydroguanosine in viral RNA as measured by HPLC with electrochemical detection. This damage might be partly responsible for inhibition of the in vitro viral RNA polymerase activity by photodynamic treatment.

Vacuolar ATPase activity is required for endosomal carrier vesicle formation.

A proton pump, the vacuolar ATPase, is known to generate the acidic lumenal environment of endosomes and lysosomes. We have investigated the role of the vacuolar ATPase in endocytic membrane traffic by combining electron microscopy in vivo with a cell-free assay that reconstitutes endosome fusion in vitro. Our observations show that inactivation of this proton pump with bafilomycin A1 has no significant effects on internalization or recycling back to the plasma membrane. However, early endosomes become highly tubular and endocytosed markers do not appear in late endosomes. Our data strongly suggest that, upon inactivation of the proton pump, the formation of a vesicular intermediate between early and late endosomes, which we term endosomal carrier vesicle, is impaired.
