Acibenzolar-S-methyl promotes wound healing of harvested sweet potatoes (Ipomoea batatas) by regulation of reactive oxygen species metabolism and phenylpropanoid pathway.

Rapid wound healing is crucial in protecting sweet potatoes (Ipomoea batatas ) against infection, water loss and quality deterioration during storage. The current study investigated how acibenzolar-S-methyl (ASM) treatment influenced wound healing in harvested sweet potatoes by investigating the underlying mechanism. It was found that ASM treatment of wounded sweet potatoes induced a significant accumulation of lignin at the wound sites, which effectively suppressed weight loss. After 4days of healing, the lignin content of ASM-treated sweet potatoes was 41.8% higher than that of untreated ones, and the weight loss rate was 20.4% lower. Moreover, ASM treatment increased the ability of sweet potatoes to defend against wounding stress through enhancing processes such as increased production of reactive oxygen species (ROS), activation of enzymes involved in the ROS metabolism (peroxidase, superoxide dismutase and catalase) and phenylpropanoid pathway (phenylalanine ammonia lyase, cinnamate-4-hydroxylase, 4-coumarate-CoA ligase and cinnamyl alcohol dehydrogenase), and intensive synthesis of phenolics and flavonoids. These results suggest that treating harvested sweet potatoes with ASM promotes wound healing through the activation of the ROS metabolism and phenylpropanoid pathway.

Chemical composition and antioxidant properties of beetroot parasitized by Meloidogyne javanica and treated with elicitors

Beetroot is rich in nutrients and bioactive compounds, such as, betalains and phenolics. The objective of this work was to analyze the chemical composition and antioxidant properties of beetroot parasitized by Meloidogyne javanica and treated with elicitors. Beet seedlings were inoculated with 1000 eggs and second-stage juveniles of M. javanica and treated with elicitors based on mannan oligosaccharides, citrus biomass, or acibenzolar-S-methyl (ASM). At 60 days after inoculation, plants were evaluated for nematode reproduction factor (RF), proximate composition, betalains, phenolic compounds, and antioxidant activity. Beetroot was found to be susceptible to M. javanica, with RF values ranging from 15.26 to 27.94. ASM and citrus biomass treatments increased ash content by 15–25% in nematode-inoculated plants. There was no treatment effect on moisture or protein content, but uninoculated plants had higher total phenolic content (~35%) than inoculated plants. Nematode infection was found to compromise betalain production but did not impact antioxidant activity. Nematode-inoculated plants showed reduced sucrose content and increased glucose and fructose levels in all treatments. Regardless of the elicitor used, beetroots inoculated M. javanica exhibit deformities that render them unmarketable in fresh form. However, the results indicate that nematode-infected beetroot may be suitable for industrial processing and compound extraction.

Optimizing the application of acibenzolar-S-methyl and copper to control bacterial spot on fresh-market tomato

This study aimed to adjust the number of applications of acibenzolar-S-methyl (ASM) and copper in an integrated program for the control of bacterial spot on fresh-market tomato. Two field trials were carried out during 2018 and 2019. Four sets of number of weekly applications of ASM were evaluated (7, 9, 11, and 13 applications). In addition to these, four other treatments were included: two as a tank mixture compound [13 weekly applications of ASM + biostimulant and nine applications of ASM + copper oxychloride]. Copper oxychloride was applied after ASM treatments in order to complete 13 applications per growing season. Copper oxychloride and untreated plots were used as a control. The variables evaluated were bacterial spot severity, total yield, and yield per classes of fruit according to the Brazilian market standards. Benefit-cost ratio (B/C) was calculated considering the income increase of a given treatment in relation to the untreated control. The disease ratings and yield data were subjected to ANOVA and the means tested for significance by Fisher, Dunnett, and orthogonal contrasts comparison tests. Regression analysis were carried out with the disease ratings to evaluate the ASM number of applications. As the number of ASM applications increased, disease severity decreased, although, the best B/C was obtained with seven applications of ASM, followed by copper applications until the end of the cycle. The tank mixture compound treatments did not result in economic gain increase. Moreover, disease severity was significantly high when ASM was applied in tank mixture with copper.

Acibenzolar-S-methyl activates calcium signalling to mediate lignin synthesis in the exocarp of Docteur Jules Guyot pears

‘Docteur Jules Guyot’ pears were immersed in acibenzolar-S-methyl (ASM) and 0.01 mol L−1 ethyl glycol tetra acetic acid (EGTA) to investigate the changes of Ca2+ receptor proteins and phenylpropanoid pathway. Results showed that ASM treatment increased the activities of phenylalanine ammonia-lyase (PAL), cinnamate-4-hydroxylase (C4H), 4-coumarate coenzyme A ligase (4CL), polyphenol oxidase (PPO), and cinnamyl alcohol dehydrogenase (CAD) in the exocarp of pears, whereas EGTA pre-treatment inhibited the activities of these enzymes. ASM treatment also enhanced the transcription of PcPAL, PcC4H, Pc4CL, PcC3H, PcCOMT, PcCCoAOMT, PcCCR, PcPOD, PcCDPK1, PcCDPK2, PcCDPK5, PcCDPK11, PcCDPK13, PcCBL1, PcCBL9, PcCIPK14, and PcCML27 in pears. EGTA + ASM treatments inhibited the transcription of PcPAL, PcC4H, Pc4CL, PcC3H, PcCCR, PcF5H, PcCAD, PcCDPK11, PcCDPK26, PcCDPK32, PcCBL1, PcCIPK14, PcCIPK23, and PcCaM in the fruit. All these results indicated that ASM induced the gene expressions of Ca2+ receptor proteins, the key enzyme activities and gene expressions in phenylpropanoid pathway; Ca2+ mediated phenylpropane metabolism in pears after ASM treatment.

Acibenzolar-S-methyl activates mitogen-activated protein kinase cascade to mediate chlorophyll and carotenoid metabolisms in the exocarp of Docteur Jules Guyot pears.

Acibenzolar-S-methyl (ASM), a well-known plant activator, has been used to protect fruit and vegetable from fungal invasion and maintain quality. However, little is known about the molecular mechanism of ASM in regulating chlorophyll and carotenoid metabolisms. Therefore, Docteur Jules Guyot pears were used as the materials to study the changes of hydrogen peroxide (H2 O2 ) production, mitogen-activated protein kinase (MAPK) cascade, transcription factors, chlorophyll, and carotenoid metabolisms after ASM and PD98059 (a MAPK cascade blocker) treatments. ASM increased NADPH oxidase (NOX) and superoxide dismutase (SOD) activities, and H2 O2 content, promoted PcMAPKKK1, PcMAPKK3, and PcMAPK6 expressions, and down-regulated PcMYC2, PcPIF1, PcPIF3, and PcPIF4 expressions in exocarp of pears. ASM also delayed the decrease of chlorophyll a and b contents, and inhibited the accumulation of β-carotene, lycopene and lutein, PcNYC1, PcHCAR, PcPPH, PcSGR1/2, PcPAO, PcPSY, PcLCYB, PcCRTZ2, PcCCS1 expressions, and promoted PcLCYE expression. PD98059 + ASM treatments depressed SOD and NOX activities and H2 O2 content, inhibited PcMAPKKK1, PcMAPKK3, PcMAPK6, PcPIF1, and PcPIF3 expressions, and promoted PcMYC2 and PcPIF4 expressions in exocarp of pears. Additionally, PD98059 + ASM accelerated PcNYC1, PcHCAR, PcPPH, PcSGR1/2, PcPAO, PcPSY, PcCYB, PcCRTZ2, and PcCCS1 expressions, thereby reducing chlorophyll a and b contents, and promoting β-carotene, lycopene and lutein contents. Postharvest ASM treatment promoted the production of H2 O2 to activate the MAPK cascade, then phosphorylated/dephosphorylated transcription factors expression, and delayed chlorophyll decomposition and carotenoid synthesis in pears. © 2022 Society of Chemical Industry.

Combination of yeast antagonists and Acibenzolar-S-Methyl reduced the severity of Fusarium head blight of wheat incited by Fusarium graminearum sensu stricto

The combination of yeast antagonists and Acibenzolar-S-Methyl (ASM) was tested against Fusarium graminearum on a spring wheat cultivar PAN3471. Two strains of Papiliotrema flavescens (Strains WL3 and WL6) and a strain of Pseudozyma sp. (MGO1) were combined with full strength ASM at anthesis, half strength ASM at anthesis and quarter strength ASM at late boot stages. The yeast and ASM treatments were applied prior to F. graminearum inoculation and disease progress was assessed over time. The combination of yeast and ASM treatments effectively reduced Fusarium Head Blight (FHB) severity and deoxynivalenol (DON) concentration compared to when the treatments were used alone. A positive correlation was observed between the Area Under Disease Progress Curve (AUDPC) and Percentage Seed Infection (PSI) (r = 0.44) whereas a negative correlation was observed between AUDPC and Hundred Seed Weight (HSW) (r = -0.77) and PSI and HSW (r = -0.44). The best combination treatment providing the highest reduction in final disease severity (41.83%), high HSW and moderate PSI was 0.075 g/l ASM at anthesis plus P. flavescens strain WL3. The highest DON reduction (19.35%) was by the treatment 0.075 g/l ASM at anthesis plus P. flavescens strain WL6. The best treatment was P. flavescens combined with 0.075 g/l ASM at anthesis. Although Pseudozyma sp. strain MGO1 did not provide the best FHB and DON reduction, its combination with ASM application improved disease control efficacy. To the best of our knowledge, this study presents the first report of the combination of P. flavescens and ASM in the management of FHB caused by F. graminearum in wheat plants.

Defence Responses Associated with Elicitor-Induced, Cultivar-Associated Resistance to Latania Scale in Kiwifruit.

Latania scale insect is a pest of global significance affecting kiwifruit. The sessile insect (life stage: settled crawler—mature adult) is covered with a waxy cap that protects it from topical pesticides, so increasingly, a selection of resistant cultivars and application of elicitors are being used in pest control. Thus far, the application of a salicylic acid (SA) phytohormone pathway elicitor, acibenzolar-S-methyl (ASM), has been shown to reduce insect development (as indicated by cap size) on one kiwifruit cultivar (‘Hayward’). To investigate how cultivar-associated resistance is affected by the ability to respond to different elicitors, we measured phytohormones (by LCMS) and gene expression (by qPCR and NanoString) on latania scale-tolerant ‘Hort16A’ and susceptible ‘Hayward’ kiwifruit over two seasons. Potted plants in the presence/absence of settled latania scales were treated with ASM (0.2 g/L) or methyl jasmonate (MeJA, 0.05% v/v), representing elicitors of the SA and JA signalling pathways, respectively. ‘Hort16A’ cultivar resistance to latania scale was associated with elevated expression of SA and SA-related defence genes (PR1 and two PR2 family genes) in the ASM treatment. MeJA treatments did not significantly affect insect development in ‘Hayward’ (latania scale did not survive on ‘Hort16A’) and did not correlate with phytohormone and gene expression measurements in either cultivar. ‘Hayward’ had greater concentrations than ‘Hort16A’ of inert storage forms of both SA and JA across all treatments. This information contributes to the selection of tolerant cultivars and the effective use of elicitors for control of latania scale in kiwifruit.

Acibenzolar-S-methyl-mediated restriction of loading of plantago asiatica mosaic virus into vascular tissues of Nicotiana benthamiana

Long-distance movement via vascular tissues is an essential step for systemic infection by plant viruses. We previously reported that pre-treatment of Nicotiana benthamiana with acibenzolar-S-methyl (ASM) both suppressed the accumulation of plantago asiatica mosaic virus (PlAMV) in inoculated leaves and delayed the long-distance movement to uninoculated upper leaves. These two effects occurred independently of each other. However, it remained unclear where and when the viral long-distance movement is inhibited upon ASM treatment. In this study, we found that ASM treatment restricted the loading of GFP-expressing PlAMV (PlAMV-GFP) into vascular tissues in the inoculated leaves. This led to delays in viral translocation to the petiole and the main stem, and to untreated upper leaves. We used cryohistological fluorescence imaging to show that ASM treatment affected the viral localization and reduced its accumulation in the phloem, xylem, and mesophyll tissues. A stem girdling experiment, which blocked viral movement downward through phloem tissues, demonstrated that ASM treatment could inhibit viral systemic infection to upper leaves, which occurred even with viral downward movement restricted. Taken together, our results showed that ASM treatment affects the loading of PlAMV-GFP into the vascular system in the inoculated leaf, and that this plays a key role in the ASM-mediated delay of viral long-distance movement.

Acibenzolar-S-methyl induces resistance against cassava mosaic geminiviruses in Nicotiana benthamiana and their vector Bemisia tabaci in cassava (Manihot esculenta)

Cassava mosaic disease (CMD), caused by cassava mosaic geminiviruses (CMGs), is a major constraint to the cassava crop in Africa and southeastern Asia. Here, we investigated the ability of acibenzolar-S-methyl (ASM), a functional analog of salicylic acid (SA), to trigger systemic acquired resistance (SAR) against two CMGs, namely African cassava mosaic virus (ACMV) and East African cassava mosaic Cameroon virus (EACMCV) in Nicotiana benthamiana. ASM treatment delayed the time to first viral symptoms appearance, reduced virus infection rate, and attenuated symptoms. Furthermore, ASM caused an enhanced recovery from symptoms of both viruses and inhibited plant death observed in N. benthamiana plants infected by EACMCV. This study further showed that ASM induced resistance to the whitefly Bemisia tabaci (Gennadius), the vector of CMGs, in cassava. Notably, we observed that ASM treatment affected adult whitefly feeding preference and oviposition in cassava under both choice and no-choice conditions. A significant reduction in whitefly adult, egg, and nymph populations was observed irrespective of ASM treatment. The results of this study show that ASM has the potential to control both CMGs and their whitefly vector which is an important first step toward managing whitefly and cassava viruses.

Acibenzolar-S-Methyl and Resistance Quantitative Trait Loci Complement Each Other to Control Apple Scab and Fire Blight.

Diversifying disease control methods is a key strategy to sustainably reduce pesticides. Plant genetic resistance has long been used to create resistant varieties. Plant resistance inducers (PRI) are also considered to promote crop health, but their effectiveness is partial and can vary according to the environment and the plant genotype. We investigated the putative interaction between intrinsic (genetic) and PRI-induced resistance in apple when affected by scab and fire blight diseases. A large F1 mapping population was challenged by each disease after a pre-treatment with acibenzolar-S-methyl (ASM) and compared with the water control. Apple scab and fire blight resistance quantitative trait loci (QTLs) were detected in both conditions and compared. ASM exhibited a strong effectiveness in reducing both diseases. When combined, QTL-controlled and ASM-induced resistance acted complementarily to reduce the symptoms from 85 to 100%, depending on the disease. In our conditions, resistance QTLs were only slightly or rarely affected by ASM treatment, despite their probable implication in various stages of the resistance buildup. Implications of these results are discussed considering already known results, the underlying mechanisms, cross protection of both types of resistance against pathogen adaptation, and practical application in orchard conditions.

Roles of calcium-dependent protein kinases mediated reactive oxygen species homeostasis in inducing resistance of apples by acibenzolar-S-methyl

This study was carried out to investigate the effect of acibenzolar-S-methyl (ASM) and ethylenebis (oxyethylenenitrilo) tetraacetic acid (EGTA) treatments on calcium-dependent protein kinases (CDPKs) and reactive oxygen species (ROS) metabolism in apples. Postharvest ASM treatment increased H2O2 content, reduced glutathione and ascorbic acid contents, and NADPH oxidase, peroxidase, ascorbate peroxidase, superoxide dismutase and glutathione reductase activities and retarded catalase activity and MdCAT expression in apples. ASM treatment enhanced MdSOD, MdPOD, MdAPX, MdGR, MdCDPK1, MdCDPK4, MdCDPK5, MdCDPK7, and MdCDPK21 expressions in apples. However, EGTA + ASM treatments suppressed H2O2, glutathione and ascorbic acid contents, NADPH oxidase, peroxidase, superoxide dismutase, ascorbate peroxidase and glutathione reductase activities. EGTA + ASM treatments suppressed the selected genes expressions in ROS metabolism and CDPKs, but up-regulated MdCAT expression in apples. These findings suggest that CDPKs play a vital role in regulating ROS metabolism and involve in inducing resistance in apples by ASM.

Acibenzolar-S-Methyl Activates Stomatal-Based Defense Systemically in Japanese Radish.

Acibenzolar-S-methyl (ASM) is a well-known plant activator, which is a synthetic analog of salicylic acid (SA). Recently, copper fungicides and antibiotics are major strategies for controlling bacterial diseases. However, resistant strains have already been found. Therefore, there is an increasing demand for sustainable new disease control strategies. We investigated the ASM disease control effect against Pseudomonas cannabina pv. alisalensis (Pcal), which causes bacterial blight on Japanese radish. In this study, we demonstrated that ASM effectively suppressed Pcal disease symptom development associated with reduced bacterial populations on Japanese radish leaves. Interestingly, we also demonstrated that ASM activated systemic acquired resistance (SAR), including stomatal-based defense on ASM-untreated upper and lower leaves. Reactive oxygen species (ROS) are essential second messengers in stomatal-based defense. We found that ASM induced stomatal closure by inducing ROS production through peroxidase. These results indicate that stomatal closure induced by ASM treatment is effective for preventing Pcal pathogen invasion into plants, and in turn reduction of disease development.

Mitogen-Activated Protein Kinase Cascade and Reactive Oxygen Species Metabolism are Involved in Acibenzolar-S-Methyl-Induced Disease Resistance in Apples.

Apple fruits were subjected to dipping treatment to explore the effects of acibenzolar-S-methyl (ASM) and the mitogen-activated protein kinase (MAPK) inhibitor PD98059 on lesion growth in fruits inoculated with Penicillium expansum. We investigated the roles of the MAPK cascade and reactive oxygen species metabolism in disease resistance in apples. ASM treatment inhibited lesion growth; suppressed catalase (CAT) activity; increased H2O2 content; reduced glutathione and ascorbic acid contents; and increased glutathione reductase, ascorbate peroxidase, peroxidase, superoxide dismutase, and NADPH oxidase activities. Moreover, ASM upregulated MdSOD, MdPOD, MdGR, MdAPX, MdMAPK4, MdMAPK2, and MdMAPKK1 expressions and downregulated MdCAT and MdMAPK3 expressions. PD98059 + ASM treatment increased CAT activity and MdCAT and MdMAPK3 expressions; inhibited MdSOD, MdPOD, MdGR, MdAPX, MdMAPK4, MdMAPK2, and MdMAPKK1 expressions; reduced superoxide dismutase, peroxidase, ascorbate peroxidase, and glutathione reductase activities; and reduced glutathione content in apples. These findings indicate that ASM induces disease resistance in apples by regulating the expressions of key genes involved in reactive oxygen species metabolism and the MAPK cascade.

Gene expression of salicylic acid and jasmonic acid pathways and photosynthesis parameters of sweet orange trees in response to acibenzolar-S-methyl

The use of resistance inducers may be a tool to assist in the management of citrus diseases. However, knowledge about how some inducers, such as the benzothiadiazoles, promote gene expression from the pathways involved in induced resistance and alter plant development are lacking. In this work, we evaluated the effect of the acibenzolar-S-methyl (ASM) on gene expression from the biosynthetic pathways of salicylic (SA) and jasmonic (JA) acids, as well as in physiological parameters of young sweet orange trees. After a single ASM application, gene expression assessed during six time points revealed no significant changes in the main pathway for SA synthesis: isochorismate synthases (ISC) 1 and ICS2 genes were up-regulated only at 6 days after treatment (dat). However, an increase in phenylalanine ammonia lyase (PAL) was observed. Salicylic acid carboxymethyl transferase (BSMT1) and salicylic acid glucosyltransferase I (SAGT1) were up-regulated, indicating expression of genes whose products modify SA. Pointedly up-regulation of lipoxygenase (LOX) and both up- and down-regulation of allene oxide synthase (AOS) and allene oxide cyclase (AOC) in biosynthetic pathways of JA was observed, while jasmonic acid methyl transferase (JMT), whose product modify JA, was consistently induced from three to 13 dat. Besides an increase in SA and JA modification enzymes, ASM reduced photosynthetic rate in treated plants, without affecting the stomatal conductance and the transpiration rate. Our results reveal how the SA and JA pathways are regulated soon after ASM treatment, in particular the up-regulation of BSTM1, SAGT1 and JMT.

Influence of Resistance-Inducing Chemical Elicitors against Pine Wilt Disease on the Rhizosphere Microbiome.

Pine wilt disease (PWD) caused by Bursaphelenchus xylophilus is a major threat to pine forests worldwide. Induction of resistance is a promising and safe management option that should be investigated in relation to its possible influence on the pine tree ecosystem, including the surrounding microbial communities. In this study, two main resistance-inducing chemical elicitors, methyl salicylic acid (MeSA) and acibenzolar-s-methyl (ASM), were tested for their control efficiency against PWD and their influence on the rhizosphere microbial composition. Foliar treatment of pine seedlings with the chemical elicitors resulted in a reduction in PWD severity, with ASM showing better control efficacy, reaching up to 73% compared to the untreated control. Moreover, bacterial community analysis of the rhizosphere revealed significant changes in several microbial taxa that were present at low relative abundance. In particular, ASM treatment resulted in a significant increase in specific microbial taxa, including members of the Rhodanobacter, Devosia, Bradyrhizobium, Acidibacter, Mesorhizobium, and Hyphomicrobium genera, which are known to play ecological and plant growth-promoting roles. Furthermore, chitinolytic bacteria were shown to be reduced in response to treatment with both MeSA and ASM. Altogether, the present findings demonstrate the occurrence of significant alterations in several ecologically important microbial taxa after treatment with resistance-inducing chemicals. As compared to MeSA treatment, ASM treatment was more effective at suppressing PWD and resulted in more beneficial changes in rhizosphere microbial composition.

Priming Melon Defenses with Acibenzolar-S-methyl Attenuates Infections by Phylogenetically Distinct Viruses and Diminishes Vector Preferences for Infected Hosts.

Plant virus management is mostly achieved through control of insect vectors using insecticides. However, insecticides are only marginally effective for preventing virus transmission. Furthermore, it is well established that symptoms of virus infections often encourage vector visitation to infected hosts, which exacerbates secondary spread. Plant defense elicitors, phytohormone analogs that prime the plant immune system against attack, may be a viable approach for virus control that complements insecticide use by disrupting pathologies that attract vectors. To explore this, we tested the effect of a commercial plant elicitor, acibenzolar-S-methyl (ASM), on infection rates, virus titers, and symptom development in melon plants inoculated with one of two virus species, Cucumber mosaic virus (CMV) and Cucurbit yellow stunting disorder virus (CYSDV). We also conducted behavioral assays to assess the effect of ASM treatment and virus inoculation on vector behavior. For both pathogens, ASM treatment reduced symptom severity and delayed disease progression. For CYSDV, this resulted in the attenuation of symptoms that encourage vector visitation and virion uptake. We did observe slight trade-offs in growth vs. defense following ASM treatment, but these effects did not translate into reduced yields or plant performance in the field. Our results suggest that immunity priming may be a valuable tool for improving management of insect-transmitted plant viruses.

Acibenzolar-S-methyl and β-amino butyric acid-induced upregulation of biochemical defense against Colletotrichum capsici infection in chilli (Capsicum annuum)

Abiotic inducers (AI) are synthetic/natural compound, induces systemic resistance in plants against pathogens. Present study aimed to evaluate the foliar application of AI [Acibenzolar-S-Methyl (ASM), and β-Amino Butyric Acid (BABA)] in managing chilli anthracnose under greenhouse conditions. Foliar applications of ASM-5 μg/ml and BABA-10 mM significantly decreases disease incidence to 13.15% and 14.1% respectively, when compared to respective controls (65% and 65.34%). AI treated seedlings recorded maximum H2O2 accumulation at 9 h (ASM) and 12 h (BABA) after challenged with pathogen. In AI treated seedlings, activity of defense-related enzymes (Phenylalanine ammonia-lyase, Peroxidase, Polyphenol oxidase, Lipoxygenase, and chitinase) was upregulated after challenge inoculation. Similarly, maximum phenolic (μg gallic acid/g tissue) accumulation of 147 and 71.2 was recorded with ASM and BABA treatment, respectively. The AI induced changes in biochemical parameters are in correlation with reduced disease incidence. Hence, AI can use as alternative, safe and eco-friendly candidate to manage chilli anthracnose.

Postharvest acibenzolar-S-methyl treatment maintains storage quality and retards softening of apple fruit.

"Golden delicious" apples were dipped in 100mg/L acibenzolar-S-methyl (ASM) to investigate the fruit quality and softening during 12days of storage. Weight loss, flesh firmness, ethylene release, respiratory rate, content of total soluble solids and titratable acid, the activity of pectinase, cellulase, and β-glucosidase, and water-insoluble pectin and water-soluble pectin contents were investigated. The results demonstrated that ASM treatment inhibited ethylene release and respiratory rate, reduced titratable acidity, and enhanced total soluble solids content in apples. Moreover, application of ASM suppressed the reduction of flesh firmness, activity of pectin methylesterase, and polygalacturonase. Cellulase, β-glucosidase, and degradation of protopectin in apple fruit were also suppressed by ASM treatment during storage. In conclusion, ASM could maintain fruit quality by regulating cell wall-degrading enzymes during storage. PRACTICAL APPLICATIONS: Application of acibenzolar-S-methyl after harvest has the potential of delaying fruit softening by regulating cell wall-degrading enzymes, thus retain fruit quality.

The role of glucose-6-phosphate dehydrogenase in reactive oxygen species metabolism in apple exocarp induced by acibenzolar-S-methyl

Apple exocarp was used to investigate the effect of acibenzolar-S-methyl (ASM) and dehydroepiandrosterone (DHEA) treatments on reaction oxygen species (ROS) metabolism. The results indicated that ASM enhanced the hydrogen peroxide (H2O2) content, the activities of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), glutathione reductase (GR) and glucose-6-phosphate dehydrogenase (G6PDH). ASM also increased the contents of ascorbic acid (AsA), reduced glutathione (GSH) and nicotinamide ademine dinucleotidephosphate (NADPH), MdSOD and MdAPX expression, but decreased MdMDHAR and dehydroascorbate reductase (MdDHAR) expression. DHEA suppressed H2O2 accumulation and POD, APX, MDHAR, G6PDH activities, but increased SOD, CAT and GR activities compared to the control. ASM and DHEA treatments suppressed the contents of AsA, GSH and NADPH, and expression of MdSOD, MdAPX and MdMDHAR. These results suggest that DHEA treatment prevented ROS metabolism induced by ASM which showed the important role of G6PDH in maintaining redox homeostasis in apple exocarp.

Acibenzolar-S-Methyl Restricts Infection of Nicotiana benthamiana by Plantago Asiatica Mosaic Virus at Two Distinct Stages.

Plant activators, including acibenzolar-S-methyl (ASM), are chemical compounds that stimulate plant defense responses to pathogens. ASM treatment inhibits infection by a variety of plant viruses, however, the mechanisms of this broad-spectrum and strong effect remain poorly understood. We employed green fluorescent protein (GFP)-expressing viruses and Nicotiana benthamiana plants to identify the infection stages that are restricted by ASM. ASM suppressed infection by three viral species, plantago asiatica mosaic virus (PlAMV), potato virus X (PVX), and turnip mosaic virus (TuMV), in inoculated cells. Furthermore, ASM delayed the long-distance movement of PlAMV and PVX, and the cell-to-cell (short range) movement of TuMV. The ASM-mediated delay of long-distance movement of PlAMV was not due to the suppression of viral accumulation in the inoculated leaves, indicating that ASM restricts PlAMV infection in at least two independent steps. We used Arabidopsis thaliana mutants to show that the ASM-mediated restriction of PlAMV infection requires the NPR1 gene but was independent of the dicer-like genes essential for RNA silencing. Furthermore, experiments using protoplasts showed that ASM treatment inhibited PlAMV replication without cell death. Our approach, using GFP-expressing viruses, will be useful for the analysis of mechanisms underlying plant activator-mediated virus restriction.
