Occurrence and diffusive air-seawater exchanges of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) in Fildes Bay, King George Island, Antarctica

We report the levels of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) in seawater and air, and the air-sea dynamics through diffusive exchange analysis in Fildes Bay, King George Island, Antarctica, between November 2019 and January 30, 2020. Hexachlorobenzene (HCB) was the most abundant compound in both air and seawater with concentrations around 39 ± 2.1 pg m−3 and 3.2 ± 2.4 pg L−1 respectively. The most abundant PCB congener was PCB 11, with a mean of 3.16 ± 3.7 pg m−3 in air and 2.0 ± 1.1 pg L−1 in seawater. The fugacity gradient estimated for the OCP compounds indicate a predominance of net atmospheric deposition for HCB, α-HCH, γ-HCH, 4,4′-DDT, 4,4′-DDE and close to equilibrium for the PeCB compound. The observed deposition of some OCs may be driven by high biodegradation rates and/or settling fluxes decreasing the concentration of these compounds in surface waters, which is supported by the capacity of microbial consortium to degrade some of these compounds. The estimated fugacity gradients for PCBs showed differences between congeners, with net volatilization predominating for PCB-9, a trend close to equilibrium for PCB congeners 11, 28, 52, 101, 118, 138, and 153, and deposition for PCB 180. Snow amplification may play an important role for less hydrophobic PCBs, with volatilization predominating after snow/glacier melting. As hydrophobicity increases, the biological pump decreases the concentration of PCBs in seawater, reversing the fugacity gradient to atmospheric deposition. This study highlights the potential impacts of climate change, through glacier retreat, on the biogeochemistry of POPs, remobilizing those compounds previously trapped within the cryosphere which in turn will transform the Antarctic cryosphere into a secondary source of the more volatile POPs in coastal areas, influenced by snow and ice melting.

Developmental Exposure to a Human-Relevant Polychlorinated Biphenyl Mixture Causes Behavioral Phenotypes That Vary by Sex and Genotype in Juvenile Mice Expressing Human Mutations That Modulate Neuronal Calcium.

Polychlorinated biphenyls (PCBs) are putative environmental risks for neurodevelopmental disorders. Here, we tested two hypotheses: (1) developmental exposure to a human-relevant PCB mixture causes behavioral phenotypes relevant to neurodevelopmental disorders; and (2) expression of human mutations that dysregulate neuronal Ca2+ homeostasis influence sensitivity to behavioral effects of developmental PCB exposures. To test these hypotheses, we used mice that expressed a gain-of-function mutation (T4826I) in ryanodine receptor 1 (RYR1), the X-linked fragile X mental retardation 1 (FMR1) CGG repeat expansion or both mutations (double mutant; DM). Transgenic mice and wildtype (WT) mice were exposed to the MARBLES PCB mix at 0, 0.1, 1, and 6 mg/kg/day in the maternal diet throughout gestation and lactation. The MARBLES PCB mix simulates the relative proportions of the 12 most abundant PCB congeners found in the serum of pregnant women at increased risk for having a child with a neurodevelopmental disorder. We assessed ultrasonic vocalizations at postnatal day 7 (P7), spontaneous repetitive behaviors at P25-P30, and sociability at P27-P32. Developmental PCB exposure reduced ultrasonic vocalizations in WT litters in all dose groups, but had no effect on ultrasonic vocalizations in transgenic litters. Developmental PCB exposure significantly increased self-grooming and decreased sociability in WT males in the 0.1 mg/kg dose group, but had no effect on WT females in any dose group. Genotype alone influenced ultrasonic vocalizations, self-grooming and to a lesser extent sociability. Genotype alone also influenced effects of PCBs on sociability. PCB levels in the brain tissue of pups increased in a dose-dependent manner, but within any dose group did not differ between genotypes. In summary, developmental PCB exposure phenocopied social behavior phenotypes observed in mice expressing human mutations that modify intracellular Ca2+ dynamics, and expression of these mutations alleviated PCB effects on ultrasonic vocalizations and repetitive behavior, and modified the dose-response relationships and sex-dependent effects of PCB effects on social behavior. These findings suggest that: (1) developmental PCB exposure causes behavioral phenotypes that vary by sex and genotype; and (2) sex-specific responses to environmental factors may contribute to sex biases in the prevalence and/or severity of neurodevelopmental disorders.

Sex and Genotype Modulate the Dendritic Effects of Developmental Exposure to a Human-Relevant Polychlorinated Biphenyls Mixture in the Juvenile Mouse.

While many neurodevelopmental disorders (NDDs) are thought to result from interactions between environmental and genetic risk factors, the identification of specific gene-environment interactions that influence NDD risk remains a critical data gap. We tested the hypothesis that polychlorinated biphenyls (PCBs) interact with human mutations that alter the fidelity of neuronal Ca2+ signaling to confer NDD risk. To test this, we used three transgenic mouse lines that expressed human mutations known to alter Ca2+ signals in neurons: (1) gain-of-function mutation in ryanodine receptor-1 (T4826I-RYR1); (2) CGG-repeat expansion in the 5′ non-coding portion of the fragile X mental retardation gene 1 (FMR1); and (3) a double mutant (DM) that expressed both mutations. Transgenic and wildtype (WT) mice were exposed throughout gestation and lactation to the MARBLES PCB mix at 0.1, 1, or 6 mg/kg in the maternal diet. The MARBLES mix simulates the relative proportions of the twelve most abundant PCB congeners found in serum from pregnant women at increased risk for having a child with an NDD. Using Golgi staining, the effect of developmental PCB exposure on dendritic arborization of pyramidal neurons in the CA1 hippocampus and somatosensory cortex of male and female WT mice was compared to pyramidal neurons from transgenic mice. A multilevel linear mixed-effects model identified a main effect of dose driven by increased dendritic arborization of cortical neurons in the 1 mg/kg PCB dose group. Subsequent analyses with genotypes indicated that the MARBLES PCB mixture had no effect on the dendritic arborization of hippocampal neurons in WT mice of either sex, but significantly increased dendritic arborization of cortical neurons of WT males in the 6 mg/kg PCB dose group. Transgene expression increased sensitivity to the impact of developmental PCB exposure on dendritic arborization in a sex-, and brain region-dependent manner. In conclusion, developmental exposure to PCBs present in the gestational environment of at-risk humans interfered with normal dendritic morphogenesis in the developing mouse brain in a sex-, genotype- and brain region-dependent manner. Overall, these observations provide proof-of-principle evidence that PCBs interact with heritable mutations to modulate a neurodevelopmental outcome of relevance to NDDs.

The Variability of the Concentrations of PAHs and PCBs in the Urban Air of Dalian with Ambient Temperature by Semipermeable Membrane Devices Monitoring

Bioavailable air concentration of 18 indicator and dioxin-like polychlorinated biphenyls (PCBs) and 16 EPA priority polycyclic aromatic hydrocarbons (PAHs) in the urban air of Dalian, China were simultaneously measured by semipermeable membrane devices (SPMDs). PCB#28, 52, 101, 105 and 118 were the most abundant PCB congeners in Dalian urban air. Phenanthrene, Fluoranthene and Pyrene were the predominat PAH in the air. PCBs were detected with magnitude ranging from non-detectable to 2151 pg SPMD−1. PAHs ranged from non-detectable to 12464 ng SPMD−1. The linear uptake stages of different PCBs and PAHs with SPMD sampling were different. There were much more PCBs during warmer sampling period than colder sampling period, however, there were a reverse results for PAHs. The concentration of some PCBs and PAHs sequestrated by SPMD (C SPMD) in the air has linear correlation with the reciprocal of the average ambient atmospheric temperature (T). The C SPMD of PAHs had negative correlation with T, while the C SPMD of the PCBs had positive correlation with T. The above research results provide basic reference data for the promotion and application of SPMD passive samplers.

The influence of sex, genotype, and dose on serum and hippocampal cytokine levels in juvenile mice developmentally exposed to a human-relevant mixture of polychlorinated biphenyls

Polychlorinated biphenyls (PCBs) are pervasive environmental contaminants implicated as risk factors for neurodevelopmental disorders (NDDs). Immune dysregulation is another NDD risk factor, and developmental PCB exposures are associated with early life immune dysregulation. Studies of the immunomodulatory effects of PCBs have focused on the higher-chlorinated congeners found in legacy commercial mixtures. Comparatively little is known about the immune effects of contemporary, lower-chlorinated PCBs. This is a critical data gap given recent reports that lower-chlorinated congeners comprise >70% of the total PCB burden in serum of pregnant women enrolled in the MARBLES study who are at increased risk for having a child with an NDD. To examine the influence of PCBs, sex, and genotype on cytokine levels, mice were exposed throughout gestation and lactation to a PCB mixture in the maternal diet, which was based on the 12 most abundant PCBs in sera from MARBLES subjects. Using multiplex array, cytokines were quantified in the serum and hippocampus of weanling mice expressing either a human gain-of-function mutation in ryanodine receptor 1 (T4826I mice), a human CGG premutation repeat expansion in the fragile X mental retardation gene 1 (CGG mice), or both mutations (DM mice). Congenic wildtype (WT) mice were used as controls. There were dose-dependent effects of PCB exposure on cytokine concentrations in the serum but not hippocampus. Differential effects of genotype were observed in the serum and hippocampus. Hippocampal cytokines were consistently elevated in T4826I mice and also in WT animals for some cytokines compared to CGG and DM mice, while serum cytokines were usually elevated in the mutant genotypes compared to the WT group. Males had elevated levels of 19 cytokines in the serum and 4 in the hippocampus compared to females, but there were also interactions between sex and genotype for 7 hippocampal cytokines. Only the chemokine CCL5 in the serum showed an interaction between PCB dose, genotype, and sex. Collectively, these findings indicate differential influences of PCB exposure and genotype on cytokine levels in serum and hippocampal tissue of weanling mice. These results suggest that developmental PCB exposure has chronic effects on baseline serum, but not hippocampal, cytokine levels in juvenile mice.

Dioxins and PCBs – Environment impact on freshwater fish contamination and risk to consumers

This paper reports polychlorinated dibenzo-p-dioxins, dibenzofurans (PCDD/F) and polychlorinated biphenyls (PCB) concentrations in sediment and fish collected from Polish rivers in agricultural and urban areas and the associated risk for fish consumers. Total TEQ concentrations in fish muscles were in the range 0.29–7.25 pg WHO-TEQ g−1 wet weight (w.w.). Generally, levels in sediment and fish muscles were low except for in the Vistula River in Cracow. In this fishery, total TEQ concentrations in sediments were high at 7.84 ± 1.73 pg WHO-TEQ g−1 dry matter and fish were non-compliant with Commission Regulation (EU) 1259/2011/EU (3.5 pg WHO-TEQ g−1 w.w. for PCDD/F and 6.5 pg WHO-TEQ g−1 w.w. for total TEQ). Dioxin congener profiles in sediments were dominated by OCDD, and in the most contaminated fisheries in Cracow additionally by OCDF and 1,2,3,4,7,8-HxCDF. The most accumulated PCDD/Fs congeners in fish were 2,3,7,8-TCDF and 2,3,4,7,8-PeCDF except in Cracow, where 1,2,3,4,7,8-HxCDF dominated. The most abundant dioxin-like PCB (DL-PCB) and non-dioxin-like PCB (NDL-PCB) congeners were PCB 118 and PCB 153, respectively, both in sediment and fish muscles. The levels and profiles of contaminants in fish reflected the pollution of their environment. To characterize the potential health risk associated with dioxin and DL-PCB intake, doses ingested in two 100g portions of fish by adults and children were calculated and expressed as percentages of Tolerable Weekly Intake (TWI). Both values of TWI – that of 14 pg WHO-TEQ kg−1 b.w. and the newer value of 2 pg WHO-TEQ kg−1 b.w. established by the EFSA in November 2018 – were taken into account.It appears that regular consumption of some species could pose a health risk, especially those from urban areas.

Polychlorinated biphenyls affects anaerobic methane production from waste activated sludge through suppressing hydrolysis-acidification and methanation processes

The widespread existence of polychlorinated biphenyls (PCB) in wastewater results in the retention of abundant PCB in waste activated sludge (WAS), which has become a global concern. Till now, the effects of PCB on methane production during WAS anaerobic digestion was still limited. This study aimed to investigate whether and how PCB affects methane production. Results showed that the increased PCB concentration led to the less methane produced. At the highest concentrations of PCB (100 mg/kg DS) in this study, the methane production (112 ± 6 L CH4/kg VS) was significantly reduced by 26.6 ± 0.1% compared to the control (153 ± 7 L CH4/kg VS). Correspondingly, VS destruction at the studied concentrations showed the similar trend. In addition, the dewaterability evaluation indicated that the PCB resulted in the deteriorative sludge dewaterability. The mechanism studies suggested that the decreased methane production with the increased levels of PCB was attributed to the suppression on hydrolysis-acidification and methanation processes. This also was supported by the decreasing key enzymes activities associated with methane production (protease, cellulase, acetate kinase (AK) and coenzyme F420). The relative activity of F420 at 100 mg/kg DS of PCB even reduced to 78% ± 3%.

Halogenated natural products and anthropogenic persistent organic pollutants in chokka squid (Loligo reynaudii) from three sites along the South Atlantic and Indian Ocean coasts of South Africa

Chokka squid (Loligo reynaudii) from three sites along the South African coast were analyzed for halogenated natural products (HNPs) and anthropogenic persistent organic pollutants (POPs). HNPs were generally more than one order of magnitude more abundant than POPs. The most prevalent pollutant, i.e. the HNP 2,3,3′,4,4′,5,5′-heptachloro-1′-methyl-1,2′-bipyrrole (Q1), was detected in all chokka squid samples with mean concentrations of 105, 98 and 45 ng/g lipid mass, respectively, at the Indian Ocean (site A), between both oceans (site B) and the South Atlantic Ocean (site C). In addition, bromine containing polyhalogenated 1′-methyl-1,2′-bipyrroles (PMBPs), 2,4,6-tribromophenol (2,4,6-TBP, up to 28 ng/g lipid mass), polybrominated methoxy diphenyl ethers, MHC-1, TBMP and other HNPs were also detected. Polychlorinated biphenyls (PCBs) were the predominant class of anthropogenic POPs. PCB 153 was the most abundant PCB congener in chokka squid from the Indian Ocean, and PCB 138 in samples from the South Atlantic Ocean and between both oceans.

Comparative Analyses of the 12 Most Abundant PCB Congeners Detected in Human Maternal Serum for Activity at the Thyroid Hormone Receptor and Ryanodine Receptor.

Polychlorinated biphenyls (PCBs) pose significant risk to the developing human brain; however, mechanisms of PCB developmental neurotoxicity (DNT) remain controversial. Two widely posited mechanisms are tested here using PCBs identified in pregnant women in the MARBLES cohort who are at increased risk for having a child with a neurodevelopmental disorder (NDD). As determined by gas chromatography-triple quadruple mass spectrometry, the mean PCB level in maternal serum was 2.22 ng/mL. The 12 most abundant PCBs were tested singly and as a mixture mimicking the congener profile in maternal serum for activity at the thyroid hormone receptor (THR) and ryanodine receptor (RyR). Neither the mixture nor the individual congeners (2 fM to 2 μM) exhibited agonistic or antagonistic activity in a THR reporter cell line. However, as determined by equilibrium binding of [3H]ryanodine to RyR1-enriched microsomes, the mixture and the individual congeners (50 nM to 50 μM) increased RyR activity by 2.4-19.2-fold. 4-Hydroxy (OH) and 4-sulfate metabolites of PCBs 11 and 52 had no TH activity; but 4-OH PCB 52 had higher potency than the parent congener toward RyR. These data support evidence implicating RyRs as targets in environmentally triggered NDDs and suggest that PCB effects on the THR are not a predominant mechanism driving PCB DNT. These findings provide scientific rationale regarding a point of departure for quantitative risk assessment of PCB DNT, and identify in vitro assays for screening other environmental pollutants for DNT potential.

Occurrence, sources and ecotoxicological risks of polychlorinated biphenyls (PCBs) in sediment cores from urban, rural and reclamation-affected rivers of the Pearl River Delta, China

Sediment cores were collected to a depth of 40 cm (50 cm for urban river sediments) in the Pearl River Delta of China from rural river sediments and river sediments undergoing the process of urbanization and reclamation. Polychlorinated biphenyls (PCBs) in sediment cores were determined to evaluate their levels, distribution, possible sources and potential risks aimed at providing effective information of management practices for local governments. Our results showed that the total concentrations of PCBs (∑16PCBs) in urban, rural and reclamation-affected river sediments ranged from 16.15 to 477.85 μg kg−1 (dry weight), with mean values of 121.94, 150.49 and 124.20 μg kg−1 (dry weight), respectively. The most abundant PCB congeners among the study area were light PCBs. Generally, PCBs showed a decreasing trend with depth along sediment cores at most sampling sites. Source analysis indicated that PCBs in the three types of river sediments mainly originated from Aroclor 1242, 1248, 1254 and 1016. Risks evaluation based on sediment quality guideline quotient (SQGQ) showed PCBs at most sampling sites would cause no or moderate adverse biological effects on benthic organisms except surface sediments of U4 and R5 (high adverse biological effects). However, threshold effects level (TEL) is ignored when calculating SQGQ, which might underestimate the risks of PCBs. Thus, a new SQGQ (NSQGQ) taken TEL into consideration was established. Results showed that NSQGQ could evaluate ecotoxicological risks of PCBs better. Redundancy analysis (RDA) showed that PCBs in sediments were positively correlated with sand content and microbial biomass carbon (MBC) (p < 0.05).

Polychlorinated Biphenyls in the Centralized Wastewater Treatment Plant in a Chemical Industry Zone: Source, Distribution, and Removal

Polychlorinated biphenyls (PCBs) could be dissolved in wastewater or adsorbed on particulate. The fate of PCBs in wastewater is essential to evaluate the feasibility of wastewater treatment processes and the environmental risk. Here dissolved and adsorbed concentrations of twenty concerned PCB congeners and total PCBs have been measured in the centralized wastewater treatment plant of a chemical industry zone in Zhejiang, China. It was found that the dyeing chemical processes were the main source of PCBs, which contributed more than 13.6%. The most abundant PCB was PCB-11 in the liquid and solid phase of each treatment stage, accounting for more than 60% of the total 209 PCBs. Partitioning behavior of PCBs between the dissolved and adsorbed phases suggested that Di-CBs were the dominant isomers (&gt;70%) and more than 89.8% of them was adsorbed on the particles and sludge. The total removal efficiency of∑209 PCBs was only 23.2% throughout the whole treatment process. A weak correlation was obtained between the individual PCB concentration and their log Kowin primary sedimentation, anaerobic hydrolysis, aerobic bioprocess stage, and the whole treatment process.

Spatial distribution and ecological risk of polychlorinated biphenyls in sediments from Qinzhou Bay, Beibu Gulf of South China

The residual level of polychlorinated biphenyls (PCBs) was determined in the surface sediments collected from the Qinzhou Bay, Beibu Gulf of Southern China. The results showed that the total concentration of PCBs ranged from 1.62 to 62.63ng/g dry wt, with a mean of 9.87ng/g dry wt. Generally, the average PCBs concentrations in three sample groups descended in this order: inner bay>outer bay>rivers. On a mean level, with respect to the PCBs homologue profiles, the analysis suggested that tetrachlorinated biphenyls was the most abundant PCB, followed by hexachlorinated biphenyls, pentachlorinated biphenyls. PCB profiles varied according to the nature of the site and its proximity to the sources. PCA with multiple linear regression analysis indicated that Aroclor products contributed to the PCBs in Qinzhou Bay. Risk assessments conducted on the levels indicated that PCBs in sediments of Qinzhou Bay posed no significant risk to human health.

Polychlorinated Biphenyls in Residential Dust: Sources of Variability

We characterized the variability in concentrations of polychlorinated biphenyls (PCBs) measured in residential dust. Vacuum cleaner samples were collected from 289 homes in the California Childhood Leukemia Study during two sampling rounds from 2001 to 2010 and 15 PCBs were measured by high resolution gas chromatography-mass spectrometry. Median concentrations of the most abundant PCBs (i.e., PCBs 28, 52, 101, 105, 118, 138, 153, and 180) ranged from 1.0-5.8 ng per g of dust in the first sampling round and from 0.8-3.4 ng/g in the second sampling round. For each of these eight PCBs, we used a random-effects model to apportion total variation into regional variability (6-11%), intraregional between-home variability (27-56%), within-home variability over time (18-52%), and within-sample variability (9-16%). In mixed-effects models, differences in PCB concentrations between homes were explained by home age, with older homes having higher PCB levels. Differences in PCB concentrations within homes were explained by decreasing time trends. Estimated half-lives ranged from 5-18 years, indicating that PCBs are removed very slowly from the indoor environment. Our findings suggest that it may be feasible to use residential dust for retrospective assessment of PCB exposures in studies of children's health.

Polychlorinated biphenyls residues in surface sediments of the eutrophic Chaohu Lake (China): characteristics, risk, and correlation with trophic status

Twenty-eight polychlorinated biphenyls (PCB) congeners were measured in surface sediments from Chaohu Lake to assess their characteristics, potential risk, and the correlation with lake trophic status. PCB levels ranged from 11.074 to 42.712 ng g−1 dry weight (d.w.) in the western lake and 2.017 to 20.189 ng g−1 d.w. in the eastern lake. The highest concentrations were found at the sites near the inlets of western lake tributary rivers where big cities and industrial centers are located. Congeners concentrations showed decreasing order of tetra-CB > tri-CB > deca-CB (PCB-209 detected) > penta-CB > hexa-CB > di-CB > hepta-CB > Octa-CB. It indicated that light and heavy Aroclor mixtures were simultaneously used surrounding the lake basin. PCB levels in the western lake are potentially dangerous to humans and the local fauna. There was a significant positive relationship between tetra-CB (one abundant PCB congener) concentration distribution and sediment grain size in the 16–64 μm fraction, whereas a negative correlation was found in the 4–8 μm fraction. Furthermore, PCB distributions were positively correlated with the total organic carbon of sediments and lake trophic status, especially in the more seriously polluted western lake zone. However, the correlation completely disappeared in eastern lake zone. It suggested that PCB contamination might be attributed to industrial wastewaters and domestic sewages from western lake basin, reaching the lake through rivers, rains and floods.

Organochlorine Pollutants in Western Antarctic Peninsula Sediments and Benthic Deposit Feeders

Sediments and benthic deposit feeding holothurians were collected near the Palmer Long Term Ecological Research grid during the austral winter of 2008. Polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) were measured in Western Antarctic Peninsula continental shelf sediments, porewater, and benthic biota. Concentrations and fluxes in sediments decreased sharply away from the tip of the peninsula toward its interior. Sedimentary PCB fluxes were an order of magnitude lower than reported elsewhere, supporting the notion of a pristiner Antarctic environment. Hexa-chlorinated biphenyls dominated (40-100%) the PCB profiles in the sediments, while trichlorinated biphenyl 28 was the most abundant PCB congener in the porewater. PCB and OCP concentrations in holothurians were comparable to concentrations in other low trophic level biota in the Antarctic food web (i.e., krill). The partitioning of PCBs and OCPs between the sediments and porewater can be explained by a dual-mode model, which included both organic carbon and black carbon as partitioning media. Alternatively, a simpler one-parameter prediction assuming coal tar-like organic carbon performed equally well in explaining porewater concentrations The majorities of PCBs (63-94%) in the Western Antarctic Peninsula sediments were bound to black carbon or recalcitrant tar-like organic carbon, thereby lowering porewater concentrations. PCBs and OCPs in the holothurians were in equilibrium with those in the porewater.

Simultaneous monitoring of PCB profiles in the urban air of Dalian, China with active and passive samplings

The concentration of polychlorinated biphenyls (PCBs) in the urban air of Dalian, China was monitored from November 2009 to October 2010 with active high-volume sampler and semipermeable membrane device (SPMD) passive sampler. The concentration of PCBs (particle + gas) (ΣPCBs) ranged from 18.6 to 91.0 pg/m3, with an average of 50.9 pg/m3, and the most abundant dioxin-like PCB (DL-PCBs) was PCB118. The WHO-TEQ values of DL-PCBs were 3.6-22.1 fg/m3, with an average of 8.5 fg/m3, and PCB126 was the maximum contributor to ΣTEQ. There was a much larger amount of PCBs in the gas phase than in the particulate phase. The dominant PCB components were lower and middle molecular weight PCBs. With increasing chlorination level, the concentration of the PCB congeners in the air decreased. The gas-particulate partitioning of PCBs was different for the four seasons. The gas-particulate partitioning coefficients (logKp) vs. subcooled liquid vapor pressures (logPL0) of PCBs had reasonable correlations for different sampling sites and seasons. The absorption mechanism contributed more to the gas-particulate partitioning process than adsorption. Correlation analysis of meteorological parameters with the concentration of PCBs was conducted using SPSS packages. The ambient temperature and atmospheric pressure were important factors influencing the concentration of PCBs in the air. The distribution pattern of the congeners of PCBs and the dominant contributors to DL-PCBs and TEQ in active samples and SPMDs passive samples were similar. SPMD mainly sequestrated gas phase PCBs.

Changes in eggshell ultrastructure of Falco naumanni and Tyto alba exposed to pesticides and polychlorinated biphenyls (PCBs)

Changes in the quality of eggs of birds exposed to environmental contaminants have been described, but few reports concern eggshell ultrastructure. In this study, infertile or addled Lesser Kestrels (Falco naumanni) and Barn owls (Tyto alba) eggs were collected from the polluted area of Gela plain (Sicily) during 2007, and compared in terms of organophosphate and organochlorine pesticides, and PCBs levels, and eggshell ultrastructure as determined by scanning electron microscopy. Pesticide and PCB residues in eggs were determined by Gas chromatography/ mass spectrometry (GC/MS) [GC Agilent 7890A/MS Agilent 5975C (Agilent technologies) using a DB-5 capillary column in the selected ion monitoring mode]. The GC/MS analysis revealed that eggs contained measurable amounts of some pesticides and PCBs. There was a low detection of organophosphate pesticides while the most abundant organochlorine residues detected were p,p’ DDT, p,p’ DDE, and Hexachlorobenzene. While, the most abundant PCBs detected congeners were PCB 138, 153, 170, 180, and 187. Although the general structure of the eggshell layers was maintained, the results showed ultrastructural differences in mammillary and palisade eggshell layers between high level and low level contaminated eggs in Tyto alba. Furthermore, mammillary cores of the eggshell had an increased distance between themselves with respect to well organized structures present in uncontaminated egg. In this paper we verify the presence of environmental contaminants in the eggs and document structural changes in bird of prey eggshells. The data could suggest that some contaminants can contribute to reduced reproductive performance (infertile or addled egg) by structural changes in the eggshell. The alteration in morphological disposition of mammillary cores could also suggest an impairment of gas exchange.

수유기간에 따른 모유 중 Indicator PCBs 분석을 통한 영유아의 인체노출평가

The levels of indicator PCBs were determined in 132 human milk samples (Colostrum, 30th days after delivery, 60th days after delivery) collected in 2006 according to World Health Organization’s (WHO) protocol. In addition, concentrations of indicator polychlorinated biphenyls (PCBs) were measured by the isotope dilution method with high resolution gas chromatograph/ high resolution mass detector (HRGC/HRMS), which gave accurate and precise data for estimations of international trend and comparisons. Average daily intakes (ADIs) of all analytes were calculated. The median level of total indicator PCBs was 14 ng/g lipid, which is significantly lower than the level found in individuals from other countries. PCB congeners 138, 153 and 180 accounted for about 65% of the sum of the indicator congeners analyzed. The concentration of the most abundant PCB 153 is positively correlated with the concentration of the sum indicator PCBs (p < 0.001). In this study, concentrations of total indicator PCBs significantly decreased with increasing number of children for a mother, indicating that parity and breast-feeding period an important role in the accumulation and elimination of these compounds in lactating women. The ADI for the median of total indicator PCBs for an infant was evaluated at 0.077 μg/kg·bw/day from birth to 6 months of age. The present study shows that concentrations of PCB 118 primipara breast milk have decreased 25~35 since 1998, and that this trend is continuing.

In Vitro Toxicity Profiling of Ultrapure Non–Dioxin-like Polychlorinated Biphenyl Congeners and Their Relative Toxic Contribution to PCB Mixtures in Humans

The toxic equivalency concept used for the risk assessment of polychlorinated biphenyls (PCBs) is based on the aryl hydrocarbon receptor (AhR)-mediated toxicity of coplanar dioxin-like (DL) PCBs. Most PCBs in the environment, however, are non-dioxin-like (NDL) PCBs that cannot adopt a coplanar structure required for AhR activation. For NDL-PCBs, no generally accepted risk concept is available because their toxicity is insufficiently characterized. Here, we systematically determined in vitro toxicity profiles for 24 PCBs regarding 10 different mechanisms of action. Prior to testing, NDL-PCB standards were purified to remove traces of DL compounds. All NDL-PCBs antagonized androgen receptor activation and inhibited gap junctional intercellular communication (GJIC). Lower chlorinated NDL-PCBs were weak estrogen receptor (ER) agonists, whereas higher chlorinated NDL-PCBs were weak ER antagonists. Several NDL-PCBs inhibited estradiol-sulfotransferase activity and bound to transthyretin (TTR) but with much weaker potencies than reported for hydroxylated PCB metabolites. AhR-mediated expression of uridine-glucuronyl transferase isozyme UGT1A6 was induced by DL-PCBs only. Hierarchical cluster analysis of the toxicity profiles yielded three separate clusters of NDL-PCBs and a fourth cluster of reference DL-PCBs. Due to small differences in relative potency among congeners, the highly abundant indicator PCBs 28, 52, 101, 118, 138, 153, and 180 also contributed most to the antiandrogenic, (anti)estrogenic, antithyroidal, tumor-promoting, and neurotoxic potencies calculated for PCB mixtures reported in human samples, whereas the most potent AhR-activating DL-PCB-126 contributed at maximum 0.2% to any of these calculated potencies. PCB-168 is recommended as an additional indicator congener, given its relatively high abundance and antiandrogenic, TTR-binding, and GJIC-inhibiting potencies.

In this study, breast milk levels of indicator polychlorinated biphenyls (PCBs) were estimated, and statistics drawn, for 22 Koreans in the general population, aged 26-38, who had resided in metropolitan areas for more than 5 years without occupational exposure to organochlorine pollutants. Concentrations of indicator PCBs were measured using the isotope dilution method with a high resolution gas chromatograph/high resolution mass detector, which provided accurate and precise data for investigation of trends, and international comparisons. The geometric mean of total indicator PCB levels was 22.3 ng/g lipid, which is significantly lower than the level in individuals from European countries. Overall, the geometric mean and 95th percentile of the most abundant congener PCB 153 were 8.04 ng/g lipid and 16.4 ng/g lipid, respectively. PCB congeners 138, 153 and 180 together accounted for about 75% of the indicator congeners analyzed. Breast milk total indicator PCB concentrations were significantly associated with age and parity, but not with body mass index (BMI), rate of body weight increase, or smoking habits. The geometric mean level of PCB 153 in breast milk from mothers with a pre-pregnant BMI < <TEX>$21\;kg/m^2$</TEX> was significantly higher than in pre-pregnant women with a BMI of <TEX>${\geqslant}\;21\;kg/m^2$</TEX> (p<0.05). Women who ate more fish, meat, and milk products did not have significantly higher total indicator PCB levels than those who ate less of these products, but a significant difference in PCB 153 levels was demonstrated between the subjects consuming higher (11.45 ng/g lipid) and lower (6.79 ng/g lipid) amounts of fish (p<0.05), after adjusting for confounders. These results suggest that age, parity and fish intake are the important factors affecting the concentrations of indicator PCBs in these subjects.
