Dietary 25-Hydroxycholecalciferol Supplementation from Day 85 of Gestation to Farrowing Enhances Performance, Antioxidant Capacity, and Immunoglobulins of Sows and Newborn Piglets.

In this study, the aim was to evaluate the effects of dietary 25-hydroxycholecalciferol supplementation from day 85 of gestation on performance, antioxidant capacity, and immunoglobulin level of sows and newborn piglets. On day 85 of gestation, forty Landrace × Yorkshire gestating sows (average body weight of 241 ± 6.8 kg; average parity of 3.47 ± 0.6) were allotted into two treatments (20 replicates per treatment) based on parity, body weight, and back fat thickness. From day 85 of gestation to farrowing, sows were fed a normal vitamin D3 diet as control (containing 50 μg/kg vitamin D3; CON), or a 25-hydroxycholecalciferol-supplemented diet (containing 50 μg/kg 25-hydroxycholecalciferol). Compared with CON, dietary 25-hydroxycholecalciferol supplementation increased (p < 0.05) protein and fat content in colostrum and the average birth body weight of newborn piglets. Sows fed 25-hydroxycholecalciferol showed increased (p < 0.05) apparent total tract digestibility of crude protein compared with CON. Diets supplemented with 25-hydroxycholecalciferol also increased (p < 0.05) the content of superoxide dismutase (SOD), and tended to increase (p = 0.06) the total antioxidant capacity content and reduce (p = 0.09) the malondialdehyde (MDA) level in colostrum. An increase (p < 0.05) in the content of SOD and a reduction (p < 0.05) in the content of MDA in the serum of newborn piglets was also observed in the 25-hydroxycholecalciferol treatment compared with CON. Dietary 25-hydroxycholecalciferol supplementation also enhanced (p < 0.05) the immunoglobulin G content and reduced (p < 0.05) the concentration of tumor nuclear factor-α in the serum of sows, as well as reducing (p < 0.05) the content of immunoglobulin G and immunoglobulin A in the serum of newborn piglets compared with CON. Supplementation of 25-hydroxycholecalciferol in sow diets increased (p < 0.05) the content of alkaline phosphatase in the serum and colostrum of sows, the concentration of insulin and crosslap in serum of sows, and the serum calcium content of newborn piglets compared with CON. In conclusion, dietary 25-hydroxycholecalciferol supplementation from day 85 of gestation could enhance performance, antioxidant capacity, and immunoglobulin in sows and newborn piglets.

Effects of phytase and 25-Hydroxycholecalciferol supplementation in broilers fed calcium-phosphorous deficient diets, with or without Eimeria challenge, on growth performance, body composition, bone development, and gut health

The study evaluated the effects of nutritional strategies on broilers challenged with Eimeria from d 14 to 26. A total of 840 Cobb male broilers were fed five diets in a 2 × 5 factorial arrangement: 1) nutrient adequate diet (PC; 0.84% calcium [Ca], 0.42% available phosphorus [avP]); 2) Ca-P deficient diet (NC; 0.64% Ca, 0.22% avP); 3) NC + 1500 FTU/kg phytase of diet (NC + PHY); 4) NC + 5000 IU/kg 25-Hydroxycholecalciferol of diet (NC + 25OHD); and 5) NC with both supplements (NC + PHY + 25OHD), with and without Eimeria challenge. All treatments had six replicate cages with 14 birds per cage. At 5 days post inoculation (DPI), the challenged birds exhibited higher serum fluorescein isothiocyanate-d (FITC-d) levels than the unchallenged birds (P < 0.001). The NC + PHY and NC + PHY + 25OHD groups exhibited lower FITC-d levels compared to the NC + 25OHD group (P = 0.012). Significant interaction effects between Eimeria challenge and dietary treatments were observed on various parameters. During 0 to 6 and 0 to 12 DPI, Eimeria challenge resulted in decreased the body weight gain (BWG) (P < 0.05) but had a negative effect on the feed conversion ratio (FCR) in birds compared to the unchallenged group (P < 0.05). Reducing Ca and avP levels in the diet (NC) did not adversely affect BWG, but negatively impacted FCR, bone ash weight, ash concentration, and femur bone microstructure parameters (P < 0.05). On 12 DPI, Eimeria challenge led to decreased tibia bone weight, bone volume, fat-free bone weight (FFBW), and ash weight of birds (P < 0.05). Supplementation with phytase alone or in combination with 25OHD improved growth performance, gut permeability, bone ash and bone microstructure parameters in birds (P < 0.05). However, the group fed 25OHD alone showed enhancements on growth performance, mineral apposition rate (MAR), bone ash concentration and ash percentage of the birds (P < 0.05). In conclusion, lowering Ca and avP levels in the diet negatively affected FCR and bone development but did not affect intestinal integrity in broilers. Dietary supplementation of phytase, 25OHD, or phytase in combination of 25OHD could enhance the growth performance and bone quality of broilers infected with Eimeria. Notably, the benefits of phytase supplementation were generally more pronounced than those associated with 25OHD supplementation; however, the combination of phytase and 25OHD could induce optimum effects.

172 Impact of maternal and postnatal 25-hydroxycholecalciferol supplementation on porcine and avian satellite cell mitotic activity and skeletal muscle growth characteristics

Abstract Vitamin D is an essential micronutrient most well-known for regulating calcium and phosphorus homeostasis and bone mineralization. However, over the last 20 yr, the extra-skeletal, pleotropic effects of vitamin D and its metabolites have gained greater attention. For livestock species reared indoors, vitamin D supplementation is especially important and commonly achieved by dietary addition of cholecalciferol, also known as vitamin D3 (D3). Cholecalciferol is hydroxylated in the liver to produce the circulating form, 25-hydroxycholecalciferol (25OHD3), also called calcidiol. Circulating 25OHD3 is subsequently converted to 1α,25-hydroxycholecalciferol (1,25OHD3) which binds a nuclear receptor found in many tissues, including skeletal muscle, to elicit its effects. This presentation will focus on the recent work regarding the effects of both maternal and postnatal 25OHD3 supplementation on porcine and avian satellite cell mitotic activity and skeletal muscle growth characteristics. Feeding broilers 25OHD3 posthatch results in a satellite cell-mediated skeletal muscle hypertrophic response in the fast-twitch pectoralis major but not the slow-twitch biceps femoris. Maternal dietary supplementation of gilts and hens with 25OHD3 in place of the majority of D3 increases circulating 25OHD3 concentrations in both dams and their offspring. The improvements in vitamin D status coincide with positive effects on skeletal muscle fiber number in fetal piglets as well as satellite cell mitotic activity and hypertrophic growth of fibers in both swine and broiler chickens. Though, interestingly, combining maternal and posthatch 25OHD3 supplementation in broilers does not appear to produce additive or synergistic effects on SC-mediated muscle growth. However, increasing circulating concentrations of 25OHD3 by supplementing either the maternal or postnatal diet with 25OHD3 has positive effects on meat yield in both porcine and avian models. Overall, these data shed light on the cellular mechanisms behind the efficacy of this vitamin D supplementation strategy using commercially available 25OHD3 sources in increasing meat yield and yet, leaves enough unanswered questions remaining to warrant further exploration.

Dietary 25-Hydroxycholecalciferol Supplementation as a Vitamin D3 Substitute Improves Performance, Egg Quality, Blood Indexes, Jejunal Morphology, and Tibia Quality in Late-Phase Laying Hens.

This study aimed to investigate whether a dietary 25-OHD3 addition improved the performance, egg quality, blood indexes, antioxidant status, jejunal morphology, and tibia quality of aged laying hens compared to a dietary VD3 addition. A total of 270 Hy-Line Brown laying hens at 55 wk of age were randomly assigned to three dietary treatments with six replicates (15 birds per replicate with 3 birds per cage). Chickens were fed a corn-soybean meal diet supplementation of 4000 IU/kg VD3 (control group), 50 μg/kg 25-OHD3 and 2000 IU/kg VD3 (experimental group 1), or 50 μg/kg 25-OHD3 and 4000 IU/kg VD3 (experimental group 2) for 12 weeks. The results demonstrated that 25-OHD3 caused a significant increase in the laying rate, especially in the 50 μg/kg 25-OHD3 + 2000 IU/kg VD3 group; the laying rate reached the maximum compared with other groups after 12 weeks (p < 0.05). However, there were no significant effects on the average egg weight, average daily feed intake, or feed-to-egg ratio (p > 0.05). A dietary supplementation of 50 μg/kg 25-OHD3 and 2000 IU/kg VD3 provided an improved eggshell strength, thick albumen height, and Haugh unit after 12 weeks (p < 0.05). Further analysis of the blood indexes showed that alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, calcium, and phosphorus were enhanced significantly in the 50 μg/kg 25-OHD3 + 2000 IU/kg VD3 group, while the content of total bilirubin decreased significantly (p < 0.05). In addition, the 25-OHD3 addition in diets improved the calcium and phosphorus contents in the serum (p < 0.05). The concentrations of 25-OHD3, parathyroid hormones, follicle-stimulating hormone, and progesterone were increased in the 50 μg/kg 25-OHD3 + 2000 IU/kg VD3 group, and the levels of cortisol, calcitonin, bone gla protein, and endotoxin in the serum reached a minimum in the 50 μg/kg 25-OHD3 + 4000 IU/kg VD3 group (p < 0.05), which constitutes an advantage for the aged laying hens. The antioxidant enzyme activities and free radical scavenging abilities in the 50 μg/kg 25-OHD3 + 2000 IU/kg VD3 group increased markedly, and the MDA level decreased significantly in the 50 μg/kg 25-OHD3 + 4000 IU/kg VD3 group (p < 0.05). Improvements in jejunal morphology and intestinal integrity resulted in an increased villi-length-to-crypt-depth ratio in the 50 μg/kg 25-OHD3 + 2000 IU/kg VD3 group (p < 0.05). Dietary 50 μg/kg 25-OHD3 and 2000 IU/kg VD3 additions improved the tibia quality, including fresh tibia weight, strength, mineral content (Ca), and trabeculae area (p < 0.05). Taken together, compared with the dietary VD3 addition, dietary supplementation of 25-OHD3 supported a stable physiological status for sustained egg production, egg quality, and bone quality in late-phase laying hens, and the addition levels of 50 μg/kg 25-OHD3 and 2000 IU/kg VD3 had the best effect. Therefore, this could provide a theoretical basis for the use of 25-OHD3 as a substitute forVD3.

Effect of 25-hydroxycholecalciferol supplementation with different dietary available phosphorus levels for broilers

ABSTRACT 1. Based on the hypothesis that 25-hydroxycholecalciferol (25-OH-D3) inclusion would optimise dietary mineral digestibility and ameliorate growth performance and bone mineralisation in available phosphorus (AvP) deficient-fed broilers, a trial was conducted to evaluate its effect on diets with different levels of AvP. 2. Broilers aged 1–21 d were randomly assigned one of the eight treatments, consisting of four dietary levels of AvP (0.45%, 0.42%, 0.39%, and 0.36%) and with or without supplementation with 25-OH-D3 at 69 μg/kg of feed. All diets contained 100 µg/kg of vitamin D3 (cholecalciferol). 3. The addition of 25-OH-D3 resulted in higher feed intake and body weight gain, and lower FCR (P < 0.05) compared to non-supplemented diets, whereas AvP levels had a quadratic effect only on feed intake. There were no interactions between treatment factors. 4. Increasing AvP levels linearly reduced the ileal digestibility of Ca and P (P < 0.01) and supplementing 25-OH-D3 increased both Ca and P ileal digestibility (P < 0.05), without any interactions observed for ileal digestibility. 5. There was an interaction, whereby 25-OH-D3 inclusion increased serum metabolites in broilers fed 0.36% to 0.42% AvP compared to the non-supplemented diets (P < 0.001), whereas, at 0.45% AvP, diets with or without 25-OH-D3 had similar results. 6. The P content in bone linearly increased in line with AvP levels (P < 0.05) and supplementation of 25-OH-D3 increased ash bone content (P < 0.001). 7. Broilers can benefit from 25-OH-D3 supplementation combined with cholecalciferol with regard to Ca and P utilisation and vitamin D status, allowing for a reduction of dietary AvP levels down to 0.36% without impairing growth performance or bone status.

Dietary cholecalciferol and 25-hydroxycholecalciferol supplementation interact to modulate reproductive performance, egg quality, serum antioxidant capacity, intestinal morphology and tibia quality of breeder geese

ABSTRACT 1. A study was conducted to evaluate the effects of dietary cholecalciferol (vitamin D3) and 25-hydroxycholecalciferol (25-OH-D3) supplementation on the reproductive performance, egg quality, eggshell ultrastructure, serum hormone level and antioxidant capacity, intestinal morphology and tibia quality of breeder geese during the laying period. 2. The trial was designed as a 3 × 3 factorial arrangement with three levels (300, 400 and 500 IU/kg) of vitamin D3 supplementation and three levels (25, 50 and 75 μg/kg) of 25-OH-D3 supplementation in a 10-wk feeding trial. 3. The results showed that the combined supplementation of 400 IU/kg vitamin D3 and 50 μg/kg 25-OH-D3 had a better feed conversion ratio and a higher egg laying rate than the other groups. Vitamin D3 supplementation significantly increased the rate of qualified eggs for hatching, eggshell strength and thickness, serum testosterone and progesterone levels, serum total superoxide dismutase and glutathione peroxidase activities, tibia ash content and bone mineral density (P < 0.05). Dietary 25-OH-D3 supplementation significantly increased serum glutathione peroxidase activity and duodenal villus height and villus height-to-crypt-depth ratio (P < 0.05). The geese receiving 500 IU/kg vitamin D3 and 75 µg/kg 25-OH-D3 had the highest tibia calcium and phosphorous content among all groups (P < 0.05). 4. Feeding 400 IU/kg vitamin D3 plus 50 µg/kg 25-OH-D3 gave optimal effects on feed conversion ratio and egg laying rate. This combination could be a nutritional strategy for increasing the laying rate, eggshell quality, serum hormone levels and serum antioxidant function regardless of 25-OH-D3 supplementation. Supplementation of 50 μg/kg 25-OH-D3 could be a recommended dose for improving the serum antioxidant capacity and intestinal morphology regardless of vitamin D3 supplementation.

Effect of combined maternal and post-hatch dietary 25-hydroxycholecalciferol supplementation on broiler chicken Pectoralis major muscle growth characteristics and satellite cell mitotic activity.

Skeletal muscle growth is largely dependent on the proliferation and differentiation of muscle-specific stem cells known as satellite cells (SC). Previous work has shown that dietary inclusion of the vitamin D3 metabolite, 25-hydroxycholecalciferol (25OHD3), also called calcidiol, can promote skeletal muscle growth in post-hatch broiler chickens. Improving vitamin D status of broiler breeder hens by feeding 25OHD3 in addition to vitamin D3 has also been shown to positively impact progeny. Yet, whether combined pre- and post-hatch supplementation with 25OHD3 produces an additive or synergistic SC-mediated, skeletal muscle growth response remains unanswered. To evaluate the effect of combined maternal and post-hatch dietary 25OHD3 supplementation on the growth and SC mitotic activity of the Pectoralis major (PM) muscles in broiler chickens, a randomized complete block design experiment with the main effects of maternal diet (MDIET) and post-hatch diet (PDIET) arranged in a 2 × 2 factorial treatment structure was conducted. From 25 to 36 wk of age, broiler breeder hens were fed 1 of 2 MDIET formulated to provide 5,000 IU D3 (MCTL) or 2,240 IU of D3 + 2,760 IU of 25OHD3 per kg of feed (M25OHD3). Their male broiler chick offspring (n = 400) hatched from eggs collected from 35 to 36 wk of age were reared in raised floor pens. Broilers were fed 1 of 2 PDIET formulated to provide 5,000 IU of D3 per kg of feed (PCTL) or 2,240 IU of D3 + 2,760 IU of 25OHD3 per kg of feed (P25OHD3). Muscle was collected at days 4, 8, 15, 22, and 29 and stored until immunofluorescence analysis. Data were analyzed as a 2-way ANOVA with SAS GLIMMIX. Dietary 25OHD3 was effectively transferred from hen plasma to egg yolks (P = 0.002) and to broiler progeny plasma (days 4 to 22; P ≤ 0.044). Including 25OHD3 in either MDIET or PDIET altered PM hypertrophic growth prior to day 29 (P ≥ 0.001) and tended to reduce Wooden Breast severity (P ≤ 0.089). Mitotic SC populations were increased in PM of MCTL:P25OHD3 and M25OHD:PCTL-fed broilers at d 4 (P = 0.037). At d 8, the PM mitotic SC populations were increased 33% by P25OHD3 (P = 0.054). The results of this study reveal that combined maternal and post-hatch 25OHD3 supplementation does not produce additive or synergistic effects on SC-mediated broiler muscle growth. However, vitamin D status improvement through dietary 25OHD3 inclusion in either the maternal or post-hatch diet stimulated broiler breast muscle growth by increasing proliferating SC populations.

Combining Maternal and Post-Hatch Dietary 25-Hydroxycholecalciferol Supplementation on Broiler Chicken Growth Performance and Carcass Characteristics

Dietary inclusion of the vitamin D3 (D3) metabolite, 25-hydroxycholecalciferol (25OHD3), was demonstrated to improve broiler growth performance and breast meat yield. To assess the effect of combined maternal (MDIET) and post-hatch (PDIET) dietary 25OHD3 inclusion on broiler growth performance and carcass characteristics, a randomized complete block design experiment with a 2 × 2 factorial treatment structure was conducted. From 25 to 38 weeks of age, broiler breeder hens were provided with 1 of 2 MDIET formulated to contain: 5000 IU D3 (MCTL), or 2240 IU of D3 + 2760 IU of 25OHD3 per kg of feed (M25OHD3). Their chick offspring (n = 448; 224 per MDIET) hatched from eggs collected from 37 to 38 weeks of age were reared in 16 replicate pens with 7 birds per pen and fed 1 of 2 PDIET in 3 phases up to day 40 formulated to contain: 5000 IU of D3 per kg of feed (PCTL), or 2240 IU of D3 + 2760 IU of 25OHD3 per kg of feed (P25OHD3). No additive or synergistic effects of combining 25OHD3 inclusion in MDIET and PDIET were observed. Broilers from 25OHD3-fed hens (M25OHD3) were heavier on day 40 than those from hens fed only D3 (MCTL; 2.911 vs. 2.834 kg; p = 0.040). Tender weight (123 vs. 117 g) and yield (5.63 vs. 5.44%) were greater in the M25OHD3 broilers than the MCTL broilers (p = 0.006). Broilers fed 25OHD3 (P25OHD3) tended to have heavier breasts (637 vs. 615 g; p = 0.050), bone-in wings (215 vs. 210 g; p = 0.070), and boneless thighs (279 vs. 270 g; p = 0.078) compared with those fed only D3 (PCTL). Neither MDIET nor PDIET altered the severity of Wooden Breast and White Striping (p ≥ 0.106). Overall, including 25OHD3 in either the maternal or broiler diet increased broiler meat yield.

Effects of 25-hydroxycholecalciferol supplementation on breast meat quality and histomorphometric characteristics in broiler chickens

This experiment was carried out to elucidate the influences of 25-hydroxycholecalciferol status in the diet and gender of the chicken on the sensory and histomorphometric characteristics in breast muscle (Pectoralis major) of broilers. The experiment was a randomized design consisting 2×2 factorial arrangement with 25-hydroxycholecalciferol (3000 and 5000 IU kg/diet) and two gender. Breast weight was not affected by dietary 25-hydroxycholecalciferolstatus and gender of the bird. Cooking loss decreased in females but not in male chickens with additionally 25-Hydroxycholecalciferol supplementation. However, increasing 25-hydroxycholecalciferol level in the diet reduced drip loss in male chickens but no change was observed in females ones. A significant increase in breast meat pH24 was observed with increasing dietary level of 25-Hydroxycholecalciferol; however, squeezable water ratio, lightness (L*), redness (a*) and yellowness (b*) values were comparable in the groups. The histomorphometric characteristics of chicken breast meat throughout the growth period, except for the cross sectional area and number of fibers, were interactively influenced by 25-hydroxycholecalciferol and gender of the bird. In conclusion, an improvement of 25-hydroxycholecalciferol status could be a practical tool to increase chicken breast meat quality with beneficial effects on water holding capacity.

Organic zinc and manganese and 25-hydroxycholecalciferol improves eggshell thickness in late-phase laying hens.

The aim of this study was to evaluate the effects of organic micromineral zinc (Zn) and manganese (Mn) and 25-hydroxycholecalciferol supplementation in late-phase laying hens' diets on performance, egg quality, lipid stability of fresh and stored eggs, and bone quality. The treatments were a basal diet and diets supplemented with 32mg Zn-Met/kg diet; 26mg Mn-Met/kg diet; 32mg Zn-Met/kg diet and 26mg Mn-Met/kg diet; 1500IU 25-hydroxycholecalciferol/kg diet; 32mg Zn-Met/kg diet; 26mg Mn-Met/kg diet; and 1500IU 25-hydroxycholecalciferol/kg diet. On performance, the birds supplemented with organic manganese had the lowest feed intake. Regarding egg quality, the birds supplemented with Zn-Met and Mn-Met, with 25-hydroxycholecalciferol alone, and with Zn-Met, Mn-Met and 25-hydroxycholecalciferol presented a greater eggshell thickness than those receiving the basal diet. Lipid stability of the yolk varied only according to storage time. No effect of supplementation was observed on bone quality. Supplementation with Zn-Met and Mn-Met, or associated with 25-hydroxycholecalciferol, or 25-hydroxycholecalciferol alone, improved eggshell thickness in aged white layers. However, the associated or isolated supplementation with these nutrients did not influence performance, lipid stability of fresh and stored egg yolk or bone quality.

PSVII-6 Effects of 25-hydroxycholecalciferol supplementation on gene expression of feedlot cattle

Abstract The objective was to evaluate the effects of 25-hydroxycholecalciferol (HyD®, DSM Produtos Nutricionais Brasil S.A.) supplementation in expression of genes related to anabolism and catabolism of feedlot cattle. A total of 120 Nellore bulls (IBW = 370±20 kg) were distributed in 24 fully roofed-pens with concrete floor (5 animals/pen) in a randomized complete block design (8 pens/treatment). The treatments were T1 (control): no supplementation of HyD®; T2: HyD® supplementation at 1 mg/animal/day; T3: HyD® supplementation at 3 mg/animal/day. Basal diet was formulated to meet requirements of finishing bulls, considering an ADG of 1.6 kg/day (Level 2 Nutrition System, Fox et al., 2004). Adaptation to diets followed a step-up scheme for 14 days, with concentrate inclusion being gradually increased from 76 to 91% DM. Finishing diets were offered from 15th to 100th day of feedlot period and animals fed twice a day (0800 and 1500), with bunks managed for a maximum of 5% orts. After slaughter, muscle samples were collected for quantitative evaluation of gene expression using RT-qPCR method, considering the following genes: SOD1 (antioxidant marker), IGF1, IGF2 and MTOR (anabolism), FOXO1, MURF1, Atrogin-1 and MSTN (catabolism). Statistical analysis was performed using PROC MIXED of SAS® and means compared by Tukey test at 5% probability. No significant differences among treatments were observed for SOD1, FOXO1, MURF1 and Atrogin-1 expression (P &amp;gt;0.05). However, tendencies (0.05 &amp;gt; P ≤0.10) could be observed for IGF1, IGF2, MTOR and MSTN expression in animals receiving HyD®, regardless of dosage. These results indicate a positive effect of HyD® on muscular anabolism and protein synthesis on feedlot finishing cattle. Moreover, the greater expression on MSTN suggest a higher protein turnover and muscular growth regulation. In conclusion, HyD® supplementation increased expression of genes correlated to muscular growth and protein synthesis, being a viable technology for beef cattle finished in feedlot systems.

Effects of normal and low calcium and phosphorus levels and 25-hydroxycholecalciferol supplementation on performance, serum antioxidant status, meat quality, and bone properties of broilers.

To determine the effects of normal and low dietary calcium (Ca) and phosphorus (P) levels and 25-hydroxycholecalciferol (25-OH-D3) supplementation on performance, serum antioxidant status, meat quality, and bone properties of broilers, 224 1-day-old Arbor Acre male broilers were used in this study. Broilers were allotted randomly to 1 of 4 treatments in a 2 × 2 factorial arrangement that included normal or low Ca and P diet with or without 69 μg/kg 25-OH-D3. The trial consists of a starter phase from day 1 to 21 and a grower phase from day 22 to 42. Dietary 25-OH-D3 supplementation increased (P < 0.05) average daily weight gain from day 22 to 42 and decreased feed conversation ratio from day 22 to 42 and day 0 to 42. On day 21, 25-OH-D3 increased serum concentrations of total antioxidant capacity (T-AOC), catalase (CAT), and glutathione peroxidase in broilers fed low Ca and P diet (Interaction, P < 0.05). 25-hydroxycholecalciferol significantly decreased serum malondialdehyde concentration. Dietary Ca and P deficiencies significantly decreased serum Ca and P concentrations and increased serum parathyroid hormone (PTH) concentration, and serum Ca and 25-OH-D3 concentrations were significantly increased by 25-OH-D3 supplementation. On day 42, serum T-AOC and CAT concentrations were decreased by dietary Ca and P deficiencies without 25-OH-D3 (Interaction, P < 0.05) and unaffected by dietary Ca and P deficiencies with 25-OH-D3. Dietary Ca and P deficiencies significantly decreased Ca, P, and alkaline phosphatase concentrations and increased PTH concentration in serum. Dietary 25-OH-D3 increased (P < 0.05) serum Ca and 25-OH-D3 concentrations and decreased (P < 0.05) serum tartrate-resistant acid phosphatase concentration. The interaction between CaP level and 25-OH-D3 was observed (P < 0.05) for tibial Ca content and femoral bone density. 25-hydroxycholecalciferol significantly increased tibial breaking strength. These data indicated that 25-OH-D3 supplementation at 69 μg/kg increased growth performance in some periods, enhanced serum antioxidant capacity, and improved bone mineralization and breaking strength of broilers.

Effect of dietary 25-hydroxycholecalciferol supplementation and high stocking density on performance, egg quality, and tibia quality in laying hens

This study was conducted to determine the effects of 25-hydroxycholecalciferol (25-OH-D3) on performance, egg quality, tibia quality, and serum hormones concentration in laying hens reared under high stocking density. A total of 800 45-week-old Lohmann laying hens were randomly allotted into a 2 × 2 factorial design with 2 levels of dietary 25-OH-D3 levels (0 and 69 μg/kg) and 2 rates of stocking densities [506 (low density) and 338 (high density) cm2/hen]. Laying hens were monitored for 16 wk. High stocking density decreased laying rate, egg weight, and feed intake compared with low stocking density (P < 0.01) during 1 to 8 wk and 1 to 16 wk. Overall, high stocking density increased eggshell lightness value and decreased shell redness and yellowness value, strength, thickness, and relative weight compared with low stocking density (P < 0.05). Dietary supplementation with 25-OH-D3 reduced the value of the eggshell lightness and increased its yellowness and eggshells weight (P ≤ 0.05). The increase in eggshell thickness was more pronounced when 25-OH-D3 was supplemented to layers under high stocking density (interaction, P < 0.05). Layers under high stocking density had lower ash content and calcium content in the tibia than layers under low stocking density (P = 0.04); dietary 25-OH-D3 increased tibia strength compared with no addition (P = 0.05). Layers under high stocking density had higher serum concentrations of 25-OH-D3, corticosterone (CORT), lipopolysaccharide (LPS), and osteocalcin (OC; P < 0.05), lower content of parathyroid hormone (PTH) compared with layers under low stocking density (P < 0.01). Dietary 25-OH-D3 increased serum concentration of 25-OH-D3, carbonic anhydrase (CA), and calcitonin (CT) (P < 0.01) and reduced corticosterone, lipopolysaccharide and osteocalcin concentration (P ≤ 0.05). The increase effect in PTH was more pronounced when 25-OH-D3 was supplemented to layers under high stocking density (interaction, P = 0.05). Overall, the results gathered in this study indicate that high stocking density result in reducing production performance, shell color and quality, and tibia health, whereas dietary 25-OH-D3 was able to maintain tibia health and to mitigate the negative impact of high stocking density on productive performance.

Effect of 25-hydroxycholecalciferol supplementation on turkey performance and immune cell parameters in a coccidial infection model

This study was conducted to identify the effects of 25-OH cholecalciferol supplementation to turkeys on the immune cells parameters, fecal coccidial oocyst shedding, macrophage nitric oxide production, T regulatory cell cytokine production, and production parameters during a coccidial challenge. A total of 200 1-day-old turkey poults were supplemented with 27.5, 55, 82.5, or 110 μg/kg 25-OH cholecalciferol and challenged or not challenged with coccidial oocysts in a 4 × 2 factorial set up of treatments. Birds fed 110 μg/kg of 25-OH cholecalciferol and infected with coccidial oocyst had 41% lower (P < 0.05) fecal oocyst and 53% higher (P < 0.05) macrophage nitric oxide production than the birds fed 27.5 μg/kg of 25-OH cholecalciferol and infected with coccidial oocyst at 5 d post-coccidial infection. Birds fed 82.5 μg/kg 25-OH cholecalciferol and infected with coccidial oocyst had 5-fold higher (P < 0.05) IL-1 mRNA amounts than the birds fed 27.5 μg/kg of 25-OH cholecalciferol and infected with coccidial oocyst. Birds fed 110 μg/kg 25-OH cholecalciferol and infected with coccidial oocyst had 5.3-fold higher (P < 0.05) IL-10 mRNA amounts than the birds fed 27.5 μg/kg of 25-OH cholecalciferol and infected with coccidial oocyst at 5 d post-coccidial infection. CD4+CD25+ cells from birds fed 110 μg of 25-OH cholecalciferol and infected with coccidial oocyst had 12-fold higher (P < 0.05) IL-10 mRNA than that from the birds fed 27.5 μg/kg of 25-OH cholecalciferol and infected with coccidial oocyst. In conclusion, supplementing birds with 101 μg/kg 25-OH cholicalciferol decreases coccidial oocyst shedding in the feces and could be a nutritional strategy to reduce the coccidial infection and spread in turkeys.

Cholecalciferol or 25-Hydroxycholecalciferol Supplementation Does Not Affect Muscle Strength and Physical Performance in Prefrail and Frail Older Adults

Vitamin D supplementation is proposed as a potential treatment strategy to counteract functional decline in older adults. However, data from randomized trials are either limited or inconsistent. This study investigated the effect of daily supplementation with 25-hydroxycholecalciferol [25(OH)D3] or cholecalciferol (vitamin D3) on muscle strength and physical performance in older adults. This was a randomized, double-blind, placebo-controlled trial of 6 mo including 78 prefrail or frail (according to the Fried criteria), community-dwelling older adults (n=43 men) aged ≥65 y, with a baseline 25-hydroxyvitamin D [25(OH)D] concentration between 20 and 50 nmol/L. Participants were supplemented daily with 10 µg 25(OH)D3, 20 µg vitamin D3, or a placebo capsule. Serum 25(OH)D was measured by liquid chromatography-tandem mass spectrometry. The primary outcome was maximal isometric knee-extension strength (Biodex System 4); secondary outcomes included knee-flexion and hand grip strength, Short-Physical Performance Battery score, Timed Up and Go score, postural sway, muscle mass (dual-energy X-ray absorptiometry), and muscle fiber type and size. The mean baseline serum 25(OH)D concentration was 37.7 nmol/L (95% CI: 35.4, 39.9 nmol/L). After 6 mo of supplementation, concentrations increased to 98.7 nmol/L (95% CI: 93.1, 104.4 nmol/L) in the 25(OH)D3 group and to 72.0 nmol/L (95% CI: 66.1, 77.8 nmol/L) in the vitamin D3 group, compared with 47.5 nmol/L (95% CI: 41.8, 53.3 nmol/L) in the placebo group (P-interaction < 0.01). Knee-extension strength did not significantly change in the 25(OH)D3 group (5.9 Nm; 95% CI: -6.2, 18.0 Nm), in the vitamin D3 group (5.5 Nm; 95% CI: -6.8, 17.8 Nm), or in the placebo group (1.8 Nm; 95% CI: -10.7, 14.4 Nm) (P-interaction=0.74). Furthermore, mean changes in physical performance tests, muscle mass, and muscle fiber type and size did not differ between the groups. Increasing the serum 25(OH)D concentration over a period of 6 mo did not significantly change muscle strength and physical performance in prefrail and frail older adults. Effects of maternal dietary canthaxanthin and 25-hydroxycholecalciferol supplementation on antioxidant status and calcium-phosphate metabolism of progeny ducks

Three experiments were conducted to investigate the effects of maternal dietary canthaxanthin (CX) and 25-hydroxycholecalciferol (25-OH-D3) supplementation on antioxidant status and calcium-phosphate metabolism of progeny ducks. Cherry Valley duck breeders (38 wk old) were fed either a control diet or the same diet plus CX (6mg/kg) and 25-OH-D3 (0.069mg/kg) for 32 weeks. Experiments 1, 2, and 3 were conducted with progeny ducks hatched from eggs laid by duck breeder hens at 54, 62, and 70 wk of age, respectively. Progeny ducks from both maternal treatments were fed with the same NRC (1994) vitamin regimen starter (1 to 14 d) and finisher (15 to 35 d) diets in experiments 1 and 2, and fed with the same high vitamin regimen starter (1 to 14 d) and finisher (15 to 35 d) diets in experiments 3. High vitamin regimen had higher levels of all vitamins, except biotin, than the NRC (1994) vitamin regimen. In experiment 1, maternal CX and 25-OH-D3 increased (P < 0.05) shank pigmentation and tibiotarsus ash and tended to decrease (P < 0.1) liver total superoxide dismutase activity (T-SOD) of one-day-old progeny ducks; and increased (P < 0.05) shank pigmentation, decreased (P = 0.05) liver protein carbonyl, and tended to increase (P < 0.1) liver total antioxidant capacity (T-AOC) of 14-day-old progeny ducks. In experiment 2, maternal CX and 25-OH-D3 increased (P < 0.05) shank pigmentation and liver T-AOC and decreased (P < 0.05) liver protein carbonyl of one-day-old progeny ducks, but increased (P < 0.05) the serum phosphate level of 14-day-old progeny ducks. In experiment 3, maternal CX and 25-OH-D3 increased (P < 0.05) shank pigmentation of one-, 14-, and 35-day-old progeny ducks and tended to increase (P < 0.1) liver T-SOD and tibiotarsus ash, but decrease (P < 0.1) liver malondialdehyde of one-day-old progeny ducks. It can be concluded that progeny dietary high vitamin regimen could partially prevent maternal CX-derived progeny shank pigmentation from bleaching. Maternal CX- and 25-OH-D3-derived effects are influenced by the hen's age and progeny's dietary vitamin regimen.

Effects of enhancing vitamin D status by 25-hydroxycholecalciferol supplementation, alone or in combination with calcium and phosphorus, on sternum mineralisation and breast meat quality in broilers

ABSTRACT1. The aim of the present study was to examine the effects of improving vitamin D status in broiler diets by supplementary 25-hydroxycholecalciferol (25OHD3), alone or in combination with calcium (Ca) and available phosphorus (aP), on live performance, sternum mineralisation and breast meat quality in broilers.2. A total of 936 1-d-old Ross 308 broilers were used in the study. After gender determination at the hatchery, chicks from each sex were randomly distributed into three dietary treatments. The following dietary treatments were used in the experiment from hatch to 38 d: (1) A control diet formulated to meet all of the nutrient requirements of broiler chicks according to the management guide; (2) The control diet supplemented with 18.7–15.0 µg/kg of 25OHD3; and (3) The control diet supplemented with 18.7–15.0 µg/kg of 25OHD3 plus Ca + aP.3. Improvement in vitamin D status by 25OHD3 supplementation, alone or in combination with Ca and aP, had no effect on body weight and feed conversion ratio of broilers.4. The serum 25OHD3 concentration significantly increased with 25OHD3 and 25OHD3 plus Ca + aP supplementation (P < 0.05), whereas the ionised Ca and Mg concentrations remained unchanged.5. Sternum absolute weight, ash content and the concentrations of Ca and P significantly increased (P < 0.01) with supplementation of 25OHD3, alone or in combination with Ca + aP.6. Supplemental 25OHD3, alone or in combination with Ca + aP, slightly increased pH24 (P = 0.05) and decreased (P < 0.01) squeezable water loss in breast meat, whereas it had no significant effect on lightness, yellowness and sarcoplasmic protein solubility.7. In conclusion, the results suggested that enhancing vitamin D status by 25OHD3 supplementation alone or in combination with Ca + aP may improve sternum structure and mineral accretion. Furthermore, supplemental 25OHD3, even in a nutritionally complete diet, may offer an effective way to improve protein solubility in female broilers.

Effects of dietary canthaxanthin and 25-hydroxycholecalciferol supplementation on the antioxidant status and tibia quality of duck breeders and newly hatched ducklings

This study evaluated the effects of dietary canthaxanthin (CX) and 25-hydroxycholecalciferol (25-OH-D3) supplementation on the antioxidant status and tibia quality of duck breeders and newly hatched ducklings. In total, 780 female and 156 male duck breeders were randomly allotted to 2 treatments. Duck breeders were fed either a commercial diet (containing 3,000 IU/kg vitamin D3) or the same diet plus a mixture of CX (6 mg/kg) and 25-OH-D3 (0.069 mg/kg) for 40 wk. The antioxidant status of duck breeders, egg yolk, and ducklings; tibia quality of duck breeders and ducklings; and shell quality of breeder eggs were investigated. The total antioxidant capacity of breeder female liver (P = 0.028), breeder male testis (P = 0.049), egg yolk (P = 0.032), one-day-old duckling liver (P = 0.024), and one-day-old duckling yolk sac (P = 0.012) were increased by dietary supplementation of the mixture of CX and 25-OH-D3 The inclusion of CX and 25-OH-D3 decreased liver protein carbonyl of breeder females (P = 0.030), and liver malonaldehyde (P = 0.050) and protein carbonyl (P = 0.030) of breeder males. Yolk (P < 0.001), shank (P < 0.001), and yolk sac pigmentation (P < 0.001) of one-day-old ducklings were increased by the supplementation of the CX and 25-OH-D3 mixture. No differences (P > 0.05) were observed in tibia quality or eggshell quality between treatments. In conclusion, the inclusion of the mixture of CX and 25-OH-D3 in a diet sufficient in vitamin D3 increased antioxidant status but not tibia quality of duck breeders and newly hatched ducklings.
