Abstract
Eukaryotic cells finely control their size and, typically, nuclear volume scales with cell volume. The corresponding nuclear to cell volume ratio (N/C ratio) is narrowly distributed for a given cell-type and deviation from this distribution is a well-known hallmark of senescence or cancer. However, mainly for the lack of methods to simultaneously measure both cellular and nuclear volumes, the mechanisms underlying their size regulation and coupling remain unclear.To overcome this limitation, we developed an imaging method for measuring in real time both cytoplasmic and nuclear volumes of living cells.
