Abstract

Surface structure of yeast protoplasts with a transmission electron microscopy using plasma polymerization replica method has been observed by Hirano and Tanaka in 1979. This is the newest replica method for transmission electron microscopy devised by Tanaka in 1978. The present authers further developed the metal extraction replica method. This method is applicable forimmunoelectron microscopy using ferritin or gold labelled with Concana-valin A. The localization of metal particles, such as ferritin or gold, was observed at the surface of protoplasts and cell wall remnants respectively. Horisberger et al. have already described the use of colloidal gold labelled with to localize mannan in the cell wall of Candida utilis by transmission electron microscopy and at the surface of yeast cells by scanning electron microscopy in 1977. We have observed gold particles more clearly at the surface of protoplasts and cell wall remnants by extraction replica method.
