Abstract

The first total synthesis of the antimalarial natural product chaetoxanthone B is described. The synthesis was completed in five steps from commercially available materials in 21% overall yield. A Heck coupling reaction provided the key aryl alkene intermediate that yielded chaetoxanthone B upon acid catalyzed intramolecular ketalization to form the dioxane/tetrahydropyran ring system.
