Abstract

Thin sections of rapidly frozen and freeze-substituted rabbit glycerinated muscle fibres loaded with myosin subfragment-1 were used to examine a three-dimensional arrangement of thin filaments in vertebrate skeletal muscle. Clearer images of the “arrowhead” structure were obtained when specimens were freeze-substituted first in a tannic acid solution and then in an OsO 4 solution. The images obtained showed that the arrowheads were aligned laterally. This indicates that all the thin filaments have the same rotational orientation in a half sarcomere of rabbit skeletal muscle in the rigor state.
