Abstract
The effect of 8-azaguanine (AG) on the appearance of AG-resistant (AG r ) mutants was investigated by selecting mutants at two concentrations of AG, namely 1.2 μg/ml and 5 μg/ml. If AG r cells arise independently of AG the number of mutants with no residual enzyme activity should be the same at the two concentrations, whereas a higher mutant frequency at 1.2 μg AG/ml should be due to a larger number of mutants with residual activity. The mutant frequencies at the two concentrations were found to be the same, while the number of mutants with no residual enzyme activity was higher at 5 μg/ml. Moreover, for some of the mutants the degree of residual enzyme activity in mutant clones was dependent on the presence or absence of AG in the medium. The results, therefore, do not coincide with an exclusive selective action of AG.
