Abstract
This study aimed to compare the effectiveness of Tris-Egg Yolk extender (TEY I) with one step of mixing and and two step of mixing (TEY II) on the quality of dog sperm frozen semen. Semen was collected from three dogs by manual massage and each semen fraction was evaluated. Rich sperm fraction was diluted with TEY I and TEY II, equilibrated at 5°C for three hours, frozen with liquid nitrogen vapour for 10 minutes, and stored in a liquid nitrogen container for 24 hours. Sperm motility and viability after dilution, equilibration, and thawing were evaluated microscopically. The results showed that no differences (P>0.05) on sperm motility after thawed in TEY II (one step) and TEY I with the sperm motility 49.44±3.38 and 46.67±3.84% respectively, but the advantage of TEY II is more easier to prepare. Sperm viability after being thawed in both extenders were 88.96±1.55% (TEY I) and 86.45±2.84% (TEY II). It was concluded that TEY II and TEY I have the same ability to maintain the quality of sperm during the freezing process for dog semen. One step mixing of TEY extender can be used for semen freezing in dogs.
