Abstract

1. 1. The presence of Na +ATPase activity was demonstrated in a rat brain microsomal fraction. 2. 2. In conventionally prepared microsomes Na +ATPase activity was about 10% of that of the (Na + - K +) stimulated ATPase, and 70% of this Na +ATPase activity was insensitive to 5 × 10 −4M ouabain. 3. 3. Treatment of microsomes with 0.1% DOC increased Na + ATPase activity 3-fold. Higher concentrations of DOC were inhibitory to all ATPase activities. Lubrol treatment removed all Na + ATPase activity from microsomes. 4. 4. Microsomes prepared using Nal possessed very little Na + ATPase activity which was all ouabainsensitive. 5. 5. In conventionally prepared microsomes the ouabain-insensitive Na + ATPase was thermolabile, both at 37°C and in cold storage. 6. 6. The possible cellular origin and role of the Na + ATPase is discussed.
