Abstract

Simian pituitary growth hormone was prepared in homogeneous form by sequential application of exclusion chromatography on Sephadex G-100, ion exchange chromatography on diethylaminoethyl Sephadex with a single eluent, and density gradient column electrophoresis. DEAE-Sephadex chromatography with a single eluent was also used to separate two biologically active components of human pituitary growth hormone. All three primate growth hormone preparations possessed similar activity in the tibia test. The homogeneity of these preparations was shown by disc electrophoresis on polyacrylamide gel under both acidic and alkaline conditions and also by analytical exclusion chromatography on Sephadex G-100. The latter technique also indicated a close similarity in molecular size of all three hormonal preparations.
