Abstract

Objective Study of rats with subarachnoid hemorrhage (SAH) after delayed cerebral vasospasm (DCVS) and the mechanism of neural function defect. Methods DCVS after SAH in rats model was established. Hematoxylin and eosin (HE) dyed then measuring inner diameter and vessel wall thickness of the middle cerebral artery; testing each rat cerebral hypoxia inducible factor-1α (HIF-1α) and the change of vascular endothelial growth factor (VEGF) after Immunohistochemical staining; and the concentration of nitric oxide (NO) and endothelin-1 (ET-1) in the arterial plasma and the cerebrospinal fluid was measured by the use of nitrate reductase and enzyme-linked immunosorption. Results The artery diameter of the control group and the experimental group A [(218.4±27.8), (212.7±29.6) μm] was significantly higher than the experimental group B [(133.9±17.5) μm] and experimental group C [(163.7±14.8) μm]; MAD detection of the HIF-1 alpha and the VEGF of the control group and the experimental group A in the cerebral cortex (0.000 91±0.000 53, 0.001 08±0.000 37 and 0.002 58±0.000 63, 0.002 67±0.000 59) were significantly lower than the experimental group B and the experimental group C (0.014 28±0.000 42, 0.001 39±0.000 44 and 0.027 96±0.000 98, 0.003 79±0.000 72); The value of ET-1 of the control group and the experimental group A in plasma and cerebrospinal fluid [(26.47±4.82), (28.81±5.16) and (64.32±9.47), (67.86±10.24) ng/L] were significantly lower than that of experimental group B and the experimental group C [(63.44±6.73), (41.35±6.06) and (108.62±8.26), (89.46±8.15) ng/L]; The value of NO of the experimental group A and the control group in plasma and cerebrospinal fluid [(49.28±1.69), (48.96±1.60) and (29.65±1.20), (29.05±1.17) μmo/L] were significantly higher than that of experimental group B and the experimental group C [(24.57±1.73), (33.42±1.59) and (15.73±1.06), (20.83±1.16) μmo/L]. Conclusion Microcirculation disorder, the value of HIF-1 alpha and VEGF in the cerebral cortex increased, the level of NO increased and the level of ET-1 decreased after SAH in rats, which may be related to the incidence of DCVS and neural function defect.
