Abstract
The cytokinins benzyladenine (BA), N 6-( Δ 2-isopentenyl)-adenine (2iP) and kinetin were incorporated into a low-salt medium and tested for effects on stimulation of shoot proliferation in vitro using different explant sources of gardenia ( Gardenia jasminoides Ellis). BA was superior to 2iP and kinetin for promotion of shoot formation from shoot tip and in vitro-derived shoot explants. Longer shoots were produced in cultures grown with 2iP than with BA or kinetin. In re-cultures of basal mass explants, the 3 cytokinins had the same effect on shoot proliferation. Addition of gibberellic acid (GA 3) to the medium in conjunction with 25 μM 2iP increased the number, length and quality rating of shoots produced. Microcuttings harvested from cultures grown in medium with 2iP or 2iP plus GA 3 rooted readily in a peat—perlite mixture (2:1 v/v).
