Abstract

Objective To investigate the expression profiles of gene and microRNA during the course of immature dendritic cells (DCs) differentiation induced by sodium butyrate in vitro. Methods The human monocyte-derived DCs were induced and total RNA of the cells was collected 24 h later. Gene array analysis of gene expression and microarray analysis of miRNA expression were performed on DCs and described with statistics. Results Thirty-four differentgenes were chosen by genearray analysis of DCs induced by sodium butyrate, including 14 up-regulated genes, and 20. Fourteen genes exhibited higher expressions, and 20 down-regulated genes as compared with control group. The function of these genes involved the secretion of the cytokines and the expression of their receptors, antigen uptake, antigen presentation, cell surface receptors, and signal transduction. The surface marker CD83 of DCs induced with sodium butyrate was (8.90±6.63), decreased significantly as compared with control group (t=3.037, P<0.05). Thirty-four different miRNAs were chosen by microarray analysis of DCs induced by sodium butyrate, including 23 miRNAs with higher expression, and 11 miRNAs with lower expression than the control group. Further analysis revealed that 17 miRNAs among the different miRNAs may keep close interaction with histone deacetylase. Conclusion During the course of immature DCs differentiation induced by sodium butyrate, multi-expression of the genes, accompanied with histone acetylase and related miRNAs, may play an important role in cell differentiation.
