Abstract

Objective To detect the express of CDC25A protein and gene in cholangioncarcinoma and to elucidate its role. Methods The express of CDC25A protein was detected by FCM in 58 cases of cholangioncarcinoma and normal bile duct, and the express of CDC25A gene was detected in 44 cases of cholangioncarcinoma,adjacent to eholangioncarcinoma and normal bile duct by RT-PCR. The correlation was analysed statistically between the expression of CDC25A protein and mRNA and chnicopathological parameters in cholangioncarcinoma. Results The express of CDC25A mRNA in normal bile duct, adjacent to cholangioncarcinoma, cholangioncarcinoma respectively was 0. 3832 ± 0. 1440,0. 4550 ± 0. 1744, 0.5802±0. 4188, The express of CDC25A mRNA was higher in cholangioncarcinoma than others (P <0.05). The protein of CDC25A in cholangioncarcinoma was higher than normal bile duct (P < 0.05). Conclusion cdc25a gene transcription and protein expression is mostly like involved in the proliferation and malignant transformation of bile epithelial and then malignant progression of bile duct carcinomas. The detection of cdc25a expression may elucidate the carcinogenesis of bile duct. Key words: Cholangioncarcinoma; cdc25a; FCM; RT-PCR
