Abstract

Objective To detect the expression of proliferating cell nuclear antigen (PCNA) protein and gene in eholangionearcinoma and to elucidate its role. Methods The express of PCNA protein was detected by Immunohistochemical technique (SP) in 42 cases of eholangionearcinoma and 20 cases normal bile duct,and the express of PCNA gene was detected in 42 cases of eholangionearcinoma, adjacent to eholangionearcinoma and normal bile duct by reverse transcription-polymerase chain reaction (RT-PCR). The correlation was analysed statistically between the express of PCNA protein and mRNA and clinicopatho-logical parameters in eholangionearcinoma. Results The express of PCNA mRNA in normal bile duct, adjacent to eholangionearcinoma,eholangionearcinoma respectively was 0. 5605 ±0. 0331,0. 5692 ± 0.0408, 0. 6303 ±0.0773,the express of PCNA mRNA was higher in eholangionearcinoma than the others (P< 0.05). the protein expression of PCNA was higher in eholangionearcinoma than normal bile duct tissue (P<0.05). Conclusion PCNA gene transcription and protein expression is mostly like involved in the proliferation and malignant transformation of bile epithelial and then malignant progression of bile duct carcinomas. The detection of PCNA expression may elucidate the carcinogenesis of bile duct. Key words: eholangionearcinoma; PCNA; Immunohistochemistry; RT-PCR
