Abstract

Objective To study the mechanism of proliferating cell nuclear antigen (PCNA) in the pathogenesis and progression of hepatocellular carcinoma (HCC).Methods Reverse transcriptasepolymerase chain reaction (RT-PCR) and immunohistochemical technique were used to detect the mRNA and protein expression of PCNA in HCC tissues,adjacent tissues and normal liver tissues.The positive rates of protein expression of PCNA in HCC tissues,adjacent tissues and normal liver tissues were 87%,49% and 20%.The expression levels of PCNA were analyzed combined with the clinicopathological factors including age,gender,tumor size,differentiation degree and lymph node metastasis.The expression of PCNA was correlated with differentiation degree and lymph node metastasis (P < 0.05).Results The positive rates of protein expression of PCNA in HCC tissues,adjacent tissues and normal liver tissues were 87%,49% and 20%.The mRNA and protein expression level of PCNA in HCC tissues was significantly higher than adjacent tissues and normal liver tissues (P < 0.05).The expression of PCNA was correlated with differentiation degree and lymph node metastasis (P <0.05) and had no correlation with age,gender,tumor size (P > 0.05).Conclusion The expression level of PCNA in HCC tissues was significantly higher than adjacent tissues and normal liver tissues which increase with the malignant degree of HCC.
