Abstract

The effects of a single intraperitoneal injection of Adriamycin (10 mg/kg of body weight) on the cellular proliferation of murine duodenal crypt cells were measured by 3H-TdR incorporation into duodenal DNA and 3H-TdR autoradiography. This Adriamycin treatment maximally inhibited the 3H-TdR incorporation at 16 hr postinjection and the inhibition was directly related to a reduction in the cellular rate of DNA synthesis, not to a change in the relative number of cells in the S-phase. The rate of DNA synthesis returned to the control mean by 48 hr and then exceeded it between 74 and 120 hr. The Adriamycin treatment also produced cytotoxicity in the duodenal crypts, and pyknosis and karyorrhexis were still evident 96 hr after the injection; however, there was no evidence to suggest a significant Adriamycin-induced perturbation in the cell-cycle kinetics of those cells which appeared viable, morphologically.
