Abstract

Since ochratoxin A produces accumulation of glycogen in rat liver, the effect of this toxin on nucleic acids and their derivatives was studied. Analyses of spectra and difference spectra in the visible region, column chromatography on Sephadex G-25, and thermal denaturation determinations indicated no interaction of ochratoxin with nucleic acids and their derivatives. It was observed, however, that ochratoxin A inhibited the phosphorylase enzyme system. At a concentration of 0.108 mg per milliliter reaction mixture, the inhibition was about 70%. No inhibitory effect of ochratoxin A on enzymes glucomutase, glucose-6-phosphate dehydrogenase, phosphorylase a or hexokinase was encountered. In this paper it is postulated that the inhibitory effect might be due to competition of the toxin with 3′-5′-cyclic AMP for the enzyme phosphorylase b kinase.
