Synthesis of N-(2-deoxy-β- d-ribosyl) 4-thiomethylindole: A purine surrogate for incorporation into oligonucleotides.

Abstract

Efficient, high-yielding syntheses of S-acetyl- N-(2-deoxy-β- d-ribosyl) 4-thiomethylindole ( 9) and the corresponding 5′- O-trityl-3′- O-phosphoramidite ester 11 are reported, suitable for direct incorporation into oligodeoxynucleotides by standard solid-phase synthesis techniques.
