Abstract

Poly (Adenosine Diphospho Ribose) is a nucleic acid polymer that has been found predominantly in eukaryotes and has been implicated in the regulation of DNA synthesis (1,2). The repeating subunit of this polymer is the adenosine diphospho ribose (ADPR) moiety derived from nicotinamide adenine dinucleotide. The ADPR moieties can also be covalently linked to acceptors such as proteins. The synthesis of poly(ADPR) has usually been studied in isolated nuclei, in chromatin preparations, or with enzyme extracts. These systems have all provided a great deal of information on the activity of poly(ADPR) polymerase; however, in order to understand the physiological relation of poly(ADPR) to DNA synthesis, it is desirable to work with a system which is as close to physiological as possible.
