Abstract
1. 2,4-Dihydroxy-6-methylpyrimidine analog of folic acid and 2,4-dihydroxy-6-methyl-,2,4-dihydroxy-2,4-diamino-,2-methyl-4-aminopyrimidine analogs have been synthesized and tested for antimetabolic activity. 2. The high activity of 2,4-dihydroxy-6-methylpyrimidine analogs of pteroic and folic acids with the inhibition of dihydrofolic acid reductase from rat liver has been demonstrated. 3. The presence of a glutamic acid residue in 2,4-dihydroxy- and 2,4-diamino analogs somewhat decreases the antimetabolic activity, and in a compound with a methyl group in position 2 of the pyrimidine ring, the residue increases the activity.
