Abstract

Thiaminase I in the kidney and spleen of carp was found to be localized in the lysosomes. It was verified with the following evidence: (1) With density gradient fractionation, the enzyme was recovered in the same fraction containing three lysosomal marker enzymes. (2) The equilibrium density of thiaminase I was identical to that of lysosomal marker enzymes and higher than that of mitochondrial marker enzymes. (3) Electron microscopic observation revealed that the fraction containing the highest thiaminase I activity was rich in lysosomes.
