Abstract

The resonance light scattering (RLS) spectrum of meso‐tetra‐(3,5‐dibromo‐4‐hydroxyphenyl) porphyrin (T(DBHP)P) with calf thymus DNA (ctDNA) has been studied. In the pH 3 buffer solution of potassium acid phthalate‐HCl, nucleic acids can enhance the intensity of resonance light scattering of meso‐tetra‐(3,5‐dibromo‐4‐hydroxyphenyl) porphyrin. The enhanced intensity of RLS at 449 nm is proportional to the concentration of ctDNA in the range 0∼0.36 µg mL−1 and the limit of detection is 0.58 ng mL−1. Based on this, an RLS method for the determination of nucleic acids, at nanogram levels, was accordingly established. This method has good selectivity and high sensitivity and it has been applied to the determination of DNA in the synthetic samples with satisfactory results.
