Abstract

A previous report from our laboratory demonstrated that the extract of Lactobacillus fermenti strain 36(IFO No.3071, ATCC No.9338) contained high activity of phosphotransacetylase (acetyl-CoA : orthophosphate acetyltransferase, EC 2.3.1.8). In the present work, reaction products of the phosphotransacetylase reaction were isolated through DEAE-cellulose column chromatography and liophilized, and they were identified as either acetyl-CoA or propionyl-CoA through ultraviolet absorption spectra, paper partition chromatography, nuclear magnetic resonance spectra, and arylamine acetyl-transferase assay. On the basis of these facts, it is concluded that the enzyme from Lactobacillus fermenti can be used for preparation of acetyl-CoA and propionyl-CoA. Since these organismus are easily cultivated and the partially purified enzyme is easily obtainable, the method described in this article can be recommended as useful and inexpensive method to obtain acyl-CoA.
