Abstract

The optimal hydrolysis conditions of soy protein isolate (SPI) with Flavourzyme for triacylglycerols accumulation (TA) inhibitory activity in differentiated 3T3-L1 adipocytes were investigated by a response surface methodology (RSM). A central composite rotatable design for three variables at five levels was applied in this study. The independent variables were pH 5.32-8.68, reaction temperature (RT) 33.2-66.8℃ and hydrolysis time (HT) 19.2-220.8 min. According to the results of response surface and contour plots, the optimal hydrolysis of SPI with Flavourzyme for minimizing the TA in the cells were: pH = 7.41, RT = 49.48℃ and HT = 140.63 min. The results also implied that the second order model was appropriate for description of the response surface since the F-value for lack of fit was not significant (p > 0.05). In addition, the model had a satisfactory coefficient of R2 (= 0.904) and was verified experimentally.
